2Nucleoporin 88 expression in hepatitis B and C virus-related liver diseases显示文摘AIM: To investigate the expression of nucleoporin 88 (Nup88) in hepatitis B virus (HBV) and C virus (HCV)-related liver diseases. METHODS: We generated a new monoclonal Nup88 antibody to investigate the Nup88 protein expression by immunohistochemistry (IHC) in 294 paraffin-embedded liver specimens comprising all stages of hepatocellular carcinogenesis. In addition, in cell culture experiments HBV-positive (HepG2.2.15 and HB611) and HBV-negative (HepG2) hepatoma cell lines were tested for the Nup88 expression by Western-immunoblotting to test data obtained by IHC.RESULTS: Specific Nup88 expression was found in chronic HCV hepatitis and unspecific chronic hepatitis, whereas no or very weak Nup88 expression was detected in normal liver. The Nup88 expression was markedly reduced or missing in mild chronic HBV infection and inversely correlated with HBcAg expression. Irrespective of the HBV- or HCV-status, increasing Nup88 expression was observed in cirrhosis and dysplastic nodules, and Nup88 was highly expressed in hepatocellular carcinomas. The intensity of Nup88 expression significantly increased during carcinogenesis (P < 0.0001) and correlated with dedifferentiation (P < 0.0001). Interestingly, Nup88 protein expression was significantly downregulated in HBV-positive HepG2.2.15 (P < 0.002) and HB611 (P < 0.001) cell lines as compared to HBV-negative HepG2 cells. CONCLUSION: Based on our immunohistochemical data, HBV and HCV are unlikely to influence the expression of Nup88 in cirrhotic and neoplastic liver tissue, but point to an interaction of HBV with the nuclear pore in chronic hepatitis. The expression of Nup88 in nonneoplastic liver tissue might reflect enhanced metabolic activity of the liver tissue. 3100 GHz silicon–organic hybrid modulator显示文摘Electro-optic modulation at frequencies of 100 GHz and beyond is important for photonic-electronic signal processing at the highest speeds.To date,however,only a small number of devices exist that can operate up to this frequency.In this study,we demonstrate that this frequency range can be addressed by nanophotonic,silicon-based modulators.We exploit the ultrafast Pockels effect by using the silicon–organic hybrid(SOH)platform,which combines highly nonlinear organic molecules with silicon waveguides.Until now,the bandwidth of these devices was limited by the losses of the radiofrequency(RF)signal and the RC(resistor-capacitor)time constant of the silicon structure.The RF losses are overcome by using a device as short as 500 μm,and the RC time constant is decreased by using a highly conductive electron accumulation layer and an improved gate insulator.Using this method,we demonstrate for the first time an integrated silicon modulator with a 3dB bandwidth at an operating frequency beyond 100 GHz.Our results clearly indicate that the RC time constant is not a fundamental speed limitation of SOH devices at these frequencies.Our device has a voltage–length product of only V_(π)L=11 V mm,which compares favorably with the best silicon-photonic modulators available today.Using cladding materials with stronger nonlinearities,the voltage–length product is expected to improve by more than an order of magnitude.Luca Alloatti Robert Palmer Sebastian Diebold Kai Philipp Pahl Baoquan Chen Raluca Dinu Maryse Fournier Jean-Marc Fedeli Thomas Zwick Wolfgang Freude Christian Koos Juerg Leuthold 2014Light(Science & Applications)2014,3,1:3

7Acute appendicitis–advances and controversies显示文摘Being one of the most common causes of the acute abdomen,acute appendicitis(AA)forms the bread and butter of any general surgeon’s practice.With the recent advancements in AA’s management,much controversy in diagnostic algorithms,possible differential diagnoses,and weighing the management options has been generated,with no absolute consensus in the literature.Since Alvarado described his eponymous clinical scoring system in 1986 to stratify AA risk,there has been a burgeoning of additional scores for guiding downstream management and mortality assessment.Furthermore,advancing literature on the role of antibiotics,variations in appendicectomy,and its adjuncts have expanded the surgeon’s repertoire of management options.Owing to the varied presentation,diagnostic tools,and management of AA have also been proposed in special groups such as pregnant patients,the elderly,and the immunocompromised.This article seeks to raise the critical debates about what is currently known about the above aspects of AA and explore the latest controversies in the field.Considering the ever-evolving coronavirus disease 2019 situation worldwide,we also discuss the pandemic’s repercussions on patients and how surgeons’practices have evolved in the context of AA.Thomas Zheng Jie Teng Xuan Rong Thong Kai Yuan Lau Sunder Balasubramaniam Vishal G Shelat 2021World Journal of Gastrointestinal Surgery2021,13,11:2
