1Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. 2Baicalin prevents LPS-induced activation of TLR4/NF-κB p65 pathway and inflammation in mice via inhibiting the expression of CD14显示文摘Previous studies have shown that baicalin,an active ingredient of the Chinese traditional medicine Huangqin,attenuates LPS-induced inflammation by inhibiting the activation of TLR4/NF-κBp65 pathway,but how it affects this pathway is unknown.It has been shown that CD14 binds directly to LPS and plays an important role in sensitizing the cells to minute quantities of LPS via chaperoning LPS molecules to the TLR4/MD-2 signaling complex.In the present study we investigated the role of CD14 in the anti-inflammatory effects of baicalin in vitro and in vivo.Exposure to LPS(1μg/mL)induced inflammatory responses in RAW264.7 cells,evidenced by marked increases in the expression of MHC II molecules and the secretion of NO and IL-6,and by activation of MyD88/NF-κB p65 signaling pathway,as well as the expression of CD14 and TLR4.These changes were dose-dependently attenuated by pretreatment baicalin(12.5–50μM),but not by baicalin post-treatment.In RAW264.7 cells without LPS stimulation,baicalin dose-dependently inhibit the protein and mRNA expression of CD14,but not TLR4.In RAW264.7 cells with CD14 knockdown,baicalin pretreatment did not prevent inflammatory responses and activation of MyD88/NF-κB p65 pathway induced by high concentrations(1000μg/mL)of LPS.Furthermore,baicalin pretreatment also inhibited the expression of CD14 and activation of MyD88/NF-κB p65 pathway in LPS-induced hepatocyte-derived HepG2 cells and intestinal epithelial-derived HT-29 cells.In mice with intraperitoneal injection of LPS and in DSS-induced UC mice,oral administration of baicalin exerted protective effects by inhibition of CD14 expression and inflammation.Taken together,we demonstrate that baicalin pretreatment prevents LPS-induced inflammation in RAW264.7 cells in CD14-dependent manner.This study supports the therapeutic use of baicalin in preventing the progression of LPS-induced inflammatory diseases.Ya-jun Fu Bo Xu Shao-wei Huang Xia Luo Xiang-liang Deng Shuang Luo Chang Liu Qing Wang Jin-yan Chen Lian Zhou 2021Acta Pharmacologica Sinica2021,42,1:31
3Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. 6First light for the sodium laser guide star adaptive optics system on the Lijiang 1.8 m telescope显示文摘A first generation sodium Laser Guide Star Adaptive Optics System(LGS-AOS) was developed and integrated into the Lijiang 1.8 m telescope in 2013. The LGS-AOS has three sub-systems:(1) a 20 W long pulsed sodium laser,(2) a 300-millimeter-diameter laser launch telescope, and(3) a 37-element compact adaptive optics system. On 2014 January 25, we obtained high resolution images of an mV8.18 star,HIP 43963, during the first light of the LGS-AOS. 7Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 8Ternary Ni-based Prussian blue analogue with superior sodium storage performance induced by synergistic effect of Co and Fe显示文摘Prussian blue analogue Na2Ni[Fe(CN)6](Ni-PB)has been widely studied as a cathode material for sodium-ion battery due to its excellent cycling performance.However,Ni-PB has a low theoretical capacity of 85 mAh g^(−1) because of the electrochemical inertness of Ni.Herein,ternary Ni-PB is successfully synthesized by double doping with Co and Fe at Ni-site,and the effect of doping with Co and Fe on the electrochemical performance of Ni-PB is systematically investigated through theoretical calculations and electrochemical tests.The first principles calculations confirm that double doping with Co and Fe can significantly reduce the energy barrier and bandgap of Ni-PB.X-ray diffraction and composition analysis results indicate that ternary NiCoFe-PB composite not only has good crystallinity and high Na content but also has low defects and crystal water.Electrochemical tests reveal that,besides the capacity contribution of high-spin Co/Fe and low-spin Fe,Co-doping enhances the electrochemical activity of low-spin Fe and Fe-doping improves the activity of high-spin Co;moreover,double doping can decrease the diffusion resistance of Na+ions through solid electrolyte interface film,accelerate the kinetics for both ion diffusion process and Faradic reaction,and increase active sites.Under the synergistic effect of Co and Fe,this ternary NiCoFe-PB exhibits outstanding electrochemical performance with a high initial discharge capacity of 120.4 mAh g^(−1) at 20mA g^(−1) and an extremely low capacity fading rate of 0.0044%per cycle at a high current density of 2 A g^(−1) even after 10,000 cycles,showing great application potential of ternary NiCoFe-PB in the field of large-scale energy storage.Lu-Lu Zhang Cheng Wei Xin-Yuan Fu Zhao-Yao Chen Bo Yan Pan-Pan Sun Kai-Jun Chang Xue-Lin Yang 2021Carbon Energy2021,3,5:2
9Quadratic Stabilization of Switched Uncertain Linear Systems: A Convex Combination Approach显示文摘We consider quadratic stabilization for a class of switched systems which are composed of a finite set of continuoustime linear subsystems with norm bounded uncertainties. Under the assumption that there is no single quadratically stable subsystem, if a convex combination of subsystems is quadratically stable, then we propose a state-dependent switching law, based on the convex combination of subsystems, such that the entire switched linear system is quadratically stable. 13Distortion mapping correction in the AIMS primary mirror testing by a computer-generated hologram显示文摘The National Solar Observatory is currently developing the Accurate Infrared Magnetic Field Measurements of the Sun(AIMS). The primary mirror of the AIMS solar telescope is an off-axis parabolic with a diameter of 1 m and with a large off-axis amount of 1 m. Due to the surface figure of the primary mirror under the used state is directly related to image quality of the whole system, a computer-generated hologram(CGH) is carried out to test the primary mirror, and the test results are used to polish the mirror to a higher surface accuracy. However, the fact that the distortion exists in the testing results leads to the failure of a further guide to deterministic optical processing. In this paper, a distortion correction method is proposed, which uses an orthogonal set of vector polynomials to mapping the coordinates of the mirror and the pixels of fringes, and then an interpolation method is adopted to obtain the corrected results. The testing accuracy by using CGH is also verified by an auto-collimate test experiment. 18Nuclear PLD1 combined with NPM1 induces gemcitabine resistance through tumorigenic IL7R in pancreatic adenocarcinoma显示文摘Objective:Pancreatic ductal adenocarcinoma(PDAC)is a highly malignant gastrointestinal cancer with a 5-year survival rate of only 9%.Of PDAC patients,15%-20%are eligible for radical surgery.Gemcitabine is an important chemotherapeutic agent for patients with PDAC;however,the efficacy of gemcitabine is limited due to resistance.Therefore,reducing gemcitabine resistance is essential for improving survival of patients with PDAC.Identifying the key target that determines gemcitabine resistance in PDAC and reversing gemcitabine resistance using target inhibitors in combination with gemcitabine are crucial steps in the quest to improve survival prognosis in patients with PDAC.Methods:We constructed a human genome-wide CRISPRa/dCas 9 overexpression library in PDAC cell lines to screen key targets of drug resistance based on sgRNA abundance and enrichment.Then,co-IP,ChIP,ChIP-seq,transcriptome sequencing,and qPCR were used to determine the specific mechanism by which phospholipase D1(PLD1)confers resistance to gemcitabine.Results:PLD1 combines with nucleophosmin 1(NPM1)and triggers NPM1 nuclear translocation,where NPM1 acts as a transcription factor to upregulate interleukin 7 receptor(IL7R)expression.Upon interleukin 7(IL-7)binding,IL7R activates the JAK1/STAT5 signaling pathway to increase the expression of the anti-apoptotic protein,BCL-2,and induce gemcitabine resistance.The PLD1 inhibitor,Vu0155069,targets PLD1 to induce apoptosis in gemcitabine-resistant PDAC cells.Conclusions:PLD1 is an enzyme that has a critical role in PDAC-associated gemcitabine resistance through a non-enzymatic interaction with NPM1,further promoting the downstream JAK1/STAT5/Bcl-2 pathway.Inhibiting any of the participants of this pathway can increase gemcitabine sensitivity.Danqi Fu Jingrui Yan Zhaoyu Zhang Yang Liu Xiaoqing Ma Jinsheng Ding Shengyu Yang Ran Zhao Antao Chang Chuntao Gao Jing Liu Tiansuo Zhao Xiuchao Wang Chongbiao Huang Song Gao Ying Ma Bo Tang Yukuan Feng Hongwei Wang Jihui Hao 2023Cancer Biology & Medicine2023,20,8:0
19Mechanism of activation and biased signaling in complement receptor C5aR1显示文摘The complement system plays an important role in the innate immune response to invading pathogens.The complement fragment C5a is one of its important effector components and exerts diverse physiological functions through activation of the C5a receptor 1(C5aR1)and associated downstream G protein andβ-arrestin signaling pathways.Dysfunction of the C5a-C5aR1 axis is linked to numerous inflammatory and immune-mediated diseases,but the structural basis for activation and biased signaling of C5aR1 remains elusive.Here,we present cryo-electron microscopy structures of the activated wild-type C5aR1–Gi protein complex bound to each of the following:C5a,the hexapeptidic agonist C5apep,and the G protein-biased agonist BM213.The structures reveal the landscape of the C5a–C5aR1 interaction as well as a common motif for the recognition of diverse orthosteric ligands.Moreover,combined with mutagenesis studies and cell-based pharmacological assays,we deciphered a framework for biased signaling using different peptide analogs and provided insight into the activation mechanism of C5aR1 by solving the structure of C5aR1I116A mutant–Gi signaling activation complex induced by C089,which exerts antagonism on wild-type C5aR1.In addition,unusual conformational changes in the intracellular end of transmembrane domain 7 and helix 8 upon agonist binding suggest a differential signal transduction process.Collectively,our study provides mechanistic understanding into the ligand recognition,biased signaling modulation,activation,and Gi protein coupling of C5aR1,which may facilitate the future design of therapeutic agents.Yuying Feng Chang Zhao Yue Deng Heli Wang Liang Ma Sicen Liu Xiaowen Tian Bo Wang Yan Bin Peipei Chen Wei Yan Ping Fu Zhenhua Shao 2023Cell Research2023,33,4:0
20Study on Ability of Ground Cover Plant of Taiyuan to Adsorb PM(2.5)显示文摘Four ground cover plant species of Taiyuan( Iris,Hemerocallis,Sedum,Hosta) were selected to study their capacities in adsorbing PM_(2.5). Meanwhile,the concentration of PM_(2.5) in Taiyuan between April 2017 and August 2017 was recorded,and the characteristics of PM_(2.5) pollution in summer and autumn in Taiyuan were studied. The results showed that for the 4 plants,the ability to adsorb PM_(2.5) was in the order from great to small of Hosta,Iris,Hemerocallis,Sedum,in which H. plantaginea had the best effect to adsorb PM_(2.5). The fresh weight and dry weight per gram of H. plantaginea were 4. 4 times and 2 times higher than those of S. spectabile,while the mass quality of dust adsorption was 2.8 times higher. The sorting result was the same as the ability to adsorb PM_(2.5) of unit leaf area. The result of the coefficient of purification showed that the purification coefficients of 3 vegetation structure were positive,and the combination of trees and shrubs in university campus had strong PM_(2.5) adsorption capacity. By comparing the seasonal variation of PM_(2.5) concentration in Taiyuan city,it found that the PM_(2.5) concentration was particularly high in late spring and early summer of Taiyuan,when most of the ground cover plants were not fully grown.
