10Estimation of genetic parameters and breeding values in shrimp F enneropenaeus chinensis using the REML/BLUP procedure显示文摘An analysis of a selection experiment was used to assess the impact of various animal model structures on REML estimates of variance components.The analyses were carried out based on 162 d body mass (BM) of 1 287 animals from 21 paternal half-sib groups of Fenneropenaeus chinensis.Estimated breeding values (EBV) of BM of all individuals were estimated using eight statistical models (A,AB,ABC,ABDC,ABMFC,ABMDC,ABFDC and ABMFDC) and BLUP (best linear unbiased prediction).These models were designed involving factors such as sex,spawn date as fixed effects,maternal genetic effects,full-sib family effects as random effects,mean BM of families at tagging and age at recording (covariate).The results demonstrate the importance of correct interpretation of effects in the data set,particularly those that can influence resemblance between relatives.The data structure and the particular model that was applied markedly influenced the magnitude of variance component estimates.Models based on few effects obtained upward biased estimates of additive genetic variance.The accuracy of genetic parameters and breeding value estimated by ABFDC model was higher than other models.The results imply that additive genetic direct value,full-sib family effects,and covariance effects besides sex and spawn date as fixed effects were very important for estimating genetic parameters and breeding value of body mass.This model had a heritability estimate of 162 d BM of 0.44.The comparison of the efficiency of selection based on breeding values or phenotypic value revealed great difference:average breeding value of the best 24 families selected by the 162 d BM breeding value and phenotype were 0.577 g and 0.366 g,respectively,representing a 36.57% higher efficiency in the former.In conclusion,selection based on breeding value was more effective than selection based on phenotypic value.Our results indicate that effects influencing the magnitude of estimates should be taken into account when estimating heritability and breeding values for BM.ZHANG Tianshi KONG Jie LUAN Sheng WANG Qingyin LUO Kun TIAN Yia 2011Acta Oceanologica Sinica2011,30,1:12

14AtMGT7:An Arabidopsis Gene Encoding a Low-Affinity Magnesium Transporter显示文摘Magnesium(Mg2+) is one of the essential cations in all cells. Although the Mg2+ transport mechanism has been welldocumented in bacteria,less is known about Mg2+ transporters in eukaryotes. The AtMGT gene family encoding putative magnesium transport proteins had been described previously. We report here that one of the Arabidopsis MGT family members,the AtMGT7 gene,encodes two mRNAs that have resulted from alternative splicing variants,designated AtMGT7a and AtMGT7b. Interestingly,the two mRNA variants were expressed with different patterns with AtMGT7a expressing in all organs,but AtMGT7b appearing only in root and flowers. The AtMGT7a variant functionally complemented a bacterial mutant lacking Mg2+ transport capacity,whereas AtMGT7b did not. The 63Ni2+ tracer uptake analysis in the bacterial model showed that AtMGT7a mediated low-affinity transport of Mg2+. Consistent with the complementation assay result,63Ni2+ tracer uptake analysis revealed that AtMGT7b did not transport Mg2+. 16Genetic parameter estimation for juvenile growth and upper thermal tolerance in turbot(Scophthalmus maximus Linnaeus)显示文摘Twenty-six half-sib groups(53 full-sib families) of turbot, Scophthalmus maximus Linnaeus, were obtained by artificial insemination. We measured growth in the offspring(40–50 individuals/family) and subjected them to a thermal tolerance challenge over a period of 34 d. There was no significant difference in daily mortality(range: 0.580%–1.391%) between Days 1–13 during the thermal tolerance challenge. However, daily cumulative mortality increased rapidly between Days 14 and 29, especially on Days 15 and 16(20.232% and 34.377%, respectively). Mortality was highest on Day 16(14.145%). We estimated the genetic parameters using the average information restricted maximum likelihood method. We used a likelihood ratio test to evaluate the significance of effects in models with and without identity as an effect, and compared the final log-likelihoods(maximum log L). Lastly, we estimated phenotypic and genetic correlation between the upper thermal tolerance limit(UTT) and body weight(BW). In this study, the positive phenotypic correlation was low between UTT and BW(0.093±0.029). The genetic correlation between UTT and BW was negative(-0.044±0.239). The heritability for upper thermal tolerance was low(0.087±0.032), which is of approximately moderate heritability. The heritability for body weight was high(0.303±0.074). 20A transceptor-channel complex couples nitrate sensing to calcium signaling in Ambidopsis显示文摘Nitrate-induced Ca^(2+) signaling is crucial for the primary nitrate response in plants.However,the molecular mechanism underlying the generation of the nitrate-specific calcium signature remains unknown.We report here that a cyclic nucleotide-gated channel(CNGC)protein,CNGC15,and the nitrate transceptor(NRT1.1)constitute a molecular switch that controls calcium influx depending on nitrate levels.The expression of CNGC15 is induced by nitrate,and its protein is localized at the plasma membrane after establishment of young seedlings.We found that disruption of CNGC15 results in the loss of the nitrate-induced Ca^(2+) signature(primary nitrate response)and retards root growth,reminiscent of the phenotype observed in the nrt1.1 mutant.We further showed that CNGC15 is an active Ca^(2+)-permeable channel that physically interacts with the NRT1.1 protein in the plasma membrane.Importantly,we discovered that CNGC15-NRT1.1 interaction silences the channel activity of the heterocomplex,which dissociates upon a rise in nitrate levels,leading to reactivation of the CNGC15 channel.The dynamic interactions between CNGC15 and NRT1.1 therefore control the channel activity and Ca^(2+) influx in a nitrate-dependent manner.Our study reveals a new nutrient-sensing mechanism that utilizes a nutrient transceptor-channel complex assembly to couple nutrient status to a specific Ca^(2+) signature.Xiaohan Wang Changxin Feng LiLi Tian Congcong Hou Wang Tian Bin Hu Qian Zhang Zhijie Ren Qi Niu Jiali Song Dongdong Kong Liangyu Liu Yikun He Ligeng Ma Chengcai Chu Sheng Luan Legong Li 2021Molecular Plant2021,14,5:5
