1Silvicultural alternatives to conventional even-aged forest man-agement-what limits global adoption?显示文摘Background: The development of forestry as a scientific and management discipline over the last two centuries has mainly emphasized intensive management operations focused on increased commodity production, mostly wood. This 'conventional' forest management approach has typically favored production of even-aged, single-species stands. While alternative management regimes have generally received less attention, this has been changing over the last three decades, especially in countries with developed economies. Reasons for this change include a combination of new information and concerns about the ecological consequences of intensive forestry practices and a willingness on the part of many forest owners and society to embrace a wider set of management objectives. Alternative silvicultural approaches are characterized by a set of fundamental principles, including avoidance of clearcutting, an emphasis on structural diversity and small-scale variability, deployment of mixed species with natural regeneration,and avoidance of intensive site-preparation methods.Methods: Our compilation of the authors' experiences and perspectives from various parts of the world aims to initiate a larger discussion concerning the constraints to and the potential of adopting alternative silvicultural practices.Results: The results suggest that a wider adoption of alternative silvicultural practices is currently hindered by a suite of ecological, economic, logistical, informational, cultural, and historical constraints. Individual contexts display their own unique combinations and relative significance of these constraints, and accordingly, targeted efforts, such as regulations and incentives, may help to overcome specific challenges.Conclusions: In a broader context, we propose that less emphases on strict applications of principles and on stand structures might provide additional flexibility and facilitate the adoption of alternative silvicultural regimes in a broader set of circumstances. At the same time, the acceptance of alternative silvicultural systems as the 'preferred or default mode of management' wil necessitate and benefit from the continued development of the scientific basis and valuation of a variety of ecosystem goods and services. 5Chronic exposure to high-density polyethylene microplastic through feeding alters the nutrient metabolism of juvenile yellow perch (Perca flavescens)显示文摘Microplastics are emergent contaminants threatening aquatic organisms including aquacultured fish.This study investigated the effects of high-density polyethylene(HDPE,100 to 125 mm)on yellow perch(Perca flavescens)based on integrative evaluation including growth performance,nutritional status,nutrient metabolism,fish health,and gut microbial community.Five test diets(0,1,2,4,or 8 g HDPE/100 g diet)containing 41%protein and 10.5%lipid were fed to juvenile perch(average body weight,25.9±0.2 g;n=15)at a feeding rate of 1.5%to 2.0%body weight daily.The feeding trial was conducted in a flow-through water system for 9 wk with 3 tanks per treatment and 15 yellow perch per tank.No mortality or HDPE accumulation in the fish was found in any treatments.Weight gain and condition factor of fish were not significantly impacted by HDPE(P>0.05).Compared to the control group,fish fed the 8%HDPE diet had significantly decreased levels of protein and ash(P<0.05).In response to the increasing levels of HDPE exposure,the hepatosomatic index value,hepatocyte size,and liver glycogen level were increased,but lipid content was reduced in the liver tissues.Compared to the control treatment,fish fed the 8%HDPE diet had significant accumulations of total bile acids and different metabolism pathways such as bile acid biosynthesis,pyruvate metabolism,and carnitine synthesis.Significant enterocyte necrosis was documented in the foregut of fish fed the 2%or 8%HDPE diet;and significant cell sloughing was observed in the midgut and hindgut of fish fed the 8%HDPE diet.Fish fed the 2%HDPE diet harbored different microbiota communities compared to the control fish.This study demonstrates that HDPE ranging from 100 to 125 mm in feed can be evacuated by yellow perch with no impact on growth.However,dietary exposure to HDPE decreased whole fish nutrition quality,altered nutrient metabolism and the intestinal histopathology as well as microbiota community of yellow perch.The results indicate that extended exposure may pose a risk to fish health and jeopardize the nutrition quality of aquacultured end product.This hypothesis remains to be investigated further.Xing Lu Dong-Fang Deng Fei Huang Fabio Casu Emma Kraco Ryan J.Newton Merry Zohn Swee J.Teh Aaron M.Watson Brian Shepherd Ying Ma Mahmound A.O.Dawood Lorena M.Rios Mendoza 2022Animal Nutrition2022,,2:1

10Unraveling female reproductive senescence to enhance healthy longevity显示文摘In a society where women often want successful careers and equal opportunities to men,the early nature of ovarian aging often forces women to make difficult life choices between career and family development.Fertility in women begins to decline after the age of 37 years and it is rare for pregnancies to occur after 45.This reproductive decline in women is inevitable and culminates in menopause,which is a major driver of age-related diseases.In a world where biomedical advances are leading to modifiable biological outcomes,it is time to focus on mitigating female reproductive senescence to maintain fertility and preserve age-related hormonal functions,with the goal of providing increased life choices and enhancing healthspan.To date,reproductive longevity research remains an understudied field.More needs to be done to unravel the biology of the ovarian follicles,which are the functional units of reproductive lifespan and are comprised of cell types including the oocyte(female gamete)and a group of specialized supporting somatic cells.Biological attempts to maintain the quality and quantity of follicles in animal models through manipulating pathways involved in aging can potentially prolong female reproductive lifespan and healthspan.Here,we summarize the molecular events driving ovarian aging and menopause and the interventional strategies to offset these events.Developing solutions to female reproductive senescence will open doors to discover ways to enhance true healthy longevity for both men and women.Lu Dong Daniel Boon Loong Teh Brian Keith Kennedy Zhongwei Huang 2023Cell Research2023,33,1:1

16Upregulation of RIN3 induces endosomal dysfunction in Alzheimer’s disease显示文摘Background In Alzheimer’s Disease(AD),about one-third of the risk genes identified by GWAS encode proteins that function predominantly in the endocytic pathways.Among them,the Ras and Rab Interactor 3(RIN3)is a guanine nucleotide exchange factor(GEF)for the Rab5 small GTPase family and has been implicated to be a risk factor for both late onset AD(LOAD)and sporadic early onset AD(sEOAD).However,how RIN3 is linked to AD pathogenesis is currently undefined.Methods Quantitative PCR and immunoblotting were used to measure the RIN3 expression level in mouse brain tissues and cultured basal forebrain cholinergic neuron(BFCNs).Immunostaining was used to define subcellular localization of RIN3 and to visualize endosomal changes in cultured primary BFCNs and PC12 cells.Recombinant flag-tagged RIN3 protein was purified from HEK293T cells and was used to define RIN3-interactomes by mass spectrometry.RIN3-interacting partners were validated by co-immunoprecipitation,immunofluorescence and yeast two hybrid assays.Live imaging of primary neurons was used to examine axonal transport of amyloid precursor protein(APP)andβ-secretase 1(BACE1).Immunoblotting was used to detect protein expression,processing of APP and phosphorylated forms of Tau.Results We have shown that RIN3 mRNA level was significantly increased in the hippocampus and cortex of APP/PS1 mouse brain.Basal forebrain cholinergic neurons(BFCNs)cultured from E18 APP/PS1 mouse embryos also showed increased RIN3 expression accompanied by early endosome enlargement.In addition,via its proline rich domain,RIN3 recruited BIN1(bridging integrator 1)and CD2AP(CD2 associated protein),two other AD risk factors,to early endosomes.Interestingly,overexpression of RIN3 or CD2AP promoted APP cleavage to increase its carboxyl terminal fragments(CTFs)in PC12 cells.Upregulation of RIN3 or the neuronal isoform of BIN1 increased phosphorylated Tau level.Therefore,upregulation of RIN3 expression promoted accumulation of APP CTFs and increased phosphorylated Tau.These effects by RIN3 was rescued by the expression of a dominant negative Rab5(Rab5S34N)construct.Our study has thus pointed to that RIN3 acts through Rab5 to impact endosomal trafficking and signaling.Conclusion RIN3 is significantly upregulated and correlated with endosomal dysfunction in APP/PS1 mouse.Through interacting with BIN1 and CD2AP,increased RIN3 expression alters axonal trafficking and procession of APP.Together with our previous studies,our current work has thus provided important insights into the role of RIN3 in regulating endosomal signaling and trafficking.Ruinan Shen Xiaobei Zhao Lu He Yongbo Ding Wei Xu Suzhen Lin Savannah Fang Wanlin Yang Kijung Sung Brian Spencer Robert A.Rissman Ming Lei Jianqing Ding Chengbiao Wu 2020Translational Neurodegeneration2020,9,2:1
17Dynamic phenotypic heterogeneity and the evolution of multiple RNA subtypes in hepatocellular carcinoma:the PLANET study显示文摘Intra-tumor heterogeneity(ITH) is a key challenge in cancer treatment, but previous studies have focused mainly on the genomic alterations without exploring phenotypic(transcriptomic and immune)heterogeneity. Using one of the largest prospective surgical cohorts for hepatocellular carcinoma(HCC)with multi-region sampling, we sequenced whole genomes and paired transcriptomes from 67 HCC patients(331 samples). We found that while genomic ITH was rather constant across stages, phenotypic ITH had a very different trajectory and quickly diversified in stage Ⅱ patients. Most strikingly, 30% of patients were found to contain more than one transcriptomic subtype within a single tumor. Such phenotypic ITH was found to be much more informative in predicting patient survival than genomic ITH and explains the poor efficacy of single-target systemic therapies in HCC.
