1鸡DMRT1基因CRISPR/Cas9载体构建及打靶效率的检测显示文摘DMRT1(Double sex and Mab-3 related transcription factor 1)是鸡性腺分化和发育的关键候选基因,其详细的功能和作用途径还未被阐明。为了解明DMRT1在鸡性腺分化和发育过程中的功能,研究首次利用在线sg RNA平台设计4条鸡DMRT1 sg RNA序列,构建了4个sg RNA表达载体:Guide-1、Guide-2、Guide-3和Guide-4。转染鸡DF-1成纤维细胞后,利用T7E1内切酶的方法检测4个表达载体的打靶效率。结果显示,成功构建了4条正确的鸡DMRT1 sg RNA表达载体,并且都能高效地在DF-1细胞中表达,其中Guide-4表达载体能有效地打靶DMRT1基因,打靶效率为24.82%。研究结果为进一步在鸡胚敲除DMRT1基因,研究其功能奠定了基础。杨秀荣 邓继贤 赵德彪 Mike McGrew Sunil Nandi 蒋和生 Michael Clinton 2017中国家禽2017,39,2:9
2Elevated basal intestinal mucosal cytokine levels in asymptomatic first-degree relatives of patients with Crohn's disease显示文摘AIM To determine levels of cytokines incolonic mucosa of asymptomatic first degreerelatives of Crohn’s disease patients.METHODS Cytokines(Interleukin(IL)1-Beta,IL-2,IL-5 and IL-8)were measured using ELISAin biopsy samples of normal looking colonicmucosa of first degree relatives of Crohn’sdisease patients(n = 9)and from normalcontrols(n = 10)with no family history ofCrohn’s disease.RESULTS Asymptomatic first degree relativesof patients with Crohn’s disease had significantlyhigher levels of basal intestinal mucosalcytokines(IL-2,IL-5 and IL-8)than normalcontrols.Whether these increased cytokinelevels serve as phenotypic markers for a geneticpredisposition to developing Crohns diseaselater on,or whether they indicate early(pre-clinical)damage has yet to be further defined.CONCLUSION Asymptomatic first degreerelatives of Crohn’s disease patients have higherlevels of cytokines in their normal-lookingintestinal mucosa compared to normal controls,This supports the hypothesis that increasedcytokines may be a cause or an early event inthe inflammatory cascade of Crohns disease andare not merely a result of the inflammatoryprocess.Indaram AVK Nandi S Weissman S Lam S Bailey B Blumstein M Greenberg R Bank S 2000World Journal of Gastroenterology2000,6,1:8
3Detection of hepatitis B virus infection: A systematic review显示文摘AIM: To review published methods for detection of hepatitis B virus(HBV) infection.METHODS: A thorough search on Medline database was conducted to find original articles describing different methods or techniques of detection of HBV, which are published in English in last 10 years. Articles outlining methods of detection of mutants or drug resistance were excluded. Full texts and abstracts(if full text not available) were reviewed thoroughly. Manual search of references of retrieved articles were also done. We extracted data on different samples and techniques of detection of HBV, their sensitivity(Sn), specificity(Sp) and applicability.RESULTS: A total of 72 studies were reviewed. HBV was detected from dried blood/plasma spots, hepatocytes, ovarian tissue, cerumen, saliva, parotid tissue, renal tissue, oocytes and embryos, cholangiocarcinoma tissue, etc. Sensitivity of dried blood spot for detecting HBV was > 90% in all the studies. In case of seronegative patients, HBV DNA or serological markers have been detected from hepatocytes or renal tissue in many instances. Enzyme linked immunosorbent assay and Chemiluminescent immunoassay(CLIA) are most commonly used serological tests for detection. CLIA systems are also used for quantitation. Molecular techniques are used qualitatively as well as for quantitative detection. Among the molecular techniques version 2.0 of the Cobas Ampliprep/Cobas Taq Man assay and Abbott's real time polymerase chain reaction kit were found to be most sensitive with a lower detection limit of only 6.25 IU/m L and 1.48 IU/m L respectively. CONCLUSION: Serological and molecular assays are predominant and reliable methods for HBV detection. 4Pre-treatment of rocks prior to comminution--A critical review of present practices显示文摘It has been an established fact that comminution processes, crushing and grinding, are most energy intensive processes which account for more than half of the total energy consumed in mineral industries.Various alternative pre-treatment methods have been tried by experts around the globe. Although these methods yielded positive results in terms of reduction in energy consumption in crushing and particularly, in grinding operations at laboratory scale, their industrial application still remains an unresolved issue and challenge. Present review paper describes each one of these methods along with outcome of earlier studies and issues that need to be addressed through further rigorous experimental investigation.It also suggests the direction in which future studies can be carried out to meet the primary objective of making comminution processes more energy efficient than today they are.Somani Aditya Nandi Tapas K. 5Effect of pyroxenite and olivine minerals as source of MgO in hematite pellet on improvement of metallurgical properties显示文摘Pelletization of hematite ore requires high fineness and very high induration temperature(~1325 °C) owing to its poor diffusion bonding unlike magnetite ore. Further, high-alumina hematite pellets show very high reduction degradation index(RDI) during low temperature(500-650 °C) reduction due to their volume expansion and lattice distortion. Noamundi(India) hematite ore contains very high Al2O3(2.3%) with adverse ratio of alumina to silica(~2) for which, it shows very high RDI. In this work, the acid pellets prepared from Noamundi ore fines of optimum Blaine fineness show good cold crushing strength(CCS). However, it shows very high RDI(77%). In order to reduce RDI, Mg O in form of two different gangue-containing fluxes, such as pyroxenite and olivine in varying quantities has been added. The optimum requirement and performance of these fluxes has been examined and compared. Both pyroxenite and olivine fluxed pellets show significant lowering of RDI(26% and 23%, respectively) and improvement of other properties, viz CCS, swelling indices etc with good reducibility(70%-77%). 6Intrusion Detection System for PS-Poll DoS Attack in 802.11 Networks Using Real Time Discrete Event System显示文摘Wi-Fi devices have limited battery life because of which conserving battery life is imperative. The 802.11 Wi-Fi standard provides power management feature that allows stations(STAs) to enter into sleep state to preserve energy without any frame losses. After the STA wakes up, it sends a null data or PS-Poll frame to retrieve frame(s) buffered by the access point(AP), if any during its sleep period. An attacker can launch a power save denial of service(PS-DoS) attack on the sleeping STA(s) by transmitting a spoofed null data or PS-Poll frame(s) to retrieve the buffered frame(s) of the sleeping STA(s) from the AP causing frame losses for the targeted STA(s). Current approaches to prevent or detect the PS-DoS attack require encryption,change in protocol or installation of proprietary hardware. These solutions suffer from expensive setup, maintenance, scalability and deployment issues. The PS-DoS attack does not differ in semantics or statistics under normal and attack circumstances.So signature and anomaly based intrusion detection system(IDS) are unfit to detect the PS-DoS attack. In this paper we propose a timed IDS based on real time discrete event system(RTDES) for detecting PS-DoS attack. The proposed DES based IDS overcomes the drawbacks of existing systems and detects the PS-DoS attack with high accuracy and detection rate. 7Effect of weathering on physico-chemical properties and combustion behavior of an Indian thermal coal显示文摘An open air stockpile of conical shape was formed with 3.5 m base diameter and 5 m height using 500 tons of -200 mm size ROM thermal coal on a concrete floor. Coal sample from the stockpile were collected at 30 days interval for analysis of various chemical properties like;proximate analysis, ultimate analysis, gross calorific value (GCV) and various combustion related properties such as ignition temperature, peak temperature, burnout temperature, maximum combustion rate, ignition index, burnout index, combustion performance index and combustion rate intensity index. Experimental results show that, due to weathering of coal fixed carbon decreased from 35.6% to 19.9%, elemental carb on (C) decreased from 46.6% to 28.6%, hydrogen (H) decreased from 3.3% to 2.9% and GCV decreased by up to 55% of original value during 330 days of storage of coal in an open atmosphere. Ash con tent of coal increased form 29.2% to 46.6% due to loss of combustibles. Sulfur (S) of coal was found to get increased from 0.33% to 1.08% during storage. The activation energy of coal combustion increased from 22 kJ/mol to 54 kJ/mol. 8墨西哥野生马铃薯Solanum pinnatisectum抗晚疫病及抗马铃薯甲虫新基因的遗传分析与分子标记(英文)显示文摘马铃薯晚疫病(Phytophthora infestans)和科罗拉多马铃薯甲虫(CPB)是马铃薯生产中最为严重的病虫害。培育高抗晚疫病和甲虫的马铃薯品种是加拿大马铃薯育种工作的重要组成部分。目前,我们实验室在二倍体1EBN墨西哥野生种中已鉴定出抗马铃薯晚疫病和甲虫的新基因,并利用原生质体融合技术成功的将其转移到栽培品种中。但是,培育出抗晚疫病和抗甲虫的马铃薯新品种仍然是一项艰难而繁杂的工作。为了加快分离抗性基因,建立与抗性基因紧密关联的DNA分子标记至关重要。本研究以感病的二倍体马铃薯品种S.cardiophyllum作为父本,与带有抗性基因的墨西哥野生种S.pinnatisectum杂交。用叶片离体鉴定的方法测试F1和BC1代群体的抗病性,从而筛选抗晚疫病和抗甲虫的植株。US-8/A2交配型病菌测试显示所有的F1代植株都表现出抗晚疫病,而在BC1群体中抗病与感病植株的比例为1:1。这个结果证明,在墨西哥野生种S.pinnatisectum中存在一个抗晚疫病的单显性基因Rpi1。马铃薯甲虫抗性检测中,BC1群体的抗虫性分离比例为1:3.这表明其对甲虫的抗性是由多基因遗传控制的。在F1和BC1群体中利用分子标记结合集团分离分析法(BSA)对S.pinnatisectum中的晚疫病抗性基因Rpi1进行精细作图。根据马铃薯第7条染色体上RFLP标记TG20A和CP56之间的EST和STS标记的序列信息,合成了27对特异性PCR引物。获得一些与抗晚疫病基因Rpi1相关联的新的DNA标记。对BC1群体中大量的个体植株进行的分析表明,在马铃薯第7条染色体上位于抗晚疫病基因Rpi1两侧的两个标记S1c9和GP127-300,它们与Rpi1基因的遗传距离分别为1.17cM和3.89cM。这些标记被用来筛选两个细菌人工染色体(BAC)文库,并分离出与晚疫病抗性相关的90-125kb的BAC克隆,这些克隆将在后续的工作中通过图位克隆的方法而用于分离晚疫病抗性基因。同时分离与甲虫抗性紧密相关的分子标记的工作正在进行中。陈勤 孙慧生 杨继良 李海燕 Sanjib Nandy Debbie Beasley 王冬冬 Mark Goettel 2009中国马铃薯2009,23,5:4
9Discrete Event System Framework for Fault Diagnosis with Measurement Inconsistency:Case Study of Rogue DHCP Attack显示文摘Fault detection and diagnosis(FDD) facilitates reliable operation of systems. Various approaches have been proposed for FDD like Analytical redundancy(AR), Principal component analysis(PCA), Discrete event system(DES) model etc., in the literature. Performance of FDD schemes greatly depends on accuracy of the sensors which measure the system parameters.Due to various reasons like faults, communication errors etc.,sensors may occasionally miss or report erroneous values of some system parameters to FDD engine, resulting in measurement inconsistency of these parameters. Schemes like AR, PCA etc.,have mechanisms to handle measurement inconsistency, however,they are computationally heavy. DES based FDD techniques are widely used because of computational simplicity, but they cannot handle measurement inconsistency efficiently. Existing DES based schemes do not use Measurement inconsistent(MI)parameters for FDD. These parameters are not permanently unmeasurable or erroneous, so ignoring them may lead to weak diagnosis. To address this issue, we propose a Measurement inconsistent discrete event system(MIDES) framework, which uses MI parameters for FDD at the instances they are measured by the sensors. Otherwise, when they are unmeasurable or erroneously reported, the MIDES invokes an estimator diagnoser that predicts the state(s) the system is expected to be in, using the subsequent parameters measured by the other sensors. The efficacy of the proposed method is illustrated using a pumpvalve system. 10Binding interaction of phosphorus heterocycles with bovine serum albumin:A biochemical study显示文摘Interaction between bovine serum albumin(BSA) and phosphorus heterocycles(PHs) was studied using multispectroscopic techniques. The results indicated the high binding affinity of PHs to BSA as it quenches the intrinsic fluorescence of BSA. The experimental data suggested the fluorescence quenching mechanism between PHs and BSA as a dynamic quenching. From the UV–vis studies, the apparent association constant(K_(app)) was found to be 9.25×10~2, 1.27×10~4and 9.01×10~2L/mol for the interaction of BSA with PH-1, PH-2 and PH-3,respectively. According to the F?rster's non-radiation energy transfer(FRET) theory, the binding distances between BSA and PHs were calculated. The binding distances(r) of PH-1, PH-2 and PH-3 were found to be2.86, 3.03, and 5.12 nm, respectively, indicating energy transfer occurs between BSA and PHs. The binding constants of the PHs obtained from the fluorescence quenching data were found to be decreased with increase of temperature. The negative values of the thermodynamic parameters ΔH, ΔS and ΔG at different temperatures revealed that the binding process is spontaneous; hydrogen bonds and van der Waals interaction were the main force to stabilize the complex. The microenvironment of the protein-binding site was studied by synchronous fluorescence and circular dichroism(CD) techniques and data indicated that the conformation of BSA changed in the presence of PHs. 12Genetic variation in relation to adaptability of three mangrove species from the Indian Sundarbans assessed with RAPD and ISSR markers显示文摘Rich genetic polymorphism is important for plants to adapt to changes because it enables the plant to make anatomical,physiological and biochemical changes in response to abiotic stress.Geomorphologic characteristics,demographic interference and a cumulative decrease in freshwater influx in the Indian Sundarbans region have proved detrimental to some economically important plants.In this study,genetic polymorphism of three mangrove species,Xylocarpus granatum,Excoecaria agallocha,and Phoenix paludosa,was assessed using RAPD and ISSR molecular markers.X.granatum,already in distress in the Sundarbans,had the least genetic polymorphism,14.56%in the RAPD analysis and 12.92% in the ISSR.Relatively higher genetic polymorphism was recorded for the profusely growing E.agallocha and P.paludosa: 24.66 and26.4% in RAPD; 24.87 and 20.32% in ISSR analysis respectively.A UPGMA dendrogram constructed using the similarity matrix from RAPD,ISSR and combined datashowed that for X.granatum,the least and highest salinity zones clustered together,whereas for E.agallocha and P.paludosa,higher and lower salinity areas clustered in different clades.Nei's genetic diversity,calculated from RAPD and ISSR data,was also in accordance with 0.0637 and 0.0583 for X.granatum,respectively,much lower than0.0794 and 0.0818 for E.agallocha and 0.0799 and 0.0688 for P.paludosa.This opposing degree of polymorphism might be attributed to the profusely growing E.agallocha and P.paludosa and precarious status of X.granatum throughout the Indian Sundarbans.Nirjhar Dasgupta Paramita Nandy Chandan Sengupta Sauren Das 2018Journal of Forestry Research2018,29,2:3
13Development of a Polyclonal Antibody-based AC-ELISA and Its Comparison with PCR for Diagnosis of Canine Parvovirus Infection显示文摘A polyclonal antibody-based antigen-capture ELISA (AC-ELISA) has been developed for detection of Canine parvovirus (CPV) antigens in faecal samples of dogs. The assay uses rabbit anti-CPV polyclonal antibody as the capture antibody, guinea pig anti-CPV polyclonal antibody as tracing antibody and anti-guinea pig HRPO conjugate as the detection system. The optimum dilution of the capture antibody and the tracing antibody capable of detecting the CPV-2 antigens was found to be 1:1 600 and 1:400, respectively, in the check-board titration. In this study, a total of 152 samples (129 faecal samples and 23 cell culture supernatant) were tested both by AC-ELISA and by polymerase chain reaction (PCR). Of the samples tested, 69 and 78 samples were found positive by AC-ELISA and PCR, respectively. The AC-ELISA had relative sensitivity, relative specificity and accuracy of 88.4%, 100.0% and 91.4% respectively. The analytical sensitivity of AC-ELISA was estimated to be 102.8 TCID50/mL whereas PCR sensitivity was 100.8 TCID50/mL. 14In-Situ Hydrothermal Synthesis of Bi–Bi2O2CO3 Heterojunction Photocatalyst with Enhanced Visible Light Photocatalytic Activity显示文摘Bismuth containing nanomaterials recently received increasing attention with respect to environmental applications because of their low cost, high stability and nontoxicity. In this work, Bi–Bi_2O_2CO_3 heterojunctions were fabricated by in-situ decoration of Bi nanoparticles on Bi_2O_2CO_3 nanosheets via a simple hydrothermal synthesis approach. X-ray diffraction(XRD), scanning electron microscopy(SEM), transmission electron microscopy(TEM) and high-resolution TEM(HRTEM) were used to confirm the morphology of the nanosheet-like heterostructure of the Bi–Bi_2O_2CO_3 composite. Detailed ultrafast electronic spectroscopy reveals that the in-situ decoration of Bi nanoparticles on Bi_2O_2CO_3 nanosheets exhibit a dramatically enhanced electron-hole pair separation rate, which results in an extraordinarily high photocatalytic activity for the degradation of a model organic dye, methylene blue(MB) under visible light illumination. Cycling experiments revealed a good photochemical stability of the Bi–Bi_2O_2CO_3 heterojunction under repeated irradiation. 15Inhibition of Cdk5 increases osteoblast differentiation and bone mass and improves fracture healing显示文摘Identification of regulators of osteoblastogenesis that can be pharmacologically targeted is a major goal in combating osteoporosis,a common disease of the elderly population. Here, unbiased kinome RNAi screening in primary murine osteoblasts identified cyclin-dependent kinase 5(Cdk5) as a suppressor of osteoblast differentiation in both murine and human preosteoblastic cells. Cdk5 knockdown by si RNA, genetic deletion using the Cre-lox P system, or inhibition with the small molecule roscovitine enhanced osteoblastogenesis in vitro. Roscovitine treatment significantly enhanced bone mass by increasing osteoblastogenesis and improved fracture healing in mice. Mechanistically, downregulation of Cdk5 expression increased Erk phosphorylation, resulting in enhanced osteoblast-specific gene expression. Notably, simultaneous Cdk5 and Erk depletion abrogated the osteoblastogenesis conferred by Cdk5 depletion alone, suggesting that Cdk5 regulates osteoblast differentiation through MAPK pathway modulation.
