12-D P-wave velocity structure of lithosphere in the North China tectonic zone: Constraints from the Yancheng-Baotou deep seismic profile显示文摘We obtained the 2-D P-wave velocity structure of the lithosphere in the eastern North China Craton, Shanxi fault subsidence zone, and Yinchuan-Hetao fault subsidence zone by ray tracking technology based on six groups of clearly identified crustal phases and one group of lithospheric interface reflection phases from seismic recording sections of 21 shots along the 1300-km-long Yancheng-Baotou deep seismic wide-angle reflection/refraction profile. The results indicate significant differences between the lithospheric structure east and west of the Taihang Mountains, which is a gravity-gradient zone as well as a zone of abrupt change in lithospheric thickness and a separation zone of different rock components. East of the Taihang Mountains, the Mesozoic and Cenozoic lithospheric structure of the North China Craton has undergone strong reformation and destruction, resulting in the lithosphere thickness decreasing to 70–80 km. The North China Basin has a very thick Cenozoic sedimentary cover and the deepest point of crystalline basement is about 7.0 km, with the crustal thickness decreasing to about 31.0 km. The crystalline basement of the Luxi uplift zone is relatively shallow with a depth of 1.0–2.0 km and crustal thickness of 33.0–35.0 km. The Subei Basin has a thicker Cenozoic sedimentary cover and the bottom of its crystalline basement is at about 5.0–6.0 km with a crustal thickness of 31.0–32.0 km. The Tanlu fault is a deep fracture which cuts the lithosphere with a significant velocity structure difference on either side of the fault. The Tanlu fault plays an important role in the lithospheric destruction in the eastern part of the North China Craton. West of the Taihang Mountains, the crustal thickness increases significantly. The crust thickness beneath the Shanxi fault depression zone is about 46 km, and there is a low-velocity structure with a velocity of less than 6.1 km s?? in the upper part of the middle crust. Combined with other geophysical study results, our data shows that the lithospheric destruction at the Shaanxi-Shanxi fault depression zone and the Yinchuan-Hetao rift surrounding the Ordos block is non-uniform. The lithosphere thickness is about 80–90 km in the Datong-Baotou area, 75–137 km at the Dingxiang-Shenmu region, and about 80–120 km in the Anyang-Yichuan area. The non-uniform lithospheric destruction may be related to the ancient tectonic zone surrounding the Ordos block. This zone experienced multi-period tectonic events in the long-term process of its tectonic evolution and was repeatedly transformed and weakened. The weakening level is related to the interactions with the Ordos block. 2Upregulation of RIN3 induces endosomal dysfunction in Alzheimer’s disease显示文摘Background In Alzheimer’s Disease(AD),about one-third of the risk genes identified by GWAS encode proteins that function predominantly in the endocytic pathways.Among them,the Ras and Rab Interactor 3(RIN3)is a guanine nucleotide exchange factor(GEF)for the Rab5 small GTPase family and has been implicated to be a risk factor for both late onset AD(LOAD)and sporadic early onset AD(sEOAD).However,how RIN3 is linked to AD pathogenesis is currently undefined.Methods Quantitative PCR and immunoblotting were used to measure the RIN3 expression level in mouse brain tissues and cultured basal forebrain cholinergic neuron(BFCNs).Immunostaining was used to define subcellular localization of RIN3 and to visualize endosomal changes in cultured primary BFCNs and PC12 cells.Recombinant flag-tagged RIN3 protein was purified from HEK293T cells and was used to define RIN3-interactomes by mass spectrometry.RIN3-interacting partners were validated by co-immunoprecipitation,immunofluorescence and yeast two hybrid assays.Live imaging of primary neurons was used to examine axonal transport of amyloid precursor protein(APP)andβ-secretase 1(BACE1).Immunoblotting was used to detect protein expression,processing of APP and phosphorylated forms of Tau.Results We have shown that RIN3 mRNA level was significantly increased in the hippocampus and cortex of APP/PS1 mouse brain.Basal forebrain cholinergic neurons(BFCNs)cultured from E18 APP/PS1 mouse embryos also showed increased RIN3 expression accompanied by early endosome enlargement.In addition,via its proline rich domain,RIN3 recruited BIN1(bridging integrator 1)and CD2AP(CD2 associated protein),two other AD risk factors,to early endosomes.Interestingly,overexpression of RIN3 or CD2AP promoted APP cleavage to increase its carboxyl terminal fragments(CTFs)in PC12 cells.Upregulation of RIN3 or the neuronal isoform of BIN1 increased phosphorylated Tau level.Therefore,upregulation of RIN3 expression promoted accumulation of APP CTFs and increased phosphorylated Tau.These effects by RIN3 was rescued by the expression of a dominant negative Rab5(Rab5S34N)construct.Our study has thus pointed to that RIN3 acts through Rab5 to impact endosomal trafficking and signaling.Conclusion RIN3 is significantly upregulated and correlated with endosomal dysfunction in APP/PS1 mouse.Through interacting with BIN1 and CD2AP,increased RIN3 expression alters axonal trafficking and procession of APP.Together with our previous studies,our current work has thus provided important insights into the role of RIN3 in regulating endosomal signaling and trafficking.Ruinan Shen Xiaobei Zhao Lu He Yongbo Ding Wei Xu Suzhen Lin Savannah Fang Wanlin Yang Kijung Sung Brian Spencer Robert A.Rissman Ming Lei Jianqing Ding Chengbiao Wu 2020Translational Neurodegeneration2020,9,2:1
3Comparative transcriptome analysis of Phascolosoma esculenta under different salinities显示文摘Salinity is an important parameter that influences the developmental progress of aquatic organisms.The larvae of Phascolosoma esculenta developed into the early stage trochosphere,the late stage trochosphere and the early stage pelagosphere in the low-salinity group(15),the middle group(22.5)and the high-salinity group(30),respectively,after being incubated for 50 h,according to morphological observation.The results showed that high salinity could accelerate the cellular development of P.esculenta.To elucidate the molecular mechanism of larval development of P.esculenta under different salinities,we performed transcriptome analysis of the larvae in P.esculenta.Nine transcriptome libraries of P.esculenta larvae were constructed,i.e.,the low-salinity group(A1,A2 and A3),the middle group(B1,B2 and B3)and the high-salinity group(C1,C2 and C3),and each group with three repeats.These nine paired-end libraries were sequenced using the Illumina HiSeq 2500 platform.In total,57.63 Gb clean data were obtained and de novo assembled into 320,527 unigenes with mean lengths of 482.62 bp and N50 of 529 bp.A total of 249,162 unigenes were significantly matched to known unique proteins.The gene ontology(GO)annotation and the Kyoto Encyclopedia of Genes and Genomes(KEGG)were used to determine the regulation pathways and candidate genes that are associated with the process of larval development.A total number of 12,811,16,991 and 1735 transcripts were identified as differentially expressed genes(DEGs)from the comparison of A vs.B,A vs.C,and B vs.C,respectively.Of these differentially expressed genes,many DEGs that were upregulated in the high-salinity group were involved in the function that controls the larval development of P.esculenta,such as the neuroactive ligand-receptor interaction and ECM-receptor interaction.In addition,the results of the transcriptome analysis were verified by quantitative real-time PCR(qRT-PCR).After morphological observation and transcriptome analysis,we think high salinity(30)is more suitable than medium(22.5)or low(15)salinity for the development of P.esculenta larvae.This information provides guidance for artificial breeding and paves the way to the further research on the individual development of P.esculenta under different salinities.Xiangyang Lin Suzhen Deng Xiande Liu Chenhui Zhong Kun Qiao Leibin Li 2020Aquaculture and Fisheries2020,5,6:1

53-[3-(3-florophenyl-2-propyn-1-ylthio)-1,2,5-thiadiazol-4-yl]-1,2,5,6-tetrahydro-1-methylpyridine oxalate,a novel xanomeline derivative,improves neural cells proliferation and survival in adult mice显示文摘The present study analyzed the influence of 3-[3-(3-florophenyl-2-propyn-1-ylthio)-1, 2, 5-thiadiazol-4-yl]-1, 2, 5, 6-tetrahydro-1-methylpyridine oxalate (EUK1001), a novel xanomeline derivative of the M1/M4 receptor agonist, on hippocampal neurogenesis in adult C57BL6 mice. Results showed that 15-day EUK1001 treatment via intraperitoneal injection promoted neural cell proliferation in the dentate gyrus, although cell differentiation did not change. The majority of bromodeoxyuridine-positive cells co-expressed the immature neuronal marker doublecortin. In addition, the level of neurogenesis in the subventricular zone was not altered. Brain-derived neurotrophic factor mRNA expression was up-regulated following EUK1001 treatment, but no change was observed in expression of camp-responsive element binding protein 1, paired box gene 6, vascular endothelial growth factor alpha, neurogenic differentiation factor 1, and wingless-related mouse mammary tumor virus integration site 3A mRNA. 6Noninherited Factors in Fetal Congenital Heart Diseases Based on Bayesian Network:A Large Multicenter Study显示文摘Background:Current studies have confirmed that fetal congenital heart diseases(CHDs)are caused by various factors.However,the quantitative risk of CHD is not clear given the combined effects of multiple factors.Objective:This cross-sectional study aimed to detect associated factors of fetal CHD using a Bayesian network in a large sample and quantitatively analyze relative risk ratios(RRs).Methods:Pregnant women who underwent fetal echocardiography(N=16,086 including 3,312 with CHD fetuses)were analyzed.Twenty-six maternal and fetal factors were obtained.A Bayesian network is constructed based on all variables through structural learning and parameter learning methods to find the environmental factors that directly and indirectly associated with outcome,and the probability of fetal CHD in the two groups is predicted through a junction tree reasoning algorithm,so as to obtain RR for fetal CHD under different exposure factor combinations.Taking into account the effect of gestational week on the accuracy of model prediction,we conducted sensitivity analysis on gestational week groups.Results:The single-factor analysis showed that the RRs for the numbers of births,spontaneous abortions,and parental smoking were 1.50,1.38,and 1.11(P<0.001),respectively.The risk gradually increased with the synergistic effect of ranging from one to more environmental factors above.The risk was higher among subjects with five synergistic factors,including the number of births,upper respiratory tract infection during early pregnancy,anemia,and mental stress as well as a history of spontaneous abortions or parental smoking,than in those with less than 5 factors(RR=2.62 or 2.28,P<0.001).This result was consistent across the participants grouped by GWs.Conclusion:We identified six factors that were directly associated with fetal CHD.A higher number of these factors led to a higher risk of CHD.These findings suggest that it is important to strengthen healthcare and prenatal counseling for women with these factors.Yanping Ruan Xiangyu Liu Haogang Zhu Yijie Lu Xiaowei Liu Jiancheng Han Lin Sun Ye Zhang Xiaoyan Gu Ying Zhao Lei Li Suzhen Ran Jingli Chen Qiong Yu Yan Xu Hongmei Xia Yihua He 2021Congenital Heart Disease2021,16,6:0
