| 1 | Genomic prediction of fruit texture and training population optimization towards the application of genomic selection in apple显示文摘Texture is a complex trait and a major component of fruit quality in apple.While the major effect of MdPG1,a gene controlling firmness,has already been exploited in elite cultivars,the genetic basis of crispness remains poorly understood.To further improve fruit texture,harnessing loci with minor effects via genomic selection is therefore necessary.In this study,we measured acoustic and mechanical features in 537 genotypes to dissect the firmness and crispness components of fruit texture.Predictions of across-year phenotypic values for these components were calculated using a model calibrated with 8,294 SNP markers.The best prediction accuracies following cross-validations within the training set of 259 genotypes were obtained for the acoustic linear distance(0.64).Predictions for biparental families using the entire training set varied from low to high accuracy,depending on the family considered.While adding siblings or half-siblings into the training set did not clearly improve predictions,we performed an optimization of the training set size and composition for each validation set.This allowed us to increase prediction accuracies by 0.17 on average,with a maximal accuracy of 0.81 when predicting firmness in the‘Gala’בPink Lady’family.Our results therefore identified key genetic parameters to consider when deploying genomic selection for texture in apple.In particular,we advise to rely on a large training population,with high phenotypic variability from which a‘tailored training population’can be extracted using a priori information on genetic relatedness,in order to predict a specific target population. | Morgane Roth Hélène Muranty Mario Di Guardo Walter Guerra Andrea Patocchi Fabrizio Costa | 2020 | Horticulture Research2020,7,1: | 4 |

| 3 | Repeated Multiview Imaging for Estimating Seedling Tiller Counts of Wheat Genotypes Using Drones显示文摘Early generation breeding nurseries with thousands of genotypes in single-row plots are well suited to capitalize on high throughput phenotyping.Nevertheless,methods to monitor the intrinsically hard-to-phenotype early development of wheat are yet rare.We aimed to develop proxy measures for the rate of plant emergence,the number of tillers,and the beginning of stem elongation using drone-based imagery.We used RGB images(ground sampling distance of 3mm pixel-1)acquired by repeated flights(≥2 flights per week)to quantify temporal changes of visible leaf area.To exploit the information contained in the multitude of viewing angles within the RGB images,we processed them to multiview ground cover images showing plant pixel fractions.Based on these images,we trained a support vector machine for the beginning of stem elongation(GS30).Using the GS30 as key point,we subsequently extracted plant and tiller counts using a watershed algorithm and growth modeling,respectively.Our results show that determination coefficients of predictions are moderate for plant count(R^(2)=0:52),but strong for tiller count(R^(2)=0:86)and GS30(R^(2)=0:77).Heritabilities are superior to manual measurements for plant count and tiller count,but inferior for GS30 measurements.Increasing the selection intensity due to throughput may overcome this limitation.Multiview image traits can replace hand measurements with high efficiency(85-223%).We therefore conclude that multiview images have a high potential to become a standard tool in plant phenomics. | Lukas Roth Moritz Camenzind Helge Aasen Lukas Kronenberg Christoph Barendregt Karl-Heinz Camp Achim Walter Norbert Kirchgessner Andreas Hund | 2020 | Plant Phenomics2020,2,1: | 2 |
| 4 | Multi-analyte biosensor interface for real-time monitoring of 3D microtissue spheroids in hanging-drop networks显示文摘Microfluidics is becoming a technology of growing interest for building microphysiological systems with integrated read-out functionalities.Here we present the integration of enzyme-based multi-analyte biosensors into a multi-tissue culture platform for‘body-on-a-chip’applications.The microfluidic platform is based on the technology of hanging-drop networks,which is designed for the formation,cultivation,and analysis of fluidically interconnected organotypic spherical three-dimensional(3D)microtissues of multiple cell types.The sensor modules were designed as small glass plug-ins featuring four platinum working electrodes,a platinum counter electrode,and an Ag/AgCl reference electrode.They were placed directly into the ceiling substrate from which the hanging drops that host the spheroid cultures are suspended.The electrodes were functionalized with oxidase enzymes to enable continuous monitoring of lactate and glucose through amperometry.The biosensors featured high sensitivities of 322±41 nA mM^(−1) mm^(−2) for glucose and 443±37 nA mM^(−1) mm^(−2) for lactate;the corresponding limits of detection were below 10μM.The proposed technology enabled tissue-size-dependent,real-time detection of lactate secretion from single human colon cancer microtissues cultured in the hanging drops.Furthermore,glucose consumption and lactate secretion were monitored in parallel,and the impact of different culture conditions on the metabolism of cancer microtissues was recorded in real-time. | Patrick M.Misun Jörg Rothe Yannick R.F.Schmid Andreas Hierlemann Olivier Frey | 2016 | Microsystems & Nanoengineering2016,2,1: | 1 |
