1Predictive value of Ki67 and p53 in locally advanced rectal cancer:Correlation with thymidylate synthase and histopathological tumor regression after neoadjuvant 5-FU-based chemoradiotherapy显示文摘AIM: To investigate the predictive value of Ki67 and p53 and their correlation with thymidylate synthase (TS) gene expression in a rectal cancer patient cohort treated according to a standardized recommended neoadjuvant treatment regimen.METHODS: Formalin fixed, paraffin embedded pre-therapeutical tumor biopsies (n = 22) and post-therapeutical resection specimens (n = 40) from patients with rectal adenocarcinoma (clinical UICC stage Ⅱ/Ⅲ) receiving standardized neoadjuvant 5-fluorouracil (5-FU) based chemoradiotherapy were studied for Ki67 and p53 expression by immunohistochemistry and correlated with TS mRNA expression by quantitative TaqMan real-time PCR after laser microdissection. The results were compared with histopathological tumor regression according to a standardized semiquantitative score grading system.RESULTS: Responders (patients with high tumor regression) showed a significantly lower Ki67 expression than non-responders in the pre-therapeutical tumor biopsies (81.2% vs 16.7%; P < 0.05) as well as in the post-therapeutical resection specimens (75.8% vs 14.3%; P < 0.01). High TS mRNA expression was significantly correlated with a high Ki67 index and low TS mRNA expression was significantly correlated with a low Ki67 index in the pre-therapeutical tumor biopsies (corr. coef. = 0.46; P < 0.01) as well as in the post-therapeutical resection specimens (corr. coef. = 0.40; P < 0.05). No significant association was found between p53 and TS mRNA expression or tumor regression.CONCLUSION: Ki67 has, like TS, predictive value in rectal cancer patients after neoadjuvant 5-FU based chemoradiotherapy. 3Intraductal ultrasound substantiates diagnostics of bile duct strictures of uncertain etiology显示文摘AIM:To report the largest patient cohort study investigating the diagnostic yield of intraductal ultrasound (IDUS) in indeterminate strictures of the common bile duct.METHODS:A patient cohort with bile duct strictures of unknown etiology was examined by IDUS.Sensitivity,specificity and accuracy rates of IDUS were calculated relating to the definite diagnoses proved by histopathology or long-term follow-up in those patients who did not undergo surgery.Analysis of the endosonographic report allowed drawing conclusions with respect to the T and N staging in 147 patients.IDUS staging was compared to the postoperative histopathological staging data allowing calculation of sensitivity,specificity and accuracy rates for T and N stages.The endoscopic retrograde cholangio-pancreatography and IDUS procedures were performed under fluoroscopic guidance using a side-viewing duodenoscope (Olympus TJF 160,Olympus,Ltd.,Tokyo,Japan).All procedures were performed under conscious sedation (propofol combined with pethidine) according to the German guidelines.For IDUS,a 6 F or 8 F ultrasound miniprobe was employed with a radial scanner of 15-20 MHz at the tip of the probe (Aloka Co.,Tokyo,Japan).RESULTS:A total of 397 patients (210 males,187 females,mean age 61.43 ± 13 years) with indeterminate bile duct strictures were included.Two hundred and sixty-four patients were referred to the department of surgery for operative exploration,thus surgical histopathological correlation was available for those patients.Out of 264 patients,174 had malignant disease proven by surgery,in 90 patients benign disease was found.In these patients decision for surgical exploration was made due to suspicion for malignant disease in multimodal diagnostics (computed tomography scan,endoscopic ultrasound or magnetic resonance imaging).Twenty benign bile duct strictures were misclassified by IDUS as malignant while 14 patients with malignant strictures were initially misdiagnosed by IDUS as benign resulting in sensitivity,specificity and accuracy ratesof 93.2%,89.5% and 91.4%,respectively.In the subgroup analysis of malignancy prediction,IDUS showed best performance in cholangiocellular carcinoma as underlying disease (sensitivity rate,97.6%) followed by pancreatic carcinoma (93.8%),gallbladder cancer (88.9%) and ampullary cancer (80.8%).A total of 133 patients were not surgically explored.32 patients had palliative therapy due to extended tumor disease in IDUS and other imaging modalities.Ninety-five patients had benign diagnosis by IDUS,forceps biopsy and radiographic imaging and were followed by a surveillance protocol with a follow-up of at least 12 mo;the mean follow-up was 39.7 mo.Tumor localization within the common bile duct did not have a significant influence on prediction of malignancy by IDUS.The accuracy rate for discriminating early T stage tumors (T1) was 84% while for T2 and T3 malignancies the accuracy rates were 73% and 71%,respectively.Relating to N0 and N1 staging,IDUS procedure achieved accuracy rates of 69% for N0 and N1,respectively.Limitations:Pretest likelihood of 52% may not rule out bias and overinterpretation due to the clinical scenario or other prior performed imaging tests.CONCLUSION:IDUS shows good results for accurate diagnostics of bile duct strictures of uncertain etiology thus allowing for adequate further clinical management.Tobias Meister Hauke S Heinzow Carina Woestmeyer Philipp Lenz Josef Menzel Torsten Kucharzik Wolfram Domschke Dirk Domagk 2013World Journal of Gastroenterology2013,19,6:19
4Cenozoic plant diversity of Yunnan: A review显示文摘Yunnan in southwestern China is renowned for its high plant diversity.To understand how this modern botanical richness formed,it is critical to investigate the past biodiversity throughout the geological time.In this review,we present a summary on plant diversity,floristics and climates in the Cenozoic of Yunnan and document their changes,by compiling published palaeobotanical sources.Our review demonstrates that thus far a total of 386 fossil species of ferns,gymnosperms and angiosperms belonging to 170 genera within 66 families have been reported from the Cenozoic,particularly the Neogene,of Yunnan.Angiosperms display the highest richness represented by 353 species grouped into 155 genera within 60 families,with Fagaceae,Fabaceae,Lauraceae and Juglandaceae being the most diversified.Most of the families and genera recorded as fossils still occur in Yunnan,but seven genera have disappeared,including Berryophyllum,Cedrelospermum,Cedrus,Palaeocarya,Podocarpium,Sequoia and Wataria.The regional extinction of these genera is commonly referred to an aridification of the dry season associated with Asian monsoon development.Floristic analyses indicate that in the late Miocene,Yunnan had three floristic regions:a northern subtropical floristic region in the northeast,a subtropical floristic region in the east,and a tropical floristic region in the southwest.In the late Pliocene,Yunnan saw two kinds of floristic regions:a subalpine floristic region in the northwest,and two subtropical floristic regions separately in the southwest and the eastern center.These floristic concepts are verified by results from our areal type analyses which suggest that in the Miocene southwestern Yunnan supported the most Pantropic elements,while in the Pliocene southwestern Yunnan had abundant Tropical Asia(Indo e Malaysia)type and East Asia and North America disjunct type that were absent from northwestern Yunnan.From the late Miocene to late Pliocene through to the present,floristic composition and vegetation types changed markedly,presumably in response to altitude changes and coeval global cooling.An integration of palaeoclimate data suggests that during the Neogene Yunnan was warmer and wetter than today.Moreover,northern Yunnan witnessed a pronounced temperature decline,while southern Yunnan experienced only moderate temperature changes.Summer precipitation was consistently higher than winter precipitation,suggesting a rainfall seasonality.This summary on palaeoclimates helps us to understand under what conditions plant diversity occurred and evolved in Yunnan throughout the Cenozoic.Yongjiang Huang Linbo Jia QiongWang Volker Mosbrugger Torsten Utescher Tao Su Zhekun Zhou 2016Plant Diversity2016,38,6:8
5Developing More Effective Enhanced Biochar Fertilisers for Improvement of Pepper Yield and Quality显示文摘Utilization of biochar at high application rates can increase soil C and crop yields, decrease greenhouse gas emissions and reduce nutrient run-off from soils. However, the high application rate of 10 t ha-1may not return a profit to the farmer due to the high cost of biochar. In this study biochar was modified through pre-treating the biomass and post-treating with phosphoric acid, minerals and different chemical fertilisers to study the effects of two new enhanced biochar fertilisers on the yield and quality of green pepper in a field experiment with 5 fertilisation treatments and 3 replications. The two new biochar fertilisers significantly(P < 0.05) increased the yield of green pepper(11.33–11.47 t ha-1), compared with the conventional chemical fertiliser(9.72 t ha-1). The biochar fertiliser treatments improved the vitamin C content of green pepper from 236.99 to 278.28 mg kg-1, and also significantly(P < 0.05) reduced the nitrate content from 132.32 to 101.92 mg kg-1, compared with chemical fertiliser. 7The Muruntau gold deposit(Uzbekistan)--A unique ancient hydrothermal system in the southern Tien Shan显示文摘The Muruntau gold deposit in the Central Kyzylkum,Uzbekistan is one of the largest single gold deposits worldwide.Data available from the literature are reviewed with the aim to(1) integrate the present knowledge on this unique deposit from Russian and English literature;(2) show the considerable progress made in the understanding of the genesis of the Muruntau deposit during the last decades;and(3) point to problems still open for future research.Deposit formation occurred through a multi-stage process involving sedimentation,regional metamorphism including thrusting,magmatism with formation of hornfels aureoles and several stages of hydrothermal activity.According to recent knowledge,synsedimentary or pure metamorphic formation of gold mineralization seems unlikely.The role of granite magmatism occurring roughly within the same time interval as the main hydrothermal gold precipitation remains uncertain.There are no signs of interaction of matter between the magma(s) and the hydrothermal system(s).On the other hand,there was an intense,high-temperature(above 400 ℃)fluid- wall rock interaction resulting in the formation of gold-bearing,cone-like stockworks with veins,veinlets and gold-bearing metasomatites.Several chemical and isotope indicators hint at an involvement of lower-crustal or mantle-related sources as well as of surface waters in ore formation.Deposit formation through brecciation involving explosion,hydrothermal or tectonic breccias might explain these data.Further investigations on breccia formation as well as on the exact timing of relevant sedimentary,metamorphic,magmatic and hydrothermal events are recommended.Ulf Kempe Torsten Graupner Reimar Seltmann Hugo de Boorder Alla Dolgopolova Maarten Zeylmans van Emmichoven 2016Geoscience Frontiers2016,7,3:7
8Sea surface temperature record from the north of the East China Sea since late Holocene显示文摘Using the alkenone paleotemperature index U k 37′, a high-resolution sea surface temperature (SST) record since 3600 a BP was reconstructed from the mud area in the north of the East China Sea. Combining with the grain size distribution curve of sensitive grain size group, which may reflect the East Asia Winter Monsoon activity, the palaeoenvironmental evolution cycle throughout the late Holocene in the area was obtained. The marine environment evolution during the last 3600 years displays a five-stage trend. (1) Temperature descending period from 0.85 cal. ka BP to present. The maximum temperature decrease amplitude is 2℃. The winter monsoon intensified and ‘Little Ice Age’ were recorded in this period. (2) Warming period from 1.90 to 0.85 cal. ka BP. The mean temperature increase amplitude is 0.8℃. The Sui-Tang warming period was recorded at about 0.85―1.35 cal. ka BP and a prominent cooling event was recorded at 1.4 cal. ka BP in this period. (3) Temperature descending period from 2.55 to 1.90 cal. ka BP. Temperature cooling amplitude is 0.9℃. This period is coincident with an integrated temperature circle recorded in the Antarctic ice core, with the temperature changes from a slow cooling stage to a rapid warming stage. (4) Temperature comparatively stable with a little ascending period from 3.2 to 2.55 cal. ka BP. Temperature warming amplitude is 0.3℃. This period is coincident with the temperature fluctuant ascending period recorded in Antarctic ice core. (5) Temperature comparatively stable with little descending period from 3.6 to 3.2 cal. ka BP. This period corresponds with the temperature fluctuant cooling period recorded in Antarctic ice core. Basically, those five periods were coincident with the Antarctic ice core record. 9How to perform gastrointestinal ultrasound: anatomy and normal findings显示文摘Gastrointestinal ultrasound is a practical, safe, cheap and reproducible diagnostic tool in inflammatorybowel disease gaining global prominence amongst clinicians. Understanding the embryological processes of the intestinal tract assists in the interpretation of abnormal sonographic findings. In general terms, the examination principally comprises interrogation of the colon, mesentery and small intestine using both lowfrequency and high-frequency probes. Interpretation of findings on GIUS includes assessment of bowel wall thickness, symmetry of this thickness, evidence of transmural changes, assessment of vascularity using Doppler imaging and assessment of other specific features including lymph nodes, mesentery and luminal motility. In addition to B-mode imaging, transperineal ultrasonography, elastography and contrast-enhanced ultrasonography are useful adjuncts. 13Multilayer optics for the EUV and soft X-rays显示文摘The demand to enhance the optical resolution, to structure and observe ever smaller details, has pushed the way towards the EUV and soft X-rays. Induced mainly by the production of more powerful electronic circuits with the aid of projection lithography, optics developments in recent years can be characterized by the use of electromagnetic radiation with smaller wavelength. The good prospects of the EUV and soft X-rays for next generation lithography systems (λ=13.5 nm), microscopy in the “water window” (λ=2.3~4.4 nm), astronomy (λ=5~31 nm), spectroscopy, plasma diagnostics and EUV/soft X-ray laser research have led to considerable progress in the development of different multilayer optics. Since optical systems in the EUV/soft X-ray spectral region consist of several mirror elements a maximum reflectivity of each multilayer is essential for a high throughput. 15A Demand Response System for Wind Power Integration: Greenhouse Gas Mitigation and Reduction of Generator Cycling显示文摘A smart grid power system for a small region consisting of 1,000 residential homes with electric heating appliances from the demand side,and a generic generation mix of nuclear,hydro,coal,gas and oil-based generators representing the supply side,is investigated using agent-based simulations.The simulation includes a transactive load control in a real-time pricing electricity market.The study investigates the impacts of adding wind power and demand response(DR)on both greenhouse gas(GHG)emissions and generator cycling requirements.The results demonstrate and quantify the effectiveness of DR in mitigating the variability of renewable generation.The extent to which greenhouse gas emissions can be mitigated is found to be highly dependent on the mix of generators and their operational capacity factors.It is expected that the effects of demand response on electricity use can reduce dependency on fossil fuel-based electricity generation.However,the anticipated mitigation of GHG emissions is found to dependent on the number and efficiency of fossil fuel generators,and especially on the capacity factor at which they operate.Therefore,if a generator(the marginal seller)is forced to use less efficient fossil fuel power generation schemes,it will result in higher GHG emissions.The simulations show that DR can yield a small reduction in GHG emissions,but also lead to a smaller increase in emissions in circumstances when,for example,a generator(the marginal seller)is forced to use less efficient fossil fuel power generation schemes.Nonetheless,DR is shown to enhance overall system operation,particularly by facilitating increased penetration of variable renewable electricity generation without jeopardizing grid operation reliability.DR reduces the amount of generator cycling by an increased order of magnitude,thereby reducing wear and tear,improving generator efficiency,and avoiding the need for additional operating reserves.The effectiveness of DR for these uses depends on the participation of responsive loads,and this study highlights the need to maintain a certain degree of diversity of loads to ensure they can provide adequate responsiveness to the changing grid conditions.Torsten Broeer Francis K.Tuffner Anaissia Franca Nedjib Djilali 2018CSEE Journal of Power and Energy Systems2018,4,2:3
16联合分析血小板特异性抗体和瞬逝生物传感器技术检测血小板抗体的方法比较显示文摘目的应用联合分析血小板特异性抗体(specificpplateletantibodies,SASPA)和瞬逝生物传感器技术(evanescentbiosensortechnology,EVA)分别检测血小板特异性同种抗体和自身抗体,对两者进行方法学比较。方法收集94例存在HPA-1a、HPA-Sb、HLA-I和自身抗体的患者血浆标本,制备血小板膜糖蛋白-血小板特异性抗体复合物,分别用SASPA和EVA方法对复合物进行检测,对SASPA检测所得平均荧光强度值(MFI)和EVA检测所得斜率值(Slope)作统计学分析和比较。结果SASPA方法对HPA-1a、HPA-Sb、HLA-I和自身抗体检测的敏感度分别为74.1%、86.9%、86.9%和40.9%,EVA方法的敏感度分别为88.9%、78.3%、60.9%和72.7%。人类血小板抗原(HPA)-1a抗体检测中,MFI与Slope表现为强相关性,相关性系数为r=0.95(P〈0.05)。结论SASPA和EVA技术对传统的单克隆抗体特异性血小板抗原固定实验(MAIPA)作了继承和改良,SASPA方法可以用极少量的血小板同时检测多种血小板特异性抗体,提高了检测效率;EVA方法将血小板裂解后的实验时间缩短为10min,极大地简化了操作过程,减少了检测时间。SASPA和EVA检测方法的建立对临床血小板免疫相关疾病的诊断和血小板输血领域的发展具有深远意义。宋文倩 Torsten J Schulze Margit Baade Harald Kluter Peter Bugert 2017国际免疫学杂志2017,40,1:3

20新型糖氨基酸类化合物的合成研究显示文摘糖氨基酸(sugar amino acids,SAAs)是一类同时含有氨基和羧基的糖类衍生物,具有糖和氨基酸的结构特征和化学反应特性,常作为多功能合成砌块用于组合化学的研究、糖模拟物和多肽模拟物的构建等.因此,该类化合物的设计与合成受到国内外化学家的持续关注.在本文中,我们发展了一种简单、高效的合成新型糖氨基酸的方法,从廉价易得的3,4,6-O-三乙酰基-D-葡萄糖烯和3,4,6-O-三乙酰基-D-半乳糖烯出发,在硝酸铈铵(CAN)的作用下通过一步自由基加成反应分别得到2-C-二羧酸酯类化合物gluco-2和galacto-2,再经过脱羧、脱乙酰、碘代、叠氮化钠取代和一锅法还原氨化反应,以最短直线步骤6步实现了糖氨基酸gluco-7(总收率34%)和galacto-7(总收率19%)的合成.所有合成的化合物均经过IR,1H NMR,13C NMR和HRMS的表征.汪学彬 王晓丽 胡静 王兆亚 Pimpalpalle Tukaram M Linker Torsten 尹健 2015化学学报2015,73,7:3
