| 1 | Increased hepcidin expression in colorectal carcinogenesis显示文摘AIM:To investigate whether the iron stores regulator hepcidin is implicated in colon cancer-associated anae- mia and whether it might have a role in colorectal car- cinogenesis. METHODS: Mass spectrometry (MALDI-TOF MS and SELDI-TOF MS) was employed to measure hepcidin in urine collected from 56 patients with colorectal cancer. Quantitative Real Time RT-PCR was utilised to determine hepcidin mRNA expression in colorectal cancer tissue. Hepcidin cellular localisation was determined using im- munohistochemistry. RESULTS: We demonstrate that whilst urinary hepcidin expression was not correlated with anaemia it was posi- tively associated with increasing T-stage of colorectal cancer (P < 0.05). Furthermore, we report that hepcidin mRNA is expressed in 34% of colorectal cancer tissue specimens and was correlated with ferroportin repres- sion. This was supported by hepcidin immunoreactivity in colorectal cancer tissue. CONCLUSION: We demonstrate that systemic hepcidin expression is unlikely to be the cause of the systemic anaemia associated with colorectal cancer. | 2 | Chemoprotective effects of curcumin in esophageal epithelial cells exposed to bile acids显示文摘AIM:To investigate the ability of curcumin to counteract the impact of bile acids on gene expression of esophageal epithelial cells.METHODS:An esophageal epithelial cell line(HET1A)was treated with curcumin in the presence of deoxycholic acid.Cell proliferation and viability assays were used to establish an appropriate dose range for curcumin.The combined and individual effects of curcumin and bile acid on cyclooxygenase-2(COX-2)and superoxide dismutase(SOD-1 and SOD-2)gene expression were also assessed.RESULTS:Curcumin in a dose range of 10-100μmol/L displayed minimal inhibition of HET-1A cell viability.Deoxycholic acid at a concentration of 200μmol/L caused a 2.4-fold increase in COX-2 gene expression compared to vehicle control.The increased expression of COX-2 induced by deoxycholic acid was partially reversed by the addition of curcumin,and curcumin reduced COX-2 expression 3.3-to 1.3-fold.HET-1A cells exposed to bile acid yielded reduced expression of SOD-1 and SOD-2 genes with the exception that high dose deoxycholic acid at 200μmol/L led to a 3-fold increase in SOD-2 expression.The addition of curcumin treatment partially reversed the bile acid-induced reduction in SOD-1 expression at all concentrations of curcumin tested.CONCLUSION:Curcumin reverses bile acid suppression of gene expression of SOD-1.Curcumin is also able to inhibit bile acid induction of COX-2 gene expression. | Matthew R Bower Harini S Aiyer Robert CG Martin Yan Li | 2010 | World Journal of Gastroenterology2010,16,33: | 10 |

| 4 | Impact and clinical usefulness of genetic data in the surgical management of colorectal cancer liver metastasis: a narrative review显示文摘Importance:In patients who undergo surgery for colorectal cancer liver metastases(CRLM),a number of somatic mutations have been associated with worse overall(OS)and recurrence-free survival(RFS).Although useful,an association with prognosis does not necessarily equate to an impact on surgical management.Objective:The aim of this review was to investigate whether the best-studied somatic mutations impact surgical management of CRLM by informing:(I)post-hepatectomy surveillance;(II)selection of surgical technique;(III)selection of optimal margin width;and(IV)selection of patients for surgery.Lastly,we discuss the refinement of genetic data from overall mutation status to specific variants,as well as lesser studied somatic mutations.Evidence Review:We conducted a computerized search using PubMed and Google Scholar for reports published so far,using mesh headings and keywords related to genetic data and CRLM.Findings:Genetic data may impact surgical management of CRLM in three ways.Firstly,KRAS mutations can predict lung recurrences.Secondly,KRAS mutations may help tailor margin width.Thirdly,KRAS mutations may help tailor surgical technique.Conclusions:Although genetic data may impact post-hepatectomy surveillance,selection of surgical technique and optimal margin width,their use to guide surgical selection remains elusive,as the data cannot support denying surgery to patients according to their somatic mutation profile. | Georgios Antonios Margonis Martin E.Kreis Jaeyun Jane Wang Carsten Kamphues Christopher L.Wolfgang Matthew J.Weiss | 2020 | Hepatobiliary Surgery and Nutrition2020,9,6: | 3 |
| 5 | Domestication and crop evolution of wheat andbarley: Genes, genomics, and future directions显示文摘Wheat and barley are two of the founder crops of the agricultural revolution that took place 10,000 years ago in the Fertile Crescent and both crops remain among the world's most important crops. Domestication of these crops from their wild ancestors required the evolution of traits useful to humans, rather than survival in their natural environment. Of these traits, grain retention and threshability, yield improvement, changes to photoperiod sensitivity and nutritional value are most pronounced between wild and domesticated forms. Knowledge about the geographical origins of these crops and the genes responsible for domestication traits largely pre-dates the era of nextgeneration sequencing, although sequencing will lead to new insights. Molecular markers were initially used to calculate distance(relatedness), genetic diversity and to generate genetic maps which were useful in cloning major domestication genes. Both crops are characterized by large,complex genomes which were long thought to be beyond the scope of whole-genome sequencing. However, advances in sequencing technologies have improved the state of genomic resources for both wheat and barley. | 6 | Role of post-translational modification of the Y box binding protein 1 in human cancers显示文摘Y box binding protein-1(YBX1)belongs to a DNA-and RNA-binding family of transcription factors,containing the highly conserved cold shock domain(CSD).YBX1 is involved in a number of cellular functions including transcription,translation,DNA damage repair etc.,and it is upregulated during times of environmental stress.YBX1 is localized in both the cytoplasm and the nucleus.There,its nuclear translocation is observed in a number of cancers and is associated with poor prognosis and disease progression.Additionally,YBX1 expression is upregulated in a variety of cancers,pointing towards its role as a potential oncogene.Under certain circumstances,YBX1 also promotes the expression of multidrug resistance 1(MDR1)gene,which is involved in the development of drug resistance.Thus,it is critical to understand the mechanism of YBX1 regulation and its downstream effects on promoting cancer development.A number of recent studies have highlighted the mechanisms of YBX1 regulation.Mass spectrometric analyses have reported several post-translational modifications that possibly play an important role in modulating YBX1 function.Phosphorylation is the most widely occurring post-translational modification in YBX1.In vivo analyses of sites like S102 and more recently,S165 illustrate the relationship of post-translational regulation of YBX1 in promoting cell proliferation and tumor growth.This review provides a comprehensive and up-to-date account of post-translational modifications identified in YBX1.This knowledge is a key in allowing us to better understand the mechanism of YBX1 regulation,which will aid in development of novel therapeutic strategies to target YBX1 in many types of cancer in the future. | Lakshmi Prabhu Antja-Voy Hartley Matthew Martin Fadumo Warsame Emily Sun Tao Lu | 2015 | Genes & Diseases2015,2,3: | 3 |
| 7 | Mitigation of atrazine,S-metolachlor,and diazinon using common emergent aquatic vegetation显示文摘By the year 2050, the population of the United States is expected to reach over 418 million,while the global population will reach 9.6 billion. To provide safe food and fiber, agriculture must balance pesticide usage against impacts on natural resources. Challenges arise when storms cause runoff to be transported to aquatic receiving systems. Vegetated systems such as drainage ditches and constructed wetlands have been proposed as management practices to alleviate pesticide runoff. Twelve experimental mesocosms(1.3 × 0.71 × 0.61 m) were filled with sediment and planted with a monoculture of one of three wetland plant species(Typha latifolia, Leersia oryzoides, and Sparganium americanum).Three mesocosms remained unvegetated to serve as controls. All mesocosms were amended with 9.2 ± 0.8 μg/L, 12 ± 0.4 μg/L, and 3.1 ± 0.2 μg/L of atrazine, metolachlor, and diazinon, respectively, over a 4 hr hydraulic retention time to simulate storm runoff.Following the 4 hr amendment, non-amended water was flushed through mesocosms for an additional 4 hr. Outflow water samples were taken hourly from pre-amendment through8 hr, and again at 12, 24, 48, 72, and 168 hr post-amendment. L. oryzoides and T. latifolia had mean atrazine, metolachlor, and diazinon retentions from 51%–55% for the first 4 hr of the experiment. Aside from S. americanum and atrazine(25% retention), unvegetated controls had the lowest pesticide retention(17%–28%) of all compared mesocosms. | 14 | Generation and preservation of continental crust in the Grenville Orogeny显示文摘Detrital zircons from modern sediments display an episodic temporal distribution of U-Pb crystallization ages forming a series of 'peaks' and 'troughs'.The peaks are interpreted to represent either periods of enhanced generation of granitic magma perhaps associated with mantle overturn and superplume events,or preferential preservation of continental crust during global collisional orogenesis.The close association of those peaks with the assembly of supercontinents implies a causal relationship between collisional orogenesis and the presence of zircon age peaks.Here these two end-member models(episodic periodicity of increased magmatism versus selective preservation during collisional orogenesis) are assessed using U-Pb,Hf,and O analysis of detrital zircons from sedimentary successions deposited during the ~ 1.3-1.1Ca accretionary.~1.1-0.9 Ga collisional,and < 0.9 Ga extensional collapse phases of the Grenville orogenic cycle in Labrador and Scotland.The pre-collisional,accretionary stage provides a baseline of continental crust present prior to orogenesis and is dominated by Archean and Paleoproterozoic age peaks associated with pre-1300 Ma Laurentian geology.Strata deposited during the Grenville Orogeny display similar Archean and Paleoproterozoic detrital populations along with a series of broad muted peaks from~ 1500 to 1100 Ma.However,post-collisional sedimentary successions display a dominant age peak between 1085 and 985 Ma,similar to that observed in modern North American river sediments.Zircons within the post-orogenic sedimentary successions have progressively lower εHf and higherδ^(18)O values from ~ 1800 to ~ 1200 Ma whereupon they have higher εHf and δ^(18) within the dominant1085-985 Ma age peak.Furthermore,the Lu-Hf isotopic profile of the Grenville-related age peak is consistent with significant assimilation and contamination by older crustal material.The timing of this dominant age peak coincides with the peak of metamorphism and magmatism associated with the Crenville Orogeny,which is a typical collisional orogenic belt.The change from broad muted age peaks in the syn-orogenic strata to a single peak in the post-orogenic sedimentary successions and in the modern river sediments implies a significant shift in provenance following continental collision.This temporal change in provenance highlights that the source(s),from which detrital zircons within syn-orogenic strata were derived,was no longer available during the later stages of the accretionary and collisional stages of the orogenic cycle.This may reflect some combination of tectonic burial,erosion,or possibly recycling into the mantle by tectonic erosion of the source(s).During continental collision,the incorporated continental crust is isolated from crustal recycling processes operative at subduction margins.This tectonic isolation combined with sedimentaiy recycling likely controls the presence of the isotopic signature associated with the Grenville Orogeny in the modern Mississippi and Appalachian river sediments.These results imply that zircon age peaks,which developed in conjunction with supercontinents,are the product of selective crustal preservation resulting from collisional orogenesis. | Christopher J.Spencer Peter A.Cawood Chris J.Hawkesworth Anthony R.Prave Nick M.W.Roberts Matthew S.A.Horstwood Martin J.Whitehouse EIMF | 2015 | Geoscience Frontiers2015,6,3: | 1 |
