| 3 | Discovering the roles of subsurface microorganisms: Progress and future of deep biosphere investigation显示文摘The discovery of the marine 'deep biosphere'-microorganisms living deep below the seafloor-is one of the most significant and exciting discoveries since the ocean drilling program began more than 40 years ago. Study of the deep biosphere has become a research frontier and a hot spot both for geological and biological sciences. Here, we introduce the history of the discovery of the deep biosphere, and then we describe the types of environments for life below the seafloor, the energy sources for the living creatures, the diversity of organisms within the deep biosphere, and the new tools and technologies used in this research field. We will highlight several recently completed Integrated Ocean Drilling Program Expeditions, which targeted the subseafloor deep biosphere within the crust and sediments. | 5 | Engineering an enthesis-like graft for rotator cuff repair:An approach to fabricate highly biomimetic scaffold capable of zone-specifically releasing stem cell differentiation inducers显示文摘Rotator cuff(RC)attaches to humerus across a triphasic yet continuous tissue zones(bone-fibrocartilage-tendon),termed“enthesis”.Regrettably,rapid and functional enthesis regeneration is challenging after RC tear.The existing grafts bioengineered for RC repair are insufficient,as they were engineered by a scaffold that did not mimic normal enthesis in morphology,composition,and tensile property,meanwhile cannot simultaneously stimulate the formation of bone-fibrocartilage-tendon tissues.Herein,an optimized decellularization approach based on a vacuum aspiration device(VAD)was developed to fabricate a book-shaped decellularized enthesis matrix(O-BDEM).Then,three recombinant growth factors(CBP-GFs)capable of binding collagen were synthesized by fusing a collagen-binding peptide(CBP)into the N-terminal of BMP-2,TGF-β3,or GDF-7,and zone-specifically tethered to the collagen of O-BDEM to fabricate a novel scaffold(CBP-GFs/O-BDEM)satisfying the above-mentioned requirements.After ensuring the low immunogenicity of CBP-GFs/O-BDEM by a novel single-cell mass cytometry in a mouse model,we interleaved urine-derived stem cell-sheets into this CBP-GFs/O-BDEM to bioengineer an enthesis-like graft.Its high-performance on regenerating enthesis was determined in a canine model.These findings indicate this CBP-GFs/O-BDEM may be an excellent scaffold for constructing enthesis-like graft to patch large/massive RC tears,and provide breakthroughs in fabricating graded interfacial tissue. | Can Chen Qiang Shi Muzhi Li Yang Chen Tao Zhang Yan Xu Yunjie Liao Shulin Ding Zhanwen Wang Xing Li Chunfeng Zhao Lunquan Sun Jianzhong Hu Hongbin Lu | 2022 | Bioactive Materials2022,7,10: | 4 |

| 7 | An improved U-Net for cell confluence estimation显示文摘Cell confluence is an important metric to determine the growth and the best harvest time of adherent cells. At present, the evaluation of cell confluence mainly relies on experienced labor, and thus it is not conducive to the automated cell culture. In this paper, we proposed an improved U-Net algorithm(called DU-Net) for the segmentation of adherent cells. First, the general convolution was replaced by the dilated convolution to expand the receptive fields for feature extraction. Then, the convolutional layers were combined with the batch normalization layers to reduce the dependence of the network on initialization. As a result, the segmentation accuracy and F1-score of the proposed DU-Net for adherent cells with low confluence(<50%) reached 96.94% and 93.87%, respectively, and for those with high confluence(≥50%), they reached 98.63% and 98.98%, respectively. sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of Analysis of computational complexity and stability are presented. Simulation results of system parameter identification show that the DFP algorithm has fast convergence and excellent robustness than LMS and RLS algorithm. Prediction results of applying DFPSOVF model to single step predictions for Lorenz chaotic time series illustrate stability and convergence and there have not divergence problems.
