| 1 | Inuence of oxytetracycline on the structure and activity of microbial community in wheat rhizosphere soil显示文摘The microbial community composition in wheat rhizosphere was analyzed by detecting colony forming units (CFUs) in agar plates. The total CFUs in rhizosphere were 1.04 × 109/g soil with 9.0 × 108/g bacteria, 1.37 × 108/g actinomyces and 3.6 × 106/g fungi. The 10 dominant bacteria were isolated from wheat rhizosphere and were grouped into genus Bacillus according to their full length 16S rRNA gene sequences. Although belonging to the same genus, the isolated strains exhibited dierent sensitivities to oxytetracycline. When a series of the rhizosphere soil was exposed under various concentrations of oxytetracycline, the microbial community structure was highly aected with significant decline of CFUs of bacteria and actinomyces (22.2% and 31.7% at 10 mg/kg antibiotic, respectively). This inhibition was clearly enhanced with the increase exposure dosage of antibiotic and could not be eliminated during 30 d incubation. There was no obvious inuence of this treatment on fungi population. | 3 | Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. It compartmental-izes the research district land into three spatial parts: unchanged part,converted part and increased part. By this method we can evaluate the numerical model and dynamic degree model for existing calculating changing speed of land-use. Fur-thermore the paper raises reviving the calculating analysis model of spatial infor-mation in order to predict the dynamic changing level of all sorts of land. More concretely speaking,the model is mainly to know the changing area and changing speed (increased or decreased) of different land classifications from the micro-cosmic angle and to clearly show the spatial distribution and spatio-temporal law for changing urban lands. We discover why the situation has taken place by com-bining social and economic conditions. The result indicates that the calculating analysis model of spatial information can derive more accurate procedure of spatial transference and increase of all kinds of land from the microcosmic angle. By this model and technology,we can make the research of spatio-temporal structure evolution in land-use be more systematical and deeper. | 13 | Deposition features of Ni on self-assembled microtubule template from biolipid by electroless method显示文摘Diacetylenic glycero-phosphatidylcholine is a chiral molecule with amphiphilic property, and it can self-assembly into a lipid microtubular structure. The lipid microtubule is a stable structure formed by tightly wound helical ribbons, and the ribbon-wrapping patterns have a significant effect on their chemical deposition on the microtubules. The deposition of colloidal Pd catalyst occurs mainly on the helical edge of the wound helical ribbons to form helical deposition lines of colloidal Pd particles in the interior and exterior of the lipid microtubules, resulting in an uneven chemical deposition of Ni on the microtubules. Catalyzed by as-deposited colloidal Pd, metallized Ni microtubules are characterized by a helical form, which may be in relation to inner stress due to the thickness difference or the different deposition processes. The observation of microtom shows that metallized tubules have a hollow structure. Some metallized tubules have a kind of coaxial double layer structure observed in the direct experiment evidence, indicating that metallization can occur in the inner and outer surface of the lipid tubules. | 17 | Potential of capillary electrophoresis mass spectrometry for the characterization and monitoring of amine-derivatized naphthenic acids from oil sands process-affected water显示文摘Capillary electrophoresis coupled to mass spectrometry(CE–MS) was used for the analysis of naphthenic acid fraction compounds(NAFCs) of oil sands process-affected water(OSPW). A standard mixture of amine-derivatized naphthenic acids is injected directly onto the CE column and analyzed by CE–MS in less than 15 min. Time of flight MS analysis(TOFMS), optimized for high molecular weight ions, showed NAFCs between 250 and 800 m/z. With a quadrupole mass analyzer, only low-molecular weight NAFCs(between 100 and 450 m/z) are visible under our experimental conditions. Derivatization of NAFCs consisted of two-step amidation reactions mediated by 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(EDC), or mediated by a mixture of EDC and N-hydroxysuccinimide, in dimethyl sulfoxide, dichloromethane or ethyl acetate. The optimum background electrolyte composition was determined to be 30%(V/V) methanol in water and 2%(V/V) formic acid. | 20 | Identification of Rice Al-responsive Genes by Semi-quantitative Polymerase Chain Reaction using Sulfite Reductase as a Novel Endogenous Control显示文摘Based on the evidence that Al resistance is an inducible process and rice is an Al-resistant crop,identification of Al-responsive genes from rice may help to further clone Al-resistant genes in plants.Semi-quantitative and real-time polymerase chain reaction (PCR) is widely applied in gene transcriptional analyses,particularly for those genes with low transcript abundance.Normalization with proper endogenous control (EC) genes is critical for these two approaches in terms of reliability and precision.We first noticed that the expression of several commonly-used EC genes was depressed under Al stress,while sulfite reductase gene (SR) was stable throughout the Al treatment.The reliability of SR as an EC gene was further tested by analyzing the expression of a number of genes in response to Al challenge.Except for the consistent results obtained for the four previously-identified genes,nine additional genes were newly defined as Al-responsive in this study.Collectively,our results suggest that SR can be used as a novel EC gene for semi-quantitative and real-time PCR analysis of Al responsive genes,and that activated transport of silicon and stimulated metabolism of carotenoid and terpenoid could be involved in Al resistance in rice plants. | Jianjun Zhang Ying Yin Yuqi Wang Xinxiang Peng | 2010 | Journal of Integrative Plant Biology2010,52,5: | 1 |
