| 1 | Osteoarthritis:toward a comprehensive understanding of pathological mechanism显示文摘Osteoarthritis(OA) is the most common degenerative joint disease and a major cause of pain and disability in adult individuals. The etiology of OA includes joint injury, obesity, aging, and heredity. However, the detailed molecular mechanisms of OA initiation and progression remain poorly understood and, currently,there are no interventions available to restore degraded cartilage or decelerate disease progression. The diathrodial joint is a complicated organ and its function is to bear weight, perform physical activity and exhibit a joint-specific range of motion during movement. During OA development, the entire joint organ is affected, including articular cartilage, subchondral bone, synovial tissue and meniscus. | 4 | Mantle dynamics of the reactivating North China Craton:Constraints from the topographies of the 410-km and 660-km discontinuities显示文摘The topographies of the 410-km and 660-km discontinuities have important implications for mantle dynamics.Here we present high-resolution seismic imaging of the 410-km and 660-km discontinuities beneath the North China Craton (NCC) employing the receiver function method.Depth anomalies (deeper or shallower than the global average depths) at both discontinuities were detected by introducing a three-dimensional regional velocity model.The depressions of the 410-km discontinuity are mostly located in the eastern NCC.A local elevation of the 660-km discontinuity appeared in the northwest of the NCC and a significant depression of the 660-km discontinuity is located in the southeast of the NCC.Two dynamic mantle regimes are speculated to explain the formation of the anomalous depth zones in the NCC.One possibility is a complex mantle upwelling linked to edge-derived convection between the stagnant slab and the thick cratonic root.The other potential dynamic regime is slab stagnating,sinking,and induced upwelling at the neighboring slab front.These regimes hint that the mantle flow was possibly dominated by dynamic interactions among the subducting slab,cratonic root,and ambient mantle beneath the NCC. | XU WeiWei ZHENG TianYu ZHAO Liang | 2011 | Science China Earth Sciences2011,54,6: | 17 |
| 5 | Preparation of activated carbon from cattail and its application for dyes removal显示文摘Activated carbon was prepared from cattail by H3PO4 activation. The effects influencing the surface area of the resulting activated carbon followed the sequence of activated temperature > activated time > impregnation ratio > impregnation time. The optimum condition was found at an impregnation ratio of 2.5, an impregnation time of 9 hr, an activated temperature of 500°C, and an activated time of 80 min. The Brunauer-Emmett-Teller surface area and average pore size of the activated carbon were 1279 m2/g and 5.585 nm, respectively. A heterogeneous structure in terms of both size and shape was highly developed and widely distributed on the carbon surface. Some groups containing oxygen and phosphorus were formed, and the carboxyl group was the major oxygen-containing functional group. An isotherm equilibrium study was carried out to investigate the adsorption capacity of the activated carbon. The data fit the Langmuir isotherm equation, with maximum monolayer adsorption capacities of 192.30 mg/g for Neutral Red and 196.08 mg/g for Malachite Green. Dye-exhausted carbon could be regenerated effectively by thermal treatment. | 6 | Cardiac autonomic nerve fiber regeneration in chronic heart failure Do Akt gene-transduced mesenchymal stem cells promote repair?显示文摘BACKGROUND: Transplantation of Akt-over-expressing mesenchymal stem cells (Akt-MSCs) has been shown to repair infarcted myocardium and improve cardiac function. However, little is known about the therapeutic effects of Akt-MSCs on cardiac autonomic neuropathy in chronic heart failure (CHF). OBJECTIVE: The present study used adriamycin-induced CHF rat models to observe the effect of Akt-MSCs on cardiac autonomic nervous regeneration and the factors mediating this effect. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Central Laboratory of Basic Medical College, China Medical University, between September 2008 and April 2009. MATERIALS: Rabbit anti-choline acetyltransferase (ChAT), growth associated protein-43 (GAP-43), synaptophysin (SYN) polyclonal antibodies and the secondary antibody (goat anti-rabbit IgG) were purchased from Boster, China. Cat-A-Kit assay system was provided by Amersham, USA. METHODS: (1) Adult rat MSCs were isolated and cultured for the preparation of Akt-MSCs. (2) Forty male Wistar rats were intramyocardially administered adriamycin at 2 mg/kg over 3 days for a total of five times and once a week for additional five times thereafter to establish CHF models. At 2 weeks after final adriamycin treatment, 34 successful CHF rat models were randomized to three groups: Akt-MSCs (n = 11), simple MSCs (s-MSCs, n =11), and control (n = 12). Each group was intravenously administered Akt-MSCs (2×106 cells in 100 μL PBS), s-MSCs (2×106 cells in 100 μL PBS) or equal volume of phosphate buffered saline, once a day for a total of three times. MAIN OUTCOME MEASURES: At 4 weeks after final adriamycin treatment, myocardial norepi- nephrine (NE) content was detected using a Cat-A-Kit assay system. Myocardial ChAT, SYN and GAP-43 were performed by immunohistochemistry and Western blot analysis. Prior to, 2 and 4 weeks after adriamycin treatment, echocardiographic examination was performed and left ventricular ejection fraction (LVEF) was determined. RESULTS: Myocardial NE content, as well as SYN-positive and GAP-43-positive nerve fiber density and expression, and LVEF, was the greatest in the Akt-MSCs group, followed by the s-MSCs group, and lastly the control group (P < 0.05 or P < 0.01). ChAT expression was similar between Akt-MSCs and s-MSCs groups, but it was higher compared with the control group (P < 0.05). NE contents were negatively correlated to LVEF (r = -0.64, P = 0.015). | 7 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 9 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 10 | IL-25 stimulates M2 macrophage polarization and thereby promotes mitochondrial respiratory capacity and lipolysis in adipose tissues against obesity显示文摘Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially macrophages.Immune molecules,including some cytokines,have a close relationship with metabolism.Interleukin(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively clarified.Human serum and liver biopsy specimens,high-fat diet-induced obesity mice and DB/DB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLD).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in vitro.IL-25 was decreased in NAFLD patients and obese mice.In addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating macrophages.IL-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated macrophages.Furthermore,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more NAD+/NADH and ATP.In conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes. | Juan Feng Lingyi Li Zhiying Ou Qiao Li Baoyong Gong Zhenxian Zhao Weiwei Qi Ti Zhou Jun Zhong Weibin Cai Xia Yang Aiping Zhao Guoquan Gao Zhonghan Yang | 2018 | Cellular & Molecular Immunology2018,15,5: | 10 |

| 12 | A coupled convolutional neural network for small and densely clustered ship detection in SAR images显示文摘Ship detection from synthetic aperture radar(SAR) imagery plays a significant role in global marine surveillance. However, a desirable performance is rarely achieved when detecting small and densely clustered ship targets, and this problem is difficult to solve. Recently, convolutional neural networks(CNNs)have shown strong detection power in computer vision and are flexible in complex background conditions,whereas traditional methods have limited ability. However, CNNs struggle to detect small targets and densely clustered ones that exist widely in many SAR images. To address this problem while preserving the good properties for complex background conditions, we develop a coupled CNN for small and densely clustered SAR ship detection. The proposed method mainly consists of two subnetworks: an exhaustive ship proposal network(ESPN) for ship-like region generation from multiple layers with multiple receptive fields, and an accurate ship discrimination network(ASDN) for false alarm elimination by referring to the context information of each proposal generated by ESPN. The motivation in ESPN is to generate as many ship proposals as possible, and in ASDN, the goal is to obtain the final results accurately. Experiments are evaluated on two data sets. One is collected from 60 wide-swath Sentinel-1 images and the other is from20 GaoF en-3(GF-3) images. Both data sets contain many ships that are small and densely clustered. The quantitative comparison results illustrate the clear improvements of the new method in terms of average precision(AP) and F 1 score by 0.4028 and 0.3045 for the Sentinel-1 data set compared with the multi-step constant false alarm rate(CFAR-MS) method. The values are verified as 0.2033 and 0.1522 for the GF-3 data set. | 14 | Roles of dietary supplementation with arginine or N-carbamylglutamate in modulating the inflammation, antioxidant property, and m RNA expression of antioxidant-relative signaling molecules in the spleen of rats under oxidative stress显示文摘This study evaluated the effects of arginine(Arg) or N-carbamylglutamate(NCG) on inflammation, antioxidant property, and antioxidant-related gene expression in rat spleen under oxidative stress. A total of 52 rats were randomly distributed into 4 treatment groups with 13 replicates per group. Rats were fed a basal diet(BD) or BD supplemented with Arg or NCG for 30 days. On day 28, half of the BD-fed rats were intraperitoneally injected with sterile saline(control group), and the other half with 12 mg/kg body weight of diquat(DT; DT group). The other 2 diet groups were intraperitoneally injected with 12 mg/kg body weight of DT with either Arg(1%)(DT t Arg) or NCG(0.1%)(DT t NCG). Rat spleen samples were collected for analysis at 48 h after DT injection. Results showed that DT damaged the antioxidant defense in rats compared with the control group(P < 0.05). Compared with the DT group, the DT t Arg and DT t NCG groups manifested improved anti-hydroxyl radical, catalase, and total superoxide dismutase(T-SOD) activities, increased glutathione content(P < 0.05), and decreased malondialdehyde content(P < 0.05).Moreover, compared with the DT group, the DT t Arg and DT t NCG groups enhanced mRNA expression of superoxide dismutase(SOD), glutathione peroxidase 1(GPx1), glutathione reductase(GR), nuclear factor erythroid 2-related factor 2(Nrf2), Kelch-like ECH-associated protein 1(Keap-1), and mammalian target of rapamycin(m TOR)(P < 0.05). | 15 | Inflammasome activation in mouse inner ear in response to MCMV induced hearing loss显示文摘Objective: To identify presence of inflammasome activated in mouse cochlea with sensorineural hearing loss(SNHL) caused by cytomegalovirus(CMV) infection.Method: MCMV was injected into the right cerebral hemisphere in neonatal BALB/c mice at 2000 pfu virus titers. Auditory brainstem responses(ABRs) were tested to evaluate hearing at 21 days. Histopathological studies were conducted to confirm localizations of MCMV infected cells in the inner ear. Expression of inflammasome related factors was assessed by immunofluorescence, Quantitative real-time PCR and Western blotting.Results: In the mouse model of CMV induced SNHL, inflammasome related kinase Caspase-1 and downstream inflammatory factor IL-1b and IL-18 were found increased and activated after CMV infection in the cochlea. These factors could further up-regulate expression of IL-6 and TNFa. These inflammatory factors are neurotoxicity and may contribute to hearing impairment. Furthermore, we also detected significantly increased AIM2 protein that accumulated in the SGN of cochleae with CMV infection.Significance: We have shown that inflammasome as a novel inherent immunity mechanism may contribute to hearing impairment.Conclusion: Our data indicate that imflammasome assemble in mouse inner ear in response to CMV infection. | 16 | Cenozoic tectono-sedimentary characteristics and extension model of the Northwest Sub-basin,South China Sea显示文摘Based on the interpretations of three seismic profiles and one wide-angle seismic profile across the Northwest Sub-basin,South China Sea.stratigraphic sequences,deformation characteristics and an extension model for this sub-basin have been worked out.Three tectonic-stratigraphic units are determined.Detailed analyses of extension show that the event occurred mainly during the Paleogene and resulted in the formation of half-grabens or grabens distributed symmetrically around the spreading center.Sediments are characterized by chaotic and discontinuous reflectors,indicating clastic sediments. Farther to the southwest,the sub-basin features mainly continental rifting instead of sea-floor spreading. | 19 | Normal Structure and Function of Endothecium Chloroplasts Maintained by ZmMs33-Mediated Lipid Biosynthesis in Tapetal Cells Are Critical for Anther Development in Maize显示文摘Genic male sterility(GMS)is critical for heterosis utilization and hybrid seed production.Although GMS mutants and genes have been studied extensively in plants,it has remained unclear whether chloroplast-associated photosynthetic and metabolic activities are involved in the regulation of anther development.In this study,we characterized the function of ZmMs33/ZmGPAT6,which encodes a member of the glycerol-3-phosphate acyltransferase(GPAT)family that catalyzes the first step of the glycerolipid synthetic pathway.We found that normal structure and function of endothecium(En)chloroplasts maintained by ZmMs33-mediated lipid biosynthesis in tapetal cells are crucial for maize anther development.ZmMs33 is expressed mainly in the tapetum at early anther developmental stages and critical for cell proliferation and expansion at late stages.Chloroplasts in En cells of wild-type anthers function as starch storage sites before stage 10 but as photosynthetic factories since stage 10 to enable starch metabolism and carbohydrate supply.Loss of ZmMs33 function inhibits the biosynthesis of glycolipids and phospholipids,which are major components of En chloroplast membranes,and disrupts the development and function of En chloroplasts,resulting in the formation of abnormal En chloroplasts containing numerous starch granules.Further analyses reveal that starch synthesis during the day and starch degradation at night are greatly suppressed in the mutant anthers,leading to carbon starvation and low energy status,as evidenced by low trehalose-6-phosphate content and a reduced ATP/AMP ratio.The energy sensor and inducer of autophagy,SnRK1,was activated to induce early and excessive autophagy,premature PCD,and metabolic reprogramming in tapetal cells,finally arresting the elongation and development of mutant anthers.Taken together,our results not only show that ZmMs33 is required for normal structure and function of En chloroplasts but also reveal that starch metabolism and photosynthetic activities of En chloroplasts at different developmental stages are essential for normal anther development.These findings provide novel insights for understanding how lipid biosynthesis in the tapetum,the structure and function of En chloroplasts,and energy and substance metabolism are coordinated to maintain maize anther development. | Taotao Zhu Ziwen Li Xueli An Yan Long Xiaofeng Xue Ke Xie Biao Ma Danfeng Zhang Yijian Guan Canfang Niu Zhenying Dong Quancan Hou Lina Zhao Suowei Wu Jinping Li Weiwei Jin Xiangyuan Wan | 2020 | Molecular Plant2020,13,11: | 9 |
| 20 | Mortality outcomes of low-dose computed tomography screening for lung cancer in urban China:a decision analysis and implications for practice显示文摘Background: Mortality outcomes in trials of low-dose computed tomography(CT) screening for lung cancer are inconsistent. This study aimed to evaluate whether CT screening in urban areas of China could reduce lung cancer mortality and to investigate the factors that associate with the screening effect.Methods: A decision tree model with three scenarios(low-dose CT screening, chest X-ray screening, and no screening) was developed to compare screening results in a simulated Chinese urban cohort(100,000 smokers aged45-80 years). Data of participant characteristics were obtained from national registries and epidemiological surveys for estimating lung cancer prevalence. The selection of other tree variables such as sensitivities and specificities of low-dose CT and chest X-ray screening were based on literature research. Differences in lung cancer mortality(primary outcome), false diagnoses, and deaths due to false diagnosis were calculated. Sensitivity analyses were performed to identify the factors that associate with the screening results and to ascertain worst and optimal screening effects considering possible ranges of the variables.Results: Among the 100,000 subjects, there were 448,541, and 591 lung cancer deaths in the low-dose CT, chest X-ray, and no screening scenarios, respectively(17.2% reduction in low-dose CT screening over chest X-ray screening and 24.2% over no screening). The costs of the two screening scenarios were 9387 and 2497 false diagnoses and 7and 2 deaths due to false diagnosis among the 100,000 persons, respectively. The factors that most influenced death reduction with low-dose CT screening over no screening were lung cancer prevalence in the screened cohort, lowdose CT sensitivity, and proportion of early-stage cancers among low-dose CT detected lung cancers.
