2Air quality monitoring using mobile microscopy and machine learning显示文摘Rapid,accurate and high-throughput sizing and quantification of particulate matter(PM)in air is crucial for monitoring and improving air quality.In fact,particles in air with a diameter of≤2.5μm have been classified as carcinogenic by the World Health Organization.Here we present a field-portable cost-effective platform for high-throughput quantification of particulate matter using computational lens-free microscopy and machine-learning.This platform,termed c-Air,is also integrated with a smartphone application for device control and display of results.This mobile device rapidly screens 6.5 L of air in 30 s and generates microscopic images of the aerosols in air.It provides statistics of the particle size and density distribution with a sizing accuracy of~93%.We tested this mobile platform by measuring the air quality at different indoor and outdoor environments and measurement times,and compared our results to those of an Environmental Protection Agency–approved device based on beta-attenuation monitoring,which showed strong correlation to c-Air measurements.Furthermore,we used c-Air to map the air quality around Los Angeles International Airport(LAX)over 24 h to confirm that the impact of LAX on increased PM concentration was present even at 47 km away from the airport,especially along the direction of landing flights.With its machinelearning-based computational microscopy interface,c-Air can be adaptively tailored to detect specific particles in air,for example,various types of pollen and mold and provide a cost-effective mobile solution for highly accurate and distributed sensing of air quality.Yi-Chen Wu Ashutosh Shiledar Yi-Cheng Li Jeffrey Wong Steve Feng Xuan Chen Christine Chen Kevin Jin Saba Janamian Zhe Yang Zachary Scott Ballard Zoltán Göröcs Alborz Feizi Aydogan Ozcan 2017Light(Science & Applications)2017,6,1:5
3Prevalence and virological profiles of hepatitis B infection in human immunodeficiency virus patients显示文摘AIM: To determine the prevalence of hepatitis B virus (HBV) in adult human immunodeficiency virus (HIV) patients with CD4+ T-cell count less than 500/mm 3 and without antiretroviral therapy; to describe different HBV-HIV coinfection virological profiles; and to search for factors associated with HBs antigen (HBsAg) presence in these HIV positive patients.METHODS: During four months (June through September 2006), 491 patients were received in four HIV positive monitoring clinical centers in Abidjan. Inclusion criteria: HIV-1 or HIV-1 and 2 positive patients, age ≥ 18 years, CD4+ T-cell count < 500/mL and formal and signed consent of the patient. Realized blood tests included HIV serology, CD4+ T-cell count, quantitative HIV RNA load and HBV serological markers, such as HBsAg and HBc antibody (anti-HBcAb). We performed HBeAg, anti-HBe antibody (anti-HBeAb), anti-HBc IgM and quantitative HBV DNA load in HBsAg positive patients. Anti-HBsAb had been tested in HIV patients with HBsAg negative and anti-HBcAb-positive. HBV DNA was also tested in 188 anti-HBcAb positive patients with HBsAg negative status and without anti-HBsAb. Univariate analysis (Pearsonχ 2 test or Fischer exact test) and multivariate analysis (backward step-wise selection logistic regression) were performed as statistical analysis. RESULTS: Mean age of 491 patients was 36 ± 8.68 years and 73.3% were female. Type-1 HIV was found in 97% and dual-type HIV (type 1 plus type 2) in 3%. World Health Organization (WHO) clinical stage was 1, 2, 3 and 4 respectively in 61 (12.4%), 233 (47.5%), 172 (35%) and 25 patients (5.1%). Median CD4+ T-cell count was 341/mm 3 (interquartile range: 221-470). One hundred and twelve patients had less than 200 CD4+ T-cell/mm 3 . Plasma HIV-1 RNA load was elevated (≥ 5 log 10 copies/mL) in 221 patients (45%). HBsAg and anti-HBcAb prevalence was respectively 13.4% and 72.9%. Of the 66 HBsAg positive patients, 22 were inactive HBV carriers (33.3%), 21 had HBeAg positive hepatitis (31.8%) and 20 had HBeAg negative hepatitis (30.3%). HBeAg and anti-HBeAb were indeterminate in 3 of them. Occult B infection prevalence (HBsAg negative, anti-HBcAb positive, anti-HBsAb negative and detectable HBV DNA) was 21.3%. Three parameters were significantly associated with the presence of HBsAg: male [odds ratio (OR): 2.2;P = 0.005; 95% confidence interval (CI): 1.3-3.8]; WHO stage 4 (OR: 3.2;P = 0.01;95% CI: 1.3-7.9); and aspartate aminotransferase (AST) level higher than the standard (OR: 1.9;P = 0.04; 95% CI: 1.02-3.8). CONCLUSION: HBV infection prevalence is high in HIV-positive patients. HBeAg positive chronic hepatitis and occult HBV infection are more frequent in HIVpositive patients than in HIV negative ones. 20Triose phosphate export from chloroplasts and cellular sugar content regulate anthocyanin biosynthesis during high light acclimation显示文摘Plants have evolvedmultiple strategies to cope with rapid changes in the environment.During high light(HL)acclimation,the biosynthesis of photoprotective flavonoids,such as anthocyanins,is induced.However,the exact nature of the signal and downstreamfactors forHL induction of flavonoid biosynthesis(FB)is still under debate.Here,we show that carbon fixation in chloroplasts,subsequent export of photosynthates by triose phosphate/phosphate translocator(TPT),and rapid increase in cellular sugar content permit the transcriptional and metabolic activation of anthocyanin biosynthesis during HL acclimation.In combination with genetic and physiological analysis,targeted and whole-transcriptome gene expression studies suggest that reactive oxygen species and phytohormones play only a minor role in rapid HL induction of the anthocyanin branch of FB.In addition to transcripts of FB,sugar-responsive genes showed delayed repression or induction in tpt-2 during HL treatment,and a significant overlapwith transcripts regulated by SNF1-related protein kinase 1(SnRK1)was observed,including a central transcription factor of FB.Analysis of mutants with increased and repressed SnRK1 activity suggests that sugar-induced inactivation of SnRK1 is required for HL-mediated activation of anthocyanin biosynthesis.Our study emphasizes the central role of chloroplasts as sensors for environmental changes as well as the vital function of sugar signaling in plant acclimation.Max-Emanuel Zirngibl Galileo Estopare Araguirang Anastasia Kitashova Kathrin Jahnke Tobias Rolka Christine Kühn Thomas Nägele Andreas S.Richter 2023Plant Communications2023,4,1:1
