3Quantitative trait loci for Fusarium head blight resistance in wheat cultivars Yangmai 158 and Zhengmai 9023显示文摘Fusarium head blight(FHB)is one of the prevalent fungal diseases of wheat worldwide.Exploring new FHB resistance quantitative trait loci(QTL)in adapted wheat cultivars is a critical step for breeding new FHB-resistant cultivars.In this study,we developed a population of 236 F5:7 recombinant inbred lines(RILs)using two popular Chinese wheat cultivars,Yangmai 158 and Zhengmai 9023,with moderate FHB resistance to identify the QTL for FHB type II resistance.This population was evaluated for percentage of symptomatic spikelets per spike(PSS)using single floret injection in repeated greenhouse experiments.Mean PSSs were 33.2%for Yangmai 158 and 30.3%for Zhengmai 9023.A genetic linkage map of 1002 single nucleotide polymorphisms(SNPs)generated by genotyping-by-sequencing(GBS)was constructed for the RIL population.Six QTL were identified for FHB resistance,and three of them were repeatable in the both experiments.Zhengmai 9023 contributed the resistance allele at one repeatable QTL,designated as Qfhb.7D,whereas Yangmai 158 contributed the resistance alleles at the other two repeatable QTL,Qfhb.3AL and Qfhb.2DS.The additional QTL,Qfhb.4AS was significant in the mean PSS,and Qfhb.2DL and Qfhb.7AS were significant in only one experiment.Replacement of each allele individually at the three repeatable QTL significantly changed PSSs.Qfhb.3AL,Qfhb.2DS,and Qfhb.7D explained 8.35%to 9.89%,5.13%to 7.43%,and 6.15%to 9.32%of the phenotypic variations,respectively.The three repeatable QTL contributed by the two parents were additive and stacking the resistance alleles from all the three repeatable QTL showed the highest level of resistance in the current RIL population.Ten SNPs in the QTL regions of Qfhb.3AL,Qfhb.2DS,and Qfhb.7D were converted into KBioscience competitive allele-specific PCR(KASP)assays.One KASP marker for Qfhb.3AL was validated in a panel of wheat cultivars from China.Some of these KASP markers could be useful for marker-assisted selection to stack these QTL.Pingping Zhang Chenjin Guo Zhao Liu Amy Bernardo Hongxiang Maa Hongxiang Ma Peng Jiang Guicheng Song Guihua Bai 2021The Crop Journal2021,9,1:8
4Association of serum lipids and severity of epithelial ovarian cancer:an observational cohort study of 349 Chinese patients显示文摘While obesity and fat intake have been associated with the risk and prognosis of epithelial ovarian cancer, the association between the lipid levels and epithelial ovarian cancer phenotype remains controversial. We conducted a retrospective study of 349 epithelial ovarian cancer patients who received treatment at Jiangsu Cancer Hospital, China between 2011 and 2017. We analyzed age at diagnosis, blood pressure, plasma glucose content, body mass index(BMI), lipid levels and clinical parameters. Severity of epithelial ovarian cancer was classified according to the International Federation of Gynecology and Obstetrics(FIGO) grading system. Univariate analysis of the clinical factors according to the severity of epithelial ovarian cancer was followed by logistic regression analysis to identify clinical factors significantly associated with epithelial ovarian cancer severity. Univariate analysis indicated that age,BMI, triglyceride(TG), and high density lipoproteins(HDL) differed significantly among different stages of epithelial ovarian cancer(P<0.05). In the logistic regression model, elevated TG(OR: 1.883; 95% CI= 1.207-2.937), and low HDL(OR: 0.497; 95% CI = 0.298-0.829) levels were significantly associated with the high severity epithelial ovarian cancer. Our data indicate that high TG and low HDL levels correlate with a high severity of epithelial ovarian cancer. 5The Arabidopsis catalase triple mutant reveals important roles of catalases and peroxisome-derived signaling in plant development显示文摘Hydrogen peroxide(H_2O_2)is generated in many metabolic processes.As a signaling molecule,H_2O_2plays important roles in plant growth and development,as well as environmental stress response.In Arabidopsis,there are three catalase genes,CAT1,CAT2,and CAT3.The encoded catalases are predominately peroxisomal proteins and are critical for scavenging H_2O_2.Since CAT1 and CAT3 are linked on chromosome 1,it has been almost impossible to generate cat1/3 and cat1/2/3 mutants by traditional genetic tools.In this study,we constructed cat1/3 double mutants and cat1/2/3 triple mutants by CRISPR/Cas9 to investigate the role of catalases.The cat1/2/3 triple mutants displayed severe redox disturbance and growth defects under physiological conditions compared with wild-type and the cat2/3 double mutants.Transcriptome analysis showed a more profound transcriptional response in the cat1/2/3 triple mutants compared to the cat2/3 mutants.These differentially expressed genes are involved in plant growth regulation as well as abiotic and biotic stress responses.In addition,expression of OXI1(OXIDATIVE SIGNAL INDUCIBLE 1)and several MAPK cascade genes were changed dramatically in the catalase triple mutant,suggesting that H_2O_2produced in peroxisomes could serve as a peroxisomal retrograde signal.Tong Su Pingping Wang Huijuan Li Yiwu Zhao Yao Lu Peng Dai Tianqi Ren Xiaofeng Wang Xuezhi Li Qun Shao Dazhong Zhao Yanxiu Zhao Changle Ma 2018Journal of Integrative Plant Biology2018,60,7:3
6Clinical significance of extended high-risk human papillomavirus genotyping and viral load in cervical cancer and precancerous lesions显示文摘Persistent infections with specific high-risk human papillomavirus(HR-HPV)strains are the leading cause of cervical cancer and precancerous lesions.HPV-16 and HPV-18 are associated with more than 70%of cervical cancer.However,with recent widespread vaccination efforts against cervical cancer,the infection rates of HPV-16 and HPV-18 have decreased across all age groups,while the infection rates of other HR-HPV strains have increased.The non-16/18 HR-HPV strains play an important role in cervical lesions.These strains can be identified with extended genotyping,and the 2019 American Society for Colposcopy and Cervical Pathology(ASCCP)guidelines recommended an HPV-based testing to assess the risk of cervical disease in patients.We reviewed and analyzed the clinical benefits of applying extended HR-HPV genotyping,which was published by the International Agency for Research on Cancer(HPV-16,18,31,33,35,39,45,51,52,56,58,59,66,and 68),to cervical cancer screening.This review concluded that cervical cancer screening needs to include extended HR-HPV genotyping.The examination of extended HR-HPV genotyping in cervical intraepithelial lesions and cervical cancers can help guide clinical practices.Pingping Su Jincheng Ma Lirui Yu Shuting Tang Pengming Sun 2023Gynecology and Obstetrics Clinical Medicine2023,3,1:2
7Thermal evolution effects on the properties of converting Cs-polluted soil into pollucite-base glass-ceramics for radioactive cesium immobilization显示文摘The Fukushima nuclear accident in Japan on March 11,2011,produced large amounts of Cs-polluted soil which must be controlled to prevent the spread of hazardous Cs into the environment.In this paper,the effects of heat treatment on the structure and properties of Cs-containing glass-ceramics,as a simulated nuclear waste form,were systematically investigated.Cesium atoms are chemically bonded in the pollucite structure,and the amorphous phase further encapsulates the pollucite crystals in the glassceramics,thus providing an extra protective layer for the immobilized Cs.XRD analysis,Raman and FT-IR studies on the glass-ceramics synergistically indicated that the optimum crystallization temperature for pollucite is around 1000℃.The pollucite is predominantly the main crystalline phase with a narrow crystal size distribution between 0.5 and 2 mm.Standard leaching test results show that the leaching rate of Cs was very low(3.0×10^(-3) g/(m^(2)·d)).The study offers a practical method for immobilizing Cs in pollucite-base glass-ceramics.Moreover,the experimentally obtained data may provide some important references for converting Cs-polluted soil into pollucite-base glass-ceramics waste form.Yong Yang Xin Cao Lifen Shi Zhengyi Zhang Pingping Wang Jinwei Li Yangshan Sun Shuyong Chen Tianhe Wang Liyun Ma Shou Peng 2021Journal of Materiomics2021,7,6:2
8A murine model for human immune thrombocytopenic purpura and comparative analysis of multiple gene expression in bone marrow and spleen显示文摘Homeostasis of platelet number in human and other mammals is well maintained for prevention of minor bleeding and for other immunological functions,but the exact molecular mechanism responsible for immune thrombocytopenic purpura(ITP) has not been fully understood.In an effort to identify genetic factors involved in initiation of platelet production in response to bleeding injury or platelet destruction,we have successfully generated an animal model of human ITP via intraperitoneal injection of anti-platelet antibody into the Balb/c mouse.Platelet counts were dropped dramatically in animals that received antibody injection within 4 h,maintained at the mini-mum level for a period of 44 h,started to rebound after 48 h,and reached to the maximum at 144 h(6 days).Final homeostasis reached at approximately 408 h(17 days),following a minor cycle of platelet number fluctuation.Using semi-quantitative RT-PCR,we assessed and compared mRNA level of CD41,c-myb,c-mpl,caspase-3,caspase-9,GATA-1,and Bcl-xl in bone marrow and spleen.Alteration of mRNA expression was correlated with the change of platelet level,and an inverse relationship was found for expression of the genes between bone marrow and spleen.No transcription was detectable for any of the seven genes in bone marrow at the time when platelet number reached the maximum(144 h).In contrast,mRNA transcripts of the seven genes were found to be at the highest level in spleen tissue.This is the first study of simultaneous detection of multiple platelet related genes in a highly reproducible ITP animal model.Our results provided the supportive evidence that expression of the above seven genes are more related to negative regulation of platelet number in spleen tissue,at least in the model animals.Hong Wei Xinchun Ding Jiangong Ren Ka Liu Pingping Tan Daquan Li Runlin Z. 9High proportion strontium-doped micro-arc oxidation coatings enhance early osseointegration of titanium in osteoporosis by anti-oxidative stress pathway显示文摘The excessive accumulation of reactive oxygen species(ROS)under osteoporosis precipitates a microenvironment with high levels of oxidative stress(OS).This could significantly interfere with the bioactivity of conventional titanium implants,impeding their early osseointegration with bone.We have prepared a series of strontium(Sr)-doped titanium implants via micro-arc oxidation(MAO)to verify their efficacy and differences in osteoinduction capabilities under normal and osteoporotic(high OS levels)conditions.Apart from the chemical composition,all groups exhibited similar physicochemical properties(morphology,roughness,crystal structure,and wettability).Among the groups,the low Sr group(Sr25%)was more conducive to osteogenesis under normal conditions.In contrast,by increasing the catalase(CAT)/superoxide dismutase(SOD)activity and decreasing ROS levels,the high Sr-doped samples(Sr75% and Sr100%)were superior to Sr25% in inducing osteogenic differentiation of MC3T3-E1 cells and the M2 phenotype polarization of RAW264.7 cells,thus enhancing early osseointegration.Furthermore,the results of both in vitro cell co-culture and in vivo studies also showed that the high Sr-doped samples(especially Sr100%)had positive effects on osteoimmunomodulation under the OS microenvironment.Ultimately,the collated findings indicated that the high proportion Sr-doped MAO coatings were more favorable for osteoporosis patients in implant restorations.Xinkun Shen Kai Fang Kendrick Hii Ru Yie Zixin Zhou Yiding Shen Shuyi Wu Yue Zhu Zhennan Deng Pingping Ma Jianfeng Ma Jinsong Liu 2022Bioactive Materials2022,7,4:2

12The CREB Regulated Transcription Coactivator 2 Suppresses HIV-1 Transcription by Preventing RNA PolⅡfrom Binding to HIV-1 LTR显示文摘The CREB-regulated transcriptional co-activators(CRTCs),including CRTC1,CRTC2 and CRTC3,enhance transcription of CREB-targeted genes.In addition to regulating host gene expression in response to cAMP,CRTCs also increase the infection of several viruses.While human immunodeficiency virus type 1(HIV-1)long terminal repeat(LTR)promoter harbors a cAMP response element and activation of the cAMP pathway promotes HIV-1 transcription,it remains unknown whether CRTCs have any effect on HIV-1 transcription and HIV-1 infection.Here,we reported that CRTC2 expression was induced by HIV-1 infection,but CRTC2 suppressed HIV-1 infection and diminished viral RNA expression.Mechanistic studies revealed that CRTC2 inhibited transcription from HIV-1 LTR and diminished RNA PolⅡoccupancy at the LTR independent of its association with CREB.Importantly,CRTC2 inhibits the activation of latent HIV-1.Together,these data suggest that in response to HIV-1 infection,cells increase the expression of CRTC2 which inhibits HIV-1 gene expression and may play a role in driving HIV-1 into latency.Ling Ma Shumin Chen Zhen Wang Saisai Guo Jianyuan Zhao Dongrong Yi Quanjie Li Zhenlong Liu Fei Guo Xiaoyu Li Pingping Jia Jiwei Ding Chen Liang Shan Cen 2021Virologica Sinica2021,36,4:1

16Genetic Analysis of Grain Yield in a 7×7 Diallel Cross F_2 Population of Wheat(T.Aestivum L.)显示文摘In order to obtain genetic information for grain yield, seven genetically diverse wheat cultivars were crossed in an incomplete diallel to study the inheritance of grain yield using F_2 progenies for two years. Significant differences were observed among genotype, year and genotype × year interaction for grain yield, and both general combining ability(GCA) and specific combining ability(SCA) were also highly significant for grain yield, suggesting that the trait was controlled by both additive and non-additive effect. The GCA estimates revealed that the best combiners for grain yield were Yangmai 5 and Ningmai 9. Adequacy tests revealed that data of grain yield was fully adequate for genetic interpretation. Over-dominance genetic effects were important for the expression of grain yield. 17Research on Forecasting Flowering Phase of Pear Tree Based on Neural Network显示文摘Predicting the blooming season of ornamental plants is significant for guiding adjustments in production decisions and providing viewing periods and routes.The current strategies for observation of ornamental plant booming periods are mainly based on manpower and experience,which have problems such as inaccurate recognition time,time-consuming and energy sapping.Therefore,this paper proposes a neural network-based method for predicting the flowering phase of pear tree.Firstly,based on the meteorological observation data of Shijiazhuang Meteorological Station from 2000 to 2019,three principal components(the temperature factor,weather factor,and humidity factor)with high correlation coefficient with the flowering phase of pear tree are obtained by using the principal component analysis method.Then,the three components are used as input factors for the BP neural network.A BP neural network prediction model is constructed based on genetic algorithm optimization.The crossover operator and mutation operator in the adaptive genetic algorithm are improved.Finally,the meteorological sample data from 2013 to 2019 are used to test and verify the algorithm in this paper.The results demonstrate that,the model can solve the local optimization problem of the BP neural network model.The prediction results of the flowering phase of pear tree are evaluated in terms of relevance and prediction accuracy.Both are superior to the traditional effective accumulated temperature and the prediction results of the stepwise regression method.This method can provide more reliable forecast information for the blooming period,which can provide decision-making reference for improving the development of tourism industry.Zhenzhou Wang Yinuo Ma Pingping Yu Ning Cao Heiner Dintera 2021Computers, Materials & Continua2021,,9:0
18Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 19Identification of prognostic indicators,diagnostic markers, and possible therapeutic targets among LIM homeobox transcription factors in breast cancer显示文摘Background:Breast cancer(BRCA)is the most common malignant tumor among women worldwide.Despite advances in treatment,many patients still die from a lack of effective diagnostic and prognostic markers and powerful therapeutic targets.LIM homeobox genes(LHXs)play vital roles in regulating the development of various organisms.However,there are limited reports regarding their roles in the diagnosis,prognosis,and treatment of BRCA.Methods:UALCAN,Kaplan–Meier plotter,cBioPortal,GeneMANIA,STRING,DAVID 6.8,TRRUST v2,LinkedOmics,and TIMER were utilized to analyze differential expression,prognostic value,genetic alteration,neighbor gene network,transcription factor targets,kinase targets,and immune cell infiltration of LHXs in BRCA patients.Results:LHX gene expression patterns are clear in BRCA and its different subtypes.Further analyses indicated that this altered expression is possibly affected by genetic and/or epigenetic changes.The prognostic and diagnostic values of certain LHXs are unique to different BRCA subtypes.LHXs are mainly involved in the regulation of differentiation and development,and their neighbor genes are primarily involved in cancer‐related pathways.Moreover,most LHXs are closely correlated with immune cell infiltration.Furthermore,LHXs may exert their functions by regulating a series of transcription factor and kinase targets.Conclusions:LHXs are unique diagnostic and prognostic markers and participate in cancer through different signaling pathways and/or regulatory mechanisms in BRCA.This study provides potential applications of LHXs for the diagnosis,prognosis,and treatment of BRCA and its different subtypes.Pingping Gao Na Sun Tingting Zhao Yuanyuan Sun Jing Gu Dandan Ma Hao Tian Zaihui Peng Yi Zhang Fei Han Xiaowei Qi 2022Cancer Innovation2022,1,3:0
20The origin of arc basalts:New advances and remaining questions显示文摘Whether arc magmatism occurs above oceanic subduction zones is the forefront of studies on convergent plate margins.The most important petrologic issue related to the evolution of arc systems is the origin of arc magmatism,among which arc basalts are the most important one because they provide insights into mantle enrichment mechanism and crust-mantle interaction at oceanic subduction zones.Fluids or melts released either by dehydration or by melting of subducting oceanic slab infiltrate and metasomatize the overlying mantle wedge at varying depth,leading to the formation of source regions of arc basalts.Such processes make most of arc basalts commonly enriched in large ion lithosphile elements and light rare earth elements,but depleted in high-field strength elements and heavy rare earth elements.Small amounts of arc basalts are characterized by relatively high Nb contents or by Nb enrichment.Rare basalts with compositions similar to ocean island basalts or mid-ocean ridge basalt also occur in arc systems.For these peculiar rocks,it remains debated whether their source is affected by subduction-related components.During their ascent and before their eruption,arc basaltic magmas are subjected to crystal fractionation,mixing and crustal contamination.In addition to the contribution of subducting slab components to the mantle source of arc basalts,the materials above the subducting slab at forearc depths would have been transported either by drag or by subduction erosion into the subarc mantle and into the source of arc magmas.Heats and materials brought by corner flows also play important roles in the generation of arc basalts.Despite the important progresses made in recent studies of arc basalts,further efforts are needed to investigate subarc mantle metasomatism,material recycling,the formation of arc magma sources,geodynamic mechanism in generating arc basalts,and their implicationd s for the initiation of plate tectonics on Earth.Yigang Xu Qiang Wang Gongjian Tang Jun Wang Hongyan Li Jinsheng Zhou Qiwei Li Yue Qi Pingping Liu Lin Ma Jingjing Fan 2020Science China Earth Sciences2020,63,12:0
