Perspectives on the Application of Genome-Editing Technologies in Crop Breeding显示文摘Most conventional and modern crop-improvement methods exploit natural or artificially induced genetic variations and require laborious characterization of the progenies of multiple generations derived from time-consuming genetic crosses.Genome-editing systems,in contrast, provide the means to rapidly modify genomes in a precise and predictable way, making it possible to introduce improvements directly into elite varieties. Here,we describe the range of applications available to agricultural researchers using existing genome-editing tools. In addition to providing examples of genome-editing applications in crop breeding,we discuss the technical and social challenges faced by breeders using genome-editing tools for crop improvement. | Kai Hua Jinshan Zhang Jose Ramon Botella Changle Ma Fanjiang Kong Baohui Liu Jian-Kang Zhu | 2019 | Molecular Plant2019,12,8: | 19
Synthetical Efficiency-based Optimization for the Power Distribution of Power-split Hybrid Electric Vehicles显示文摘Now the optimization strategies for power distribution are researched widely, and most of them are aiming to the optimal fuel economy and the driving cycle must be preknown. Thus if the actual driving condition deviates from the scheduled driving cycle, the effect of optimal results will be declined greatly. Therefore, the instantaneous optimization strategy carried out on-line is studied in this paper. The power split path and the transmission efficiency are analyzed based on a special power-split scheme and the efficiency models of the power transmitting components are established. The synthetical efficiency optimization model is established for enhancing the transmission efficiency and the fuel economy. The identification of the synthetical efficiency as the optimization objective and the constrain group are discussed emphatically. The optimization is calculated by the adaptive simulated annealing(ASA)algorithm and realized on-line by the radial basis function(RBF)-based similar models. The optimization for power distribution of the hybrid vehicle in an actual driving condition is carried out and the road test results are presented. The test results indicate that the synthetical efficiency optimization method can enhance the transmission efficiency and the fuel economy of the power-split hybrid electric vehicle(HEV)observably.Compared to the rules-based strategy the optimization strategy is optimal and achieves the approximate global optimization solution for the power distribution. The synthetical efficiency optimization solved by ASA algorithm can give attentions to both optimization quality and calculation efficiency, thus it has good application foreground for the power distribution of power-split HEV. | WANG Weida HAN Lijin XIANG Changle MA Yue LIU Hui | 2014 | Chinese Journal of Mechanical Engineering2014,27,1: | 12

Novel Wx alleles generated by base editing for improvement of rice grain quality显示文摘Amylose content(AC),which is regulated by the Waxy(Wx)gene,is a major indicator of eating and cooking quality(ECQ)in rice(Oryza sativa). Thus far, only a limited number of mutations in the N-terminal domain of Wx were found to have a major impact on the AC of rice grains and no mutations with such effects were reported for other regions of the Wx protein. Here, nucleotide substitutions in the middle region of Wx were generated by adenine and cytosine base editors. The nucleotide substitutions led to changes in 15 amino acid residues of Wx,and a series of novel Wx alleles with ACs of 0.3%-29.43%(wild type with AC of 19.87%)were obtained.Importantly, the waxy~(abe2)allele showed a'soft rice'AC,improved ECQ,favorable appearance, and no undesirable agronomic traits. The transgenes were removed from the waxy~(abe2)progeny,generating a promising breeding material for improving rice grain quality. | Xiaorui Huang Fei Su Sheng Huang Fating Mei Xiaomu Niu Changle Ma Hui Zhang Xiaoguo Zhu Jian-Kang Zhu Jinshan Zhang | 2021 | Journal of Integrative Plant Biology2021,63,9: | 4
Creation of aromatic maize by CRISPR/Cas显示文摘Aroma is an important quality parameter for breeding in rice(Oryza sativa). For example, the aromatic rice varieties basmati and jasmine rice, with a popcorn-like scent, are popular worldwide and routinely command a price premium.2-acetyl-1-pyrroline(2AP)is a key flavor compound among over 200 volatiles identified in fragrant rice.A naturally fragrant germplasm exists in multiple plant species besides rice, which all exhibit lower activity of BETAINE ALDEHYDE DEHYDROGENASE 2(BADH2).However,no equivalent aromatic germplasm has been described in maize(Zea mays). Here,we characterized the two maize BADH2 homologs,ZmBADH2a and ZmBADH2b.We generated zmbadh2a and zmbadh2b single mutants and the zmbadh2a-zmbadh2b double mutant by CRISPR/Cas in four inbred lines.A popcorn-like scent was only noticeable in seeds from the double mutant, but not from either single mutant or in wild type.In agreement,we only detected 2AP in fresh kernels and dried mature seeds from the double mutant, which accumulated between 0.028 and 0.723 mg/kg 2AP.These results suggest that ZmBADH2a and ZmBADH2b redundantly participate in 2AP biosynthesis in maize, and represent the creation of the world's first aromatic maize by simultaneous genome editing of the two BADH2 genes. | Yanxiao Wang Xiaoqin Liu Xiuxiu Zheng Wenxia Wang Xunqing Yin Haifeng Liu Changle Ma Xiaomu Niu Jian-Kang Zhu Fei Wang | 2021 | Journal of Integrative Plant Biology2021,63,9: | 3
A CRISPR/Cas12a-empowered surface plasmon resonance platform for rapid and specific diagnosis of the Omicron variant of SARS-CoV-2显示文摘The outbreak of the COVID-19 pandemic was partially due to the challenge of identifying asymptomatic and presymptomatic carriers of the virus, and thus highlights a strong motivation for diagnostics with high sensitivity that can be rapidly deployed. On the other hand, several concerning SARS-CoV-2 variants, including Omicron, are required to be identified as soon as the samples are identified as‘positive’.Unfortunately,a traditional PCR test does not allow their specific identification.Herein, for the first time,we have developed MOPCS(Methodologies of Photonic CRISPR Sensing),which combines an optical sensing technology-surface plasmon resonance(SPR)with the‘gene scissors’clustered regularly interspaced short palindromic repeat(CRISPR)technique to achieve both high sensitivity and specificity when it comes to measurement of viral variants.MOPCS is a low-cost,CRISPR/Cas12a-systemempowered SPR gene-detecting platform that can analyze viral RNA,without the need for amplification, within 38 min from sample input to results output, and achieve a limit of detection of 15 fM.MOPCS achieves a highly sensitive analysis of SARS-CoV-2,and mutations appear in variants B.1.617.2(Delta),B.1.1.529(Omicron)and BA.1(a subtype of Omicron). This platform was also used to analyze some recently collected patient samples from a local outbreak in China, identified by the Centers for Disease Control and Prevention. This innovative CRISPR-empowered SPR platform will further contribute to the fast, sensitive and accurate detection of target nucleic acid sequences with single-base mutations. | Zhi Chen Jingfeng Li Tianzhong Li Taojian Fan Changle Meng Chaozhou Li Jianlong Kang Luxiao Chai Yabin Hao Yuxuan Tang Omar AAl-Hartomy Swelm Wageh Abdullah GAl-Sehemi Zhiguang Luo Jiangtian Yu Yonghong Shao Defa Li Shuai Feng William J.Liu Yaqing He Xiaopeng Ma Zhongjian Xie Han Zhang | 2022 | National Science Review2022,9,8: | 3
The Arabidopsis catalase triple mutant reveals important roles of catalases and peroxisome-derived signaling in plant development显示文摘Hydrogen peroxide(H_2O_2)is generated in many metabolic processes. As a signaling molecule,H_2O_2plays important roles in plant growth and development,as well as environmental stress response. In Arabidopsis, there are three catalase genes,CAT1,CAT2,and CAT3.The encoded catalases are predominately peroxisomal proteins and are critical for scavenging H_2O_2.Since CAT1 and CAT3 are linked on chromosome 1,it has been almost impossible to generate cat1/3 and cat1/2/3 mutants by traditional genetic tools. In this study,we constructed cat1/3 double mutants and cat1/2/3 triple mutants by CRISPR/Cas9 to investigate the role of catalases. The cat1/2/3 triple mutants displayed severe redox disturbance and growth defects under physiological conditions compared with wild-type and the cat2/3 double mutants.Transcriptome analysis showed a more profound transcriptional response in the cat1/2/3 triple mutants compared to the cat2/3 mutants. These differentially expressed genes are involved in plant growth regulation as well as abiotic and biotic stress responses. In addition, expression of OXI1(OXIDATIVE SIGNAL INDUCIBLE 1)and several MAPK cascade genes were changed dramatically in the catalase triple mutant, suggesting that H_2O_2produced in peroxisomes could serve as a peroxisomal retrograde signal. | Tong Su Pingping Wang Huijuan Li Yiwu Zhao Yao Lu Peng Dai Tianqi Ren Xiaofeng Wang Xuezhi Li Qun Shao Dazhong Zhao Yanxiu Zhao Changle Ma | 2018 | Journal of Integrative Plant Biology2018,60,7: | 3
P-T stability conditions of methane hydrate in sediment from South China Sea显示文摘For reasonable assessment and safe exploitation of marine gas hydrate resource, it is important to determine the stability conditions of gas hydrates in marine sediment. In this paper, the seafloor water sample and sediment sample (saturated with pore water) from Shenhu Area of South China Sea were used to synthesize methane hydrates, and the stability conditions of methane hydrates were investigated by multi-step heating dissociation method. Preliminary experimental results show that the dissociation temperature of methane hydrate both in seafloor water and marine sediment, under any given pressure, is depressed by approximately -1.4 K relative to the pure water system. This phenomenon indicates that hydrate stability in marine sediment is mainly affected by pore water ions. | Shicai Sun Yuguang Ye Changling Liu Fengkui Xiang Yan Ma | 2011 | Journal of Natural Gas Chemistry2011,20,5: | 2
A classification-based method to estimate event-related potentials from single trial EEG显示文摘A novel method based on machine learning is developed to estimate event-related potentials from single trial electroencephalography. This paper builds a basic framework using classification and an optimization model based on this framework for estimating event-related potentials. Then the SingleTrialEM algorithm is derived by introducing a logistic regression model, which could be obtained by training before SingleTrialEM is used, to instantiate the optimization model. The simulation tests demonstrate that the proposed method is correct and solid. The advantage of this method is verified by the comparison between this method and the Woody filter in simulation tests. Also, the cognitive test results are consistent with the conclusions of cognitive science. | HUANG ZhiHua LI MingHong ZHOU ChangLe MA YuanYe | 2012 | Science China(Life Sciences)2012,55,1: | 2
Ribonuclease H-like gene SMALL GRAIN2 regulates grain size in rice through brassinosteroid signaling pathway显示文摘Grain size is a key agronomic trait that determines the yield in plants.Regulation of grain size by brassinosteroids(BRs)in rice has been widely reported. However, the relationship between the BR signaling pathway and grain size still requires further study. Here,we isolated a rice mutant, named small grain2(sg2),which displayed smaller grain and a semi-dwarf phenotype. The decreased grain size was caused by repressed cell expansion in spikelet hulls of the sg2 mutant.Using map-based cloning combined with a Mut Map approach,we cloned SG2,which encodes a plant-specific protein with a ribonuclease H-like domain.SG2 is a positive regulator downstream of GLYCOGEN SYNTHASE KINASE2(GSK2)in response to BR signaling, and its mutation causes insensitivity to exogenous BR treatment.Genetical and biochemical analysis showed that GSK2 interacts with and phosphorylates SG2.We further found that BRs enhance the accumulation of SG2 in the nucleus, and subcellular distribution of SG2 is regulated by GSK2 kinase activity. In addition,Oryza sativa OVATE family protein 19(OsOFP19),a negative regulator of grain shape, interacts with SG2 and plays an antagonistic role with SG2 in controlling gene expression and grain size. Our results indicated that SG2 is a new component of GSK2-related BR signaling response and regulates grain size by interacting with Os OFP19. | Yunshuai Huang Hui Dong Changling Mou Ping Wang Qixian Hao Min Zhang Hongmin Wu Fulin Zhang Tengfei Ma Rong Miao Kai Fu Yaping Chen Ziyan Zhu Cheng Chen Qikai Tong Zhuoran Wang Shirong Zhou Xi Liu Shijia Liu Yunlu Tian Ling Jiang Jianmin Wan | 2022 | Journal of Integrative Plant Biology2022,64,10: | 1

Py-GC/MS study of lignin pyrolysis and effect of catalysts on product distribution显示文摘Fast pyrolysis is one of the most promising methods to convert lignin into fuels and chemicals. In the present study,pyrolysis-gas chromatography/mass spectrometry(Py-GC/MS)was used to evaluate vapor phase product distribution of lignin fast pyrolysis.During the non-catalytic pyrolysis process, lignin was pyrolyzed at 400℃,500℃ and 600℃ respectively, finding that the highest yield of aromatic hydrocarbons was obtained at 600℃.Catalytic pyrolysis experiments were also conducted to investigate the effects of catalyst pore structure and acidity on the product distributions.Five different catalysts(HZSM-5,MCM-41,TiO_(2),ZrO_(2) and Mg(Al)O)were applied to lignin catalytic pyrolysis, and the catalytic performance was estimated by analyzing the pyrolytic products. The catalysts were characterized by using X-ray diffraction(XRD),BET,and NH3(CO_(2))temperature programmed desorption.Based on these characterizations, discussion was carried out to explain the formation of the produc distributions.Among the five catalysts,HZSM-5 exhibited the best performance on the formation of aromatic hydrocarbons. | Si Zhan Wang Chenguang Bi Kang Zhang Xinghua Yu Chiling Dong Renjie Ma Longlong Pang Changle | 2017 | International Journal of Agricultural and Biological Engineering2017,10,5: | 1
Observation of electron weak localization and correlation effects in disordered graphene显示文摘We have studied the electron transport properties of a disordered graphene sample, where the disorder was intentionally strengthened by Ga+ ion irradiation. The magneto-conductance of the sample exhibits a typical two-dimensional electron weak localization behavior, with electron-electron interaction as the dominant dephasing mechanism. The absence of electron anti-weak localization in the sample implies strong intersublattice and/or intervalley scattering caused by the disorders. The temperature and bias-voltage dependencies of conductance clearly reveal the suppression of conductance at low energies, indicating opening of a Coulomb gap due to electron-electron interaction in the disordered graphene sample. | TAN ChangLing TAN ZhenBing MA Li QU FanMing YANG Fan CHEN Jun LIU GuangTong YANG HaiFang YANG ChangLi LU Li | 2009 | Science China(Physics,Mechanics & Astronomy)2009,52,9: | 1
A Transcriptome Analysis of Neural Tissue of Litopenaeus vannamei After Acute Exposure to Alexandrium pacificum显示文摘Alexandrium pacificum(A.pacificum)is a typical paralytic shellfish poisonous dinoflagellate.Harmful algal blooms(HABs)caused by this species can bring serious environmental problems and economic losses to the aquaculture industry. In this study, transcriptome sequencing and analyses were performed on the neural tissue of Litopenaeus vannamei(L.vannamei)after acute exposure to A.pacificum disrupted solution for 72 h,and differentially expressed genes(DEGs)were identified. The results showed that, compared with the control samples,300 DEGs were identified in the experimental group,of which 194 were up-regulated,and 106 down-regulated.The gene ontology(GO)functional enrichment analysis showed that DEGs were significantly enriched in the cortical cytoskeleton organization, troponin complex,amylo-alpha-1,6-glucosidase and thymidine phosphorylase.Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis found that DEGs were mainly enriched in the oxidative phosphorylation process, intercellular tight junctions and mitophagy. The results showed that the proteoglycans, signaling pathways, and various metabolic processes that regulate cell proliferation, differentiation, and apoptosis all played an essential role in the response of L.vannamei to A.pacificum toxins. | YANG Huijie HUANG Xianghu MA Yukun HU Zhangxi ZHANG Ning LI Changling ZHANG Yulei | 2024 | Journal of Ocean University of China2024,23,2: | 0

Dual-salt assisted synergistic synthesis of Prussian white cathode towards high-capacity and long cycle potassium ion battery显示文摘Prussian blue analogues(PBAs) are considered as superior cathode materials for potassium-ion batteries(PIBs) because of their three-dimensional open framework structure, high stability, and low cost. However, the intrinsic lattice defects and low potassium content typically results in poor rate and cycling performance, thus limited their practical applications. In this work,high-quality K1.64FeFe(CN)6(PW-HQ)material with less crystalline water(6.21%) and high potassium content(1.64 mol^(-1)) was successfully synthesized by a novel coprecipitation method with potassium citrate(K-CA) and potassium chloride(KCl) addition.Specifically, the electrode delivers a reversible capacity of 113.1 mA h g^(-1)at the current rate of 50 mA g^(-1)with~100% coulombic efficiency.Besides, the electrode retained 90% reversible capacity at 500 mA g^(-1)current density after 1000 cycles, indicating only 0.01% capacity decay per cycle. Moreover,we have revealed that the introduction of K-CA controlled the chelating rate of Fe(Ⅱ) and the addition of KCl increased the K+content, hence improving the capacity and stability of the asprepared electrodes.Structural evolution and potassium storage mechanism were further investigated by detailed ex-situ X-ray diffraction and in-situ Raman measurements, which demonstrated reversible potassiation/depotassiation behavior and negligible volume change during the electrochemical process. In general, this work provides an efficient strategy to eliminate water contents in Prussian blue cathode and improve its electrochemical performance, which plays a key role in promoting the industrialization of potassium ion batteries. | Rui Ma Zixing Wang Qingfeng Fu Wang Zhou Ying Mo Jian Tu Zhiyong Wang Peng Gao Changling Fan Jilei Liu | 2023 | Journal of Energy Chemistry2023,,8: | 0
A novel single-trial event-related potential estimation method based on compressed sensing显示文摘Cognitive functions are often studied using eventrelated potentials(ERPs)that are usually estimated by an averaging algorithm.Clearly, estimation of single-trial ERPs can provide researchers with many more details of cognitive activity than the averaging algorithm.A novel method to estimate single-trial ERPs is proposed in this paper. This method includes two key ideas.First, singular value decomposition was used to construct a matrix, which mapped singletrial electroencephalographic recordings(EEG)into a low-dimensional vector that contained little information from the spontaneous EEG.Second,we used the theory of compressed sensing to build a procedure to restore single-trial ERPs from this low-dimensional vector.ERPs are sparse or approximately sparse in the frequency domain. This fact allowed us to use the theory of compressed sensing. We verified this method in simulated and real data. Our method and dVCA(differentially variable component analysis),another method of single-trial ERPs estimation, were both used to estimate single-trial ERPs from the same simulated data. Results demonstrated that our method significantly outperforms dVCA under various conditions of signal-to-noise ratio. Moreover, the single-trial ERPs estimated from the real data by our method are statistically consistent with the theories of cognitive science. | Zhihua Huang Minghong Li Shangchuan Yang Yuanye Ma Changle Zhou | 2013 | Neuroscience Bulletin2013,29,6: | 0
