1Circulating tumor cells:biology and clinical significance显示文摘Circulating tumor cells(CTCs)are tumor cells that have sloughed off the primary tumor and extravasate into and circulate in the blood.Understanding of the metastatic cascade of CTCs has tremendous potential for the identification of targets against cancer metastasis.Detecting these very rare CTCs among the massive blood cells is challenging.However,emerging technologies for CTCs detection have profoundly contributed to deepening investigation into the biology of CTCs and have facilitated their clinical application.Current technologies for the detection of CTCs are summarized herein,together with their advantages and disadvantages.The detection of CTCs is usually dependent on molecular markers,with the epithelial cell adhesion molecule being the most widely used,although molecular markers vary between different types of cancer.Properties associated with epithelial-tomesenchymal transition and stemness have been identified in CTCs,indicating their increased metastatic capacity.Only a small proportion of CTCs can survive and eventually initiate metastases,suggesting that an interaction and modulation between CTCs and the hostile blood microenvironment is essential for CTC metastasis.Single-cell sequencing of CTCs has been extensively investigated,and has enabled researchers to reveal the genome and transcriptome of CTCs.Herein,we also review the clinical applications of CTCs,especially for monitoring response to cancer treatment and in evaluating prognosis.Hence,CTCs have and will continue to contribute to providing significant insights into metastatic processes and will open new avenues for useful clinical applications.Danfeng Lin Lesang Shen Meng Luo Kun Zhang Jinfan Li Qi Yang Fangfang Zhu Dan Zhou Shu Zheng Yiding Chen Jiaojiao Zhou 2021Signal Transduction and Targeted Therapy2021,6,12:8
2High proportion strontium-doped micro-arc oxidation coatings enhance early osseointegration of titanium in osteoporosis by anti-oxidative stress pathway显示文摘The excessive accumulation of reactive oxygen species(ROS)under osteoporosis precipitates a microenvironment with high levels of oxidative stress(OS).This could significantly interfere with the bioactivity of conventional titanium implants,impeding their early osseointegration with bone.We have prepared a series of strontium(Sr)-doped titanium implants via micro-arc oxidation(MAO)to verify their efficacy and differences in osteoinduction capabilities under normal and osteoporotic(high OS levels)conditions.Apart from the chemical composition,all groups exhibited similar physicochemical properties(morphology,roughness,crystal structure,and wettability).Among the groups,the low Sr group(Sr25%)was more conducive to osteogenesis under normal conditions.In contrast,by increasing the catalase(CAT)/superoxide dismutase(SOD)activity and decreasing ROS levels,the high Sr-doped samples(Sr75% and Sr100%)were superior to Sr25% in inducing osteogenic differentiation of MC3T3-E1 cells and the M2 phenotype polarization of RAW264.7 cells,thus enhancing early osseointegration.Furthermore,the results of both in vitro cell co-culture and in vivo studies also showed that the high Sr-doped samples(especially Sr100%)had positive effects on osteoimmunomodulation under the OS microenvironment.Ultimately,the collated findings indicated that the high proportion Sr-doped MAO coatings were more favorable for osteoporosis patients in implant restorations.Xinkun Shen Kai Fang Kendrick Hii Ru Yie Zixin Zhou Yiding Shen Shuyi Wu Yue Zhu Zhennan Deng Pingping Ma Jianfeng Ma Jinsong Liu 2022Bioactive Materials2022,7,4:2

6Smart ZnS@C filler for super-anticorrosive self-healing zinc-rich epoxy coating显示文摘The zinc-rich epoxy cathodic protection coating is the most widely used anticorrosion material for marine steel.However,traditional conductive fillers lack the intelligent self-healing effect,which limits the long-term anticorrosion performance.Herein,with uniform carbon-coated ZnS(ZnS@C)nanoballs as the smart active release filler,we propose an anticorrosive and self-healing zinc-rich maleic anhydride epoxy coating.Due to the high pore filling efficiency of the nanoballs,the water vapor transmission rate of the coating with an initial corrosion efficiency of 99.92%and a low-frequency impedance of|Z|f=10mHz=3.88×10^(10) Ω·cm^(2),was reduced by 52%.The carbon-shell of the nanoball increases electron transmission paths in the coating and improves conductivity by nearly two orders of magnitude,which effectively activates more Zn-sites and extends the cathodic protection time.Moreover,once the steel-substrate undergoes regional corrosion,the SO_(4)^(2-)hydrolyzes from the ZnS-core of the nanoball and reacts with iron ions on the corroded area accurately and intelligently to fill the gap and self-heals into a new dense barrier layer(Fe_(2)(SO_(4))_(3),etc.),which significantly improves the shielding protection ability during the long-term usage of the coating.The effective anticorrosion time of the proposed coating could be up to 3,400 h.Kai Yang Yixue Duan Guicheng Liu Guoyan Ma Hao Fu Xuyong Chen Manxiang Wang Gangqiang Zhu Woochul Yang Yiding Shen 2022Nano Research2022,15,5:1

11The histone methyltransferase SUVR2 promotes DSB repair via chromatin remodeling and liquidliquid phase separation显示文摘Maintaining genomic integrity and stability is particularly important for stem cells,which are at the top of the cell lineage origin.Here,we discovered that the plant-specific histone methyltransferase SUVR2 maintains the genome integrity of the root tip stem cells through chromatin remodeling and liquid-liquid phase separation(LLPS)when facing DNA double-strand breaks(DSBs).The histone methyltransferase SUVR2(MtSUVR2)has histone methyltransferase activity and catalyzes the conversion of histone H3 lysine 9 monomethylation(H3K9me1)to H3K9me2/3 in vitro and in Medicago truncatula.Under DNA damage,the proportion of heterochromatin decreased and the level of DSB damage marker y-H2AX increased in suvr2 mutants,indicating that MtSUVR2 promotes the compaction of the chromatin structure through H3K9 methylation modification to protect DNA from damage.Interestingly,MtSUVR2 was induced by DSBs to phase separate and form droplets to localize at the damage sites,and this was confirmed by immunofluorescence and fluorescence recovery after photobleaching experiments.The IDR1 and lowcomplexity domain regions of MtSUVR2 determined its phase separation in the nucleus,whereas the IDR2 region determined the interaction with the homologous recombinase MtRAD51.Furthermore,we found that MtSUVR2 drove the phase separation of MtRAD51 to form'DNA repair bodies,'which could enhance the stability of MtRAD51 proteins to facilitate error-free homologous recombination repair of stem cells.Taken together,our study reveals that chromatin remodeling-associated proteins participate in DNA repair through LLPS.Qianwen Liu Peng Liu Tuo Ji Lihua Zheng Chen Shen Shasha Ran Jinling Liu Yafei Zhao Yiding Niu Tao Wang Jiangli Dong 2022Molecular Plant2022,15,7:1

15Rheological Behaviour for Polymer Melts and Concentrated Solutions Part Ⅲ: A New Multiple Entanglement Model to Predict the Dependence of Linear Viscoelastic Function (η_0, Ψ_(10)~0,η_(ext)~0) on the Ranges of Primary Molecular Weights and th显示文摘It is shown theoretically that the viscoelasticity of polymer melts is determined by three combining factorst they are the primary molecular weight and its distribution, the number of entanglement sites on polymer chain and the sequence distribution of constituent chains in entanglement spacings. A unified quantity for the three combing factors is the average constrained dimensional number of constituent chains in the long entanglement spacings (v). A new relation of v to the primary molecular weight and the number of testing polymers were derived from the multiple entanglement and reptation model, and a new method for determining v was proposed. The dependences of linear viscoelastic functions on the primary molecular weight and its distribution were derived by the statistical method. When Mn=6Me to 18 Me, the values of (v) can range from 3.33 to 3.70.
