5Isolation and Characterization of Bacillus Amyloliquefaciens L-1 for Biocontrol of Pear Ring Rot显示文摘Considering the heavy losses caused by pear ring rot(Botryosphaeria berengeriana) disease, the potential biocontrol agent against B. berengeriana was isolated and characterized in this work. Bacteria, isolated from pear orchard rhizosphere soil, were screened for the biological control of pear ring rot caused by B. berengeriana. Among them, strain L-1 showed noticeable inhibitory activities against B. berengeriana and six other commonly occurring postharvest pathogens. Molecular methods indicated strain L-1 was Bacillus amyloliquefaciens. The potential of strain L-1 as an effective biocontrol agent was further estimated. Results showed strain L-1 could successfully colonize in pear wounds, its colonies reached 142.35 folds on 4 days post inoculation, and maintained at a high level during storage. In addition, strain L-1 caused abnormal hyphae growth of B. berengeriana,and its inhibitory percentage against pear ring rot reached 76.55% in vivo on 11 days post inoculation. Strain L-1 significantly induced the peroxidase(POD) and catalase(CAT) activities and delayed the accumulation of malondialdehyde(MDA) in pears. What's more, strain L-1 did not impair the fruit quality. 6Prevalence of HIV Indeterminate Western Blot Tests and Follow-up of HIV Antibody Sero-Conversion in Southeastern China显示文摘HIV-indeterminate Western blotting(WB)results are typically obtained in WB confirmatory assays,and the number of indeterminate samples may increase with the detection of HIV infections,which will present considerable challenges for the management of HIV/AIDS.Nucleic acid detection has been used as a laboratory test for screening suspected or indeterminate samples.However,the effectiveness of these assays for the differential diagnosis of HIV-indeterminate WB samples remained undetermined.In this study,210 subjects with HIV-indeterminate WB results were detected from 6360 positive HIV screening samples between 2015 and 2016 in southeastern China,in which HIV-indeterminate WB results accounted for 3.30%.The highest proportion of indeterminate results was observed in pregnant and lying-in women receiving physical examinations(16.67%),followed by that in voluntary blood donors(8.82%).The most common WB band patterns were p24,gpl60 and p24,and gpl60.The follow-up study revealed that the highest negative and positive conversion rates of HIV antibodies were in samples with a single p24 band(80.28%),and with gpl60 and p24 bands(86.21%),respectively.Among the Env,Gag,and Pol antibodies,samples with a Gag band showed the highest negative conversion rate(81.25%),whereas the highest positive conversion rate was observed in samples with an Env band(56.76%).In addition,quantitative and qualitative HIV nucleic acid testing exhibited the highest sensitivity(96.3%)and specificity(97.85%),respectively.Our results indicate a lower proportion of HIV indeterminate WB results in southeastern China compared to previous reports,and the follow-up re-examination of patients with HIV indeterminate results should be performed.Nucleic acid testing facilitates the identification of HIV infections.Shouli Wu Min Gao Jian Zheng Pingping Yan Wei Wang Xiaoli Lu Yuefeng Qiu Yansheng Yan 2019Virologica Sinica2019,34,4:15
7Correlation between tongue manifestations and glucose, total cholesterol, and high-density lipoprotein cholesterol in patients with acute cerebral infarction显示文摘OBJECTIVE: To analyze the association between tongue manifestations and the levels of glucose (GLU), total cholesterol(TCH), and high-density lipoprotein cholesterol(HDL-C) in subjects with acute cerebral infarction.METHODS: Hospitalized patients with first unilateral cerebral infarction in the Neurological Department of Xuanwu Hospital were included and the correlation between tongue fur color, fur nature,and the levels of GLU,TCH, HDL-C were analyzed.RESULTS: HDL level in the thin fur group was higher than that in the thick fur group(P=0.02). The difference in the levels of GLU,TCH, and HDL-C among the groups was significant(P<0.05), classified in terms of slippery, moist, and dry fur. Further comparison between the groups by Student-Newman-Keuls test showed that GLU level in the dry fur group was the highest. 8Prognostic evaluation of postoperative adjuvant therapy for operable cervical cancer:10 years'experience of National Cancer Center in China显示文摘Objective: The aim of this study was to investigate the prognostic factors and to evaluate the impact of adjuvant therapy on clinical outcome for early-stage cervical cancer.Methods: The clinical-pathological data of all 1,335 patients with the International Federation of Gynecology and Obstetrics(FIGO) Ib–IIa cervical cancer treated with primary radical surgery at the Chinese National Cancer Center between May 2007 and Dec 2013 were retrospectively reviewed.The median follow-up was 70 months.Results: Of all the patients,61.6% of the cases received adjuvant therapy,with 5-year disease-free survival(DFS)of 92.1% and 5-year overall survival(OS) of 95.0%.In multivariate analysis,differentiation of G3(P<0.05),lymph node metastasis(LNM,P<0.05) and lymphovascular space invasion(LVSI,P<0.05) were independent predictors for OS,while LNM(P<0.05),deep stroma invasion(DSI,P<0.05) and LVSI(P<0.05) were independent factors for DFS.The samples were stratified by histologic type,and cervical squamous cell carcinoma(SCC) was found to share the same independent factors except for differentiation of OS.As to patients with cervical adenocarcinoma/adenosquamous carcinoma(AC/ASC),differentiation was the independent predictor of OS(P<0.05); and LVSI of DFS(P<0.05).Of 236 patients with high-risk factors,there was no significant difference in survival between concurrent chemoradiotherapy(CCRT,n=195),radiotherapy(RT,n=24),and chemotherapy(CT,n=17).Among the 190 patients with LNM who underwent CCRT,124 cases showed improved DFS after sequential CT(P=0.118),with a recurrence rate decrease of 14%,though the difference was not statistically significant.Patients with single intermediate-risk factors like DSI or LVSI were found to partially benefit from adjuvant therapy,but the difference was not statistically significant.Conclusions: LNM,LVSI,DSI and differentiation were found to be independent prognostic factors for operable cervical cancer.Aggressive postoperative adjuvant therapy based on single risk factors in Chinese National Cancer Center could benefit survival.CCRT+CT outperformed CCRT in high-risk patients.For patients with single non-high-risk factor,the role of adjuvant therapy needs to be further discussed.Tong Shu Dan Zhao Bin Li Yating Wang Shuanghuan Liu Pingping Li Jing Zuo Ping Ba i RongZhang Lingying Wu 2017Chinese Journal of Cancer Research2017,29,6:12
9Intestinal lysozyme liberates Nod1 ligands from microbes to direct insulin trafficking in pancreatic beta cells显示文摘Long-range communication between intestinal symbiotic bacteria and extra-intestinal organs can occur through circulating bacterial signal molecules, through neural circuits, or through cytokines or hormones from host cells. Here we report that Nod1 ligands derived from intestinal bacteria act as signal molecules and directly modulate insulin trafficking in pancreatic beta cells. The cytosolic peptidoglycan receptor Nod1 and its downstream adapter Rip2 are required for insulin trafficking in beta cells in a cell-autonomous manner. Mechanistically, upon recognizing cognate ligands, Nod1 and Rip2 localize to insulin vesicles, recruiting Rab1a to direct insulin trafficking through the cytoplasm. Importantly, intestinal lysozyme liberates Nod1 ligands into the circulation, thus enabling long-range communication between intestinal microbes and islets. The intestine-islet crosstalk bridged by Nod1 ligands modulates host glucose tolerance. 10Adsorption characteristics of a novel ceramsite for heavy metal removal from stormwater runoff显示文摘Urban sediments have rapidly increased in recent years around the world,and their effective management has become an important problem.To remove heavy metals from stormwater runoff and use sediments as a resource,a novel ceramsite was developed using sewer pipe sediments(SPS),river bed sediments(RBS),urban water supply treatment sludge(WSTS),and wastewater treatment plant excess sludge(WWTS).The optimal composition was determined based on the Brunauer–Emmett–Teller specific surface area and an orthogonal test design.The adsorption characteristics of the novel ceramsite for dissolved heavy metals(Cu^(2+)and Cd^(2+)) were investigated through adsorption isotherms and kinetic experiments at(25±1)℃.Both Cu^(2+) and Cd^(2+) were effectively removed by the novel ceramsite,and their equilibrium adsorption was 4.96 mg·g^(-1) and 3.84 mg·g^(-1),respectively.Langmuir isotherms and a pseudo-first-order kinetic equation described the adsorption process better than other techniques.Characterization analysis of the ceramsite composition before and after heavy metal adsorption showed that the Cu^(2+) and Cd^(2+) contents in the ceramsite increased after adsorption.The results revealed that adsorption is both a physical and chemical process,and that ceramsite can be used as a bioretention medium to remove heavy metals from stormwater runoff while simultaneously converting problematic urban sediments into a resource.Jianlong wang Yuanling Zhao Pingping Zhang Liqiong Yang Huai'ao Xu Guangpeng Xi 2018Chinese Journal of Chemical Engineering2018,26,1:9
11Identification of Maximum Road Friction Coefficient and Optimal Slip Ratio Based on Road Type Recognition显示文摘The identification of maximum road friction coefficient and optimal slip ratio is crucial to vehicle dynamics and control.However,it is always not easy to identify the maximum road friction coefficient with high robustness and good adaptability to various vehicle operating conditions.The existing investigations on robust identification of maximum road friction coefficient are unsatisfactory.In this paper,an identification approach based on road type recognition is proposed for the robust identification of maximum road friction coefficient and optimal slip ratio.The instantaneous road friction coefficient is estimated through the recursive least square with a forgetting factor method based on the single wheel model,and the estimated road friction coefficient and slip ratio are grouped in a set of samples in a small time interval before the current time,which are updated with time progressing.The current road type is recognized by comparing the samples of the estimated road friction coefficient with the standard road friction coefficient of each typical road,and the minimum statistical error is used as the recognition principle to improve identification robustness.Once the road type is recognized,the maximum road friction coefficient and optimal slip ratio are determined.The numerical simulation tests are conducted on two typical road friction conditions(single-friction and joint-friction)by using CarSim software.The test results show that there is little identification error between the identified maximum road friction coefficient and the pre-set value in CarSim.The proposed identification method has good robustness performance to external disturbances and good adaptability to various vehicle operating conditions and road variations,and the identification results can be used for the adjustment of vehicle active safety control strategies.GUAN Hsin WANG Bo LU Pingping XU Liang 2014Chinese Journal of Mechanical Engineering2014,27,5:8

13Systematic optimization of the yeast cell factory for sustainable and high efficiency production of bioactive ginsenoside compound K显示文摘Ginsenoside Compound K(CK)has been recognized as a major functional component that is absorbed into the systemic circulation after oral administration of ginseng.CK demonstrates diverse bioactivities.A phase I clinical study indicated that CK was a potential candidate for arthritis therapy.However,a phase II clinical study was suspended because of the high cost associated with the present CK manufacturing approach,which is based on the traditional planting-extracting-biotransforming process.We previously elucidated the complete CK biosynthetic pathway and realized for the first time de novo biosynthesis of CK from glucose by engineered yeast.However,CK production was not sufficient for industrial application.Here,we systematically engineered Saccharomyces cerevisiae to achieve high titer production of CK from glucose using a previously constructed protopanaxadiol(PPD)-producing chassis,optimizing UGTPg1 expression,improving UDP-glucose biosynthesis,and tuning down UDP-glucose consumption.Our final engineered yeast strain produced CK with a titer of 5.74 g/L in fed-batch fermentation,which represents the highest CK production in microbes reported to date.Once scaled-up,this high titer de novo microbial biosynthesis platform will enable a robust and stable supply of CK,thus facilitating study and medical application of CK.Pingping Wang Jiali Wang Guoping Zhao Xing Yan Zhihua Zhou 2021Synthetic and Systems Biotechnology2021,6,2:7
14Anti-parkinsonian effects of octacosanol in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine-treated mice显示文摘Our previous research showed that octacosanol exerted its protective effects in 6-hydroxydopamine-induced Parkinsonian rats. The goal of this study was to investigate whether octacosanol would attenuate neurotoxicity in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine (MPTP)-treated C57BL/6N mice and its potential mechanism. Behavioral tests, tyrosine hydroxylase immunohistochemistry and western blot were used to investigate the effects of octacosanol in a mouse model of Parkinson's disease. Oral administration of octacosanol (100 mg/kg) significantly improved behavioral impairments in mice treated by MPTP and markedly ameliorated morphological appearances of tyrosine hydroxylase-positive neuronal cells in the substantia nigra. Furthermore, octacosanol blocked MPTP-induced phosphorylation of p38MAPK and JNK, but not ERK1/2. These findings implicated that the protective effects afforded by octacosanol might be mediated by blocking the phosphorylation of p38MAPK and JNK on the signal transduction in vivo. 17Effect of monovalent anions on cationic Gemini micro-emulsion显示文摘Micro-emulsion usually consists of water, oil, surfactants and co-surfactants, and each component has an effect on the phase behavior and solubilization of the micro-emulsion. When the surfactant in the micro-emulsion system is quaternary ammonium cationic Gemini surfactant, the surfactant mainly combines with the anions in the salt. With the increase of salt concentration, the phase transformation of Winsor I → Winsor III → Winsor II occurred, but the optimum salinity and salt width are different because of the type of salt. The effects of 5 different kinds of monovalent anions, including C_6H_5SO_3^-, I-, Br-, NO_3^- and Cl-, on the phase behavior and solubilization of quaternary ammonium cationic Gemini micro-emulsion are researched by Winsor phase diagram. It is found that the effects of organic anions C_6H_5SO_3-and I-on the phase behavior and solubilization of quaternary ammonium cationic Gemini micro-emulsion are most significant, and the effects of Br-, NO_3^- and Cl-are less significant. 19Variations in silicate concentration affecting photosynthetic carbon fixation by spring phytoplankton assemblages in surface water of the Strait of Malacca显示文摘The Strait of Malacca (SoM), the world's busiest sea-route, is increasingly polluted as the rapid development of world trades, affecting phytoplankton primary productivity therein. The variations of surface phytoplankton biomass, size-structure and carbon fixation were investigated across the SoM during the spring period (May 4 to 9, 2011). Chlorophyll a concentration increased from 0.12 μg/L at the northwest entrance of the SoM to a maximal 0.63 μg/L at narrowest section, and decreased to 0.10 μg/L at the southeast entrance. Photosynthetic carbon fixation by phytoplankton coincided well with Chl a biomass, and increased from 10.8 to 22.3 μg C/(L d), then decreased to 9.21 μg C/(L d); while the carbon fixation rate showed an inverse pattern to the changes of Chl a, and decreased from 87.1 to 35.5 μg C/(μg Chl a d) and increased thereafter to 95.3 μg C/(μg Chla·d). Picophytoplankton cells (<3 μm) contributed to more than 60% and 50% of the total Chl a and carbon fixation at both the entry waters; while the contributions of pico-cells decreased sharply to the minimum of 18.3% and 27.5% at the narrowest part of the SoM. 20Key CO_(2)capture technology of pure oxygen exhaust gas combustion for syngas-fueled high-temperature fuel cells显示文摘Integrated gasification fuel cells(IGFCs)integrating high-temperature solid oxide fuel cell technology with CO_(2)capture processes represents highly-efficient power systems with negligible CO_(2)emissions.Flame burning with pure oxygen is an ideal method for fuel cell exhaust gas treatment,and this report describes experimental and numerical studies regarding an oxy-combustor for treating the exhaust gas of a 10 kW IGFC system anode.The applied simulation method was verified based on experiments,and the key performance indices of the combustor were studied under various conditions.It was determined that 315 K was the ideal condensation temperature to obtain flame stability.Under these pure oxygen flame burning conditions,CO was almost completely converted,and the dry mole fraction of CO_(2)after burning was C 0.958 when there was up to 5%excess O_(2).Overall,5%excess O_(2)was recommended to maximize CO_(2)capture and promote other environmental considerations.Additionally,the optimal tangential fuel jet angle to control the liner temperature was approximately 25°.The total fuel utilization had to be high enough to maintain the oxygen flame temperature of the anode exhaust gas below 1800 K to ensure that the system was environmentally friendly.The results presented herein have great value for designing IGFCs coupled with CO_(2)capture systems.Hanlin Wang Qilong Lei Pingping Li Changlei Liu Yunpeng Xue Xuewei Zhang Chufu Li Zhibin Yang 2021International Journal of Coal Science & Technology2021,8,3:3
