1Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China,has become a public health emergency of international concern.The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models.Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs,viral replication,chest X-ray,histopathological changes and immune response.Results:Viral replication of nasopharyngeal swabs,anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema,notably,old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection.Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin 2020Animal Models and Experimental Medicine2020,3,1:22

3小鼠骨骼肌挫伤后基因表达的时间进程(英文)显示文摘Background:Though the mechanisms of skeletal muscle regeneration are deeply understood,those involved in muscle contusion,one of the most common muscle injuries in sports medicine clinics,are not.The objective of this study is to explore the mechanisms involved in muscle regeneration after contusion injury.Methods:In this study,a total of 72 mice were used.Eight of them were randomly chosen for the control group,while the rest were subjected to muscle contusion.Subsequently,their gastrocnemius muscles were harvested at different time points.The changes in muscle morphology were assessed by hematoxylin and eosin(HE) stain.In addition,the gene expression was analyzed by real-time polymerase chain reaction.Results:The data showed that the expression of many genes,i.e.,specific markers of immune cells and satellite cells,regulatory factors for muscle regeneration,cytokines,and chemokines,increased in the early stages of recovery,especially in the first 3 days.Furthermore,there were strict rules in the expression of these genes.However,almost all the genes returned to normal at 14 days post-injury.Conclusion:The sequence of immune cells invaded after muscle contusion was neutrophils,M1 macrophages and M2 macrophages.Some CC(CCL2,CCL3,and CCL4) and CXC(CXCL10) chemokines may be involved in the chemotaxis of these immune cells.HGF may be the primary factor to activate the satellite cells after muscle contusion.Moreover,2 weeks are needed to recover when acute contusion happens as used in this study.Weihua Xiao Yu Liu Beibei Luo Linlin Zhao Xiaoguang Liu Zhigang Zeng Peijie Chen 2016Journal of Sport and Health Science2016,5,1:8
4Research and application of a power-flow-calculation method in multiterminal VSC-HVDC power grid显示文摘For demonstrating a multiterminal voltage-source converter(VSC)-based high-voltage DC(HVDC)(VSCHVDC) project, this study puts forward a technical route for calculating the power flow in a 500-kV VSC-HVDC power grid in comparison with that of an AC power grid. The Jacobian matrix used in the power-flow calculation was deduced through methods such as Newton–Laphson iteration and Taylor series expansion. Further, the operation effect of powerflow calculation on a true bipolar VSC-HVDC power grid was analyzed briefly. The elements of the Jacobian matrix corresponding to VSC were studied under the mode of droop control and the control strategy of VSC-HVDC power grid was analyzed in detail. The power-flow calculation model for VSC-HVDC power grid of the master–slave control mode was simplified using the PQ decomposition method of the power-flow calculation of an AC power grid. Moreover, a four-terminal model of the Zhangbei VSC-HVDC demonstration project was established and tested on MATLAB. The simulation results under two kinds of operating conditions were analyzed and compared to the results of BPA; the deviation between the power-flow results was studied. 5Activated carbon enhanced ozonation of oxalate attributed to HO·oxidation in bulk solution and surface oxidation: Effect of activated carbon dosage and pH显示文摘Ozonation of oxalate in aqueous phase was performed with a commercial activated carbon(AC)in this work. The effect of AC dosage and solution pH on the contribution of hydroxyl radicals(HOU) in bulk solution and oxidation on the AC surface to the removal of oxalate was studied. We found that the removal of oxalate was reduced by tert-butyl alcohol(tBA) with low dosages of AC,while it was hardly affected by tBA when the AC dosage was greater than 0.3 g/L. tBA also inhibited ozone decomposition when the AC dosage was no more than 0.05 g/L, but it did not work when the AC dosage was no less than 0.1 g/L. These observations indicate that HOUin bulk solution and oxidation on the AC surface both contribute to the removal of oxalate. HOU oxidation in bulk solution is significant when the dosage of AC is low, whereas surface oxidation is dominant when the dosage of AC is high. The oxalate removal decreased with increasing pH of the solution with an AC dosage of 0.5 g/L. The degradation of oxalate occurs mainly through surface oxidation in acid and neutral solution, but through HOUoxidation in basic bulk solution. 6Transcriptomic analysis reveals key regulators of mammogenesis and the pregnancy-lactation cycle显示文摘An organ unique to mammals,the mammary gland develops 90%of its mass after birth and experiences the pregnancy-lactation-involution cycle(PL cycle)during reproduction.To understand mammogenesis at the transcriptomic level and using a ribo-minus RNA-seq protocol,we acquired greater than 50 million reads each for the mouse mammary gland during pregnancy(day 12 of pregnancy),lactation(day 14 of lactation),and involution(day 7 of involution).The pregnancy-,lactation-and involution-related sequencing reads were assembled into 17344,10160,and 13739 protein-coding transcripts and1803,828,and 1288 non-coding RNAs(ncRNAs),respectively.Differentially expressed genes(DEGs)were defined in the three samples,which comprised 4843 DEGs(749 up-regulated and 4094 down-regulated)from pregnancy to lactation and4926 DEGs(4706 up-regulated and 220 down-regulated)from lactation to involution.Besides the obvious and substantive upand down-regulation of the DEGs,we observe that lysosomal enzymes were highly expressed and that their expression coincided with milk secretion.Further analysis of transcription factors such as Trps1,Gtf2i,Tcf7l2,Nupr1,Vdr,Rb1,and Aebp1,and ncRNAs such as mir-125b,Let7,mir-146a,and mir-15 has enabled us to identify key regulators in mammary gland development and the PL cycle.ZHOU YuanYuan GONG Wei XIAO JingFa WU JiaYan PAN LinLin LI XiaoNuan WANG XuMin WANG WeiWei HU SongNian YU Jun 2014Science China(Life Sciences)2014,57,3:4
7Overexpression of miR-101 suppresses collagen synthesis by targeting EZH2 in hypertrophic scar fibroblasts显示文摘Background:MicroRNA-101(miR-101)is a tumor suppressor microRNA(miRNA)and its loss is associated with the occurrence and progression of various diseases.However,the biological function and target of miR-101 in the pathogenesis of hypertrophic scars(HS)remains unknown.Methods:We harvested HS and paired normal skin(NS)tissue samples from patients and cultured their fibroblasts(HSF and NSF,respectively).We used quantitative reverse transcriptase polymerase chain reaction(qRT-PCR),fluorescence in situ hybridization(FISH),enzyme-linked immunosorbent assays(ELISA)and Western blot analyses to measure mRNA levels and protein expression of miR-101,enhancer of zeste homolog 2(EZH2),collagen 1 and 3(Col1 and Col3)andα-smooth muscle actin(α-SMA)in different in vitro conditions.We also used RNA sequencing to evaluate the relevant signaling pathways and bioinformatics analysis and dual-luciferase reporter assays to predict miR-101 targets.We utilized a bleomycin-induced fibrosis mouse model in which we injected miR-101 mimics to evaluate collagen deposition in vivo.Results:We found low expression of miR-101 in HS and HSF compared to NS and NSF.Overexpressing miR-101 decreased Col1,Col3 andα-SMA expression in HSF.We detected high expression of EZH2 in HS and HSF.Knockdown of EZH2 decreased Col1,Col3 andα-SMA in HSF.Mechanistically,miR-101 targeted the 3-untranslated region(3UTR)of EZH2,as indicated by the decreased expression of EZH2.Overexpressing EZH2 rescued miR-101-induced collagen repression.MiR-101 mimics effectively suppressed collagen deposition in the bleomycin-induced fibrosis mouse model.Conclusions:Our data reveal that miR-101 targets EZH2 in HS collagen production,providing new insight into the pathological mechanisms underlying HS formation.Jie Li Yan Li Yunchuan Wang Xiang He Jing Wang Weixia Cai Yanhui Jia Dan Xiao Jian Zhang Ming Zhao Kuo Shen Zichao Li Wenbin Jia Kejia Wang Yue Zhang Linlin Su Huayu Zhu Dahai Hu 2021Burns & Trauma2021,9,1:3

17Experimental study on the acoustic propagation and anisotropy of coal rocks显示文摘In order to study the propagation laws of acoustic wave of coal samples from the Upper Permian Xuanwei Formation in the east of Yunnan Province,China,under saturated water and dry conditions,the basic physical parameters,acoustic parameters and anisotropic parameters were obtained through the experiments.Based on FFT and wavelet analysis theory,the spectral characteristics of coal samples under different conditions were studied.The results show that physical parameters of coal samples in different directions have different values,that is,the anisotropy of coal samples is obvious.When the coal samples are saturated with water,the acoustic velocities and the attenuation coefficient increase,whereas the dominant frequency decreases.The signal amplitude of the frequency domain significantly decreases,that is,the internal structure of coal samples is damaged.The P-wave velocity and S-wave velocity increase with the increase of the confining pressure,whereas the anisotropy parameters decrease with the increase of the confining pressure.Overall,this study provides the basis to understand basic acoustic information and anisotropy characteristics of coal samples.Linlin Huang Xiangjun Liu Sen Yan Jian Xiong Haiming He Peng Xiao 2022Petroleum2022,8,1:1
18NPRC deletion mitigated atherosclerosis by inhibiting oxidative stress, inflammation and apoptosis in ApoE knockout mice显示文摘Previous studies suggested a beneficial effect of natriuretic peptides in animal models of cardiovascular disease,but the role of natriuretic peptide receptor C(NPRC)in the pathogenesis of atherosclerosis(AS)remains unknown.This study was designed to test the hypothesis that NPRC may promote AS lesion formation and instability by enhancing oxidative stress,inflammation,and apoptosis via protein kinase A(PKA)signaling.ApoE^(−/−)mice were fed chow or Western diet for 12 weeks and NPRC expression was significantly increased in the aortic tissues of Western diet-fed mice.Systemic NPRC knockout mice were crossed with ApoE^(−/−)mice to generate ApoE^(−/−)NPRC^(−/−)mice,and NPRC deletion resulted in a significant decrease in the size and instability of aortic atherosclerotic lesions in ApoE^(−/−)NPRC^(−/−)versus ApoE^(−/−)mice.In addition,endothelial cell-specific NPRC knockout attenuated atherosclerotic lesions in mice.In contrast,endothelial cell overexpression of NPRC aggravated the size and instability of atherosclerotic aortic lesions in mice.Experiments in vitro showed that NPRC knockdown in human aortic endothelial cells(HAECs)inhibited ROS production,pro-inflammatory cytokine expression and endothelial cell apoptosis,and increased eNOS expression.Furthermore,NPRC knockdown in HAECs suppressed macrophage migration,cytokine expression,and phagocytosis via its effects on endothelial cells.On the contrary,NPRC overexpression in endothelial cells resulted in opposite effects.Mechanistically,the anti-inflammation and anti-atherosclerosis effects of NPRC deletion involved activation of cAMP/PKA pathway,leading to downstream upregulated AKT1 pathway and downregulated NF-κB pathway.In conclusion,NPRC deletion reduced the size and instability of atherosclerotic lesions in ApoE^(−/−)mice via attenuating inflammation and endothelial cell apoptosis and increasing eNOS expression by modulating cAMP/PKA-AKT1 and NF-κB pathways.Thus,targeting NPRC may provide a promising approach to the prevention and treatment of atherosclerosis.Cheng Cheng Jie Zhang Xiaodong Li Fei Xue Lei Cao Linlin Meng Wenhai Sui Meng Zhang Yuxia Zhao Bo Xi Xiao Yu Feng Xu Jianmin Yang Yun Zhang Cheng Zhang 2023Signal Transduction and Targeted Therapy2023,8,9:0
19Correlation analysis of longitudinal cracks and vertical deformation within asphalt pavement of cold regions显示文摘The asphalt pavement longitudinal crack is a common distress in cold regions,resulting from uneven deformation of the subgrade.Analysis of the correlation law between uneven deformation and crack distress is of positive significance for understanding the mechanism of crack initiation,and putting forward treatment measures.In view of the complexity of longitudinal crack inducement and road surface deformation,the grey relational method was used to analyze this relationship.Through long-term monitoring of the vertical deformation data of typical road sections,the vertical deformation law of the pavement surface and its deformation characteristics under the action of temperature field are analyzed.Parameters such as vertical relative deformation,vertical relative deformation rate and vertical differential deformation VDSr were constructed to describe vertical deformation characteristics.Typical distribution characteristics of longitudinal fractures and their length and distribution characteristics are also described.The grey correlation analysis theory was utilized to analyze the relationship between deformation characteristics of sections,cross sections and monitoring points and longitudinal crack characteristics(length and location).The analysis reveals a linear positive correlation or a high correlation between several indicators.This study can provide a deeper understanding of the occurrence and development mechanism of longitudinal cracks in asphalt pavement of cold areas,and give references for the research of road engineering structure,materials and distress prevention.Ke Chen Lun Ji ZeYu Xiao JinQi Wu LinLin Xu ZhiCe Cheng 2023Research in Cold and Arid Regions2023,15,6:0
20Genome degradation promotes Salmonella pathoadaptation by remodeling fimbriae-mediated proinflammatory response显示文摘Understanding changes in pathogen behavior(e.g.increased virulence,a shift in transmission channel)is critical for the public health management of emerging infectious diseases.Genome degradation via gene depletion or inactivation is recognized as a pathoadaptive feature of the pathogen evolving with the host.However,little is known about the exact role of genome degradation in affecting pathogenic behavior,and the underlying molecular detail has yet to be examined.Using large-scale global avian-restricted Salmonella genomes spanning more than a century,we projected the genetic diversity of Salmonella Pullorum(bvSP)by showing increasingly antimicrobial-resistant ST92 prevalent in Chinese flocks.The phylogenomic analysis identified three lineages in bvSP,with an enhancement of virulence in the two recently emerged lineages(L2/L3),as evidenced in chicken and embryo infection assays.Notably,the ancestor L1 lineage resembles the Salmonella serovars with higher metabolic flexibilities and more robust environmental tolerance,indicating stepwise evolutionary trajectories towards avian-restricted lineages.Pan-genome analysis pinpointed fimbrial degradation from a virulent lineage.The later engineeredfim-deletion mutant,and all other five fimbrial systems,revealed behavior switching that restricted horizontal fecal-oral transmission but boosted virulence in chicks.By depleting fimbrial appendages,bvSP established persistent replication with less proinflammation in chick macrophages and adopted vertical transovarial transmission,accompanied by ever-increasing intensification in the poultry industry.Together,we uncovered a previously unseen paradigm for remodeling bacterial surface appendages that supplements virulence-enhanced evolution with increased vertical transmission.Xiao Zhou Xiamei Kang Jiaqi Chen Yan Song Chenghao Jia Lin Teng Yanting Tang Zhijie Jiang Xianqi Peng Xiaoxi Tao Yiwei Xu Linlin Huang Xuebin Xu Yaohui Xu Tengfei Zhang Shenye Yu Jiansen Gong Shaohui Wang Yuqing Liu Guoqiang Zhu Corinna Kehrenberg Franois-Xavier Weill Paul Barrow Yan Li Guoping Zhao Min Yue 2023National Science Review2023,10,10:0
