Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China, has become a public health emergency of international concern. The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models. Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs, viral replication, chest X-ray,histopathological changes and immune response. Results:Viral replication of nasopharyngeal swabs, anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema, notably, old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection. | Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin | 2020 | Animal Models and Experimental Medicine2020,3,1: | 22
Glutamine synthetase as an early marker for hepatocellular carcinoma based on proteomic analysis of resected smal hepatocel ular carcinomas显示文摘BACKGROUND:Hepatocellular carcinoma(HCC)is a highly malignant tumor with a poor prognosis.Because small HCCs possess most of the characteristics of early HCC,we investigated small HCCs to screen potential biomarkers for early diagnosis.METHODS:Proteins were extracted from 10 sets of paired tissue samples from HBV-infected small-HCC patients. The extracted proteins were well resolved by two-dimensional electrophoresis. These HCC-associated proteins were then identified by MALDI-TOF/TOF MS following image analysis.Western blotting and immunohistochemistry were used to assess glutamine synthetase(GS)and phenazine biosynthesislike domain-containing protein(PBLD)expression in liver tissue.Enzyme-linked immunosorbent assays in 152 serum samples(from 49 healthy donors,24 patients with liver cirrhosis, and 79 with HCC)were used to further assess the significance of GS clinically.RESULTS:Fifteen up-regulated and three down-regulated proteins were identified.Western blotting confirmed GS overexpression and decreased PBLD expression in liver tissue.Immunohistochemistry showed that GS was expressed in 70.0%(84/120)of HCCs and 35.8%(43/120)of nontumor tissues;PBLD was expressed in 74.2%(89/120) of nontumor tissues and 40.8%(49/120)of HCCs.The Chi-square test showed significant expression differences between HCCs and adjacent tissues.Consistent with this, serum GS levels in HCC patients were significantly higher than those in liver cirrhosis patients and healthy donors, while the latter two groups were also significantly different. In addition, a diagnostic cutoff value of 2.6 mg/ml was used for GS;it was elevated in 19(76.0%)of 25 HCC patients with AFP≤20 ng/ml and 47(88.7%)of 53 HCC patients with AFP≤200 ng/ml.CONCLUSION:GS and PBLD are abnormally expressed in most HCCs.GS may be a novel serum marker for early HCC, especially for those patients with low AFP levels(≤200 ng/ml). | Jiang Long,Zhen-Wei Lang,Hua-Guang Wang,Tai-Ling Wang,Bao-En Wang and Si-Qi Liu Liver Research Center,Beijing Friendship Hospital, Capital Medical University,Beijing 100050,China Department of Hepatology,Department of Pathology, Beijing Youan Hospital,Capital Medical University,Beijing 100069,China Department of Pharmaceutical Affairs,Beijing Chaoyang Hospital,Capital Medical University,Beijing 100020,China Department of Pathology,China-Japan Friendship Hospital,Beijing 100029,China Beijing Genomics Institute,Chinese Academy of Science,Beijing Airport Industrial Zone B-6,Shunyi,Beijing 101300,China | 2010 | Hepatobiliary & Pancreatic Diseases International2010,9,3: | 21
A Deep Double-Channel Dense Network for Hyperspectral Image Classifica-tion显示文摘Hyperspectral Image(HSI)classification based on deep learning has been an attractive area in recent years. However,as a kind of data-driven algorithm, the deep learning method usually requires numerous computational resources and high-quality labelled datasets, while the expenditures of high-performance computing and data annotation are expensive. In this paper,to reduce the dependence on massive calculation and labelled samples,we propose a deep Double-Channel dense network(DDCD)for Hyperspectral Image Classification.Specifically,we design a 3D Double-Channel dense layer to capture the local and global features of the input. And we propose a Linear Attention Mechanism that is approximate to dot-product attention with much less memory and computational costs. The number of parameters and the consumptions of calculation are observably less than contrapositive deep learning methods, which means DDCD owns simpler architecture and higher efficiency.A series of quantitative experiences on 6 widely used hyperspectral datasets show that the proposed DDCD obtains state-of-the-art performance, even though when the absence of labelled samples is severe. | Kexian WANG Shunyi ZHENG Rui LI Li GUI | 2021 | Journal of Geodesy and Geoinformation Science2021,4,4: | 11
Land cover classification from remote sensing images based on multi-scale fully convolutional network显示文摘Although the Convolutional Neural Network(CNN)has shown great potential for land cover classification, the frequently used single-scale convolution kernel limits the scope of informa-tion extraction. Therefore,we propose a Multi-Scale Fully Convolutional Network(MSFCN)with a multi-scale convolutional kernel as well as a Channel Attention Block(CAB)and a Global Pooling Module(GPM)in this paper to exploit discriminative representations from two-dimensional(2D)satellite images.Meanwhile,to explore the ability of the proposed MSFCN for spatio-temporal images,we expand our MSFCN to three-dimension using three-dimensional(3D)CNN,capable of harnessing each land cover category’s time series interac-tion from the reshaped spatio-temporal remote sensing images. To verify the effectiveness of the proposed MSFCN,we conduct experiments on two spatial datasets and two spatio-temporal datasets. The proposed MSFCN achieves 60.366%on the WHDLD dataset and 75.127%on the GID dataset in terms of mIoU index while the figures for two spatio-temporal datasets are 87.753%and 77.156%.Extensive comparative experiments and abla-tion studies demonstrate the effectiveness of the proposed MSFCN. | Rui Li Shunyi Zheng Chenxi Duan Libo Wang Ce Zhang | 2022 | Geo-Spatial Information Science2022,25,2: | 4
Sequential infection with H1N1 and SARS-CoV-2 aggravated COVID-19 pathogenesis in a mammalian model, and covaccination as an effective method of prevention of COVID-19 and influenza显示文摘Influenza A virus may circulate simultaneously with the SARS-CoV-2 virus, leading to more serious respiratory diseases during this winter. However, the influence of these viruses on disease outcome when both influenza A and SARS-CoV-2 are present in the host remains unclear.Using a mammalian model, sequential infection was performed in ferrets and in K18-MCE2 mice, with SARS-CoV-2 infection following H1N1.We found that co-infection with H1N1 and SARS-CoV-2 extended the duration of clinical manifestation of COVID-19,and enhanced pulmonary damage, but reduced viral shedding of throat swabs and viral loads in the lungs of ferrets. Moreover, mortality was increased in sequentially infected mice compared with single-infection mice.Compared with singlevaccine inoculation,co-inoculation of PiCoVacc(a SARS-CoV-2 vaccine)and the flu vaccine showed no significant differences in neutralizing antibody titers or virus-specific immune responses.Combined immunization effectively protected K18-MCE2 mice against both H1N1 and SARS-CoV-2 infection. Our findings indicated the development of systematic models of co-infection of H1N1 and SARS-CoV-2,which together notably enhanced pneumonia in ferrets and mice,as well as demonstrated that simultaneous vaccination against HINT and SARS-CoV-2 may be an effective prevention strategy for the coming winter. | Linlin Bao Wei Deng Feifei Qi Qi Lv Zhiqi Song Jiangning Liu Hong Gao Qiang Wei Pin Yu Yanfeng Xu Yajin Qu Fengdi Li Jing Xue Shuran Gong Mingya Liu Guanpeng Wang Shunyi Wang Binbin Zhao Bin Cong Chuan Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 4

Enhancing the Effectiveness of Trimethylchlorosilane Purification Process Monitoring with Variational Autoencoder显示文摘In modern industry, process monitoring plays a significant role in improving the quality of process conduct. With the higher dimensional of the industrial data, the monitoring methods based on the latent variables have been widely applied in order to decrease the wasting of the industrial database.Nevertheless, these latent variables do not usually follow the Gaussian distribution and thus perform unsuitable when applying some statistics indices, especially the T^(2) on them.Variational AutoEncoders(VAE),an unsupervised deep learning algorithm using the hierarchy study method, has the ability to make the latent variables follow the Gaussian distribution. The partial least squares(PLS)are used to obtain the information between the dependent variables and independent variables. In this paper,we will integrate these two methods and make a comparison with other methods. The superiority of this proposed method will be verified by the simulation and the Trimethylchlorosilane purification process in terms of the multivariate control charts. | Jinfu Wang Shunyi Zhao Fei Liu Zhenyi Ma | 2022 | Computer Modeling in Engineering & Sciences2022,,8: | 1

ACE2 expression is regulated by AhR in SARS-CoV-2-infected macaques显示文摘During the current outbreak of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),more than 115 million people have been infected, and 2.5 million have died.1,2 Despite such great harm to human health, the pathogenesis of COVID-19 remains unclear. As the first step in the pathogenetic process, viral entry is mediated by the binding of the SARS-CoV-2 surface spike(S)protein to angiotensin-converting enzyme 2(ACE2)on host cells, such as lung epithelial cells. As an alternative to S protein-blocking strategies, manipulating host cell ACE2 expression may exert an inhibitory effect on SARS-CoV-2 infection. However, the molecular mechanism regulating ACE2 expression remains unclear. | Jiadi Lv Pin Yu Zhenfeng Wang Wei Deng Linlin Bao Jiangning Liu Fengli Li Qiangqiang Zhu Nannan Zhou Qi Lv Guanpeng Wang Shunyi Wang Yabo Zhou Jiangping Song Wei-Min Tong Yuying Liu Chuan Qin Bo Huang | 2021 | Cellular & Molecular Immunology2021,18,5: | 1

Research on internet traffic classification techniques using supervised machine learning显示文摘Internet traffic classification is vital to the areas of network operation and management. Traditionalclassification methods such as port mapping and payload analysis are becoming increasingly difficult asnewly emerged applications (e.g. Peer-to-Peer) using dynamic port numbers, masquerading techniquesand encryption to avoid detection. This paper presents a machine learning (ML) based traffic classificationscheme, which offers solutions to a variety of network activities and provides a platform of performanceevaluation for the classifiers. The impact of dataset size, feature selection, number of applicationtypes and ML algorithm selection on classification performance is analyzed and demonstrated by the followingexperiments: (1) The genetic algorithm based feature selection can dramatically reduce the costwithout diminishing classification accuracy. (2) The chosen ML algorithms can achieve high classificationaccuracy. Particularly, REPTree and C4.5 outperform the other ML algorithms when computational complexityand accuracy are both taken into account. (3) Larger dataset and fewer application types wouldresult in better classification accuracy. Finally, early detection with only several initial packets is proposedfor real-time network activity and it is proved to be feasible according to the preliminary results. | 李君 Zhang Shunyi Wang Pan Li Cuilian | 2009 | High Technology Letters2009,15,4: | 1

Injectable nanofiber-reinforced bone cement with controlled biodegradability for minimally-invasive bone regeneration显示文摘Injectable materials show their special merits in regeneration of damaged/degenerated bones in minimally-invasive approach.Injectable calcium phosphate bone cement(CPC)has attracted broad attention for its bioactivity,as compared to non-degradable polymethyl methacrylate cement. However, its brittleness, poor anti-washout property and uncontrollable biodegradability are the main challenges to limit its further clinical application mainly because of its stone-like dense structure and fragile inorganic-salt weakness.Herein,we developed a kind of injectable CPC bone cement with porous structure and improved robustness by incorporating poly(lactide-co-glycolic acid)(PLGA)nanofiber into CPC,with carboxymethyl cellulose(CMC)to offer good injectability as well as anti-wash-out capacity.Furthermore, the introduction of PLGA and CMC also enabled a formation of initial porous structure in the cements, where PLGA nanofiber endowed the cement with a dynamically controllable biodegradability which provided room for cell movement and bone ingrowth.Inter-estingly,the reinforced biodegradable cement afforded a sustainable provision of Ca^(2+)bioactive components, together with its porous structure,to improve synergistically new bone formation and osteo-integration in vivo by using a rat model of femur condyle defect.Further study on regenerative mechanisms indicated that the good minimally-invasive bone regeneration may come from the synergistic enhanced osteogenic effect of calcium ion enrichment and the improved revascularization capacity contributed from the porosity as well as the lactic acid released from PLGA nanofiber. These results indicate the injectable bone cement with high strength,anti-washout property and controllable biodegradability is a promising candidate for bone regeneration in a minimally-invasive approach. | Peihao Cai Shunyi Lu Jieqin Yu Lan Xiao Jiayi Wang Haifeng Liang Lei Huang Guanjie Han Mengxuan Bian Shihao Zhang Jian Zhang Changsheng Liu Libo Jiang Yulin Li | 2023 | Bioactive Materials2023,,3: | 1

A Stochastic Optimization Approach for Dynamic Adaptive Streaming over NDN显示文摘Nowadays, video streaming counts for the major part of network traffic over the Internet. However,on account of the host-to-host mechanism of the traditional IP network, video distribution over IP-based Internet encounters bottlenecks.Fortunately,a new proposed future Internet architecture, named data networking(NDN)can improve the performance of video distribution by its features such as in-network storage,multi-path forwarding, etc. In this paper,we design an adaptive bitrate algorithm based on Lyapunov optimization theory over NDN to optimize the long-term quality-of-experience(QoE)of video distribution while ensuring the stability of the whole system. When the network condition is abundant and stable, the problem can be simplified by approximating to a fixed-slot queuing model, but the theoretical performance will degrade when the network status is poor and fluctuate fiercely. Therefore,we divide the problem into two models of fixed time slot and non-fixed time slot and design two Lyapunov optimization algorithms to adapt different network scenarios. The proposed algorithms do not require prior knowledge of the network bandwidth and are capable of running online with the client’s available information.Simulation and realistic experiment results demonstrate that our algorithms perform better than others in NDN. | Xiaobin Tan Shunyi Wang Quan Zheng Bei Liu Yi He Xiangyang Wu | 2021 | Journal of Communications and Information Networks2021,6,3: | 0
The protection of CoronaVac against the infection of wild-type SARS-CoV-2(WH-09)or Omicron variant in nude-hACE2 mice显示文摘Background:Immunocompromised individuals have an increased risk of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection and severe outcomes, but we pay less attention to these people.Athymic nude mice are a murine strain with a spontaneous deficiency of the Foxn1 gene, which can result in thymic degeneration or its absence, leading to immunosuppression and a decrease in the number of T cells, and are widely used in preclinical evaluations of disease in immunocompromised populations. Methods:We investigated the protection of the coronavirus disease 2019(COVID-19)inactivated vaccine(CoronaVac)against the infection of wild-type SARS-CoV-2(WH-09)or Omicron variant utilizing a hybrid-type nude-hACE2 mouse model. Results:Compared with nude-hACE2/W mice, the viral load in the brain and lung tissue of nude-hACE2 mice(nude-hACE2/WV)infected with WH-09 after vaccination significantly decreased, and the histopathological changes were also reduced. The viral load in the brain and lung tissue of nude-hACE2 mice(nude-hACE2/OV)infected with the Omicron variant after vaccination was lower than that in nude-hACE2/O,but histopathological symptoms did not improve significantly. Conclusion:CoronaVac provides some protection against infection of both WH-09 and the Omicron variant in the nude-hACE2 mice. Our findings aimed to provide a reference for vaccination against SARS-CoV-2 in immunocompromised populations. | Kaili Lin Meixuan Liu Lu Sun Hanjun Fu Hongwei Qiao Shunyi Wang Sidan Pan Hong Gao | 2023 | Animal Models and Experimental Medicine2023,6,4: | 0
