Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China, has become a public health emergency of international concern. The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models. Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs, viral replication, chest X-ray,histopathological changes and immune response. Results:Viral replication of nasopharyngeal swabs, anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema, notably, old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection. | Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin | 2020 | Animal Models and Experimental Medicine2020,3,1: | 22
Sequential infection with H1N1 and SARS-CoV-2 aggravated COVID-19 pathogenesis in a mammalian model, and covaccination as an effective method of prevention of COVID-19 and influenza显示文摘Influenza A virus may circulate simultaneously with the SARS-CoV-2 virus, leading to more serious respiratory diseases during this winter. However, the influence of these viruses on disease outcome when both influenza A and SARS-CoV-2 are present in the host remains unclear.Using a mammalian model, sequential infection was performed in ferrets and in K18-MCE2 mice, with SARS-CoV-2 infection following H1N1.We found that co-infection with H1N1 and SARS-CoV-2 extended the duration of clinical manifestation of COVID-19,and enhanced pulmonary damage, but reduced viral shedding of throat swabs and viral loads in the lungs of ferrets. Moreover, mortality was increased in sequentially infected mice compared with single-infection mice.Compared with singlevaccine inoculation,co-inoculation of PiCoVacc(a SARS-CoV-2 vaccine)and the flu vaccine showed no significant differences in neutralizing antibody titers or virus-specific immune responses.Combined immunization effectively protected K18-MCE2 mice against both H1N1 and SARS-CoV-2 infection. Our findings indicated the development of systematic models of co-infection of H1N1 and SARS-CoV-2,which together notably enhanced pneumonia in ferrets and mice,as well as demonstrated that simultaneous vaccination against HINT and SARS-CoV-2 may be an effective prevention strategy for the coming winter. | Linlin Bao Wei Deng Feifei Qi Qi Lv Zhiqi Song Jiangning Liu Hong Gao Qiang Wei Pin Yu Yanfeng Xu Yajin Qu Fengdi Li Jing Xue Shuran Gong Mingya Liu Guanpeng Wang Shunyi Wang Binbin Zhao Bin Cong Chuan Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 4
ACE2 expression is regulated by AhR in SARS-CoV-2-infected macaques显示文摘During the current outbreak of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),more than 115 million people have been infected, and 2.5 million have died.1,2 Despite such great harm to human health, the pathogenesis of COVID-19 remains unclear. As the first step in the pathogenetic process, viral entry is mediated by the binding of the SARS-CoV-2 surface spike(S)protein to angiotensin-converting enzyme 2(ACE2)on host cells, such as lung epithelial cells. As an alternative to S protein-blocking strategies, manipulating host cell ACE2 expression may exert an inhibitory effect on SARS-CoV-2 infection. However, the molecular mechanism regulating ACE2 expression remains unclear. | Jiadi Lv Pin Yu Zhenfeng Wang Wei Deng Linlin Bao Jiangning Liu Fengli Li Qiangqiang Zhu Nannan Zhou Qi Lv Guanpeng Wang Shunyi Wang Yabo Zhou Jiangping Song Wei-Min Tong Yuying Liu Chuan Qin Bo Huang | 2021 | Cellular & Molecular Immunology2021,18,5: | 1
Widely tunable microwave photonic filter based on self-adaptive optical carrier regeneration显示文摘In this paper, we have designed and proofed a widely tunable microwave photonic filter (MPF) on the basis of self-adaptive optical carrier regeneration method through stimulated Brillouin scattering (SBS) amplification. The MPF system features with a single microwave passband as the result of the processed signal sideband beating with the regenerated optical carrier. Since the Brillouin-laser optical carrier and the SBS amplification are generated from one laser source, the optical carrier regeneration process of the proposed MPF is self- adaptive and carrier-frequency independent. Moreover, by simply varying the wavelength spacing between the optical carrier and the optical filter, the central passband of the MPF can be continuously tuned from 6 to 32 GHz, which is limited by the bandwidth of the modulator and the photodetector. The 3 dB bandwidth and the out-of-band suppression ratio of the MPF can reach 640 MHz and 21 dB, respectively. | LI GuanPeng WANG Hui QI XiaoQiong XIE Liang ZHANG BangHong | 2017 | Science China(Technological Sciences)2017,60,6: | 1
Vacuum wetting of Ag/TA2 to develop a novel micron porous Ti with significant biocompatibility and antibacterial activity显示文摘Porous Ti with low modulus, excellent bio-corrosion resistance, biocompatibility, and antibacterial activity is highly pursued as advanced implant materials. In this work,a new approach to prepare micron porous structures on the surface layer of a grade 2 commercially-pure Ti(TA2)was proposed, which utilized a simple vacuum wetting process of pure Ag on the surface of TA2.The microstructure, corrosion resistance, biocompatibility, mechanical properties, antibacterial ability, and formation mechanism of the asfabricated porous Ti were studied. The results show that the pores(with average pore sizes of 0.5-5μm)are distributed on the surface layer of the TA2 with a depth of~10μm.In particular,a large number of silver nanoparticles(Ag NPs)form which are dispersed on the porous structures. The formation mechanisms of the porous structures and Ag NPs were elucidated, suggesting that the volatilization/sublimation of Ag in TA2 is crucial. The porous Ti possesses excellent bio-corrosion resistance, surface wettability, biocompatibility, antibacterial activity, and a relatively low elastic modulus of 40-55 GPa,which may have a promising future in the field of orthopedic implants. This work also provides a novel idea for the development of advanced porous Ti materials for orthopedic-related basic research and biomedical applications. | Guanpeng Liu Yulong Li Ming Yan Jicai Feng Jian Cao Min Lei Quanwen Liu Xiaowu Hu Wenqin Wang Xuewen Li | 2022 | Journal of Materials Science & Technology2022,,21: | 0
Investigation on the Changing Characteristics of Flow-Induced Noise in a Centrifugal Pump显示文摘Centrifugal pumps are widely used in engineering for a variety of applications.A known drawback of these devices is the high-level noise generated during operations, which can affect their stability and adversely influence the entire working environment.By combining the Powell vortex sound theory, numerical simulations and experimental measurements, this research explores the trends of variation and the corresponding underlying mechanisms for the flow-induced noise at various locations and under different operating conditions. It is shown that the total sound source intensity(TSSI)and total sound pressure level(TSPL)in the impeller,in the region between the inlet to the outlet and along the circumferential extension of the volute, are much higher than those at pump inlet and outlet.Additionally, under various rotational speeds with the design flow rate(Condition 1),the TSSI and TSPL at pump inlet and outlet are higher than those obtained with the opening of the valve kept unchanged(Condition 2);vice versa when these two parameters are evaluated at various locations in the impeller and the volute under the Condition 2,they exceed the equivalent values obtained for the other Condition 1. | Guanpeng Li Lihui Sun Zhaoyang Wang Chunguo An Chang Guo Shen Cheng Ming Gao | 2021 | Fluid Dynamics & Materials Processing2021,17,5: | 0
Synthesis and Property of H_2La_2Ti_3O_(10)/CdS Layered Nanocomposite Photocatalyst显示文摘H2La2Ti3O10/CdS nanocomposite with higher photoactivity was synthesized by a stepwise exchange process from H2La2Ti3O 10 , which was obtained by H+-exchanging reaction of K2La2Ti3O 10 with HCl other than the normally used HNO3. The pillaring process was investigated by XRD, TEM, FT-IR and BET methods. The photocatalytic decomposition of aniline was used as the model system to evaluate the photochemical properties of H2La2Ti3O 10 /CdS, H2La2Ti3O 10 and K2La2Ti3O 10 . It is found that the photoactivity of layered H2La2Ti3O 10 is greatly improved by the intercalation of CdS in the interlayer. In general, the excellent photoactivity of the H2La2Ti3O 10 /CdS nanocomposite might be attributing to the extremely small particle size of incorporated CdS and good contact between CdS and La2Ti3O 10 2- layers. On the other hand, the coupling of two semiconductor particles with different energy gap is useful to achieve effective charge separation. In H2La2Ti3O 10 /CdS, a photo-generated electron can transfer from CdS to the La2Ti3O 10 2- layer, while the holes remain in the CdS particles. This helps to diffuse the electrons and holes before reaching the interface, and the holes and electrons can be effectively captured by the electrolyte in the solution. Simultaneously, the optimal operating condition of photodecomposition aniline was investigated. | Zhang Lili Liu Guanpeng Zhang Weiguang Lu Lude Yang Xujie Wang Xin | 2006 | Journal of Rare Earths2006,24,2: | 0
Local field potentials, spiking activity, and receptive fields in human visual cortex显示文摘The concept of receptive field(RF) is central to sensory neuroscience. Neuronal RF properties have been substantially studied in animals, while those in humans remain nearly unexplored. Here, we measured neuronal RFs with intracranial local field potentials(LFPs) and spiking activity in human visual cortex(V1/V2/V3). We recorded LFPs via macro-contacts and discovered that RF sizes estimated from lowfrequency activity(LFA, 0.5–30 Hz) were larger than those estimated from low-gamma activity(LGA, 30–60 Hz) and high-gamma activity(HGA, 60–150 Hz). We then took a rare opportunity to record LFPs and spiking activity via microwires in V1 simultaneously. We found that RF sizes and temporal profiles measured from LGA and HGA closely matched those from spiking activity. In sum, this study reveals that spiking activity of neurons in human visual cortex could be well approximated by LGA and HGA in RF estimation and temporal profile measurement, implying the pivotal functions of LGA and HGA in early visual information processing. | Lu Luo Xiongfei Wang Junshi Lu Guanpeng Chen Guoming Luan Wu Li Qian Wang Fang Fang | 2024 | Science China(Life Sciences)2024,67,3: | 0
Variable Selection in High-Dimensional Error-in-Variables Models via Controlling the False Discovery Proportion显示文摘Multiple testing has gained much attention in high-dimensional statistical theory and applications, and the problem of variable selection can be regarded as a generalization of the multiple testing. It is aiming to select the important variables among many variables.Performing variable selection in high-dimensional linear models with measurement errors is challenging. Both the influence of high-dimensional parameters and measurement errors need to be considered to avoid severely biases. We consider the problem of variable selection in error-in-variables and introduce the DCoCoLasso-FDP procedure,a new variable selection method.By constructing the consistent estimator of false discovery proportion(FDP)and false discovery rate(FDR),our method can prioritize the important variables and control FDP and FDR at a specifical level in error-in-variables models. An extensive simulation study is conducted to compare DCoCoLasso-FDP procedure with existing methods in various settings, and numerical results are provided to present the efficiency of our method. | Xudong Huang Nana Bao Kai Xu Guanpeng Wang | 2022 | Communications in Mathematics and Statistics2022,10,1: | 0
ADMM Algorithmic Regularization Paths for Sparse and Large Scale Positive-Definite Covariance Matrix Estimation显示文摘Estimating sparse positive-definite covariance matrices in high dimensions has received extensive attention in the past two decades. However, many existing algorithms are proposed for a single regularization parameter and little attention has been paid to estimating the covariance matrices over the full range of regularization parameters. In this paper we suggest to compute the regularization paths of estimating the positive-definite covariance matrices through a one-step approximation of the warm-starting Alternating Direction Method of Multipliers(ADMM)algorithm, which quickly outlines a sequence of sparse solutions at a fine resolution. We demonstrate the effectiveness and computational savings of our proposed algorithm through elaborative analysis of simulated examples. | XIA Lin WANG Guanpeng HUANG Xudong | 2022 | Wuhan University Journal of Natural Sciences2022,27,2: | 0
