| 2 | Mitochondria-associated endoplasmic reticulum membranes allow adaptation of mitochondrial metabolism to glucose availability in the liver显示文摘Mitochondria-associated endoplasmic reticulum membranes(MAM)play a key role in mitochondrial dynamics and function and in hepatic insulin action.Whereas mitochondria are important regulators of energy metabolism,the nutritional regulation of MAM in the liver and its role in the adaptation of mitochondria physiology to nutrient availability are unknown.In this study,we found that the fasted to postprandial transition reduced the number of endoplasmic reticulum-mitochondria contact points in mouse liver.Screening of potential hormonal/metabolic signals revealed glucose as the main nutritional regulator of hepatic MAM integrity both in vitro and in vivo.Glucose reduced organelle interactions through the pentose phosphate-protein phosphatase 2A(PP-PP2A)pathway,induced mitochondria fission,and impaired respiration.Blocking MAM reduction counteracted glucose-induced mitochondrial alterations.Furthermore,disruption of MAM integrity mimicked effects of glucose on mitochondria dynamics and function.This glucose-sensing system is deficient in the liver of insulin-resistant ob/ob and cyclophilin D-KO mice,both characterized by chronic disruption of MAM integrity,mitochondrial fission,and altered mitochondrial respiration.These data indicate that MAM contribute to the hepatic glucose-sensing system,allowing regulation of mitochondria dynamics and function during nutritional transition.Chronic disruption of MAM may participate in hepatic mitochondrial dysfunction associated with insulin resistance. | Pierre Theurey Emily Tubbs Guillaume Vial Julien Jacquemetton Nadia Bendridi Marie-Agnes Chauvin Muhammad Rizwan Alam Muriel Le Romancer Hubert Vidal Jennifer Rieusset | 2016 | Journal of Molecular Cell Biology2016，8，2： | 2 |

| 16 | Acoustic radiation force impulse predicts long-term outcomes in a large-scale cohort:High liver cancer,low comorbidity in hepatitis B virus显示文摘BACKGROUND Acoustic radiation force impulse(ARFI)is used to measure liver fibrosis and predict outcomes.The performance of elastography in assessment of fibrosis is poorer in hepatitis B virus(HBV)than in other etiologies of chronic liver disease.AIM To evaluate the performance of ARFI in long-term outcome prediction among different etiologies of chronic liver disease.METHODS Consecutive patients who received an ARFI study between 2011 and 2018 were enrolled.After excluding dual infection,alcoholism,autoimmune hepatitis,and others with incomplete data,this retrospective cohort were divided into hepatitis B(HBV,n=1064),hepatitis C(HCV,n=507),and non-HBV,non-HCV(NBNC,n=391)groups.The indexed cases were linked to cancer registration(1987-2020)and national mortality databases.The differences in morbidity and mortality among the groups were analyzed.RESULTS At the enrollment,the HBV group showed more males(77.5%),a higher prevalence of prediagnosed hepatocellular carcinoma(HCC),and a lower prevalence of comorbidities than the other groups(P<0.001).The HCV group was older and had a lower platelet count and higher ARFI score than the other groups(P<0.001).The NBNC group showed a higher body mass index and platelet count,a higher prevalence of pre-diagnosed non-HCC cancers(P<0.001),especially breast cancer,and a lower prevalence of cirrhosis.Male gender,ARFI score,and HBV were independent predictors of HCC.The 5-year risk of HCC was 5.9%and 9.8%for those ARFI-graded with severe fibrosis and cirrhosis.ARFI alone had an area under the receiver operating characteristic curve(AUROC)of 0.742 for prediction of HCC in 5 years.AUROC increased to 0.828 after adding etiology,gender,age,and platelet score.No difference was found in mortality rate among the groups.CONCLUSION The HBV group showed a higher prevalence of HCC but lower comorbidity that made mortality similar among the groups.Those patients with ARFI-graded severe fibrosis or cirrhosis should receive regular surveillance. | Jennifer Tai Adam P Harrison Hui-Ming Chen Chiu-Yi Hsu Tse-Hwa Hsu Cheng-Jen Chen Wen-Juei Jeng Ming-Ling Chang Le Lu Dar-In Tai | 2023 | World Journal of Gastroenterology2023，29，14： | 1 |

| 20 | Transgenerational phenotypic plasticity of diapause induction and related fitness cost in a commercial strain of the parasitoid Aphidius ervi Haliday显示文摘Diapause is an adaptation that insects have evolved to synchronize their life cycle with that of seasonal climatic changes and resources availability.However,cues for its induction are not always clear and,in some cases,a maternal effect may be involved.At the population level,just a part of the individuals may exhibit diapause with important consequences in terms of winter survival.Moreover,clear indicators of diapause state are difficult to identify.Diapause induction was thus investigated in the aphid parasitoid species Aphidius ervi Haliday(Hymenoptera:Braconidae)developing in the aphid Sitobion avenae(Hemiptera:Aphididae)at four crossed photothermal regimes(16℃and 8℃,16:8 h L:D and 8:16 h L:D),and during 2 successive generations.We analyzed the reliability of changes in mummy color to assess for the diapausing state compared to dissections,and we measured parasitoid morphological and physiological traits.We observed that the proportion of dark brown mummies increased after one generation under low photothermal regime compared to other regimes.No diapause was recorded at 16℃,16:8 h L:D,while we observed 16.2%and 67.5%diapause incidence at 8℃,8:16 h L:D,at 1st and 2nd generation,respectively.Diapause induction is thus increased by short day-length conditions and low temperatures as well as by maternal effects.All parasitoid life-history traits(weight,size,fat content,water content,egg-load,and longevity)were affected by the photothermal regime and/or the generation.These results raise new questions on the environmental thresholds needed to induce diapause and on survival and adaptation potential of commercially available parasitoid strains in different environments. | Muhammad Mubashir Saeed Kevin Tougeron Abu Bakar Muhammad Raza Muhammad Afzal Anjum Aqueel Guillaume Jean Le Goff Frangois Renoz Jennifer Pirotte Thierry Hance | 2021 | Insect Science2021，28，3： | 0 |
