| 1 | Tight junctions in inflammatory bowel diseases and inflammatory bowel disease associated colorectal cancer显示文摘Inflammatory bowel diseases are characterised by inflammation that compromises the integrity of the epithelial barrier. The intestinal epithelium is not only a static barrier but has evolved complex mechanisms to control and regulate bacterial interactions with the mucosal surface. Apical tight junction proteins are critical in the maintenance of epithelial barrier function and control of paracellular permeability. The characterisation of alterations in tight junction proteins as key players in epithelial barrier function in inflammatory bowel diseases is rapidly enhancing our understanding of critical mechanisms in disease pathogenesis as well as novel therapeutic opportunities. | 2 | Orthotopic transplantation model of human gastrointestinal cancer and detection of micrometastases显示文摘AIM To establish a relevant animal model ofhuman gastrointestinal cancer, which can beused for repetitive investigations, so as toimprove our understanding and management ofcarcinogenesis and cancer metastasis.METHODS Intact tissues of human colorectaland pancreatic cancers were transplanted innude mice. The biological characteristics of theoriginal and the corresponding transplantedtumors were investigated by HE staining, PASstaining and immunostaining. The metastases inthe livers and lungs of nude mice wereinvestigated by immunostaining withbiotinylated mab KL-1 and by RT-PCR using CK20specific primers.RESULTS There were totally 9 of 16 surgicalspecimens growing in nude mice subcutaneouslyand/.or orthotopically (4 of 6 colorectal and 5 of10 pancreatic cancer). Tumor cell content of thespecimens and freezing of tissue specimens areimportant factors influencing the growth oftransplanted tumor. In the group of fresh tumortissues with greater than 50% tumor cellcontent, the success rate of the transplantationwas 100% (3 cases of pancreatic cancer and 3cases of colorectal cancer). The orthotopicallytransplanted tumors resemble the original tumormorphologically and biologically, including TAAexpression such as CEA byimmunohistochemistry, and CEA level in theserum of mice. Ki-67 labeling index and theexpression of TAA especially K-ras, 17-lA andRA-96, are associated with the potential of tumorgrowth in nude mice. Micrometastases in thelungs and livers of tumor bearing mice can bedetected by immunostaining with biotinylatedmab KL-1 and CK20-specific RT-PCR.CONCLUSION An orthotopic transplantationmodel for human colon and pancreatic cancer innude mice has been set up. We have alsoestablished sensitive detection methods withCK-immunohistochemistry and CK20-RT-PCR tostudy xenotransplanted human cancer and itsmetastatic cancer cells in the liver and lung ofnude mice. | 5 | A MATHEMATICAL MODEL FOR RESERVOIR SEDIMENTATION AND FLUVIAL PROCESSES显示文摘This paper consists of two parts: mechanism of non-equilibrium transport of non-uniform sediment and its application in the mathematical model. Based on a statistic approach of sediment transport proposed by the authors, a 1 - D equation of non-e- quilibrium transport for various size group of sediment particles is developed. Then equations to predict change of sediment concentration and the corresponding changes in size distribution of suspended load and bed material are also derived. The conception that changes in size distribution are interrelated to sediment-carrying capacity is expounded. These results reveal the essence of sediment transport of non-uniform sediment particles. In the second part a mathematical model incorporating the mentioned equations to compute deposition and scouring in reservoirs as well as the fluvial processes of river channels has been developed. Research results show that rill erosion processes can be schematically divided into five stages, that is, concentrated flow forming process; little waterfalls in the concentrated flow forming Process; little waterfalls developing into rill head cut process; rill head cuts developing to form discontinuous rill process and discontinuous rills connecting to form continuous rill process. Rill head cuts backward erosion, rill wall collapsing and runoff washing rill bed are the main patterns of rill development. | 11 | Dabigatran etixilate and traumatic brain injury: Evolving anticoagulants require evolving care plans显示文摘AIM: To investigate the outcomes of trauma patients with traumatic brain injury(TBI) on Dabigatran Etexilate(DE). METHODS: Following IRB approval, all patients taking DE who were admitted to our level 1 trauma service were enrolled in the study. Injury complexity, length of stay(LOS), intensive care length of stay, operative intervention, therapeutic interventions and outcomes were analyzed retrospectively. RESULTS: Twenty-eight of 4310 admissions were taking DE. Eleven patients were excluded on concurrent antiplatelet therapy. Average age was 77.14 years(64-94 years), and average LOS was 4.7 d(1-35 d). Thirty-two percent were admitted with intracranial hemorrhage. Eighteen percent received factor Ⅶ, and 22% received dialysis in attempts to correct coagulopathy. Mortality was 21%.CONCLUSION: The low incidence, absence of reversal agents, and lack of practice guidelines makes managing patients with TBI taking DE frustrating and provider specific. | 12 | Growing Operational Use of FY-3 Data in the ECMWF System显示文摘This paper reviews the data quality and impact of observations from the FY-3 satellite series used operationally in the ECMWF system. This includes data from the passive microwave radiometers MWHS-1, MWHS-2 and MWRI, as well as observations from the radio occultation receiver GNOS. Evaluations against background equivalents show that the quality of the observations is broadly comparable to that of similar instruments on other polar-orbiting satellites, even though biases for the passive microwave observations can be somewhat larger and more complex for some channels. An observing system experiment shows that the FY-3 instruments jointly contribute significantly to the forecast skill in the ECMWF system. | 13 | Characterization of colonic dendritic cells in normal and colitic mice显示文摘AIM: Recent studies demonstrating the direct involvement of dendritic cells (DC) in the activation of pathogenic T cells in animal models of inflammatory bowel disease identify DC as important antigen presenting cells in the colon. However, very little is known about the properties of colonic DC.METHODS: Using immunohistochemistry, electron microscopy and flow cytometry we have characterized and compared colonic DC in the colon of healthy animals and interleukin-2-deficient (IL2-/-) mice that develop colitis.RESULTS: In the healthy colon, DC resided within the lamina propria and in close association with the basement membrane of colonic villi. Type 1 myeloid (CD11c+, CD11b+,B220-, CD8α-) DC made up the largest (40-45%) population and all DC expressed low levels of CD80, CD86, and CD40,and had high endocytic activity consistent with an immature phenotype. In colitic IL2-/- mice, colonic DC numbers increased four- to five-fold and were localized within the epithelial layer and within aggregates of T and B cells. They were also many more DC in mesenteric lymph nodes (MLN).The majority (>85%) of DC in the colon and MLN of IL2-/-mice were type 1 myeloid, and expressed high levels of MHC class Ⅱ, CD80, CD86, CD 40, DEC 205, and CCR5molecules and were of low endocytic activity consistent with mature DC.CONCLUSION: These findings demonstrate striking changes in the number, distribution and phenotype of DC in the inflamed colon. | 17 | Reactive and non-reactive transport modelling in partially water saturated argillaceous porous media around the ventilation experiment,Mont Terri显示文摘At Mont Terri Underground Research Laboratory (URL) Switzerland, a specific experiment has been performed in a tunnel, in order to investigate the hydro-mechano-chemical (HMC) perturbations induced in the argillaceous formation by forced ventilation. This experiment has been selected in the international project DECOVALEX to be used for process model development and validation. The numerical simulation of the geochemical response to the ventilation experiment (VE) is the object of the present paper, focusing on the transport of chloride as a conservative species and sulphate as a reactive species. Utilising the validated hydro-mechanical (HM) results from earlier steps of the DECOVALEX task, reactive and non-reactive transport models, incorporating the current understanding of the geochemistry at the site, were successfully constructed for the whole experimental period. The associated parametric and process uncertainty analyses clearly demonstrate that the basic HM understanding must be sound. | 18 | Assimilation of Feng-Yun-3B Satellite Microwave Humidity Sounder Data over Land显示文摘The ECMWF has been assimilating Feng-Yun-3B(FY-3B) satellite microwave humidity sounder(MWHS) data over ocean in an operational forecasting system since 24 September 2014. It is more difficult, however, to assimilate microwave observations over land and sea ice than over the open ocean due to higher uncertainties in land surface temperature, surface emissivity and less effective cloud screening. We compare approaches in which the emissivity is retrieved dynamically from MWHS channel 1 [150 GHz(vertical polarization)] with the use of an evolving emissivity atlas from 89 GHz observations from the MWHS onboard NOAA and EUMETSAT satellites. The assimilation of the additional data over land improves the fit of short-range forecasts to other observations, notably ATMS(Advanced Technology Microwave Sounder) humidity channels, and the forecast impacts are mainly neutral to slightly positive over the first five days. The forecast impacts are better in boreal summer and the Southern Hemisphere.
