| 1 | Efficacy and safety of tenofovir in chronic hepatitis B: Australian real world experience显示文摘AIM To evaluate the long-term treatment outcomes of tenofovir therapy in patients in a real world Australian tertiary care setting.METHODS We performed a retrospective analysis of treatment outcomes among treatment-na?ve and treatment-experienced patients receiving a minimum 3 mo tenofovir therapy through St Vincent's Hospital Melbourne, Australia. We included patients receiving tenofovir [tenofovir disoproxil fumarate(TDF)] monotherapy, as well as patients treated with TDF in combination with a second antiviral agent. Patients were excluded if they demonstrated human immune-deficiency virus/hepatitis C virus/hepatitis delta virus coinfection or were less than 18 years of age. We considered virological and biochemicalresponse, as well as safety outcomes. Virological response was determined by measurement of hepatitis B virus(HBV) DNA using sensitive assays; biochemical response was determined via serum liver function tests; histological response was determined from liver biopsy and fibroscan; safety analysis focused on glomerular renal function and bone mineral density. The primary efficacy endpoint was complete virological suppression over time, defined by HBV DNA < 20 IU/m L. Secondary efficacy endpoints included rates of biochemical response, and HB e antigen(HBe Ag)/HB surface antigen loss and seroconversion over time.RESULTS Ninety-two patients were identified who fulfilled the enrolment criteria. Median follow-up was 26 mo(range 3-114). Mean age was 46(24-78) years, 64(70%) were male and 77(84%) were of Asian origin. 55(60%) patients were treatment-na?ve and 62 patients(67%) were HBe Ag-negative. Complete virological suppression was achieved by 45/65(71%) patients at 12 mo, 37/46(80%) at 24 mo and 25/28(89%) at 36 mo. Partial virological response(HBV DNA 20-2000 IU/m L) was achieved by 89/92(96.7%) of patients. Multivariate analysis showed a significant relationship between virological suppression at end of follow-up and baseline HBV DNA level(OR = 0.897, 95%CI: 0.833-0.967, P = 0.0046) and HBe Ag positive status(OR = 0.373, 95%CI: 0.183-0.762, P = 0.0069). There was no difference in response comparing treatment-na?ve and treatment-experienced patients. Three episodes of virological breakthrough occurred in the setting of noncompliance. Tenofovir therapy was well tolerated.CONCLUSION Tenofovir is an efficacious, safe and well-tolerated treatment in an Australian real-world tertiary care setting. | 2 | polyketide and benzopyran compounds of an endophytic fungus isolated from Cinnamomum mollissimum:biological activity and structure显示文摘Objective:To study bioactivity and compounds produced by an endophytic Phoma sp.fungus isolated from the medicinal plant Cinnamomum mollissimum.Methods:Compounds produced by the fungus were extracted from fungal broth culture with ethyl acetate.This was followed by hioaclivity profiling of the crude extract fractions obtained via high performance liquid chromatography.The fractions were tested for cytotoxicity to P388 murine leukemic cells and antimicrobial activity against bacteria and pathogenic fungi.Compounds purified from active fractions which showed antibacterial,antifungal and cytotoxic activities were identified using capillary nuclear magnetic resonance analysis,mass spectrometry and admission to AntiMarin database.Results:Three known compounds,namely 4—hydroxymellein,4,8—dihydroxy—6—melhoxy—3—methyl—3,4-dihydro—1H-isochromen-1—one and 1—(2,6-dihydroxyphenyl) ethanone,were isolated from the fungus.The polyketide compound 4—hydroxymellein showed high inhibitory activity against P388 murine leukemic cells(94.6%) and the bacteria Bacillus sublilis(97.3%).Meanwhile.4,8—dihydroxy-6—melhoxy—3—meth) 1—3,4-dihydro—1H—isochromen—1-one,a benzopyran compound,demonstrated moderate inhibitory activity against P388 murine leukemic cells(48.8%)and the fungus Aspergillus niger(56.1%).The second polyketide compound.1(2,6—dihydroxyphenyl)ethanone was inactive against the tested targets.Conclusions:These findings demonstrate the potential of endophytes as producers ol pharmacologically important compounds,including polyketides which are major secondary metabolites in fungi. | Carolina Santiago Lin Sun Murray Herbert Gibson Munro Jacinta Santhanam | 2014 | Asian Pacific Journal of Tropical Biomedicine2014,4,8: | 2 |

| 5 | Traditional knowledge, use and conservation of plants by the communities of Tharaka-Nithi County, Kenya显示文摘Rural communities in Kenya largely depend on plant resources for their livelihood.The utilization of these resources depends on the availability of plant resources and the level of knowledge of the residents.We conducted an ethnobotanical study in Tharaka-Nithi County in Kenya to determine the knowledge and utilization of various plant species by the local communities.The study was conducted in four major administrative regions from June 2018 to February 2019,involving interview schedules using semi-structured open-ended questionnaires and guided field collections with 48 informants.A total of 214 plant species distributed in 73 families and 169 genera with 616 Use Reports(URs)were documented.Fabaceae was the highest family cited by the informants(31 species)followed by Lamiaceae and Euphorbiaceae(each with 11 species).Trees(49%)and shrubs(32%)were the top life forms of the plants frequently utilized by the local residents.The general plant uses reported were medicinal,food,fodder,construction,fuel,pesticidal,religious,live fencing,and making crafts.Zanthoxylum gilletii,Prunus africana,and Solanum incanum were found to be highly valued by the local communities.Plant utilization as food and medicinal uses against snake-bite related problems had the highest Informant Consensus Factor(ICF).Only 29(13.6%)of the species reported had their status assessed by the International Union for Conservation of Nature(IUCN).Conservation measures,alongside awareness creation in this region,are highly recommended for the species endemic to the region,highly depended on by the community,and those threatened according to IUCN standards. | Vivian Kathambi Fredrick Munyao Mutie Peninah Cheptoo Rono Neng Wei Jacinta Ndunge Munyao Peris Kamau Robert Wahiti Gituru Guang-Wan Hu Qing-Feng Wang | 2020 | Plant Diversity2020,42,6: | 2 |
