| 1 | Neutral Red Retention by Earthworm Coelomocytes: A Biomarker of Cadmium Contamination in Soil显示文摘The earthworm Metaphire posthuma were used as a model to assess the toxic potential of cadmium incorporated into the soil by environmental or human activities. The retention period of neutral red in the lysosomes of the coelomocytes was used as a biomarker. The viability of harvested coelomocytes by a non_invasive extrusion protocol was 93% with no alteration by the dye during experimentation. The control cells retained dye for 119 and 121 min in normal soil and KCl, respectively, whereas a linear decline in the retention time in the treated earthworm coelomocytes was observed. | 6 | Simultaneous quantification of prodrug oseltamivir and its metabolite oseltamivir carboxylate in human plasma by LC-MS/MS to support a bioequivalence study显示文摘A simple,precise and rapid liquid chromatography-tandem mass spectrometry(LC-MS/MS) method has been developed and validated for the simultaneous determination of oseltamivir and oseltamivir carboxylate,a neuraminidase inhibitor,using their deuterated analogs as internal standards(ISs).The method involved solid phase extraction of the analytes and ISs from 200 μL human plasma with no reconstitution and drying steps.The chromatographic separation was achieved on a Symmetry C18(100 mm 4.6 mm,5 μm) column using 10 mM ammonium formate and acetonitrile(30:70,v/v) as the mobile phase in a run time of 2.0 min.Quantitation of analytes and ISs were done by multiple reaction monitoring on a triple quadrupole mass spectrometer in the positive ionization mode.The linearity of the method was established in the concentration range of 0.5-200 ng/mL and 2.0-800 ng/mL for oseltamivir and oseltamivir carboxylate respectively.The mean extraction recovery for oseltamivir(94.4%) and oseltamivir carboxylate(92.7%) from spiked plasma samples was consistent and reproducible.The application of this method was demonstratedby a bioequivalence study in 42 healthy Indian subjects with 75 mg oseltamivir phosphate capsules.The assay reproducibility was established by reanalysis of 151 incurred subject samples. | Ajay Gupta Swati Guttikar Pranav S. | 8 | Effect of Alstonia scholaris bark extract on testicular function of Wistar rats显示文摘Aim:To evaluate the antifertility effect of Alstonia scholaris bark extract in male rats.Methods:In male Wistar rats Alstonia scholaris bark extract was given by oral route at a dose of 200mg/day for60days,The fertility and testicular function were assessed by mating tests,sperm motility,sperm concentration,biochemical indices and testicular cell population dynamics.Results:Oral feeding with the extract at a dose of 200mg/day for the period of 60days did not cause body weight loss,while the weights of testes,epididymides,seminal vesicle and ventral prostate were significantly reduced.The production of step-19 spermatids was reduced by 79.6%in treated rats.The popu-lation of preleptotene and pachytene spermatocytes were decrased by61.9%and60.1%,respectively.Spematogo-nia and Sertoli cell population were also affected.The seminiferous tubule and Leydig cell nuclear area were reduced significantly(P<0.01)when compared to the controls..Reduced sperm count and motility yesulted in a total supprs-sion of fertility.A significant fall in the protein and sialic acid content of the testes,epididymides,seminal vesicle and ventral prostate as well as glycogen content of testes were also noticed.The fructose content in the seminal vesicle was lowered whereas the testicular cholesterol was elevated as compared with the controls.The following com-pounds were obtained from the extract with chromatographic separation over Si-gel column;α-amyrin,β-amyrin,lupiol acetate,venenative,rhazine and yohimbine,Conclusion:At the dose level employed,Alstonia scholaris bark extract has a significant antifertility effect in male rast;the primary site of action may be post meiotic germ cells(Step19spermatids). | R.
