1Accuracy and responses of genomic selection on key traits in apple breeding显示文摘The application of genomic selection in fruit tree crops is expected to enhance breeding eficiency by increasing prediction accuracy,increasing selection intensity and decreasing generation interval.The objectives of this study were to assess the accuracy of prediction and selection response in commercial apple breeding programmes for key traits.The training population comprised 977 individuals derived from 20 pedigreed fllsib families.Historic phenotypic data were available on 10 traits related to productivity and fruit external appearance and genotypic data for 7829 SNPs obtained with an llumina 20K SNP array.From these data,a genome-wide prediction model was built and subsequently used to calculate genomic breeding values of five application fllsib families.The application families had genotypes at 364 SNPs from a dedicated 512 SNP array,and these genotypic data were extended to the high-density level by imputation.These five families were phenotyped for 1 year and their phenotypes were compared to the predicted breeding values.Accuracy of genomic prediction across the 10 traits reached a maximum value of 0.5 and had a median value of 0.19.The accuracies were strongly affected by the phenotypic distribution and heritability of traits.In the largest family,significant selection response was observed for traits with high heritability and symmetric phenotypic distribution.Traits that showed non-significant response often had reduced and skewed phenotypic variation or low heritability.Among the five application families the accuracies were uncorrelated to the degree of relatedness to the training population.The results underline the potential of genomic prediction to accelerate breeding progress in outbred fruit tree crops that still need to overcome long generation intervals and extensive phenotyping costs.Hélène Muranty Michela Troggio Inès Ben Sadok Mehdi Al Rifaï Annemarie Auwerkerken Elisa Banchi Riccardo Velasco Piergiorgio Stevanato W.Eric van de Weg Mario Di Guardo Satish Kumar François Laurens Marco C.A.M.Bink 2015Horticulture Research2015,2,1:14

4Is overhydration in peritoneal dialysis patients associated with cardiac mortality that might be reversible?显示文摘AIM To study the relationship between overhydration(OH) in peritoneal dialysis(PD) patients and cardiac mortality.METHODS OH, as measured by body composition monitor(BCM), is associated with increased mortality in dialysis patients. BCM has been used to guide treatment on the assumption that correcting OH will improve cardiac morbidity and mortality although data demonstrating causality that is reversible is limited. We wished to determine if OH in PD patients predicted cardiac mortality, and if there was a correlation between OH and cardiac troponin-T(cT n T) levels. Finally, we wished to determine if improving OH values would lead to a decrement in cT n T. All prevalent PD patients over the study period of 57 mo who had contemporaneous BCM and cT nT measurements were followed irrespective of transplantation or PD technique failure. We also studied a cohort of patients with who had severe OH(> +2L).The Fresenius Body Composition Monitor was used to obtain hydration parameters. cT nT levels were done as part of routine clinical care. Data was analysed using SPSS version 20.0.RESULTS There were 48 deaths in the 336 patients. The patients that died from cardiac or non-cardiac causes were similar with respect to their age, incidence of diabetes mellitus, gender, ethnicity and cause of renal failure. However, the patients with cardiac causes of death had significantly shorter dialysis vintage(10.3 mo vs 37.0 mo, P < 0.0001) and were significantly more overhydrated by BCM measurement(2.95 L vs 1.35 L, P < 0.05). The mean(standard error of the means) hydration status of the 336 patients was +1.15(0.12) L and the median [interquartile range(IQR)] cT nT level was 43.5(20-90) ng/L. The cT nT results were not normally distributed and were therefore transformed logarithmically. There was a statistically significant correlation between Log(cT nT) with the OH value(Spearman r value 0.425, P < 0.0001). We identified a sub-group of patients that were severely overhydrated; median(IQR) hydration at baseline was +2.7(2.3 to 3.7) L. They were followed up for a minimum of 6 mo. Reduction in OH values in these patients over 6 mo correlated with lowering of cT nT levels(Spearman r value 0.29, P < 0.02). CONCLUSION Patients that were overhydrated had higher cT nT, and had deaths that were more likely to be cardiac related. 5Hepatic encephalopathy before and neurological complications after liver transplantation have no impact on the employment status 1 year after transplantation显示文摘AIM To investigate the impact of hepatic encephalopathybefore orthotopic liver transplantation(OLT) and neurological complications after OLT on employment after OLT.METHODS One hundred and fourteen patients with chronic liver disease aged 18-60 years underwent neurological examination to identify neurological complications, neuropsychological tests comprising the PSE-SyndromeTest yielding the psychometric hepatic encephalopathy score, the critical flicker frequency and the Repeatable Battery for the Assessment of Neuropsychological Status(RBANS), completed a questionnaire concerning their occupation and filled in the short form 36(SF-36) to assess health-related quality of life before OLT and 12 mo after OLT, if possible. Sixty-eight(59.6%) patients were recruited before OLT, while on the waiting list for OLT at Hannover Medical School [age: 48.7 ± 10.2 years, 45(66.2%) male], and 46(40.4%) patients were included directly after OLT. RESULTS Before OLT 43.0% of the patients were employed. The patients not employed before OLT were more often non-academics(employed: Academic/non-academic 16(34.0%)/31 vs not employed 10(17.6%)/52, P = 0.04), had more frequently a history of hepatic encephalopathy(HE)(yes/no; employed 15(30.6%)/34 vs not employed 32(49.2%)/33, P = 0.05) and achieved worse results in psychometric tests(RBANS sum score mean ± SD employed 472.1 ± 44.5 vs not employed 443.1 ± 56.7, P = 0.04) than those employed. Ten patients(18.2%), who were not employed before OLT, resumed work afterwards. The patients employed after OLT were younger [age median(range, min-max) employed 47(42, 18-60) vs not employed 50(31, 29-60), P = 0.01], achieved better results in the psychometric tests(RBANS sum score mean ± SD employed 490.7 ± 48.2 vs not employed 461.0 ± 54.5, P = 0.02) and had a higher health-related quality of life(SF 36 sum score mean ± SD employed 627.0 ± 138.1 vs not employed 433.7 ± 160.8; P < 0.001) compared to patients not employed after OLT. Employment before OLT(P < 0.001), age(P < 0.01) and SF-36 sum score 12 mo after OLT(P < 0.01) but not HE before OLT or neurological complications after OLT were independent predictors of the employment status after OLT.CONCLUSION HE before and neurological complications after OLT have no impact on the employment status 12 mo after OLT. 8Acute hepatitis induced by an Aloe vera preparation: A case report显示文摘AIM: Aloe vera, plant extracts of Aloe barbadensis miller,is widely used in phytomedicine. The first case of acute hepatitis due to this compound was described.METHODS: Description of a clinical case.RESULTS: Hepatitis in a 57-year old female could be linked to the ingestion of Aloe barbadensis miller compounds. 12Tapping the tree-ring archive for studying effects of resin extraction on the growth and climate sensitivity of Scots pine显示文摘Background: In the German Democratic Republic(GDR), resin tapping in Scots pine(Pinus sylvestris L.) forests was a major economic activity, and resin-tapped stands are frequently found up until this day. In this study, we investigate how the mechanical damage caused by resin tapping affects the growth and climate sensitivity of Scots pine using a dendroecological approach.Methods: Tree-ring samples were collected from resin-tapped and non-tapped trees in two forest areas in northeastern Germany, and tree-growth patterns were analyzed. For elucidating effects of resin tapping on the climate sensitivity of pine growth, climate-growth relationships and pointer years were studied.Results: We observed that resin tapping positively affects tree growth at breast height, likely as wood formation is concentrated on the living part of the bole(i.e. after tapping there is no growth taking place on the tapping face due to the mechanical damage done to the cambium). 14Sap flow for beech(Fagus sylvatica L.)in a natural and a managed forest—effect of spatial heterogeneity显示文摘Aims Beech(Fagus sylvatica L.)is an important species in natural and managed forests in Europe.This drought-sensitive species dominates even-aged stands as well more natural stands composed of a mixture of tree species,age and size classes.This study evaluates the extent that heterogeneity in spacing and tree diameter affect the seasonal availability and use of water.Methods Two stands were evaluated:(i)a heterogeneous forest remnant(NAT)with trees up to;300 years old,a mean top height of 28.4 m and a total of 733 stems ha1with stem diameters averaging 18 cm and(ii)an even-aged 80-year old stand(MAN),with a height of 25 m,and a total of 283 stems ha1 with diameters averaging 38 cm.Stem sap flow,Js(g m2 s1),was continuously measured in 12(MAN)and 13(NAT)trees using 20-mm long heat dissipation sensors.Individual tree measures of sap flow were correlated using non-linear statistical methods with air vapour pressure deficit(D,hPa)and global radiation(Rg,J m2 day1),along with constraints imposed by reductions in soil water content(SWC).SWC was measured as volumetric%using time domain reflectometry.Important Findings The daily integrated Js(Js-sum)for trees growing in the evenly spaced MAN stand and trees in canopy and closed forest positions in NAT stand decreased as the availability of soil moisture was reduced.In the heterogeneous NAT stand,SWC in a recently formed canopy gap remained high throughout the vegetation period.Based on regression models,the predicted relative decrease in Js-sum for dry relative to moist soil water conditions in the closed forest(at mean daily D=10 hPa)was 7–11%for trees near the gap and 39–42%for trees in the closed forest.In MAN,the reduction in Js-sum was 29%in dry relative to moist conditions.Js-sum in the outer 20 mm of the xylem in NAT was lower than that in MAN and the rate of decline in Js with xylem depth was less in NAT than in MAN.In MAN,Js-sum in deep and outer xylem was negatively affected at low soil moisture availability;in NAT,this was the case for only the outer xylem indicating that deep roots could be important in supplying water at times of low soil moisture in the upper soil.Lise Dalsgaard Teis Nørgaard Mikkelsen Annemarie Bastrup-Birk 2011Journal of Plant Ecology2011,4,1:2
