| 2 | Dihydromyricetin and Salvianolic acid B inhibit alpha-synuclein aggregation and enhance chaperone-mediated autophagy显示文摘Background:Progressive accumulation ofα-synuclein is a key step in the pathological development of Parkinson’s disease.Impaired protein degradation and increased levels ofα-synuclein may trigger a pathological aggregation in vitro and in vivo.The chaperone-mediated autophagy(CMA)pathway is involved in the intracellular degradation processes ofα-synuclein.Dysfunction of the CMA pathway impairsα-synuclein degradation and causes cytotoxicity.Results:In the present study,we investigated the effects on the CMA pathway andα-synuclein aggregation using bioactive ingredients(Dihydromyricetin(DHM)and Salvianolic acid B(Sal B))extracted from natural medicinal plants.In both cell-free and cellular models ofα-synuclein aggregation,after administration of DHM and Sal B,we observed significant inhibition ofα-synuclein accumulation and aggregation.Cells were co-transfected with a Cterminal modifiedα-synuclein(SynT)and synphilin-1,and then treated with DHM(10μM)and Sal B(50μM)16 hours after transfection;levels ofα-synuclein aggregation decreased significantly(68%for DHM and 75%for Sal B).Concomitantly,we detected increased levels of LAMP-1(a marker of lysosomal homeostasis)and LAMP-2A(a key marker of CMA).Immunofluorescence analyses showed increased colocalization between LAMP-1 and LAMP-2A withα-synuclein inclusions after treatment with DHM and Sal B.We also found increased levels of LAMP-1 and LAMP-2A both in vitro and in vivo,along with decreased levels ofα-synuclein.Moreover,DHM and Sal B treatments exhibited anti-inflammatory activities,preventing astroglia-and microglia-mediated neuroinflammation in BAC-α-syn-GFP transgenic mice.Conclusions:Our data indicate that DHM and Sal B are effective in modulatingα-synuclein accumulation and aggregate formation and augmenting activation of CMA,holding potential for the treatment of Parkinson’s disease. | Jia-Zhen Wu Mustafa Ardah Caroline Haikal Alexander Svanbergsson Meike Diepenbroek Nishant N.Vaikath Wen Li Zhan-You Wang Tiago F.Outeiro Omar M.El-Agnaf Jia-Yi Li | 2019 | Translational Neurodegeneration2019,8,1: | 11 |

| 5 | Age-Dependent Alpha-Synuclein Accumulation and Phosphorylation in the Enteric Nervous System in a Transgenic Mouse Model of Parkinson's Disease显示文摘The enteric nervous system(ENS) controls the function of the gastrointestinal tract and has been implicated in various diseases, including Parkinson's disease(PD). PD is a neurodegenerative disease with Lewy bodies(LBs) and Lewy neurites(LNs) as the main pathological features. In addition to the typical motor symptoms in PD,attention has been drawn to non-motor symptoms, such as constipation, implying dysfunction of the ENS. In the present study, we characterized the age-dependent morphological alterations and aggregation of a-synuclein(asyn), the primary protein component in LBs and LNs, in the ENS in an a-syn transgenic mouse model. We found that the expression and accumulation of a-syn increased gradually in neurons of Meissner's and Auerbach's plexuses of the gastrointestinal tract with age(from 1 week to 2 years). In addition, a-syn was increasingly phosphorylated at the serine 129 residue, reflecting pathological alterations of the protein over time. Furthermore, asyn was present in different subtypes of neurons expressing vasoactive intestinal polypeptide, neuronal nitric oxide synthase, or calretinin. | 6 | Microstructural evolution of a PM TiAl alloy during heat treatment in α+γ phase field显示文摘In this study, the effect of temperatures and cooling rates of heat treatment on the microstructure of a powder metallurgy (PM) Ti-46Al-2Cr-2Nb-(B,W) (at.%) alloy was studied. Depending on the cooling rate and temperature, the different structures were obtained from the initial near-γ (NG) microstructures by heat treatment in the α+γ field. The results show that the microstructures of samples after furnace cooling (FC) consist primarily of equiaxed γ and α 2 grains, with a few grains containing lamellae. Duplex microstructures consist mainly of γ grains and lamellar colonies were obtained in the quenching into another furnace at 900°C (QFC) samples. | 11 | Determination of basal ileal endogenous losses and standardized ileal digestibility of amino acids in barley fed to growing pigs显示文摘Background: Basal ileal endogenous amino acid(AA) losses(IAA_(end)) and standardized ileal digestibility(SID) values of cereal grains, such as barley, are apparently underestimated when determined according to the nitrogen(N)-free method. Regression analysis between the dietary apparent ileal digestible content(c AID) and total crude protein(CP) and AA can be considered as alternative approach to obtain more accurate values for IAA_(end)and SID of AA in cereal grains.Methods: Eight hulled barley genotypes were used, with barley being the only source of CP and AA in the assay diets. The diets contained 95 % as-fed of these eight barley genotypes each, ranging in CP content between 109.1 and 123.8 g/kg dry matter(DM). Nine ileally T-cannulated barrows, average body weight(BW) 30 ± 2 kg, were allotted to a row-column design comprising eight periods with 6 d each and nine pigs. On d 5 and the night of d 6 of every period, ileal digesta were collected for a total of 12 h. The IAA_(end) and the SID were determined by linear regression analysis between c AID and total dietary CP and AA.Results: There exist linear relationships between cA ID and total CP and AA(P < 0.001). The IAA_(end) of CP, Lys, Met, Thr and Trp amounted to 35.34, 1.08, 0.25, 1.02 and 0.38 g/kg DM intake(DMI), respectively, which are greater compared to average IAA_(end) determined previously under N-free feeding conditions. The SID of CP, Lys, Met, Thr and Trp was 90,79, 85, 79 and 86 %, respectively, and was greater when compared to tabulated values. Moreover, these SID values were greater than those reported in literature, based on correction of apparent ileal digestibility(AID) of CP and AA for their IAA_(end)values. | 15 | Influence of lighting environment on social preferences in sticklebacks from two different photic habitats.I.mate preferences of wild-caught females显示文摘Ultraviolet(UV)A signals(320-400 nm)are important in mate choice in numerous species.The sensitivity for UV signals is not only assumed to be costly,but also expected to be a function of the prevailing ecological conditions.Generally,those signals are favored by selection that efficiently reach the receiver.A decisive factor for color signaling is the lighting environment,especially in aquatic habitats,as the visibility of signals,and thus costs and benefits,are instantaneously influenced by it.Although ecological aspects of color signal evolution are relatively well-studied,there is little data on specific effects of environmental UV-light conditions on signaling at these shorter wavelengths.We studied wild-caught gravid female 3-spined sticklebacks Gasterosteus aculeatus of 2 photic habitat types(tea-stained and clear-water lakes),possessing great variation in their UV transmission.In 2 treatments,tea-stained and clear-water,preferences for males viewed under UV-present(UV-1-)and UV-absent(UV-)conditions were tested.A preference for males under UV+conditions was found for females from both habitat types,thus stressing the significance of UV signals in stickleback's mate choice decisions.However,females from both habitat types showed the most pronounced preferences for males under UV-h conditions under clear-water test conditions.Moreover,reflectance measurements revealed that the carotenoid-based orange-red breeding coloration in wild-caught males of both habitat types differed significantly in color intensity(higher in clear-water males)and hue(more red shifted in clear-water males)while no significant differences in UV coloration were found.The differential reflection patterns in longer wavelengths suggest that sticklebacks of both habitat types have adapted to the respective water conditions.Adaptations of UV signals in a sexual context to ambient light conditions in both behavior and coloration seem less evident. | Meike Hiermes Stephanie REHER Ingolf P.RICK Theo C.M.Bakker | 2021 | Current Zoology2021,67,3: | 2 |
