1A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide.Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen 2019Molecular Plant2019,12,5:18
2The apricot(Prunus armeniaca L.)genome elucidates Rosaceae evolution and beta-carotenoid synthesis显示文摘Apricots,scientifically known as Prunus armeniaca L,are drupes that resemble and are closely related to peaches or plums.As one of the top consumed fruits,apricots are widely grown worldwide except in Antarctica.A high-quality reference genome for apricot is still unavailable,which has become a handicap that has dramatically limited the elucidation of the associations of phenotypes with the genetic background,evolutionary diversity,and population diversity in apricot.DNA from P.armeniaca was used to generate a standard,size-selected library with an average DNA fragment size of~20 kb.The library was run on Sequel SMRT Cells,generating a total of 16.54 Gb of PacBio subreads(N50=13.55 kb).The high-quality P.armeniaca reference genome presented here was assembled using long-read single-molecule sequencing at approximately 70×coverage and 171×Illumina reads(40.46 Gb),combined with a genetic map for chromosome scaffolding.The assembled genome size was 221.9 Mb,with a contig NG50 size of 1.02 Mb.Scaffolds covering 92.88%of the assembled genome were anchored on eight chromosomes.Benchmarking Universal Single-Copy Orthologs analysis showed 98.0%complete genes.We predicted 30,436 protein-coding genes,and 38.28%of the genome was predicted to be repetitive.We found 981 contracted gene families,1324 expanded gene families and 2300 apricot-specific genes.The differentially expressed gene(DEG)analysis indicated that a change in the expression of the 9-cis-epoxycarotenoid dioxygenase(NCED)gene but not lycopene beta-cyclase(LcyB)gene results in a lowβ-carotenoid content in the white cultivar“Dabaixing”.This complete and highly contiguous P.armeniaca reference genome will be of help for future studies of resistance to plum pox virus(PPV)and the identification and characterization of important agronomic genes and breeding strategies in apricot.Fengchao Jiang Junhuan Zhang Sen Wang Li Yang Yingfeng Luo Shenghan Gao Meiling Zhang Shuangyang Wu Songnian Hu Haoyuan Sun Yuzhu Wang 2019Horticulture Research2019,6,1:9
3The potential value of microRNA-4463 in the prognosis evaluation in hepatocellular carcinoma显示文摘The purpose of this study is to measure the expression of microRNA-4463 and microRNA-6087 between normal persons and patients with hepatocellular carcinoma(HCC),and to clarify the meaning of them in the prognosis evaluation in HCC.Forty-five samples from healthy people and patients,who had been diagnosed with hepatocellular carcinoma before any treatment,were collected to study respectively.Real-time PCR was used to detect the expression of miRNA-4463 and miRNA-6087 in the serum of control group and hepatocellular carcinoma patients.The expression of miR-4463 in the serum of HCC patients was significantly higher than that in control group(P<0.05),and the expression level was independent of gender,tumor size,cell types,stages,alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL)and HBsAg status(P>0.05).But there was a significant difference of different level of AFP in HCC(P<0.05),and the difference between the group of AFP lower than 400 ug/l and the control group is statistically significant(P<0.05).Besides,the survival time had showed a significant difference at the high and low expression levels(P<0.05).But the expression level of miRNA-6087 was no difference in HCC and control group.The disorder of miRNA-4463 occurred in HCC,even the AFP level doesn’t rises.What’s more,patients who get the high level of miRNA-4463 seem to have a shorter survival time.And it contributes great to the prognostic evaluation.This is the first study to illustrate the potential significance of miRNA-4463 in the prognosis in HCC.Tian Hu Jincheng Li Chuhong Zhang Xiu lv Sai Li Sha He Hanxing Yan Yixi Tan Mingsheng Lei Meiling Wen Jianhong Zuo 2017Genes & Diseases2017,4,2:3

10Cytotoxic effects of docetaxel as a candidate drug of drug-eluting stent on human umbilical vein endothelial cells and the signaling pathway of cell migration inhibition,adhesion delay and shape change显示文摘Docetaxel(DTX),a paclitaxel analogue,can efficiently inhibit proliferation of vascular smooth muscle cells and has broadly been used as an antiangiogenesis drug.However,as a candidate drug of drug-eluting stent,the effects of DTX on human umbilical vein endothelial cells(HUVECs)are still not well understood.Herein,we investigated the effects of DTX on proliferation,apoptosis,adhesion,migration and morphology of HUVECs in vitro.We found that DTX had the cytostatic and cytotoxic effects at low and high concentrations,respectively.DTX could inhibit the proliferation and migration of HUVECs,induce HUVECs apoptosis,delay HUVECs adhesion and decrease spreading area and aspect ratio of individual cells.The signaling pathway that DTX led to the migration inhibition,adhesion delay and shape change of HUVECs is the VE-cadherin mediated integrin b1/FAK/ROCK signaling pathway.The study will provide a theoretical basis for the clinical application of DTX.Tingzhang Hu Chun Yang Meiling Fu Jiali Yang Rolin Du Xiaolin Ran Tieying Yin Guixue Wang 2017Regenerative Biomaterials2017,4,3:1

12Trustworthy Service Selection Integrating Cloud Model and Possibility Degree Ranking of Interval Numbers显示文摘It has been challenging to select suitable services from abundant candidates in cloud environment.Aiming to the characteristics of batch computing mode and stream computing mode, a novel trustworthy service selection approach is proposed integrating cloud model and interval numbers theory. To facilitate potential users to understand the quality of service, the trustworthiness of service is described with interval number using reverse cloud generator, and the services with poor performance are filtered out by employing deviation degree or proximity degree. Two formulas of possibility degree of interval numbers are designed to compare the trustworthiness values between cloud services by utilizing probability zone analysis and geometrical analysis respectively, and the ranking method for possibility degree of interval numbers is exploited to select the most trustworthy service. 13A tactical nanomissile mobilizing antitumor immunity enables neoadjuvant chemo-immunotherapy to minimize postsurgical tumor metastasis and recurrence显示文摘Neoadjuvant chemotherapy has become an indispensable weapon against high-risk resectable cancers,which benefits from tumor downstaging.However,the utility of chemotherapeutics alone as a neoadjuvant agent is incapable of generating durable therapeutic benefits to prevent postsurgical tumor metastasis and recurrence.Herein,a tactical nanomissile(TALE),equipped with a guidance system(PD-L1 monoclonal antibody),ammunition(mitoxantrone,Mit),and projectile bodies(tertiary amines modified azobenzene derivatives),is designed as a neoadjuvant chemo-immunotherapy setting,which aims at targeting tumor cells,and fast-releasing Mit owing to the intracellular azoreductase,thereby inducing immunogenic tumor cells death,and forming an in situ tumor vaccine containing damage-associated molecular patterns and multiple tumor antigen epitopes to mobilize the immune system.The formed in situ tumor vaccine can recruit and activate antigen-presenting cells,and ultimately increase the infiltration of CD8^(+)T cells while reversing the immunosuppression microenvironment.Moreover,this approach provokes a robust systemic immune response and immunological memory,as evidenced by preventing 83.3%of mice from postsurgical metastasis or recurrence in the B16-F10 tumor mouse model.Collectively,our results highlight the potential of TALE as a neoadjuvant chemo-immunotherapy paradigm that can not only debulk tumors but generate a long-term immunosurveillance to maximize the durable benefits of neoadjuvant chemotherapy.Tao He Mingxing Hu Shunyao Zhu Meiling Shen Xiaorong Kou Xiuqi Liang Lu Li Xinchao Li Miaomiao Zhang Qinjie Wu Changyang Gong 2023Acta Pharmaceutica Sinica B2023,13,2:0

16Human 8-cell embryos enable efficient induction of disease-preventive mutations without off-target effect by cytosine base editor显示文摘Approximately 140 million people worldwide are homozygous carriers of APOE4(ε4),a strong genetic risk factor for late onset familial and sporadic Alzheimer’s disease(AD),91%of whom will develop AD at earlier age than heterozygous carriers and noncarriers.Susceptibility to AD could be reduced by targeted editing of APOE4,but a technical basis for controlling the off-target effects of base editors is necessary to develop low-risk personalized gene therapies.Here,we first screened eight cytosine base editor variants at four injection stages(from 1-to 8-cell stage),and found that FNLS-YE1 variant in 8-cell embryos achieved the comparable base conversion rate(up to 100%)with the lowest bystander effects.In particular,80%of AD-susceptibleε4 allele copies were converted to the AD-neutralε3 allele in humanε4-carrying embryos.Stringent control measures combined with targeted deep sequencing,whole genome sequencing,and RNA sequencing showed no DNA or RNA off-target events in FNLS-YE1-treated human embryos or their derived stem cells.Furthermore,base editing with FNLS-YE1 showed no effects on embryo development to the blastocyst stage.Finally,we also demonstrated FNLS-YE1 could introduce known protective variants in human embryos to potentially reduce human susceptivity to systemic lupus erythematosus and familial hypercholesterolemia.Our study therefore suggests that base editing with FNLS-YE1 can efficiently and safely introduce known preventive variants in 8-cell human embryos,a potential approach for reducing human susceptibility to AD or other genetic diseases.Yinghui Wei Meiling Zhang Jing Hu Yingsi Zhou Mingxing Xue Jianhang Yin Yuanhua Liu Hu Feng Ling Zhou Zhifang Li Dongshuang Wang Zhiguo Zhang Yin Zhou Hongbin Liu Ning Yao Erwei Zuo Jiazhi Hu Yanzhi Du Wen Li Chunlong Xu Hui Yang 2023Protein & Cell2023,14,6:0

Severe jaundice can cause neonatal bilirubin encephalopathy. Clinically because severe hyperbilirubinemia more causes, the cause of hemolysis and infection is more common, this also accounted for a certain proportion of unknown reasons. UGT1A1 is a key enzyme for bilirubin metabolism, and unexplained jaundice is associated with a decrease in UGT1A1 activity caused by mutations in UGT1A1 gene, leading to neonatal persistent jaundice. 19Decision Difficulty, Work-Family Conflict, and Social Support for Chinese Nurses in Guangzhou after the Two-Child Policy显示文摘Background:AlthoughChina's Mainland implemented a two-child policy (every legally married may have two children) on January 1, 2016, women in China are becoming increasingly cautious in their decision-making about family planning, due to work-family conflicts and a lack of social support. Objective:The aim of this study was to investigate Chinese nurses' intents to have a second child, and to explore the relationship between decision making difficulties in terms of childbearing intents, work-family conflict, and social support for Chinese nurses. Methods:Data were collected at three different hospital levels in four districts of Guangzhou, from 711 nurses of childbearing age (18-50 years) who had had at least one child. A structured questionnaire was used, composed of the Decisional Conflict Scale (DCS), Work-Family Conflict Scale (WFCS), and Scale of Perceived Social Support (SPSS). Results:Participants’ mean age was 33.6 years (18-50). The mean scores of decisional conflict by DCS, work-family conflict by WFCS and perceived social support by SPSS were 40.9 (SD = 14.8), 55.83 (SD = 15.18) and 57.85 (SD = 15.77), respectively. Work-family conflict was positively correlated with decisional conflict (P < 0.01), and social support was negatively related to decisional conflict (P < 0.01). By logistic regression analysis, family interference with work (FIW), especially time-based FIW, was a significant predictor of nurses having decisional conflicts in their childbearing intents (OR=1.083, 95% CI: 1.045 to 1.123). Perceived social support was another significant predictor of decisional difficulty (OR=0.981, 95% CI: 0.970 to 0.992). Discussion:This study found that female nurses, working night shifts and with temporary job contract had higher decisional conflicts in intents of having a second child. The results showed that for Chinese nurses, work-family conflict influences decisional conflicts about having a second child. 20Clinical efficacy and safety of linezolid in intensive care unit patients显示文摘Background:To characterize the population of critically ill patients and infections treated with linezolid in the intensive care unit(ICU),and to evaluate the clinical efficacy and safety of linezolid therapy.Methods::This multi-center,observational,real-world study was conducted across 52 hospitals between June 9,2018,and December 28,2019.Patients who met the following inclusion criteria were included:(1)admitted to the ICU,(2)of any age group,and(3)having a clinical or laboratory diagnosis of a Gram-positive bacterial infection.Clinical efficacy was categorized as success(cured or improved),failed,or non-evaluable.Adverse events and serious adverse events were recorded during treatment.Results::A total of 366 ICU patients who met the inclusion criteria were evaluated.Linezolid was used as second-and first-line treatment in 232(63.4%)and 134(36.6%)patients,respectively.The most common isolated strain was Staphylococcus aureus(methicillin-resistant Staphylococcus aureus:n=37/119,31.1%;methicillin-susceptible Staphylococcus aureus:n=15/119,12.6%);this was followed by Enterococci(vancomycin-resistant Enterococci:n=8/119,6.7%;vancomycin-susceptible Enterococci:n=11/119,9.2%)and Streptococcus pneumoniae(multidrug-resistant:n=4/119,3.4%;non-multidrug resistant:n=2/119,1.7%).The main infection sites where pathogens were detected included the lung(n=216/366,59.6%),skin and soft tissue(n=104/366,28.4%),and blood(n=50/366,13.7%).Clinical success was achieved in 301(82.2%)patients;34(9.3%)were cured and 267(73.0%)improved;treatment failure and non-evaluable outcomes were observed in 29(7.9%)in 36(9.8%)patients,respectively.Linezolid-related adverse events were reported in 8(2.2%)patients.No treatment-related serious adverse events were reported.Conclusions::Based on real-world results,linezolid was found to be effective and safe in the treatment of Gram-positive bacterial infections in critically ill patients.Aijia Ma Meiling Dong Jiangli Cheng Xuelian Liao Wei Dong Chang Liu Chenggong Hu Jing Yang Yan Kang 2023Journal of Intensive Medicine2023,3,1:0
