| 1 | A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide. | Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen | 2019 | Molecular Plant2019,12,5: | 18 |

On the basis of systematic analysis of ten thousands primary optical signals at the corresponding original positions, the quantitative statistic distribution information of each element was obtained. The biggest degrees of segregation of low alloy steel continuous casting billet were calculated accurately according to the quantitative distribution maps of the contents. It was suggested that the weight ratio in a certain content range was used to judge the homogeneity of the materials, and the two models —— the total weight ratio of contents (the degree of statistic homogeneity, H) within the permissive content range (C0±R) and the median value confidence extension ratio (the degree of statistic segregation, S) at 95% of confidence limit of weight ratio —— were put forward. The two models reflect the composition and state distribution regularity of the metal materials in a large region. | 8 | The effects of altered BMP4 signaling in first branchialarch- derived murine embryonic orofacial tissues显示文摘The first branchial arch(BA1),which is derived from cranial neural crest(CNC)cells,gives rise to various orofacial tissues.Cre mice are widely used for the determination of CNC and exploration of gene functions in orofacial development.However,there is a lack of Cre mice specifically marked BA1’s cells.Pax2-Cre allele was previously generated and has been widely used in the field of inner ear development.Here,by compounding Pax2-Cre and R26R-mTmG mice,we found a specific expression pattern of Pax2^(+)cells that marked BA1’s mesenchymal cells and the BA1-derivatives.Compared to Pax2-Cre;R26R-mTmG allele,GFP^(+)cells were abundantly found both in BA1 and second branchial arch in Wnt1-Cre;R26R-mTmG mice.As BMP4 signaling is required for orofacial development,we over-activated Bmp4 by using Pax2-Cre;pMes-BMP4 strain.Interestingly,our results showed bilateral hyperplasia between the upper and lower teeth.We also compare the phenotypes of Wnt1-Cre;pMes-BMP4 and Pax2-Cre;pMes-BMP4 strains and found severe deformation of molar buds,palate,and maxilla-mandibular bony structures in Wnt1-Cre;pMes-BMP4 mice;however,the morphology of these orofacial organs were comparable between controls and Pax2-Cre;pMes-BMP4 mice except for bilateral hyperplastic tissues.We further explore the properties of the hyperplastic tissue and found it is not derived from Runx2^(+)cells but expresses Msx1,and probably caused by abnormal cell proliferation and altered expression pattern of p-Smad1/5/8.In sum,our findings suggest altering BMP4 signaling in BA1-specific cell lineage may lead to unique phenotypes in orofacial regions,further hinting that Pax2-Cre mice could be a new model for genetic manipulation of BA1-derived organogenesis in the orofacial region. | Jue Xu Meiling Chen Yanan Yan Qiaoxue Zhao Meiying Shao Zhen Huang | 2021 | International Journal of Oral Science2021,13,4: | 3 |
| 9 | Genetic Analysis and Mapping of the Purple Gene in Purple Heading Chinese Cabbage显示文摘To analyze genetic linkage and map purple gene(Br Pur) controlling purple inner leaves trait of Chinese cabbage, a F2 population was constructed by selfing a F1 plant from homozygous purple heading line ‘14S839' and homozygous orange heading line ‘14S162'. The phenotype investigation of head color showed that the segregation ratio of purple to non-purple individuals was consistent with the expected ratio of 3:1,which indicated that purple inner leaves trait is controlled by a single dominant gene. A total of 297 SSRs in whole genome of Chinese cabbage were tested by modified Bulked Segregant Analysis(BSA) method. Two linked markers flanking Br Pur, A710 and A714, were obtained and Br Pur was mapped on linkage group A07 based on the sequence of these two markers. Subsequently, two new markers flanking Br Pur, CL-12 and B214-87, were developed based on two known anthocyanins-related genes, Br4CL3 and Bra004214. However, the traditional carburizing is limited by poor microstructure distribution and low rate of carburizing. The rare earth ion implantation is known to help improving the properties of tribology, corrosion resistance and oxidation resistance of metal. In this article, the RE implantation is employed to assist the carburizing. Lanthanum and cerium ion implantations are initially used to assist 12 Cr2 Ni4 A low pressure vacuum carburization.The microstructure, content of retained austenite, hardness, thickness of layer and carbon diffusion were analyzed by optical microscopy(OM), scanning electron microscopy(SEM), X-ray diffraction(XRD) and Rockwell/Vickers hardness tester, respectively. It was shown that lanthanum and cerium implantations can improve structure of the vacuum carburizing layer, and enhance the uniformity of carbon element distribution on the carburized surface. Meanwhile the RE implantation plays a positive role in promoting the surface hardness and carburized rate. The lanthanum element has more significant effect on surface hardness and content of retained austenite than cerium element. As the microporous structure of the carbon substrate is generally regarded as the active site hosts, its hostility to electron/mass transfer could lead to the incomplete fulfillment of the catalytic activity. Besides, the formation of inactive metallic Fe particles during the conventional catalyst synthesis could also decrease the active site density and complicate the identification of real active site. Herein, we developed a facial hydrogen etching methodology to yield single site Fe-N-C catalysts featured with micro/mesoporous hierarchical structure. The hydrogen concentration in pyrolysis process was designated to effectively regulate the pore structure and active site density of the resulted catalysts.The optimized sample achieves excellent ORR catalytic performance with an ultralow H2O2 yield(1%)and superb stability over 10,000 cycles.
