1The potential value of microRNA-4463 in the prognosis evaluation in hepatocellular carcinoma显示文摘The purpose of this study is to measure the expression of microRNA-4463 and microRNA-6087 between normal persons and patients with hepatocellular carcinoma(HCC),and to clarify the meaning of them in the prognosis evaluation in HCC.Forty-five samples from healthy people and patients,who had been diagnosed with hepatocellular carcinoma before any treatment,were collected to study respectively.Real-time PCR was used to detect the expression of miRNA-4463 and miRNA-6087 in the serum of control group and hepatocellular carcinoma patients.The expression of miR-4463 in the serum of HCC patients was significantly higher than that in control group(P<0.05),and the expression level was independent of gender,tumor size,cell types,stages,alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL)and HBsAg status(P>0.05).But there was a significant difference of different level of AFP in HCC(P<0.05),and the difference between the group of AFP lower than 400 ug/l and the control group is statistically significant(P<0.05).Besides,the survival time had showed a significant difference at the high and low expression levels(P<0.05).But the expression level of miRNA-6087 was no difference in HCC and control group.The disorder of miRNA-4463 occurred in HCC,even the AFP level doesn’t rises.What’s more,patients who get the high level of miRNA-4463 seem to have a shorter survival time.And it contributes great to the prognostic evaluation.This is the first study to illustrate the potential significance of miRNA-4463 in the prognosis in HCC.Tian Hu Jincheng Li Chuhong Zhang Xiu lv Sai Li Sha He Hanxing Yan Yixi Tan Mingsheng Lei Meiling Wen Jianhong Zuo 2017Genes & Diseases2017,4,2:3

4CHRT:Clustering-Based Hybrid Re-Routing System for Traffic Congestion Avoidance显示文摘Re-routing system has become an important technology to improve traffic efficiency.The traditional re-routing schemes do not consider the dynamic characteristics of urban traffic,making the planned routes unable to cope with the changing traf-fic conditions.Based on real-time traffic information,it is challenging to dynamically re-route connected vehicles to alleviate traffic congestion.Moreover,how to obtain global traffic information while reducing communication costs and improving travel efficiency poses a challenge to the re-routing system.To deal with these challenges,this paper proposes CHRT,a clustering-based hybrid re-routing system for traffic congestion avoidance.CHRT develops a multi-layer hybrid architecture.The central server accesses the global view of traffic,and the distributed part is composed of vehicles divided into clusters to reduce latency and communication overhead.Then,a clustering-based priority mechanism is proposed,which sets priorities for clusters based on realtime traffic information to avoid secondary congestion.Furthermore,to plan the optimal routes for vehicles while alleviating global traffic congestion,this paper presents a multi-metric re-routing algorithm.Through extensive simulations based on the SUMO traffic simulator,CHRT reduces vehicle traveling time,fuel consumption,and CO2 emissions compared to other systems.In addition,CHRT globally alleviates traffic congestion and improves traffic efficiency.Jie Huo Xiangming Wen Luning Liu Luhan Wang Meiling Li Zhaoming Lu 2021China Communications2021,18,7:2

6CRISPR/Cas9 screening identifies a kinetochore-microtubule dependent mechanism for Aurora-A inhibitor resistance in breast cancer显示文摘Background:Overexpression of Aurora-A(AURKA)is a feature of breast cancer and associates with adverse prognosis.The selective Aurora-A inhibitor alisertib(MLN8237)has recently demonstrated promising antitumor responses as a single agent in various cancer types but its phase III clinical trial was reported as a failure since MLN8237 did not show an apparent effect in prolonging the survival of patients.Thus,identification of potential targets that could enhance the activity of MLN8237 would provide a rationale for drug combination to achieve better therapeutic outcome.Methods:Here,we conducted a systematic synthetic lethality CRISPR/Cas9 screening of 507 kinases using MLN8237 in breast cancer cells and identified a number of targetable kinases that displayed synthetic lethality interactions with MLN8237.Then,we performed competitive growth assays,colony formation assays,cell viability assays,apoptosis assays,and xenograft murine model to evaluate the synergistic therapeutic effects of Haspin(GSG2)depletion or inhibition with MLN8237.For mechanistic studies,immunofluorescence was used to detect the state of microtubules and the localization of Aurora-B and mitotic centromere-associated kinesin(MCAK).Results:Among the hits,we observed that Haspin depletion or inhibition marginally inhibited breast cancer cell growth but could substantially enhance the killing effects of MLN8237.Mechanistic studies showed that co-treatment with Aurora-A and Haspin inhibitors abolished the recruitment of Aurora-B and mitotic centromere-associated kinesin(MCAK)to centromeres which were associated with excessive microtubule depolymerization,kinetochore-microtubule(KT-MT)attachment failure,and severe mitotic catastrophe.We further showed that the combination of MLN8237 and the Haspin inhibitor CHR-6494 synergistically reduced breast cancer cell viability and significantly inhibited both in vitro and in vivo tumor growth.Conclusions:These findings establish Haspin as a synthetic lethal target and demonstrate CHR-6494 as a potential combinational drug for promoting the therapeutic effects of MLN8237 on breast cancer.Ailin Chen Shijun Wen Fang Liu Zijian Zhang Meiling Liu Yuanzhong Wu Bin He Min Yan Tiebang Kang Eric W-F Lam Zifeng Wang Quentin Liu 2021Cancer Communications2021,41,2:1

9High coercivity Nd-Ce-Fe-B nanostructured ribbons prepared from melt spinning technique显示文摘The Ce-substituted(Nd1-xCex)12.2 Fe81.6 B6.2(x=0.0, 0.2, 0.4, 0.6) nanocrystalline ribbons were prepared by annealing amorphous ribbons from melt spinning. It is found that all ribbons are in a multiphase state consisting of a-Fe phase, Nd(Ce)-rich phases and RE2 Fe14 B(RE = Nd, Ce) phases. However, the coercivity of all annealed ribbons can reach a considerably high value without doping any heavy rare earth or other coercivity enhanced elements. A strong intergranular exchange coupling appears in these nanocrystalline ribbons. The Nd12.2 Fe81.6 B6.2 ribbons with multiphase have a coercivity of about 11.3 k Oe, and the coercivity decreases slightly with increasing Ce content. A coercivity of 7.5 kOe can be obtained when60 at% of Nd is replaced by Ce(x = 0.6) due to the grain refinement and the strong intergranular exchange coupling. 10Mechanical performance and cyocompatibility of PU/PLCL nanofibrous electrospun scaffolds for skin regeneration显示文摘Skin tissue engineering with considerable skin regeneration capability is an urgent need for the wound site.The current challenge for researchers is to develop a bionic scaffold that imitates the extracellular matrix for the regeneration of the damaged regions.In our study,poly(L-lactide-co-caprolactone)(PLCL)was blended with polyurethane(PU)to obtain nanofibrous scaffolds via electrospinning.The electrospun fibers with 50%PLCL content had a certain number of intersections and jointing points,and exhibited significantly enhanced mechani-cal properties combined with suitable porosity.Moreover,cell activities demonstrated that PU/PLCL membranes had significantly biological advantages in enhanced growth of human skin fibroblasts with spreading morphology compared with PU membranes,indicating good cytocompatibility of composite scaffolds.These findings proved that PU/PLCL electrospun membranes have great potential in applications of skin tissue engineering.Xiang Gao Meiling Wen Yang Liu Tian Hou Meiwen An 2022Engineered Regeneration2022,3,1:1

12Human 8-cell embryos enable efficient induction of disease-preventive mutations without off-target effect by cytosine base editor显示文摘Approximately 140 million people worldwide are homozygous carriers of APOE4(ε4),a strong genetic risk factor for late onset familial and sporadic Alzheimer’s disease(AD),91%of whom will develop AD at earlier age than heterozygous carriers and noncarriers.Susceptibility to AD could be reduced by targeted editing of APOE4,but a technical basis for controlling the off-target effects of base editors is necessary to develop low-risk personalized gene therapies.Here,we first screened eight cytosine base editor variants at four injection stages(from 1-to 8-cell stage),and found that FNLS-YE1 variant in 8-cell embryos achieved the comparable base conversion rate(up to 100%)with the lowest bystander effects.In particular,80%of AD-susceptibleε4 allele copies were converted to the AD-neutralε3 allele in humanε4-carrying embryos.Stringent control measures combined with targeted deep sequencing,whole genome sequencing,and RNA sequencing showed no DNA or RNA off-target events in FNLS-YE1-treated human embryos or their derived stem cells.Furthermore,base editing with FNLS-YE1 showed no effects on embryo development to the blastocyst stage.Finally,we also demonstrated FNLS-YE1 could introduce known protective variants in human embryos to potentially reduce human susceptivity to systemic lupus erythematosus and familial hypercholesterolemia.Our study therefore suggests that base editing with FNLS-YE1 can efficiently and safely introduce known preventive variants in 8-cell human embryos,a potential approach for reducing human susceptibility to AD or other genetic diseases.Yinghui Wei Meiling Zhang Jing Hu Yingsi Zhou Mingxing Xue Jianhang Yin Yuanhua Liu Hu Feng Ling Zhou Zhifang Li Dongshuang Wang Zhiguo Zhang Yin Zhou Hongbin Liu Ning Yao Erwei Zuo Jiazhi Hu Yanzhi Du Wen Li Chunlong Xu Hui Yang 2023Protein & Cell2023,14,6:0

14Effect of hydrogen impurities on hydrogen oxidation activity of Pt/C catalyst in proton exchange membrane fuel cells显示文摘High-purity of hydrogen is vital to the guarantee of end usage in proton exchange membrane fuel cell(PEMFC)electric vehicles(EVs)with superior durability and low expense.However,the currently employed hydrogen,primarily from fossil fuel,still contains some poisoning impurities that significantly affect the durability of PEMFCs.Here,we investigate the poisoning effect of several typical hydrogen impurities(S^(2-),Cl^(-),HCOO^(-)and CO_(3)^(2-))on the hydrogen oxidation reaction(HOR)of the state-of-the-art carbon-supported platinum(Pt/C)catalyst used in the PEMFC anode.Electrochemical results indicate that the electrochemically active surface area of Pt/C is hampered by these hydrogen impurities with reduced effective Pt reactive sites due to the competitive adsorption against hydrogen at Pt sites showing the extent of the poisoning on Pt sites in the order:S^(2-)>Cl^(-)>HCOO^(-)>CO_(3)^(2-).Density functional theory calculations reveal that the adsorption energy of S2-on Pt(111)is greater than that of Cl^(-),HCOO^(-)and CO_(2),and the electronic structure of Pt is found to be changed due to the adsorption of impurities showing the downshift of the d-band centre of Pt that weakens the adsorption of hydrogen on the Pt sites.This work provides valuable guidance for future optimization of hydrogen quality and also emphasizes the importance of anti-poisoning anode catalyst development,especially towards H_(2)S impurities that seriously affect the durability of PEMFCs.Yufeng Qin Chaoyong Sun Shaojie Ke Rou Wen Yifan Wang Jing Ji Meiling Dou 2023Clean Energy2023,7,1:0
15A preliminary study on 3D position reconstruction ofmonolithic crystal readout显示文摘Purpose Compared to pixelated crystal detectors,monolithic crystal detectors present more advantages.Without reflective layer between crystal pixel,monolithic crystal detectors provide higher detection efficiency while being cheaper.Another important feature is that monolithic crystal detectors can provide depth of interaction(DOI)information,which can improve image quality in nuclear imaging systems.In this article,we aim to study a new positioning scheme of this kind of detector in order to push its applications in nuclear imaging systems.Methods Fan-beam collimation and convolutional neural network are combined to reconstruct interaction points in crystal.Firstly,we built a monolithic detector model based on Geant4 and validated this positioning scheme with simulation data.Then,we constructed a monolithic GAGG(Ce)detector and evaluated its performance with experimental data.Results With proposed positioning scheme,in experiment,we obtained~1.7 mm FWHM resolution on average in x-/ydirection,while~2.5 mm FWHM resolution on average in DOI direction for a 33×33×10 mm3 monolithic GAGG(Ce)crystal coupled with 8×8 SiPM array.Conclusion Through simulations and experiments,we validated our positioning scheme,i.e.,fan-beam collimation and convolutional neural network.Convolutional neural network can reconstruct 3D positions of gamma-rays interaction points in a high resolution,and fan-beam collimation can provide high calibration efficiency;with combination of them,we established a high position resolution and high calibration efficiency positioning scheme for monolithic crystal detector.Wen He Xianchao Huang Yingjie Wang Baotong Feng Meiling Zhu Shuangquan Liu Zhiming Zhang Long Wei 2021Radiation Detection Technology and Methods2021,5,1:0
16Histone lactylation driven by mROS-mediated glycolytic shift promotes hypoxic pulmonary hypertension显示文摘Increased mitochondrial reactive oxygen species(mROS)and glycolysis have been established in pulmonary hypertension(PH).However,the effect of elevated mROS on glycolytic shift and how increased glycolysis promotes hypoxic pulmonary artery smooth muscle cell(PASMC)proliferation and vascular remodeling remain elusive.Here,we reported that hypoxia-induced mROS inhibit HIF-1αhydroxylation and further trigger PASMC glycolytic switch through the upregulated HIF-1α/PDK1&PDK2/p-PDH-E1αaxis,which facilitates lactate accumulation and histone lactylation.Through H3K18la and HIF-1αChIP–seq analysis,we found that the enhanced histone lactylation of HIF-1αtargets,such as Bmp5,Trpc5,and Kit,promotes PASMC proliferation.Knockdown of Pdk1&2 blunts lactate production,histone lactylation marks,and PASMC proliferation.Moreover,pharmacological intervention with lactate dehydrogenase inhibitor diminishes histone lactylation and ameliorates PASMC proliferation and vascular remodeling in hypoxic PH rats.Taken together,this study provides proof of concept for anti-remodeling therapy through lactate manipulation.Jian Chen Meiling Zhang Yanjie Liu Shihong Zhao Yanxia Wang Meng Wang Wen Niu Faguang Jin Zhichao Li 2022Journal of Molecular Cell Biology2022,14,12:0
