3Chromosome-scale genome assembly provides insights into the evolution and flavor synthesis of passion fruit (Passiflora edulis Sims)显示文摘Passion fruit(Passiflora edulis Sims)is an economically valuable fruit that is cultivated in tropical and subtropical regions of the world.Here,we report an~1341.7Mb chromosome-scale genome assembly of passion fruit,with 98.91%(~1327.18Mb)of the assembly assigned to nine pseudochromosomes.The genome includes 23,171 protein-coding genes,and most of the assembled sequences are repetitive sequences,with long-terminal repeats(LTRs)being the most abundant.Phylogenetic analysis revealed that passion fruit diverged after Brassicaceae and before Euphorbiaceae.Ks analysis showed that two whole-genome duplication events occurred in passion fruit at 65 MYA and 12 MYA,which may have contributed to its large genome size.An integrated analysis of genomic,transcriptomic,and metabolomic data showed that‘alpha-linolenic acid metabolism’,‘metabolic pathways’,and‘secondary metabolic pathways’were the main pathways involved in the synthesis of important volatile organic compounds(VOCs)in passion fruit,and this analysis identified some candidate genes,including GDP-fucose Transporter 1-like,Tetratricopeptide repeat protein 33,protein NETWORKED 4B isoform X1,and Golgin Subfamily A member 6-like protein 22.In addition,we identified 13 important gene families in fatty acid pathways and eight important gene families in terpene pathways.Gene family analysis showed that the ACX,ADH,ALDH,and HPL gene families,especially ACX13/14/15/20,ADH13/26/33,ALDH1/4/21,and HPL4/6,were the key genes for ester synthesis,while the TPS gene family,especially PeTPS2/3/4/24,was the key gene family for terpene synthesis.This work provides insights into genome evolution and flavor trait biology and offers valuable resources for the improved cultivation of passion fruit.Zhiqiang Xia Dongmei Huang Shengkui Zhang Wenquan Wang Funing Ma Bin Wu Yi Xu Bingqiang Xu Di Chen Meiling Zou Huanyu Xu Xincheng Zhou Rulin Zhan Shun Song 2021Horticulture Research2021,8,1:15
4The function of small RNAs in plant biotic stress response显示文摘Small RNAs(s RNAs) play essential roles in plants upon biotic stress. Plants utilize RNA silencing machinery to facilitate pathogen-associated molecular pattern-triggered immunity and effector-triggered immunity to defend against pathogen attack or to facilitate defense against insect herbivores. Pathogens, on the other hand, are also able to generate effectors and s RNAs to counter the host immune response. The arms race between plants and pathogens/insect herbivores has triggered the evolution of s RNAs,RNA silencing machinery and pathogen effectors. A great number of studies have been performed to investigate the roles of s RNAs in plant defense, bringing in the opportunity to utilize s RNAs in plant protection. Transgenic plants with pathogen-derived resistance ability or transgenerational defense have been generated, which show promising potential as solutions for pathogen/insect herbivore problems in the field. 6The effects of altered BMP4 signaling in first branchialarch- derived murine embryonic orofacial tissues显示文摘The first branchial arch(BA1),which is derived from cranial neural crest(CNC)cells,gives rise to various orofacial tissues.Cre mice are widely used for the determination of CNC and exploration of gene functions in orofacial development.However,there is a lack of Cre mice specifically marked BA1’s cells.Pax2-Cre allele was previously generated and has been widely used in the field of inner ear development.Here,by compounding Pax2-Cre and R26R-mTmG mice,we found a specific expression pattern of Pax2^(+)cells that marked BA1’s mesenchymal cells and the BA1-derivatives.Compared to Pax2-Cre;R26R-mTmG allele,GFP^(+)cells were abundantly found both in BA1 and second branchial arch in Wnt1-Cre;R26R-mTmG mice.As BMP4 signaling is required for orofacial development,we over-activated Bmp4 by using Pax2-Cre;pMes-BMP4 strain.Interestingly,our results showed bilateral hyperplasia between the upper and lower teeth.We also compare the phenotypes of Wnt1-Cre;pMes-BMP4 and Pax2-Cre;pMes-BMP4 strains and found severe deformation of molar buds,palate,and maxilla-mandibular bony structures in Wnt1-Cre;pMes-BMP4 mice;however,the morphology of these orofacial organs were comparable between controls and Pax2-Cre;pMes-BMP4 mice except for bilateral hyperplastic tissues.We further explore the properties of the hyperplastic tissue and found it is not derived from Runx2^(+)cells but expresses Msx1,and probably caused by abnormal cell proliferation and altered expression pattern of p-Smad1/5/8.In sum,our findings suggest altering BMP4 signaling in BA1-specific cell lineage may lead to unique phenotypes in orofacial regions,further hinting that Pax2-Cre mice could be a new model for genetic manipulation of BA1-derived organogenesis in the orofacial region.Jue Xu Meiling Chen Yanan Yan Qiaoxue Zhao Meiying Shao Zhen Huang 2021International Journal of Oral Science2021,13,4:3

10IncR26319/miR-2834/EndophilinA axis regulates oogenesis of the silkworm,Bombyx mori显示文摘Oocyte maturation is critical for insect reproduction.Vitellogenesis,the timely production and uptake of vitellogenin(Vg),is crucial for female fecundity.Vg is synthesized in fat body and absorbed by the oocytes through endocytosis during insect oogenesis.In the silkworm,Bombyx mori,we discovered that a nucleus-enriched long-noncoding RNA(lncRNA)lncR26319 regulates Endophilin A(EndoA)-a member of the endophilin family of endocytic proteins-through competitive binding to miR-2834.The lncR26319-miR-2834-EndoA axis was required for Vg endocytosis in the silkworm;loss of EndoA or overexpression ofmiR-2834significantlyreduced eggnumbers invirginmoths.Inaddition,accumulation of miR-2834 resulted in pupal and adult deformation and reduced fecundity in females.The expression of Vg,30-kDa(30K)protein,and egg-specific protein(Esp)decreased after knockdown of EndoA or overexpression of miR-2834,while knockdown of miR-2834 had an opposite effect on the expression of Vg,30K protein gene,and Esp.These results suggest that the lncR26319-miR-2834-EndoA axis contributes to the endocytic activity in theVguptake and leads to the normal progression of oogenesis in the silkworm.Thus,miR-2834 and EndoA are crucial for female reproduction and could be potential targets for new pest management strategies in lepidopterans.Yi Wang Yu Fu Hao Chena Chenyue Zhao Qunxia Huang Meiling Chang Wujie Qiu Yawen Shen Dandan Li 2023Insect Science2023,30,1:1

18Accelerating C_(2+)alcohols synthesis from syngas by simultaneous optimizations of CO dissociation and chain growth over CuCo alloy catalyst显示文摘With regard to the reaction of higheralcohol synthesis(HAS),the optimizations of activity and selectivity towards C_(2+)alcohol are restricted by the improper equilibrium in two different CO activation pathways and chain growth capacity.Herein,we find that delibrately controlling the compositions of catalysts is an effective strategy to achieve the equilibrium of CO activation pathways and promote the chain growth.As a result,the optimized Cu_(0.25)Co_(0.75) alloy catalyst can achieve a large proportion of higher alcohol in alcohol products(C_(2+)OH/MeOH=4.40),together with high CO conversion of 71.27%and space-time-yield of 147.65 g kg^(-1) h^(-1).The mechanistic studies suggest that the good performance of Cu_(0.25)Co_(0.75) catalyst is attributed to the synergistic effect between alloyed Cu and Co.Meiling Shui Chao Huang Peiyu Ma Wenjie Li Qun He Wenlong Wu Yisheng Tan Jun Bao 2021Chinese Chemical Letters2021,32,7:1
