| 1 | Inositol Pyrophosphate lnsP8 Acts as an Intracellular Phosphate Signal in Arabidopsis显示文摘The maintenance of cellular phosphate(Pi)homeostasis is of great importance in living organisms.The SPX domain-containing protein 1(SPX1)proteins from both Arabidopsis and rice have been proposed to act as sensors of Pi status.The molecular signal indicating the cellular Pi status and regulating Pi homeostasis in plants,however,remains to be identified,as Pi itself does not bind to the SPX domain.Here,we report the identification of the inositol pyrophosphate lnsP8 as a signaling molecule that regulates Pi homeostasis in Arabidopsis.Polyacrylamide gel electrophoresis profiling of InsPs revealed that lnsP8 level positively correlates with cellular Pi concentration.We demonstrated that the homologs of diphosphoinositol pentaki-sphosphate kinase(PPIP5K),VIH1 and VIH2,function redundantly to synthesize lnsP8,and that the vih1 vih2 double mutant overaccumulates Pi.SPX1 directly interacts with PHR1,the central regulator of Pi starvation responses,to inhibit its function under Pi-replete conditions.However,this interaction is compromised in the vih1 vih2 double mutant,resulting in the constitutive induction of Pi starvation-induced genes,indicating that plant cells cannot sense cellular Pi status without lnsP8.Furthermore,we showed that lnsP8 could directly bind to the SPX domain of SPX1 and is essential for the interaction between SPX1 and PHR1.Collectively,our study suggests that lnsP8 is the intracellular Pi signaling molecule serving as the ligand of SPX1 for controlling Pi homeostasis in plants. | Jinsong Dong Guojie Ma Liqian Sui Mengwei Wei Viswanathan Satheesh Ruyue Zhang Shenghong Ge Jinkai Li Tong-En Zhang Christopher Wittwer Henning JJessen Huiming Zhang Guo-Yong An Dai-Yin Chao Dong Liu Mingguang Lei | 2019 | Molecular Plant2019,12,11: | 8 |

| 3 | Effects of grazing intensity on seed production of Caragana stenophylla along a climatic aridity gradient in the Inner Mongolia Steppe,China显示文摘As the increases of climatic aridity and grazing intensity,shrubs play an increasingly important role in grassland ecosystem in arid and semi-arid regions,and its abundance also generally increases.However,the effects of climatic aridity and grazing intensity on sexual reproduction of shrubs in grassland remain largely unclear.In order to understand the effects of grazing intensity and climatic drought stress,and their interaction on seed production of shrub species,we examined the seed number,seed weight and seed yield of Caragana stenophylla under three grazing intensities(fenced,mild grazing and severe grazing) across a climatic aridity gradient(semi-arid,arid,very arid and intensively arid zones) in the Inner Mongolia Steppe,northern China during 2012–2013.The seed number,seed weight and seed yield gradually increased from the semi-arid to the very arid zones,but decreased from the very arid to the intensively arid zones in fenced plots.The seed number and seed yield decreased from the semi-arid to the intensively arid zones in mild and severe grazing treatment plots,therefore,grazing enhanced the suppression effect of climatic aridity on seed production of C.stenophylla.The seed number and seed yield gradually decreased as grazing intensity increased.The seed weight was highest in severe grazing plots,followed by the mild grazing plots and then the fenced plots.Precipitation varied interannually during the study period.We observed that the seed number,seed weight and seed yield were lower in the low precipitation year(2013) than in the high precipitation year(2012).As climatic drought stress increased,the negative effects of grazing on seed production of C.stenophylla also gradually increased.Our results indicated that climatic drought stress may contribute to the encroachment of C.stenophylla shrub in arid zones by promoting its seed production.However,grazing had negative effects on sexual reproduction of C.stenophylla,and the combined effects of drought stress and grazing seriously suppressed sexual reproduction of C.stenophylla in the intensively arid zone. | XIE Lina CHEN Weizhong Christopher A GABLER HAN Lei GUO Hongyu CHEN Qing MA Chengcang GU Song | 2016 | Journal of Arid Land2016,8,6: | 5 |

| 5 | Inhibition of focal adhesion kinase enhances antitumor response of radiation therapy in pancreatic cancer through CD8+ T cells显示文摘Objective:Pancreatic ductal adenocarcinoma(PDAC)is a deadly malignancy,due in large part to its resistance to conventional therapies,including radiotherapy(RT).Despite RT exerting a modest antitumor response,it has also been shown to promote an immunosuppressive tumor microenvironment.Previous studies demonstrated that focal adhesion kinase inhibitors(FAKi)in clinical development inhibit the infiltration of suppressive myeloid cells and T regulatory(T regs)cells,and subsequently enhance effector T cell infiltration.FAK inhibitors in clinical development have not been investigated in combination with RT in preclinical murine models or clinical studies.Thus,we investigated the impact of FAK inhibition on RT,its potential as an RT sensitizer and immunomodulator in a murine model of PDAC.Methods:We used a syngeneic orthotopic murine model to study the effect of FAKi on hypofractionated RT.Results:In this study we showed that IN10018,a small molecular FAKi,enhanced antitumor response to RT.Antitumor activity of the combination of FAKi and RT is T cell dependent.FAKi in combination with RT enhanced CD8+T cell infiltration significantly in comparison to the radiation or FAKi treatment alone(P<0.05).FAKi in combination with radiation inhibited the infiltration of granulocytes but enhanced the infiltration of macrophages and T regs in comparison with the radiation or FAKi treatment alone(P<0.01).Conclusions:These results support the clinical development of FAKi as a radiosensitizer for PDAC and combining FAKi with RT to prime the tumor microenvironment of PDAC for immunotherapy. | Arsen Osipov Alex B.Blair Juliane Liberto Jianxin Wang Keyu Li Brian Herbst Yao Xu Shiqi Li Nan Niu Rufiaat Rashid Ding Ding Yanan Liu Zaiqi Wang Christopher L.Wolfgang Richard A.Burkhart Daniel Laheru Lei Zheng | 2021 | Cancer Biology & Medicine2021,18,1: | 3 |

| 8 | Central role of myeloid MCPIP1 in protecting against LPSinduced inflammation and lung injury显示文摘Although systemic inflammatory responses attributable to infection may lead to significant lung injury,the precise molecular mechanisms leading to lung damage are poorly understood and therapeutic options remain limited.Here,we show that myeloid monocyte chemotactic protein-inducible protein 1(MCPIP1)plays a central role in protecting against LPS-induced inflammation and lung injury.Myeloid-specific MCPIP1 knockout mice developed spontaneous inflammatory syndromes,but at a late age compared to global MCPIP1 knockout mice.Moreover,mice with a myeloid-specific deletion of MCPIP1 were extremely sensitive to LPS-induced lung injury due to overproduction of proinflammatory cytokines and chemokines.We identified C/EBPβand C/EBPδ,two critical transcriptional factors that drive cytokine production and lung injury,as targets of MCPIP1 RNase.LPS administration caused MCPIP1 protein degradation in the lungs.Pharmacological inhibition of MALT1,a paracaspase that cleaves MCPIP1,by MI-2 selectively increased the MCPIP1 protein levels in macrophages and in the lungs.Meanwhile,administration of MI-2 protected mice from LPS-induced inflammation,lung injury and death.Collectively,these results indicate that myeloid MCPIP1 is central in controlling LPS-induced inflammation and lung injury.Pharmacological inhibition of MALT1 protease activity may be a good strategy to treat inflammatory diseases by enhancing MCPIP1 expression in myeloid cells. | Yong Li Xuan Huang Shengping Huang Hui He Tianhua Lei Fatma Saaoud Xiao-Qiang Yu Ari Melnick Anil Kumar Christopher J Papasian Daping Fan Mingui Fu | 2017 | Signal Transduction and Targeted Therapy2017,2,1: | 2 |
| 9 | Sedimentary environment controls on the accumulation of organic matter in the Upper Ordovician Wufeng–Lower Silurian Longmaxi mudstones in the Southeastern Sichuan Basin of China显示文摘The Upper Ordovician–Lower Silurian mudstones(including the Wufeng, Guanyinqiao and Longmaxi Formations) in the Sichuan Basin are some of the most important shale gas plays in China. In order to enhance our understanding of the process of formation of organic carbon up to 10%, optical, microscopy and geochemical methods have been used to investigate the petrographic and geochemical characteristics of the formation. Firstly, three mudstone lithofacies were identified based on a wide variety of mudstone laminations. These are:(a) indistinctly laminated mudstone;(b) parallellaminated mudstone; and(c) nonparallel-laminated mudstone. Then, combining with the evidence from depocenter migration, Th/U ratios and total organic carbon, the abundant organo-minerallic fabrics suggest that organic carbon was preferentially deposited and preserved in anoxic, low energy and stagnant water conditions during deposition of the Wufeng and Longmaxi Formations.
