2Dual color optogenetic control of neural populations using low-noise,multishank optoelectrodes显示文摘Optogenetics allows for optical manipulation of neuronal activity and has been increasingly combined with intracellular and extracellular electrophysiological recordings.Genetically-identified classes of neurons are optically manipulated,though the versatility of optogenetics would be increased if independent control of distinct neural populations could be achieved on a sufficient spatial and temporal resolution.We report a scalable multisite optoelectrode design that allows simultaneous optogenetic control of two spatially intermingled neuronal populations in vivo.We describe the design,fabrication,and assembly of low-noise,multisite/multicolor optoelectrodes.Each shank of the four-shank assembly is monolithically integrated with 8 recording sites and a dualcolor waveguide mixer with a 7×30μm cross-section,coupled to 405 nm and 635 nm injection laser diodes(ILDs)via gradient-index(GRIN)lenses to meet optical and thermal design requirements.To better understand noise on the recording channels generated during diode-based activation,we developed a lumped-circuit modeling approach for EMI coupling mechanisms and used it to limit artifacts to amplitudes under 100μV upto an optical output power of 450μW.We implanted the packaged devices into the CA1 pyramidal layer of awake mice,expressing Channelrhodopsin-2 in pyramidal cells and ChrimsonR in paravalbumin-expressing interneurons,and achieved optical excitation of each cell type using sub-mW illumination.We highlight the potential use of this technology for functional dissection of neural circuits.Komal Kampasi Daniel F.English John Seymour Eran Stark Sam McKenzie Mihály Vöröslakos György Buzsáki Kensall D.Wise Euisik Yoon 2018Microsystems & Nanoengineering2018,4,1:8
3Methionine adenosyltransferases in liver health and diseases显示文摘Methionine adenosyltransferases(MATs)are essential for cell survival because they catalyze the biosynthesis of the biological methyl donor S-adenosylmethionine(SAMe)from methionine and adenosine triphosphate(ATP).Mammalian cells express two genes,MAT1A and MAT2A,which encode two MAT catalytic subunits,α1 andα2,respectively.Theα1 subunit organizes into dimers(MATIII)or tetramers(MATI).Theα2 subunit is found in the MATII isoform.A third gene MAT2B,encodes a regulatory subunit b,that regulates the activity of MATII by lowering the inhibition constant(Ki)for SAMe and the Michaelis constant(Km)for methionine.MAT1A expressed mainly in hepatocytes maintains the differentiated state of these cells whereas MAT2A and MAT2B are expressed in non-parenchymal cells of the liver(hepatic stellate cells[HSCs]and Kupffer cells)and extrahepatic tissues.A switch from the liverspecific MAT1A to MAT2A has been observed during conditions of active liver growth and dedifferentiation.Liver injury,fibrosis,and cancer are associated with MAT1A silencing and MAT2A/MAT2B induction.Even though both MAT1A and MAT2A are involved in SAMe biosynthesis,they exhibit distinct molecular interactions in liver cells.This review provides an update on MAT genes and their roles in liver pathologies.Komal Ramani Shelly C.Lu 2017Liver Research2017,1,2:5

5A high methionine,low folate and vitamin B_6/B_(12) containing diet can be associated with memory loss by epigenetic silencing of netrin-1显示文摘Memory-epigenetics which is the loss of memory due to epigenetic modifications can be due to the silencing of genes involved in cognitive functions and this is the basis of the current study.We hypothesize that a diet containing high methionine and low vitamins can lead to memory impairment by increasing global DNA methylation and therefore,silencing the netrin-1 gene,which encodes the glycoprotein involved in neurogenesis,axonal guidance and maintenance of the synaptic plasticity.Wild type(C57 BL/6 J) mice were fed with a diet containing excess methionine(1.2%),low-folate(0.08 mg/kg),vitamin B_6(0.01 mg/kg),and B_(12)(10.4 mg/kg) for 6 weeks.Mice were examined weekly for the long-term memory function,using a passive avoidance test,which determined loss of fear-motivated long-term memory starting from the fourth week of diet.Similarly,an increase in brain %5-methyl cytosine was observed starting from the 4 th week of diet in mice.Mice fed with a high methionine,low folate and vitamins containing diet showed a decrease in netrin-1 protein expression and an increase in netrin-1 gene promotor methylation,as determined by methylation-sensitive restriction enzyme-polymerase chain reaction analysis.The increase in methylation of netrin-1 gene was validated by high-resolution melting and sequencing analysis.Furthermore,the association of netrin-1 with memory was established by administering netrin that considerably restored long-term fear motivated memory.Taken together,these results suggest that a diet rich in methionine and lacking in folate and vitamin B_6/B_(12) can induce defects in learning and memory.Furthermore,the data indicates that decrease in netrin-1 expression due to hyper-methylation of its gene can be associated with memory loss.The animal procedures were approved by the Institutional Animal Care and Use Committee,University of Louisville,USA(No.A3586-01) on February 2,2018.Anuradha Kalani Pankaj Chaturvedi Komal Kalani Pradip K.Kamat Poonam Chaturvedi Neetu Tyagi 2019Neural Regeneration Research2019,14,7:4

7Monitoring and modeling of urban sprawl through remote sensing and GIS in Kuala Lumpur, Malaysia显示文摘Introduction:Kuala Lumpur is currently experiencing a large-scale urban transformation.This study explores a multi-buffer ring method to investigate the spatial and temporal dynamics of urban growth in the Kuala Lumpur metropolitan area by using Geographical Information Systems(GIS)and remote sensing imageries from1989 to 2014.Methods:The first Landsat images from 1989,2001,and 2014 were classified with maximum likelihood classification to produce land cover maps and identify four types of land cover:urban/built-up,agriculture,forest,and water.Then,the area coverage for all the land use types at different points in time were measured and combined with distance from the city center.Later on,urbanization densities from the city center to the outside for every 1-km distance from 1 to 50 km are calculated.Results:City density on the distance of 5,8,15,20,and 30 km was very high in the years 1989 and 2014.Temporally,urban expansion shows fast and slow growth stages with high-speed growth shifting to the east side of the city.Conclusions:In general,the population,traffic conditions,industrialization,and policy are the major factors thatinfluenced the urban expansion.Mukesh Singh Boori Maik Netzband Komal Choudhary Vít Voženílek 2015Ecological Processes2015,4,1:2

9Correlation between the proportion of breast volume involved by locally advanced tumors and invasion of the skin and posterior structures显示文摘AIM: To evaluate any differences between the percentages of involved breast volume, pathologic attributes,and tumor marker expression of T3 and T4a-c tumors in locally advanced breast cancers(BC).METHODS: All patients with T3 N > 0 and T4a-c BC without evidence of distant metastasis(M0), presenting to the Breast Clinic from 1980 to 2010, were examinedto determine whether their BC's involved ≥ 50% of their breast volumes, defined by gross replacement of at least one hemisphere. Core needle biopsy or postmastectomy specimens from tumors involving a known percent of breast volume were evaluated for:(1)pathological grades and lympho-vascular invasion(LVI);(2) hormone receptor(ER/PR) expression > 0; and(3)epidermoid growth factor 2(her2) over-expression(3+)by immune-histochemical staining or fluorescent in situ hybridization.RESULTS: The data base included 98 patients with T3N> 0 M0 and 120 with T4a-c, any N disease, M0 disease. T3 tumor masses involved 50% or more of the breast in 23/98(24%), and T4a-c tumors 65/120(54%)(P < 0.001). Only 1% of T3 tumors and 23% of T4a-c tumors presented with total breast replacement. 10The caspase-1 inhibitor VX765 upregulates connexin 43 expression and improves cell-cell communication after myocardial infarction via suppressing the IL-1β/p38 MAPK pathway显示文摘Connexin 43 (Cx43) is the most important protein in the gap junction channel between cardiomyocytes. Abnormalities of Cx43 change the conduction velocity and direction of cardiomyocytes, leading to reentry and conduction block of the myocardium, thereby causing arrhythmia. It has been shown that IL-1β reduces the expression of Cx43 in astrocytes and cardiomyocytes in vitro. However, whether caspase-1 and IL-1β affect connexin 43 after myocardial infarction (MI) is uncertain. In this study we investigated the effects of VX765, a caspase-1 inhibitor, on the expression of Cx43 and cell-to-cell communication after MI. Rats were treated with VX765 (16 mg/kg, i.v.) 1 h before the left anterior descending artery (LAD) ligation, and then once daily for 7 days. The ischemic heart was collected for histochemical analysis and Western blot analysis. We showed that VX765 treatment significantly decreased the infarct area, and alleviated cardiac dysfunction and remodeling by suppressing the NLRP3 inflammasome/caspase-1/IL-1β expression in the heart after MI. In addition, VX765 treatment markedly raised Cx43 levels in the heart after MI. In vitro experiments were conducted in rat cardiac myocytes (RCMs) stimulated with the supernatant from LPS/ATP-treated rat cardiac fibroblasts (RCFs). Pretreatment of the RCFs with VX765 (25 μM) reversed the downregulation of Cx43 expression in RCMs and significantly improved intercellular communication detected using a scrape-loading/dye transfer assay. We revealed that VX765 suppressed the activation of p38 MAPK signaling in the heart tissue after MI as well as in RCMs stimulated with the supernatant from LPS/ATP-treated RCFs. 12Self nano-emulsifying drug delivery system for Embelin:Design,characterization and in-vitro studies显示文摘CThe objective of the present study was to prepare solid self-nanoemulsifying drug delivery system(S-SNEDDS)containing Capryol-90 as oil phase for the delivery of Embelin,a poorly water soluble herbal active ingredient.Box-Behnken experimental design was employed to optimise the formulation variables,X1(amount of oil;Capryol 90),X2(amount of surfactant;Acrysol EL 135)and X3(amount of co-surfactant;PEG 400).Systems were appraised for visual characteristics for self emulsifying time,globule size and drug release.Optimised liquid formulations were formulated into free flowing granules(S-SNEDDS)by adsorption on the porous materials like Aerosil 200 and Neusilin and thereby compressed into tablet.In vitro dissolution studies of SNEDDS revealed increased in the dissolution rate of the drug.FT-IR data revealed no physicochemical interaction between drug and excipients.Solid state characterization of S-SNEDDS by DSC and Powder XRD confirmed reduction in drug crystallinity which further supports the results of dissolution studies.TEM analysis exhibited spherical globules.Further,the accelerated stability studies for 6 months revealed that S-SNEDDS of Embelin are found to be stable without any significant change in physicochemical properties.Thus,the present studies demonstrated dissolution enhancement potential of porous carrier based S-SNEDDS for poorly water soluble herbal active ingredient,Embelin.Komal Parmar Jayvadan Patel Navin Sheth 2015Asian Journal of Pharmaceutical Sciences2015,10,5:2
13Breast carcinoma with metastasis to the gallbladder:an unusual case report with a short review of literature显示文摘Gallbladder metastases are very rare and usually arise from malignant melanoma, renal cell carcinoma and cervical carcinoma. Breast carcinoma tnetastatic to the gallbladder is extremely rare and only 4 cases have been reported in the English literature. We hereby report a 54-year-old lady who was diagnosed as having breast carcinoma and underwent modified radical mastectomy. One month after the operation, she developed acute abdomenal pain and underwent cholecystectomy after clinical investigation. Histopathological examination revealed metastasis to the gallbladder. Being considered a patient with tnetastatic breast carcinoma she was subjected to taxane and anthracycline-based palliative chemotherapy.
