2Spatiotemporal changes in vegetation coverage and its driving factors in the Three-River Headwaters Region during 2000-2011显示文摘The Three-River Headwaters Region(TRHR), which is the source area of the Yangtze River, Yellow River, and Lancang River, is of key importance to the ecological security of China. Because of climate changes and human activities, ecological degradation occurred in this region. Therefore, 'The nature reserve of Three-River Source Regions' was established, and 'The project of ecological protection and construction for the Three-River Headwaters Nature Reserve' was implemented by the Chinese government. This study, based on MODIS-NDVI and climate data, aims to analyze the spatiotemporal changes in vegetation coverage and its driving factors in the TRHR between 2000 and 2011, from three dimensions. Linear regression, Hurst index analysis, and partial correlation analysis were employed. The results showed the following:(1) In the past 12 years(2000–2011), the NDVI of the study area increased, with a linear tendency being 1.2%/10a, of which the Yangtze and Yellow River source regions presented an increasing trend, while the Lancang River source region showed a decreasing trend.(2) Vegetation coverage presented an obvious spatial difference in the TRHR, and the NDVI frequency was featured by a bimodal structure.(3) The area with improved vegetation coverage was larger than the degraded area, being 64.06% and 35.94%, respectively during the study period, and presented an increasing trend in the north and a decreasing trend in the south.(4) The reverse characteristics of vegetation coverage change are significant. In the future, degradation trends will be mainly found in the Yangtze River Basin and to the north of the Yellow River, while areas with improving trends are mainly distributed in the Lancang River Basin.(5) The response of vegetation coverage to precipitation and potential evapotranspiration has a time lag, while there is no such lag in the case of temperature.(6) The increased vegetation coverage is mainly attributed to the warm-wet climate change and the implementation of the ecological protection project.LIU Xianfeng ZHANG Jinshui ZHU Xiufang PAN Yaozhong LIU Yanxu ZHANG Donghai LIN Zhihui 2014Journal of Geographical Sciences2014,24,2:47
3Two-dimensional multimetallic sulfide nanosheets with multi-active sites to enhance polysulfide redox reactions in liquid Li2S6-based lithium-polysulfide batteries显示文摘The lithium-sulfur battery has attracted enormous attention as being one of the most significant energy storage technologies due to its high energy density and cost-effectiveness.However,the 'shuttle effect' of polysulfide intermediates represents a formidable challenge towards its wide applications.Herein,we have designed and synthesized two-dimensional Cu,Zn and Sn-based multimetallic sulfide nanosheets to construct multi-active sites for the immobilization and entrapment of polysulfides with offering better performance in liquid Li2S6-based lithium-polysulfide batteries.Both experimental measurements and theoretical computations demonstrate that the interfacial multi-active sites of multimetallic sulfides not only accelerate the multi-chained redox reactions of highly diffusible polysulfides,but also strengthen affinities toward polysulfides.By adopting multimetallic sulfide nanosheets as the sulfur host,the liquid Li2 S6-based cell exhibits an impressive rate capability with 1200 mAh/g and retains 580 mAh/g at 0.5 mA/cm^(2) after 1000 cycles.With high sulfur mass loading conditions,the cell with 2.0 mg/cm^(2) sulfur loading delivers a cell capacity of 1068 mAh/g and maintains 480 mAh/g with 0.8 mA/cm^(2) and 500 cycles.This study provides new insights into the multifunctional material design with multi-active sites for elevated lithium-polysulfide batteries.Chenyang Zha Donghai Wu Yuwei Zhao Jun Deng Jinghua Wu Rong Wu Meng Yang Lin Wang Houyang Chen 2021Journal of Energy Chemistry2021,30,1:3

14Biophysical characterization and ligand-binding properties of the elongation factor Tu from Mycobacterium tuberculosis显示文摘Mycobacterium tuberculosis(Mtb)is the key devastating bacterial pathogen responsible for tuberculosis.Increasing emergence of multi-drug-resistant,extensively drug-resistant,and rifampicin/isoniazid-resistant strains of Mtb makes the discovery of validated drug targets an urgent priority.As a vital translational component of the protein biosynthesis system,elongation factor Tu(EF-Tu)is an important molecular switch responsible for selection and binding of the cognate aminoacyl-tRNA to the acceptor site on the ribosome.In addition,EF-Tu from Mtb(MtbEF-Tu)is involved in the initial step of trans-translation which is an effective system for rescuing the stalled ribosomes from non-stop translation complexes under stress conditions.Given its crucial role in protein biosynthesis,EF-Tu is identified as an excellent molecular target for drug design.Here,we reported the recombinant expression,purification,biophysical characterization,and structural modeling of the MtbEF-Tu protein.Our results demonstrated that prokaryotic expression plasmids of pET28a-MtbEF-Tu could be expressed efficiently in Escherichia coli.We successfully purified the 6× His-tagged proteins with a yield of 16.8 mg from 1 l of Luria Bertani medium.Dynamic light scattering experiments showed that MtbEF-Tu existed in a monomeric form,and circular dichroism experiments indicated that MtbEF-Tu was well structured.Moreover,isothermal titration calorimetry experiments displayed that the purified MtbEF-Tu protein possessed intermediate binding affinities for guanosine-5′-triphosphate(GTP)and GDP.The GTP/GDP-binding sites were predicted by flexible molecular docking approach which reveals that GTP/GDP binds to MtbEF-Tu mainly through hydrogen bonds.Our work lays the essential basis for further structural and functional studies of MtbEF-Tu as well as MtbEF-Tu-related novel drug developments.Juanjuan Yang Jing Hong Ling Luo Ke Liu Chun Meng Zhi-liang Ji Donghai Lin 2019Acta Biochimica et Biophysica Sinica2019,51,2:1

17Cefotetan-bound human RKIP involves in Ras/Raf1/MEK/ERK signaling pathway显示文摘Cefotetan is widely used to treat bacterial infections in the clinic owing to its broad spectrum of antibacterial activity.In the present study,we demonstrate that cefotetan can bind to the conserved ligand-binding pocket of human Raf1 kinase inhibitory protein(hRKIP),which acts as a negative regulator of the Ras/Raf1/MEK/ERK signaling pathway.The cefotetan-bound hRKIP adopts a rigid structure with insufficient space for binding Raf1 kinase,thereby reliving the inhibitory activity of hRKIP in the Ras/Raf1/MEK/ERK signaling pathway and enhancing the phosphorylation level of ERK.Both NMR titration and molecular docking approaches show that several residues(P74,Y81,W84,P111,P112,K113,S142,G143,D144,W173,P178,Y181 and L184)play crucial roles in hRKIP binding cefotetan.NMR dynamics analysis reveals that the binding of cefotetan with hRKIP promotes ps-ns internal motion but reducesμs-ms conformational exchange for residues in the cefotetan-binding pocket of hRKIP.Our results not only disclose the structural basis of cefotetan upregulating the Ras/Raf1/MEK/ERK signaling pathway but also benefit developing novel drugs against diseases caused by the impaired Ras/Raf1/MEK/ERK pathway.Chenyun Guo Hao Xu Yu Zhou Zhihua Wu Bin Jiang Hanyu Chen Donghai Lin 2022Acta Biochimica et Biophysica Sinica2022,54,12:0
18Nucleation and growth of Fe-rich phases in Al-5Ti-1B modified Al-Fe alloys investigated using synchrotron X-ray imaging and electron microscopy显示文摘Plate-like Fe-rich intermetallic phases directly influence the mechanical properties of recycled Al alloys;thus, many attempts have been made to modify the morphology of these phases. Through synchrotron X-ray imaging and electron microscopy, the underlying nucleation and growth mechanisms of Fe-rich phases during the solidification of Al-5 Ti-1 B-modified Al-2 Fe alloys were revealed in this study. The results showed that the Al-5 Ti-1 B grain refiner as well as the applied pressure both resulted in reduction of the size and number of primary Al_(3)Fe phases and promoted the formation of eutectic Al_(6)Fe phases.The tomography results demonstrated that Al-5 Ti-1 B changed the three-dimensional(3 D) morphology of primary Fe-rich phases from rod-like to branched plate-like, while a reduction in their thickness and size was also observed. This was attributed to the fact that Ti-containing solutes in the melts inhibit the diffusion of Fe atoms and the Al_(3)Fe twins produce re-entrant corner on the twin boundaries along the growth direction. Moreover, the TiB_(2) provides possible nucleation sites for Al_6Fe phases. The nucleation mechanism of Fe-rich phases is discussed in terms of experimental observations and crystallography calculations. 19Biophysical and functional characterizations of recombinant RimI acetyltransferase from Mycobacterium tuberculosis显示文摘Nα-acetylation is a universal protein modification related to a wide range of physiological processes in eukaryotes and prokaryotes. RimI, an Nα-acetyltransferase in Mycobacterium tuberculosis, is responsible for the acetylation of the α-amino group of the N-terminal residue in the ribosomal protein S18. Despite growing evidence that protein acetylation may be correlated with the pathogenesis of tuberculosis, no structural information is yet available for mechanistically understanding the MtRimI acetylation. To enable structural studies for MtRimI, we constructed a serial of recombinant MtRimI proteins and assessed their biochemical properties. We then chose an optimal construct MtRimIC21A4-153 and expressed and purified the truncated high-quality protein for further biophysical and functional characterizations. The 2D 1H-15N heteronuclear single quantum coherence spectrum of MtRimIC21A4-153 exhibits wider chemical shift dispersion and favorable peak isolation, indicating that MtRimIC21A4-153 is amendable for further structural determination. Moreover, bio-layer interferometry experiments showed that MtRimIC21A4-153 possessed similar micromolar affinity to full-length MtRimI for binding the hexapeptide substrate Ala-Arg-Tyr-Phe-Arg-Arg. Enzyme kinetic assays also exhibited that MtRimIC21A4-153 had almost identical enzymatic activity to MtRimI, indicating insignificant influence of the recombinant variations on enzymatic functions. Furthermore, binding sites of the peptide were predicted by molecular docking approach, suggesting that this substrate binds to MtRimI primarily through electrostatic and hydrogen bonding interactions. 20Recombinant expression, biophysical and functional characterization of ClpS from Mycobacterium tuberculosis显示文摘Intracellular proteolysis is attracting more and more attention for its unique and important character in Mycobacterium tuberculosis(Mt).The ClpS protein from Mt(MtClpS)plays a critical role in intracellular proteolysis by recognizing N-end rule substrates,which makes it become a potential target for antibacterial drugs.However,the molecular mechanism of MtClpS recognizing N-end rule substrates remains unclear.Preparation of highly concentrated and pure MtClpS protein is a prerequisite for further structural and functional studies.In the present work,we tried several fusion tags and various expression conditions to maximize the production of MtClpS in Escherichia coli.We established an efficient approach for preparing the MtClpS protein with a high yield of 24.7 mg/l and a high purity of 98%.After buffer screening,we obtained a stable MtClpS protein sample concentrated at 0.63 mM in the presence of glycerol,l-Arginine,and l-Glutamate.Moreover,circular dichroism characterization indicated that the secondary structure of MtClpS consists of 38%α-helix and 24%β-sheet.The 2D 1H-15N HSQC nuclear magnetic resonance spectrum showed a good dispersion of resonance peaks with uniform intensity,indicating that the purified MtClpS protein was well folded and conformationally homogeneous.Isothermal titration calorimetry experiments revealed significant interactions of MtClpS with N-end rule peptides beginning with Leu,Tyr,Trp,or Phe.Furthermore,residues D34,D35,and H66 were confirmed as key residues for MtClpS recognizing the N-end rule peptide.The successful expression and biophysical characterization of MtClpS enabled us to gain insight into the molecular mechanism of MtClpS recognizing N-end rule substrates.The obtained stable and pure recombinant MtClpS will enable future inhibitor screening experiments.Chenyun Guo Yihang Xiao Fangkai Bs Weiliang Lin Huilin Wang Hongwei Yao Donghai Lin 2019Acta Biochimica et Biophysica Sinica2019,51,11:0
