Osteoarthritis:toward a comprehensive understanding of pathological mechanism显示文摘Osteoarthritis(OA) is the most common degenerative joint disease and a major cause of pain and disability in adult individuals. The etiology of OA includes joint injury, obesity, aging, and heredity. However, the detailed molecular mechanisms of OA initiation and progression remain poorly understood and, currently, there are no interventions available to restore degraded cartilage or decelerate disease progression. The diathrodial joint is a complicated organ and its function is to bear weight, perform physical activity and exhibit a joint-specific range of motion during movement. During OA development, the entire joint organ is affected, including articular cartilage, subchondral bone, synovial tissue and meniscus. A full understanding of the pathological mechanism of OA development relies on the discovery of the interplaying mechanisms among different OA symptoms, including articular cartilage degradation, osteophyte formation, subchondral sclerosis and synovial hyperplasia, and the signaling pathway(s) controlling these pathological processes.Di Chen Jie Shen Weiwei Zhao Tingyu Wang Lin Han John L Hamilton Hee-Jeong Im 2017Bone Research2017,5,1:121

Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently, increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression, yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that, during AD progression, the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine, which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation, contributing to AD-associated neuroinflammation.Importantly, the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China, suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation, harnesses neuroinflammation and reverses the cognition impairment.Together, our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192

A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. Further efforts should focus on identifying the etiology of the SARS-associated virus and ruling out conclusively the existence of otherpossible SARS-related pathogen(s).QIN E'de ZHU Qingyu YU Man FAN Baochang CHANG Guohui SI Bingyin YANG Bao PENG Wenming JIANG Tao LIU Bohua DENG Yongqiang LIU Hong ZHANG Yu WANG Cui LI Yuquan GAN Yonghua LI Xiaoyu L Fushuang TAN Gang CAO Wuchun, YANG Ruifu Institute of Microbiology and Epidemiology, Chinese Academy of Military Medical Sciences, Beijing 100071, China WANG Jian, LI Wei, XU Zuyuan, LI Yan, WU Qingfa, LIN Wei, CHEN Weijun, TANG Lin, DENG Yajun, HAN Yujun, LI Changfeng, LEI Meng, LI Guoqing, LI Wenjie, L Hong, SHI Jianping, TONG Zongzhong, ZHANG Feng, LI Songgang, LIU Bin, LIU Siqi, DONG Wei, WANG Jun, Gane K-S Wong, YU Jun & YANG Huanming* Beijing Genomics Institute, Chinese Academy of Sciences, Beijing 101300 National Center for Genome Information, Beijing 101300, China 2003Chinese Science Bulletin2003,48,10:121
Detection and parameter estimation of multicomponent LFM signal based on the fractional Fourier transform显示文摘This paper presents a new method for the detection and parameter estimation of multicomponent LFM signals based on the fractional Fourier transform. For the optimization in the fractional Fourier domain, an algorithm based on Quasi-Newton method is proposed which consists of two steps of searching, leading to a reduction in computation without loss of accuracy. And for multicomponent signals, we further propose a signal separation technique in the fractional Fourier domain which can effectively suppress the interferences on the detection of the weak components brought by the stronger components. The statistical analysis of the estimate errors is also performed which perfects the method theoretically, and finally, simulation results are provided to show the validity of our method.QI Lin1, 2, TAO Ran1, ZHOU Siyong1 & WANG Yue1 1. 14Production and application of a novel bioflocculant by multiple-microorganism consortia using brewery wastewater as carbon source显示文摘The flocculating activity of a novel bioflocculant MMF1 produced by multiple-microorganism consortia MM1 was investigated.MM1 was composed of strain BAFRT4 identified as Staphylococcus sp.and strain CYGS1 identified as Pseudomonas sp.Theflocculating activity of MMF1 isolated from the screening medium was 82.9%,which is remarkably higher than that of the bioflocculantproduced by either of the strains under the same condition.Brewery wastewater was also used as the carbon source for MM1,and thecost-effective production medium for MM1 mainly comprised 1.0 L brewery water(chemical oxygen demand(COD)5000 mg/L),0.5 g/L urea,0.5 g/L yeast extract, and 0.2 g/L(NH4)2SO4.The optimal conditions for the production of MMF1 was inoculum size2%,initial pH 6.0,cultivating temperature 30°C,and shaking speed 160 r/min,under which the flocculating activity of the MMF1reached 96.8%.Fifteen grams of purified bioflocculant could be recovered from 1.0 L of fermentation broth.MMF1 was identified asa macromolecular substance containing both protein and polysaccharide. It showed good flocculating performance in treating indigotinprinting and dyeing wastewater, and the maximal removal effciencies of COD and chroma were 79.2%and 86.5%,respectively.ZHANG Zhi-qiang LIN Bo XIA Si-qing WANG Xue-jiang YANG A-ming 2007Journal of Environmental Sciences2007,19,6:62
LA-(MC)-ICPMS U-Pb zircon geochronology and Lu-Hf isotope compositions of the Taihua complex on the southern margin of the North China Craton显示文摘Widely distributed on the southern margin of the North China Craton, the Taihua complex extends roughly in an east-west direction and the relatively complete successions are found in Lushan County,Henan Province.Like many other Archean terranes, the Taihua Complex can be divided into two major lithological units along the Dangze River, namely gneisses series and supracrustal rocks. The former is located on the north side of the river and chiefly composed of TTG gneisses and amphibolites;the latter on the south side of the river is mainly supracrustal rock.Coupled with the previous studies, the results obtained by this study show that the Taihua complex was formed in a large time span from Neoarchean to Palaeoproterozoic. The TTG gneisses and amphibolites are dated at Neoarchean (2794–2752 Ma).The 2.9 Ga and 3.1 Ga zircons in amphibolites could be xenocrysts. In the North China Craton, the 2.8–2.7 Ga old rocks crop out in several areas e.g.western Shandong Province,Jiaodong Peninsula and Lushan area of Henan Province. In addition,2.8–2.7 Ga detrital zircons or xenocryst zircons have been recognized in Huai’an,Fuping,Wutai areas of North China Craton and also reported in the Early Paleozoic diamondiferous kimberlites in Mengyin and Fuxian. All these age results indicate that the 2.8–2.7 Ga rocks may have been developed much broader region than today’s outcrops.Zircon Hf and whole rock Nd isotopes show that the 2.8–2.7 Ga tectono-thermal event represents an important period of crustal growth with minor ancient crust reworked in the North China Craton. However the formation of supracrustal rocks is limited to 2.2–2.0 Ga in the Palaeoproterozoic time, not the Archaean,as previously believed.Combined with the chronological data of aluminium-rich metamorphic rocks (Khondalite series) on the southern margin of the North China Craton and adjacent areas,it is suggested that the above areas have widely developed Paleoproterozoic passive continental margin environment.DIWU ChunRong SUN Yong LIN CiLuan WANG HongLiang 2010Chinese Science Bulletin2010,55,23:72
Remediation of Heavy Metal-Polluted Agricultural Soils Using Clay Minerals: A Review显示文摘Heavy metal contamination of agricultural soils poses risks and hazards to humans. The remediation of heavy metal-polluted soils has become a hot topic in environmental science and engineering. In this review, the application of clay minerals for the remediation of heavy metal-polluted agricultural soils is summarized, in terms of their remediation effects and mechanisms, influencing factors, and future focus. Typical clay minerals, natural sepiolite, palygorskite, and bentonite, have been widely utilized for the in-situ immobilization of heavy metals in soils, especially Cd-polluted paddy soils and wastewater-irrigated farmland soils. Clay minerals are able to increase soil p H, decrease the chemical-extractable fractions and bioavailability of heavy metals in soils, and reduce the heavy metal contents in edible parts of plants. The immobilization effects have been confirmed in field-scale demonstrations and pot trials. Clay minerals can improve the environmental quality of soils and alleviate the hazards of heavy metals to plants. As main factors affecting the immobilization effects, the p H and water condition of soils have drawn academic attention. The remediation mechanisms mainly include liming, precipitation, and sorption effects. However, the molecular mechanisms of microscopic immobilization are unclear. Future studies should focus on the long-term stability and improvement of clay minerals in order to obtain a better remediation effect.XU Yi LIANG Xuefeng XU Yingming QIN Xu HUANG Qingqing WANG Lin SUN Yuebing 2017Pedosphere2017,27,2:66
Impacts of chemical fertilizer reduction and organic amendments supplementation on soil nutrient, enzyme activity and heavy metal content显示文摘Excessive use of agro-chemicals(such as mineral fertilizers) poses potential risks to soil quality. Application of organic amendments and reduction of inorganic fertilizer are economically feasible and environmentally sound approaches to develop sustainable agriculture. This study investigated and evaluated the effects of mineral fertilizer reduction and partial substitution of organic amendment on soil fertility and heavy metal content in a 10-season continually planted vegetable field during 2009–2012. The experiment included four treatments: 100% chemical fertilizer(CF100), 80% chemical fertilizer(CF80), 60% chemical fertilizer and 20% organic fertilizer(CF60+OM20), and 40% chemical fertilizer and 40% organic fertilizer(CF40+OM40). Soil nutrients, enzyme activity and heavy metal content were determined. The results showed that single chemical fertilizer reduction(CF80) had no significant effect on soil organic matter content, soil catalase activity and soil heavy metal content, but slightly reduced soil available N, P, K, and soil urease activity, and significantly reduced soil acid phosphatase activity. Compared with CF100, 40 or 60% reduction of chemical fertilizer supplemented with organic fertilizer(CF60+OM20, CF40+OM40) significantly increased soil organic matter, soil catalase activity and urease activity especially in last several seasons, but reduced soil available P, K, and soil acid phosphatase activity. In addition, continuous application of organic fertilizer resulted in higher accumulation of Zn, Cd, and Cr in soil in the late stage of experiment, which may induce adverse effects on soil health and food safety.NING Chuan-chuan GAO Peng-dong WANG Bing-qing LIN Wei-peng JIANG Ni-hao CAI Kun-zheng 2017Journal of Integrative Agriculture2017,16,8:64
Clinical characteristics of 24 asymptomatic infections with COVID-19 screened among close contacts in Nanjing,China显示文摘Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infections. This study aims to present the clinical characteristics of 24 cases with asymptomatic infection screened from close contacts and to show the transmission potential of asymptomatic COVID-19 virus carriers.Epidemiological investigations were conducted among all close contacts of COVID-19 patients(or suspected patients)in Nanjing,Jiangsu Province,China, from Jan 28 to Feb 9,2020,both in clinic and in community.Asymptomatic carriers were laboratory-confirmed positive for the COVID-19 virus by testing the nucleic acid of the pharyngeal swab samples.Their clinical records, laboratory assessments, and chest CT scans were reviewed. As a result, none of the 24 asymptomatic cases presented any obvious symptoms while nucleic acid screening.Five cases(20.8%)developed symptoms(fever, cough, fatigue, etc.)during hospitalization.Twelve(50.0%)cases showed typical CT images of ground-glass chest and 5(20.8%)presented stripe shadowing in the lungs. The remaining 7(29.2%)cases showed normal CT image and had no symptoms during hospitalization. These 7 cases were younger(median age:14.0 years;P=0.012)than the rest.None of the 24 cases developed severe COVID-19 pneumonia or died. The median communicable period, defined as the interval from the first day of positive nucleic acid tests to the first day of continuous negative tests, was 9.5 days(up to 21 days among the 24 asymptomatic cases).Through epidemiological investigation,we observed a typical asymptomatic transmission to the cohabiting family members, which even caused severe COVID-19 pneumonia.Overall, the asymptomatic carriers identified from close contacts were prone to be mildly ill during hospitalization. However, the communicable period could be up to three weeks and the communicated patients could develop severe illness. These results highlighted the importance of close contact tracing and longitudinally surveillance via virus nucleic acid tests.Further isolation recommendation and continuous nucleic acid tests may also be recommended to the patients discharged.Zhiliang Hu Ci Song Chuanjun Xu Guangfu Jin Yaling Chen Xin Xu Hongxia Ma Wei Chen Yuan Lin Yishan Zheng Jianming Wang Zhibin Hu Yongxiang Yi Hongbing Shen 2020Science China(Life Sciences)2020,63,5:77
Study of liver cirrhosis over ten consecutive years in Southern China显示文摘AIM: To investigate the etiology and complications of liver cirrhosis(LC) in Southern China. METHODS: In this retrospective, cross-sectional study, we identified cases of liver cirrhosis admitted between January 2001 to December 2010 and reviewed the medical records. Patient demographics, etiologies and complications were collected, and etiological changes were illustrated by consecutive years and within twotime periods(2001-2005 and 2006-2010). All results were expressed as the mean ± SD or as a percentage. The χ2 test or Student's t-test was used to analyze the differences in age, gender, and etiological distribution, and one-way analysis of variance was applied to estimate the trends in etiological changes. We analyzed the relationship between the etiologies and complications using unconditioned logistic regression, and the risk of upper gastrointestinal bleeding(UGIB) and hepatocellular carcinoma(HCC) in the major etiological groups was evaluated as ORs. A P value less than 0.05 was considered significant. Statistical computation was performed using SPSS 17.0 software.RESULTS: In this study, we identified 6719(83.16%) male patients and 1361(16.84%) female patients. The average age of all of the patients was 50.5 years at the time of diagnosis. The distribution of etiological agents was as follows: viral hepatitis, 80.62% [hepatitis B virus(HBV) 77.22%, hepatitis C virus(HCV) 2.80%,(HBV + HCV) 0.58%]; alcohol, 5.68%; mixed etiology, 4.95%; cryptogenic, 2.93%; and autoimmune hepatitis, 2.03%; whereas the other included etiologies accounted for less than 4% of the total. Infantile hepatitis syndrome LC patients were the youngest(2.5 years of age), followed by the metabolic LC group(27.2 years of age). Viral hepatitis, alcohol, and mixed etiology were more prevalent in the male group, whereas autoimmune diseases, cryptogenic cirrhosis, and metabolic diseases were more prevalent in the female group. When comparing the etiological distribution in 2001-2005 with that in 2006-2010, the proportion of viral hepatitis decreased from 84.7% to 78.3%(P < 0.001), and the proportion of HBV-induced LC also decreased from 81.9% to 74.6%(P < 0.001). The incidence of mixed etiology, cryptogenic cirrhosis, and autoimmune diseases increased by 3.1%(P < 0.001), 0.5%(P = 0.158), and 1.3%(P < 0.001), respectively. Alcohol-induced LC remained relatively steady over the 10-year period. The ORs of the development of UGIB between HBV and other major etiologies were as follows: HCV, 1.07; alcohol, 1.89; autoimmune, 0.90; mixed etiology, 0.83; and cryptogenic, 1.76. The ORs of the occurrence of HCC between HBV and other major etiologies were as follows: HCV, 0.54; alcohol, 0.16; autoimmune, 0.05; mixed etiology, 0.58; and cryptogenic, 0.60.CONCLUSION: The major etiology of liver cirrhosis in Southern China is viral hepatitis. However, the proportions of viral hepatitis and HBV are gradually decreasing. Alcoholic LC patients exhibit a greater risk of experiencing UGIB, and HBV LC patients may have a greater risk of HCC.Xing Wang Shang-Xiong Lin Jin Tao Xiu-Qing Wei Yuan-Ting Liu Yu-Ming Chen Bin Wu 2014World Journal of Gastroenterology2014,20,37:72

The ratio of normal saline to HES in the SH and SHG groups was 3:1. The glutamine (20% glutamine dipeptide, 100 mL/d) was supplemented into the resuscitation liquid in the SHG group. Complications and outcomes including respiratory and abdominal infection, sepsis, abdominal hemorrhage, intra-abdominal hypertension, abdominal compartment syndrome (ACS), renal failure, acute respiratory distress syndrome (ARDS), multiple organ dysfunction syndrome (MODS), operation intervention, length of intensive care unit stay, length of hospital stay, and mortality at 60 d were compared. Moreover, blood oxygen saturation (SpO 2 ), gastric intramucosal pH value (pHi), intra-abdominal pressure (IAP), inflammation cytokines, urine lactulose/mannitol (L/M) ratio, and serum endotoxin were investigated to evaluate the inflammatory reaction and gut barrier. RESULTS: Compared to the NS group, patients in the SH and SHG groups accessed the endpoint more quickly (3.9 ± 0.23 d and 4.1 ± 0.21 d vs 5.8 ± 0.25 d, P < 0.05) with less fluid volume (67.26 ± 28.53 mL/kg/d, 61.79 ± 27.61 mL/kg per day vs 85.23 ± 21.27 mL/kg per day, P < 0.05). Compared to the NS group, incidence of renal dysfunction, ARDS, MODS and ACS in the SH and SHG groups was obviously lower. Furthermore, incidence of respiratory and abdominal infection was significantly decreased in the SH and SHG groups, while no significant difference in sepsis was seen. Moreover, less operation time was needed in the SH and SHG group than the NS group, but the difference was not significant. The mortality did not differ significantly among these groups. Blood SpO 2 and gastric mucosal pHi in the SH and SHG groups increased more quickly than in the NS group, while IAP was significantly decreased in the SH and SHG group. Moreover, the serum tumor necrosis factor-α, interleukin-8 and C-reactive protein levels in the SH and SHG groups were obviously lower than in the NS group at each time point. Furthermore, urine L/M ratio and serum endotoxin were significantly lower in the SH group and further decreased in the SHG group.CONCLUSION: Results indicated that combination of normal saline, HES and glutamine are more efficient in resuscitation of SAP by relieving inflammation and sustaining the intestinal barrier.Gang Zhao Jun-Gang Zhang He-Shui Wu Jin Tao Qi Qin Shi-Chang Deng Yang Liu Lin Liu Bo Wang Kui Tian Xiang Li Shuai Zhu Chun-You Wang 2013World Journal of Gastroenterology2013,19,13:56
