1Inverse-designed photonic fibers and metasurfaces for nonlinear frequency conversion[Invited]显示文摘Typically, photonic waveguides designed for nonlinear frequency conversion rely on intuitive and established principles, including index guiding and bandgap engineering, and are based on simple shapes with high degrees of symmetry. We show that recently developed inverse-design techniques can be applied to discover new kinds of microstructured fibers and metasurfaces designed to achieve large nonlinear frequency-conversion efficiencies. As a proof of principle, we demonstrate complex, wavelength-scale chalcogenide glass fibers and gallium phosphide three-dimensional metasurfaces exhibiting some of the largest nonlinear conversion efficiencies predicted thus far,e.g., lowering the power requirement for third-harmonic generation by 104 and enhancing second-harmonic generation conversion efficiency by 107. 3Improvements and New Evaluation of NIM4 Cesium Fountain Clock and Construction of NIM5 Clock in 2006显示文摘The NIM4 cesium fountain clock has been operating stably and sub-continually since Aug. 2003. In this paper we present improvements on NIM4 in 2005-2006 and the most recent evaluation for its frequency shifts with an uncertainty of 5E-15. A 220 days comparison between NIM4 and GPS showed an agreement of 2E-14. 13A high-resolution route map reveals distinct stages of chondrocyte dedifferentiation for cartilage regeneration显示文摘Articular cartilage damage is a universal health problem.Despite recent progress,chondrocyte dedifferentiation has severely compromised the clinical outcomes of cell-based cartilage regeneration.Loss-of-function changes are frequently observed in chondrocyte expansion and other pathological conditions,but the characteristics and intermediate molecular mechanisms remain unclear.In this study,we demonstrate a time-lapse atlas of chondrocyte dedifferentiation to provide molecular details and informative biomarkers associated with clinical chondrocyte evaluation.We performed various assays,such as single-cell RNA sequencing(scRNA-seq),live-cell metabolic assays,and assays for transposase-accessible chromatin with high-throughput sequencing(ATAC-seq),to develop a biphasic dedifferentiation model consisting of early and late dedifferentiation stages.Early-stage chondrocytes exhibited a glycolytic phenotype with increased expression of genes involved in metabolism and antioxidation,whereas late-stage chondrocytes exhibited ultrastructural changes involving mitochondrial damage and stress-associated chromatin remodeling.Using the chemical inhibitor BTB06584,we revealed that early and late dedifferentiated chondrocytes possessed distinct recovery potentials from functional phenotype loss.Notably,this two-stage transition was also validated in human chondrocytes.An image-based approach was established for clinical use to efficiently predict chondrocyte plasticity using stage-specific biomarkers.Overall,this study lays a foundation to improve the quality of chondrocytes in clinical use and provides deep insights into chondrocyte dedifferentiation.Yishan Chen Yeke Yu Ya Wen Juan Chen Junxin Lin Zixuan Sheng Wenyan Zhou Heng Sun Chengrui An Jiansong Chen Weiliang Wu Chong Teng Wei Wei Hongwei Ouyang 2022Bone Research2022,10,3:1
14ASER:Animal Sex Reversal Database显示文摘Sex reversal,representing extraordinary sexual plasticity during the life cycle,not only triggers reproduction in animals but also affects reproductive and endocrine system-related diseases and cancers in humans.Sex reversal has been broadly reported in animals;however,an integrated resource hub of sex reversal information is still lacking.Here,we constructed a comprehensive database named ASER(Animal Sex Reversal)by integrating sex reversal-related data of 18 species from teleostei to mammalia.We systematically collected 40,018 published papers and mined the sex reversal-associated genes(SRGs),including their regulatory networks,from 1611 core papers.We annotated homologous genes and computed conservation scores for whole genomes across the 18 species.Furthermore,we collected available RNA-seq datasets and investigated the expression dynamics of SRGs during sex reversal or sex determination processes.In addition,we manually annotated 550 in situ hybridization(ISH),fluorescence in situ hybridization(FISH),and im-munohistochemistry(IHC)images of SRGs from the literature and described their spatial expression in the gonads.Collectively,ASER provides a unique and integrated resource for researchers to query and reuse organized data to explore the mechanisms and applications of SRGs in animal breeding and human health.The ASER database is publicly available at http://gffzz680c155e21ae46dbhwk59f5op0wb969f9.ffgz.tsg.suse.edu.cn/.Yangyang Li Zonggui Chen Hairong Liu Qiming Li Xing Lin Shuhui Ji Rui Li Shaopeng Li Weiliang Fan Haiping Zhao Zuoyan Zhu Wei Hu Yu Zhou Daji Luo 2021Genomics, Proteomics & Bioinformatics2021,19,6:0
15Cognitive disorder and changes in cholinergic receptors, N-methyl-D aspartate receptors, neural cell adhesion molecule, and brain-derived neurotrophic factor following brain injury显示文摘BACKGROUND: Learning and memory damage is one of the most permanent and the severest symptoms of traumatic brain injury; it can seriously influence the normal life and work of patients. Some research has demonstrated that cognitive disorder is closely related to nicotine cholinergic receptors, N-methyl-D aspartate receptors, neural cell adhesion molecule, and brain-derived neurotrophic factor. OBJECTIVE: To summarize the cognitive disorder and changes in nicotine cholinergic receptors, N-methyl-D aspartate receptors, neural cell adhesion molecule, and brain-derived neurotrophic factor following brain injury. RETRIEVAL STRATEGY: A computer-based online search was conducted in PUBMED for English language publications containing the Keywords 'brain injured, cognitive handicap, acetylcholine, N-methyl-D aspartate receptors, neural cell adhesion molecule, brain-derived neurotrophic factor' from January 2000 to December 2007. There were 44 papers in total. Inclusion criteria: ① articles about changes in nicotine cholinergic receptors, N-methyl-D aspartate receptors, neural cell adhesion molecule, and brain-derived neurotrophic factor following brain injury; ② articles in the same researching circle published in authoritative journals or recently published. Exclusion criteria: duplicated articles. LITERATURE EVALUATION: References were mainly derived from research on changes in these four factors following brain injury. The 20 included papers were clinical or basic experimental studies. DATA SYNTHESIS: After craniocerebral injury, changes in these four factors in brain were similar to those during recovery from cognitive disorder, to a certain degree. Some data have indicated that activation of nicotine cholinergic receptors, N-methyl-D aspartate receptors, neural cell adhesion molecule, and brain-derived neurotrophic factor could greatly improve cognitive disorder following brain injury. However, there are still a lot of questions remaining; for example, how do these factors change at different time points after brain injury, and what is the relationship between associated factors and cognitive disorder. 16Recombinant expression, biophysical and functional characterization of ClpS from Mycobacterium tuberculosis显示文摘Intracellular proteolysis is attracting more and more attention for its unique and important character in Mycobacterium tuberculosis(Mt).The ClpS protein from Mt(MtClpS)plays a critical role in intracellular proteolysis by recognizing N-end rule substrates,which makes it become a potential target for antibacterial drugs.However,the molecular mechanism of MtClpS recognizing N-end rule substrates remains unclear.Preparation of highly concentrated and pure MtClpS protein is a prerequisite for further structural and functional studies.In the present work,we tried several fusion tags and various expression conditions to maximize the production of MtClpS in Escherichia coli.We established an efficient approach for preparing the MtClpS protein with a high yield of 24.7 mg/l and a high purity of 98%.After buffer screening,we obtained a stable MtClpS protein sample concentrated at 0.63 mM in the presence of glycerol,l-Arginine,and l-Glutamate.Moreover,circular dichroism characterization indicated that the secondary structure of MtClpS consists of 38%α-helix and 24%β-sheet.The 2D 1H-15N HSQC nuclear magnetic resonance spectrum showed a good dispersion of resonance peaks with uniform intensity,indicating that the purified MtClpS protein was well folded and conformationally homogeneous.Isothermal titration calorimetry experiments revealed significant interactions of MtClpS with N-end rule peptides beginning with Leu,Tyr,Trp,or Phe.Furthermore,residues D34,D35,and H66 were confirmed as key residues for MtClpS recognizing the N-end rule peptide.The successful expression and biophysical characterization of MtClpS enabled us to gain insight into the molecular mechanism of MtClpS recognizing N-end rule substrates.The obtained stable and pure recombinant MtClpS will enable future inhibitor screening experiments.Chenyun Guo Yihang Xiao Fangkai Bs Weiliang Lin Huilin Wang Hongwei Yao Donghai Lin 2019Acta Biochimica et Biophysica Sinica2019,51,11:0

18Addressing the Chengjiang conundrum: A palaeoecological view on the rarity of hurdiid radiodonts in this most diverse early Cambrian Lagerstatte显示文摘Over one hundred arthropod fossil species have been described from the famous Chengjiang Lagerst?tte(South China, Cambrian Stage 3, ca. 518 Ma) including a diverse assemblage of radiodonts–a group containing Anomalocaris and its relatives. These iconic stem-group euarthropods include some of the largest animals of the time, and some are known from hundreds of specimens. A longstanding conundrum has been the rarity or absence of hurdiids from Cambrian Series 2 Lagerst?tten like Chengjiang. This is because radiodonts are generally common in such deposits and the oldest radiodont ever discovered is a hurdiid. Furthermore, this family displays the widest geographic and temporal ranges of all radiodont families, and the highest diversity. Here we document the first hurdiid frontal appendages from Chengjiang, which display unique features within the family and may provide insights for understanding the character evolution of hurdiid appendages. The palaeoenvironmental distribution of hurdiids suggests that the rarity of hurdiids in Chengjiang may be due to a preference for deeper water environments,and the later success of this family from the Wuliuan onwards may relate to their ability to tolerate cooler water temperatures than other radiodont families.
