1Abscisic Acid-mediated Epigenetic Processes in Plant Development and Stress Responses显示文摘Abscisic acid (ABA) regulates diverse plant processes, growth and development under non-stress conditions and plays a pivotal role in abiotic stress tolerance. Although ABA-regulated genetic processes are well known, recent discoveries reveal that epigenetic processes are an integral part of ABA-regulated processes. Epigenetic mechanisms, namely, histone modifications and cytosine DNA methylation-induced modification of genome give rise to epigenomes, which add diversity and complexity to the genome of organisms. Histone monoubiquitination appears to regulate ABA levels in developing seeds through histone H2B monoubiquitination. ABA and H2B ubiquitination dependent chromatin remodeling regulate seed dormancy. Transcription factor networks necessary for seed maturation are repressed by histone deacetylases (HDACs)-dependent and PICKLE chromatin remodeling complexes (CRCs), whereas ABA induces the expression of these genes directly or through repression of HDACs. Abiotic stress-induced ABA regulates stomatal response and stress-responsive gene expression through HDACs and HOS15-dependent histone deacetylation, as well as through the ATP-dependent SWITCH/SUCROSE NONFERMENTING CRC. ABA also probably regulates the abiotic stress response through DNA methylation and short interfering RNA pathways. 4RNA-directed DNA methylation and demethylation in plants显示文摘RNA-directed DNA methylation (RdDM) is a nuclear process in which small interfering RNAs (siRNAs) direct the cytosine methylation of DNA sequences that are complementary to the siRNAs. In plants, double stranded-RNAs (dsRNAs) generated by RNA-dependent RNA polymerase 2 (RDR2) serve as precursors for Dicer-like 3 dependent biogenesis of 24-nt siRNAs. Plant specific RNA polymerase IV (Pol IV) is presumed to generate the initial RNA transcripts that are substrates for RDR2. siRNAs are loaded onto an argonaute4-containing RISC (RNA-induced silencing complex) that targets the de novo DNA methyltransferase DRM2 to RdDM target loci. Nascent RNA transcripts from the target loci are generated by another plant-specific RNA polymerase, Pol V, and these transcripts help recruit com- plementary siRNAs and the associated RdDM effector complex to the target loci in a transcrip- tion-coupled DNA methylation process. Small RNA binding proteins such as ROS3 may direct tar- get-specific DNA demethylation by the ROS1 family of DNA demethylases. Chromatin remodeling en- zymes and histone modifying enzymes also participate in DNA methylation and possibly demethylation. One of the well studied functions of RdDM is transposon silencing and genome stability. In addition, RdDM is important for paramutation, imprinting, gene regulation, and plant development. Lo- cus-specific DNA methylation and demethylation, and transposon activation under abiotic stresses suggest that RdDM is also important in stress responses of plants. 5A group of SUVH methyl-DNA binding proteins regulate expression of the DNA demethylase ROS_1 in Arabidopsis显示文摘DNA methylation is typically regarded as a repressive epigenetic marker for gene expression.Genome-wide DNA methylation patterns in plants are dynamically regulated by the opposing activities of DNA methylation and demethylation reactions. In Arabidopsis, a DNA methylation monitoring sequence(MEMS) in the promoter of the DNA demethylase gene ROS_1 functions as a methylstat that senses these opposing activities and regulates genome DNA methylation levels by adjusting ROS_1 expression. How DNA methylation in the MEMS region promotes ROS_1 expression is not known. Here, we show that several Su(var)3-9 homologs(SUVHs) can sense DNA methylation levels at the MEMS region and function redundantly to promote ROS_1 expression. The SUVHs bind to the MEMS region, and the extent of binding is correlated with the methylation level of the MEMS.Mutations in the SUVHs lead to decreased ROS_1 expression, causing DNA hypermethylation at more than 1,000 genomic regions. 6ROP11 GTPase Negatively Regulates ABA Signaling by Protecting ABI1 Phosphatase Activity from Inhibition by the ABA Receptor RCAR1/PYL9 in Arabidopsis显示文摘The phytohormone abscisic acid (ABA) regulates many key processes in plants, such as seed germina- tion, seedling growth, and abiotic stress tolerance. In recent years, a minimal set of core components of a major ABA signaling pathway has been discovered. These components include a RCAR/PYR/PYL family of ABA receptors, a group of PP2C phosphatases, and three SnRK2 kinases. However, how the interactions between the receptors and their targets are regulated by other proteins remains largely unknown. In a companion paper published in this issue, we showed that ROP11, a member of the plant- specific Rho-like small GTPase family, negatively regulates multiple ABA responses in Arabidopsis. The current work demonstrated that the constitutively active ROP11 (CA-ROP11) can modulate the RCAR1/PYL9-mediated ABA signaling pathway based on reconstitution assays in Arabidopsis thaliana protoplasts. Furthermore, using luciferase complementation imaging, yeast two-hybrid assays, co- immunoprecipitation assays in Nicotiana benthamiana and bimolecular fluorescence complementation assays, we demonstrated that CA-ROP11 directly interacts with ABI1, a signaling component downstream of RCAR1/PYL9. Finally, we provided biochemical evidence that CA-ROP11 protects ABI1 phosphatase activity from inhibition by RCAR1/PYL9 and thus negatively regulates ABA signaling in plant cells. 7Inositol Pyrophosphate lnsP8 Acts as an Intracellular Phosphate Signal in Arabidopsis显示文摘The maintenance of cellular phosphate(Pi)homeostasis is of great importance in living organisms.The SPX domain-containing protein 1(SPX1)proteins from both Arabidopsis and rice have been proposed to act as sensors of Pi status.The molecular signal indicating the cellular Pi status and regulating Pi homeostasis in plants,however,remains to be identified,as Pi itself does not bind to the SPX domain.Here,we report the identification of the inositol pyrophosphate lnsP8 as a signaling molecule that regulates Pi homeostasis in Arabidopsis.Polyacrylamide gel electrophoresis profiling of InsPs revealed that lnsP8 level positively cor­relates with cellular Pi concentration.We demonstrated that the homologs of diphosphoinositol pentaki-sphosphate kinase(PPIP5K),VIH1 and VIH2,function redundantly to synthesize lnsP8,and that the vih1 vih2 double mutant overaccumulates Pi.SPX1 directly interacts with PHR1,the central regulator of Pi star­vation responses,to inhibit its function under Pi-replete conditions.However,this interaction is compro­mised in the vih1 vih2 double mutant,resulting in the constitutive induction of Pi starvation-induced genes,indicating that plant cells cannot sense cellular Pi status without lnsP8.Furthermore,we showed that lnsP8 could directly bind to the SPX domain of SPX1 and is essential for the interaction between SPX1 and PHR1.Collectively,our study suggests that lnsP8 is the intracellular Pi signaling molecule serving as the ligand of SPX1 for controlling Pi homeostasis in plants.Jinsong Dong Guojie Ma Liqian Sui Mengwei Wei Viswanathan Satheesh Ruyue Zhang Shenghong Ge Jinkai Li Tong-En Zhang Christopher Wittwer Henning JJessen Huiming Zhang Guo-Yong An Dai-Yin Chao Dong Liu Mingguang Lei 2019Molecular Plant2019,12,11:8
8Electrochemical behavior of Co-Cr and Ni-Cr dental cast alloys显示文摘The cast structures influencing the electrochemical corrosion behavior of Co-Cr and Ni-Cr dental alloys were studied using potentiodynamic polarization and AC impedance in 0.9% (mass fraction) NaCl solution at (37±1) ℃. The phase and microstructure of the alloys that were fabricated using two different casting methods viz. centrifugal casting and high frequency induction casting, were examined using X-ray diffraction analysis, scanning electron microscopy and energy dispersive spectroscopy. The roles of alloying elements and the passive film homogeneity on the corrosion resistance of Co-Cr-Mo and Ni-Cr-Mo dental cast alloys were reviewed. The results of electrochemical study show that the dependence of corrosion resistance on the microstructure associated with the casting methods is marginal. The Co-Cr alloy exhibits more desirable corrosion resistance properties than the Ni-Cr alloy. 16Osa-miR167d facilitates infection of Magnaporthe oryzae in rice显示文摘MicroRNAs(miRNAs)play important roles in rice response to Magnaporthe oryzae,the causative agent of rice blast disease.Studying the roles of rice miRNAs is of great significance for the disease control.Osa-miR167d belongs to a conserved miRNA family targeting auxin responsive factor(ARF)genes that act in developmental and stress-induced responses.Here,we show that OsamiR167d plays a negative role in rice immunity against M.oryzae by suppressing its target gene.The expression of Osa-miR167d was significantly suppressed in a resistant accession at and after 24 h post inoculation(hpi),however,its expression was significantly increased at 24 hpi in the susceptible accession upon M.oryzae infection.Transgenic rice lines over-expressing Osa-miR167d were highly susceptible to multiple blast fungal strains.By contrast,transgenic lines expressing a target mimicry to block OsamiR167d enhanced resistance to rice blast disease.In addition,knocking out the target gene ARF12 led to hypersusceptibility to multiple blast fungal strains.Taken together,our results indicate that Osa-miR167d negatively regulate rice immunity to facilitate the infection of M.oryzae by downregulating ARF12.Thus,Osa-miR167d-ARF12 regulatory module could be valuable in improvement of blast-disease resistance.Zhi-Xue Zhao Qin Feng Xiao-Long Cao Yong Zhu He Wang Viswanathan Chandran Jing Fan Ji-Qun Zhao Mei Pu Yan Li Wen-Ming Wang 2020Journal of Integrative Plant Biology2020,62,5:3
17Protective effect of Cardiospermum halicacabum leaf extract on glycoprotein components on STZ-induced hyperglycemic rats显示文摘Objective:To investigate the protective role of Cardiospermum halicacabum(C.halicacabum) leaf extract on glycoprotein metabolism in streptozotocin(STZ)-induced diabetic rats.Methods: Diabetes was induced in male albino Wistar rats by intraperitonial administration of STZ.The C.halicacabum leaf extract(CHE) was administered orally to normal and STZ-diabetic rats for 45 days.The effects of C.halicacabum leaf extract(CHE) on plasma and tissue glycoproteins (hexose,hexosamine,fucose and sialic acid) were determined.Results:The levels of plasma and tissues glycoproteins containing hexose,hexosamine and fucose were significantly increased in STZ-induced diabetic rats.In addition,the level of sialic acid significantly increased in plasma and liver while decreased in kidney of STZ-induced diabetic rats.After administration of CHE to diabetic rats,the metabolic alteration of glycoprotein reverted towards normal levels.
