| 1 | S-Allylmercaptocysteine improves alcoholic liver disease partly through a direct modulation of insulin receptor signaling显示文摘Alcoholic liver disease(ALD) causes insulin resistance, lipid metabolism dysfunction, and inflammation. We investigated the protective effects and direct regulating target of S-allylmercaptocysteine(SAMC) from aged garlic on liver cell injury. A chronic ethanol-fed ALD in vivo model(the NIAAA model) was used to test the protective functions of SAMC. It was observed that SAMC(300 mg/kg, by gavage method) effectively ameliorated ALD-induced body weight reduction, steatosis,insulin resistance, and inflammation without affecting the health status of the control mice, as demonstrated by histological, biochemical, and molecular biology assays. By using biophysical assays and molecular docking, we demonstrated that SAMC directly targeted insulin receptor(INSR) protein on the cell membrane and then restored downstream IRS-1/AKT/GSK3 b signaling. Liver-specific knock-down in mice and siRNA-mediated knock-down in AML-12 cells of Insr significantly impaired SAMC(250 mmol/L in cells)-mediated protection. Restoration of the IRS-1/AKT signaling partly recovered hepatic injury and further contributed to SAMC’s beneficial effects. Continuous administration of AKT agonist and recombinant IGF-1 in combination with SAMC showed hepato-protection in the mice model.Long-term(90-day) administration of SAMC had no obvious adverse effect on healthy mice. | 2 | TRIB3-GSK-3β interaction promotes lung fibrosis and serves as a potential therapeutic target显示文摘Pulmonary fibrosis(PF)is a chronic,progressive,fatal interstitial lung disease with limited available therapeutic strategies.We recently reported that the protein kinase glycogen synthase kinase-3β(GSK-3β)interacts with and inactivates the ubiquitin-editing enzyme A20 to suppress the degradation of the transcription factor CCAAT/enhancer-binding protein beta(C/EBPβ)in alveolar macrophages(AMs),resulting in a profibrotic phenotype of AMs and promoting the development of PF.Here,we showed that chronic lung injury upregulated the stress response protein tribbles homolog 3(TRIB3),which interacted with GSK-3βand stabilized GSK-3βfrom ubiquitination and degradation.Elevated GSK-3βexpression phosphorylated A20 to inhibit its ubiquitin-editing activity,causing the accumulation of C/EBPβand the production of several profibrotic factors in AMs and promoting PF development.Activated C/EBPβ,in turn,increased the transcription of TRIB3 and GSK-3β,thereby establishing a positive feedback loop in AMs.The knockdown of TRIB3 expression or the pharmacologic disruption of the TRIB3-GSK-3βinteraction was an effective PF treatment.Our study reveals an intact profibrotic axis of TRIB3-GSK-3β-A20-C/EBPβin AMs,which represents a target that may provide a promising treatment strategy for PF. | Shanshan Liu Xiaoxi Lv Xupeng Wei Chang Liu Qiao Li Jiali Min Fang Hua Xiaowei Zhang Ke Li Pingping Li Yang Xiao Zhuowei Hu Bing Cui | 2021 | Acta Pharmaceutica Sinica B2021,11,10: | 2 |
| 3 | A preliminary report on adjuvant analgesic efficacy of HANS in opioid tolerant patients with cancer pain显示文摘Objective: To observe the adjuvant analgesic efficacy of Han's Acupoint Nerve Stimulator(HANS) in opioid tolerant patients with cancer pain. Methods: A prospective non-controlled study was conducted. Opioid tolerant patients with cancer pain were enrolled and treated with both routinely analgesics and adjuvant HANS(2/100 Hz for 30 min/d, 5 d on and 2 d off for two weeks). Cancer pain, quality of life(QOL), anxiety and depression were assessed before enrollment and on d 8 and d 15 with the BPI-C, EORTC QLQ-C30, and self-rating anxiety scale(SAS)/self-rating depression scale(SDS), respectively; the therapeutic frequency of breakthrough pain(BP) and daily opioid dose were also recorded. Results: Totally 47 patients meeting the inclusion criteria participated in this study; 43 patients completed the two-week treatment and assessment. The mean scores of patient's 'worst' and 'least' pain intensity assessed with BPI-C decreased significantly on d 8 and d 15; the therapeutic frequency of BP also significantly decreased; but the average daily dose of opioids did not change significantly. This paper presents the design, characterization, and realization of powder metallurgy processed metal-matrix friction materials intended for the above-mentioned tribological brakes. Selecting appropriate ingredients, which provides an effective way to tailor the properties of the friction material, is evolving as a strategy to meet the design requirements. The tribological behaviors of the friction material are experimentally investigated under different conditions, and special attention is focused on the vacuum tribology. Examinations and analyses of the friction surface and subsurface corroborate the wear mechanism. In addition, the erosion resistances of the friction material are evaluated by exposure tests of ultraviolet irradiation and atomic oxygen. | 11 | Efficient genome editing in Claviceps purpurea using a CRISPR/Cas9 ribonucleoprotein method显示文摘Claviceps purpurea produces many pharmacologically important ergot alkaloids(EAS),which are widely used to treat migraine and hypertension and to aid childbirth.Although an EAS biosynthetic cluster of C.purpurea has been discovered more than 20 years ago,the complete biosynthetic pathway of EAS has not been fully characterized until now.The main obstacle to elucidating this pathway and strain modification is the lack of efficient genome-editing tools for C.purpurea.The conventional gene manipulation method for C.purpurea relies on homologous recombination(HR),although the efficiency of HR in C.purpurea is very low(~1-5%).Consequently,the disruption of target genes is laborious and time-consuming.Although CRISPR/Cas9 genome-editing methods based on in vivo Cas9 expression and gRNA transcription have been reported recently,their gene-disruption efficiency is still very low.Here,we developed an efficient genome-editing system in C.purpurea based on in vitro assembled CRISPR/Cas9 gRNA ribonucleoprotein complexes.As proof of principle,three target genes were efficiently knocked out using this CRISPR/Cas9 ribonucleoprotein complex-mediated HR system,with editing efficiencies ranging from 50%to 100%.Inactivation of the three genes,which are closely related to uridine biosynthesis(ura5),hypha morphology(rac),and EAS production(easA),resulted in a uridine auxotrophic mutant,a mutant with a drastically different phenotype in axenic culture,and a mutant that did not produce EAS,respectively.Our ribonucleoprotein-based genome-editing system has a great advantage over conventional and in vivo CRISPR/Cas9 methods for genome editing in C.purpurea,which will greatly facilitate elucidation of the EAS biosynthetic pathway and other future basic and applied research on C.purpurea. | Lu Yu Meili Xiao Zhihua Zhu Yinmei Wang Zhihua Zhou Pingping Wang Gen Zou | 2022 | Synthetic and Systems Biotechnology2022,7,2: | 1 |

sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of | 18 | Tin transport and cassiterite precipitation from hydrothermal fluids显示文摘Cassiterite(SnO_(2))is the main ore mineral of tin in magmatic-hydrothermal tin deposits,but tin transport and precipitation mechanisms from hydrothermal fluids remain poorly understood.We critically evalu-ated aqueous tin speciation in hydrothermal fluids from extensive experimental data and thermody-namic modeling.Sn(II)chloride complexes in hydrothermal fluids exist mainly as SnCl^(+),SnCl_(2)(aq),and SnCl_(3).The revised Helgeson-Kirkham-Flowers model parameters of these three tin species and two tin ions(Sn^(4+) and Sn^(2+))were derived from the correlation algorithms among these parameters,and the standard molar properties of cassiterite were optimized to be internally consistent with the available thermodynamic dataset.These thermodynamic parameters,together with the available equilibrium con-stant equation of Sn(IV)chloride complexes,could reproduce the available solubility data of cassiterite in acidic solutions at 400-700℃under oxygen fugacity(f_(o2))levels buffered by hematite-magnetite(HM)or nickel-nickel oxide(NNO).These comparisons allow modeling chemical systems of SnO_(2)-NaCl-HCI-H_(2)O(liquid phase)to examine tin transport and cassiterite precipitation mechanisms under tin-mineralizing conditions:300--500℃,50-150 MPa,2 molal NaCI,and fo。levels from QFM(quartz-fayalite-magnetite)to HM.Sn(I)chloride complexes are commonly interpreted to dominate in aqueous tin speciation under f_(o2)=NNO,but our modeling results indicate that considerable contents of Sn(IV)chloride complexes also exist in those reduced fluids with high HCI contents,consistent with recent in situ high-temperature experiments and molecular dynamic simulations.The Sn(I)/Sn(IV)ratios in fluids depends onfo,temperature,and HCl contents.A considerable amount of Sn(IV)possibly exist in an early mineralization stage even under f_(o2)=NNO;if so,redox reactions are unnecessary to precipitate cassiterite from these mineralizing fluids.We find that even if the f_(o2) levels are constant,simple cooling can alter mineralizing fluids to be more oxidized(e.g.,from QFM to HM)and cause cassiterite precipitation,indicating that oxidizing agents are not necessary as previously thought.This explains why cassiterite can precipitate in host rocks(e.g.,sandstone or quartzite)that do not provide oxidizing agents.A simple rise in f_(o2),levels and pH neutralization(e.g.,greisenization)also cause cassiterite precipitation.Cassiterite solubility in oxidized acidic hydrothermal fluids(NNO | Xiangchong Liu Pingping Yu Changhao Xiao | 2023 | Geoscience Frontiers2023,14,6: | 0 |
