3Geochemistry and Zircon U–Pb–Hf Isotopic Systematics of the Sanchahe Quartz Monzonite Intrusion in the North Qinling Tectonic Zone, Central China: Implications for its Petrogenesis and Tectonic Setting显示文摘The Sanchahe quartz monzonite intrusion is situated in the middle segment of the North Qinling tectonic belt, Central China mainland, and consists chiefly of sanukitoid–like and granodioritic-monzogranitic rocks. The sanukitoid–like rocks are characterized by quartz monzonites, which display higher Mg#(55.0–59.0), and enrichments in Na2 O+K2 O(7.28–8.94 %), Ni(21-2312 ppm), Cr(56-4167 ppm), Sr(553-923 ppm), Ba(912-1355 ppm) and LREE((La/Yb)N =9.47–15.3), from negative to slightly positive Eu anomalies(δEu=+0.61 to +1.10), but also depletion in Nb, Ta and Ti. The granodioritic-monzogranitic rocks diaplay various Mg#of 6.00-53.0, high Na2 O+K2 O(7.20– 8.30%), Sr(455–1081 ppm) and(La/Yb)N(27.6–47.8), with positive Eu anomalies(δEu=1.03–1.57) and depleted Nb, Ta and Ti. Laser ablation inductively coupled plasma mass spectrometry(LA-ICPMS) zircon U-Pb isotopic dating reveals that the sanukitoid-like rocks were emplaced at two episodes of magmatism at 457±3 Ma and 431±2 Ma, respectively. The monzogranites were emplaced at 445±7Ma. Sanukitoid–like rocks have their εHf(t) values ranging from +0.3 to +15.1 with Hf–depleted mantle model ages of 445 to 1056 Ma, and the monzogranite shows its εHf(t) values ranging from 21.6 to +10.8 with Hf–depleted mantle model ages of 635 to 3183 Ma. Petrological, geochemical and zircon Lu –Hf isotopic features indicate that the magmatic precursor of sanukitoid–like rocks was derived from partial melting of the depleted mantle wedge materials that were metasomatized by fluids and melts related to subduction of oceanic slab, subsequently the sanukitoid magma ascended to crust level. This emplaced mantle magma caused partial melting of crustally metamorphosed sedimentary rocks, and mixing with the crustal magma, and suffered fractional crystallization, which lead to formations of quartz monzonites. However, the magmatic precursor of the granodioritic-monzogranitic rocks were derived from partial melting of subducted oceanic slab basalts. 4Target-triggered inhibiting oxidase-mimicking activity of platinum nanoparticles for ultrasensitive colorimetric detection of silver ion显示文摘The development of efficient methods for the detection of hazardous and toxic elements is extremely important for environmental security and public health. In this work, we developed a facile colorimetric assaying system for Ag+ detection in aqueous solution. Chitosan-stabilized platinum nanoparticles(ChPtNPs) were synthesized and severed as an artificial oxidase to catalyze the oxidation of the substrate3,30,5,50-tetramethylbenzidine(TMB) and generate color signal. In the presence of Ag+, due to the strong metallophilic interactions between Ag+ and Pt2+ on the surface of Ch-PtNPs, Ag+ can weaken the affinity to the substrates and inactivate the catalytic activity of Ch-PtNPs, leading to decreased absorbance signal to varying degrees depending on Ag+ amount. Combing the specific binding between Ch-PtNPs and Ag+ with signal amplification procedure based on the Ch-PtNPs-catalyzed TMB oxidation, a sensitive,selective, simple, cost-effective, and rapid detection method for Ag+ can be realized. Ag+ ions in tap and lake waters have been successfully detected. 5Beta-and Novel Delta-Coronaviruses Are Identified from Wild Animals in the Qinghai-Tibetan Plateau, China显示文摘Outbreaks of severe virus infections with the potential to cause global pandemics are increasingly concerning. One type of those commonly emerging and re-emerging pathogens are coronaviruses(SARS-Co V, MERS-Co V and SARS-Co V-2).Wild animals are hosts of different coronaviruses with the potential risk of cross-species transmission. However, little is known about the reservoir and host of coronaviruses in wild animals in Qinghai Province, where has the greatest biodiversity among the world's high-altitude regions. Here, from the next-generation sequencing data, we obtained a known beta-coronavirus(beta-Co V) genome and a novel delta-coronavirus(delta-Co V) genome from faecal samples of 29 marmots, 50 rats and 25 birds in Yushu Tibetan Autonomous Prefecture, Qinghai Province, China in July 2019. According to the phylogenetic analysis, the beta-Co V shared high nucleotide identity with Coronavirus HKU24. Although the novel delta-Co V(Mt Co V) was closely related to Sparrow deltacoronavirus ISU42824, the protein spike of the novel delta-Co V showed highest amino acid identity to Sparrow coronavirus HKU17(73.1%). Interestingly, our results identified a novel host(Montifringilla taczanowskii) for the novel delta-Co V and the potential cross-species transmission. The most recent common ancestor(t MRCA) of Mt Co Vs along with other closest members of the species of Coronavirus HKU15 was estimated to be 289 years ago. 6Evaluation of Heavy Metals and Pesticide Residues in Dendrobium officinale Produced in Guangxi显示文摘[Objectives]To establish a method for the determination of heavy metals and pesticide residues in Dendrobium officinale produced in Guangxi and its culture substrate.[Methods]The contents of five heavy metals(Pb,Cd,As,Hg and Cu)were determined by the atomic absorption spectrophotometric(AAS)method;residues of 9 organochlorine pesticides(α-BHC,β-BHC,γ-BHC,δ-BHC,p,p′-DDT,p,p′-DDE,p,p′-DDD,o,p′-DDT and pentachloronitrobenzene)were determined by gas chromatography(GC).[Results]The contents of five heavy metals in the medicinal materials of D.officinale produced in Guangxi were 0.96,0.21,0.021 mg/kg,and not detected,and 0.35 mg/kg.The contents of five heavy metals in the culture substrate were 0.03,0.19,0.21,0.004 and 1.80 mg/kg,respectively;the content of 9 organochlorine pesticide residues in the sample was not detected.[Conclusions]The detection results of heavy metal content and organochlorine pesticide residues in D.officinale produced in Guangxi and its substrate are consistent with the limit requirements of the Chinese Pharmacopoeia.This experiment is expected to provide reliable data reference for ensuring the safety of the drug resources of D.officinale in Guangxi.Hua ZHU Ruiting HE Piaoling HUANG Wenqi YANG Anda WEI Yuanjing TAO Zhonghua DAI 2019Medicinal Plant2019,10,5:3
7Effective suppression of the photodarkening effect in high-power Yb-doped fiber amplifiers by H2 loading显示文摘The radical suppression of the photodarkening effect and laser performance deterioration via H2 loading were demonstrated in high-power Yb-doped fiber(YDF)amplifiers.The photodarkening loss at equilibrium was114.4 d B/m at 702 nm in the pristine fiber,while it vanished in the H2-loaded fiber.To obtain a deeper understanding of the impact of photodarkening on laser properties,the evolution of the mode instability threshold and output power in fiber amplifiers was investigated.After pumping for 300 min,the mode instability threshold of the pristine fiber dropped from 770 to 612 W,and the periodic fluctuation of the output power became intense,finally reaching 100 W.To address the detrimental effects originating from photodarkening,H2 loading was applied in contrast experiments.The output power remained stable,and no sign of mode instability was observed in the H2-loaded fiber.Moreover,the transmittance at 638 nm confirmed the absence of the photodarkening effect.The results pave the way for the further development of high-power fiber lasers.RUITING CAO GUI CHEN YISHA CHEN ZHILUN ZHANG XIANFENG LIN BIN DAI LUYUN YANG JINYAN LI 2020Photonics Research2020,8,3:2

9Dual-AAV split prime editor corrects the mutation and phenotype in mice with inherited retinal degeneration显示文摘The prime editor(PE)can edit genomes with almost any intended changes,including all 12 possible types of base substitutions,small insertions and deletions,and their combinations,without the requirement for double strand breaks or exogenous donor templates.PE demonstrates the possibility of correcting a variety of disease-causing mutations and might expand the therapeutic application of gene editing.In this study,PE was optimized based on a dual-adeno-associated virus(AAV)split-intein system in vitro by screening different split sites and split inteins.We found that splitting PE before amino acid 1105(Ser)of SpCas9 with Rma intein resulted in the highest on-target editing.The orientations of pegRNA and nicking sgRNA in the AAV vector were further optimized.To test the in vivo performance of the optimized dual-AAV split-PE3,it was delivered by subretinal injection in rd12 mice with inherited retinal disease Leber congenital amaurosis.The prime editors corrected the pathogenic mutation with up to 16%efficiency in a precise way,with no detectable off-target edits,restored RPE65 expression,rescued retinal and visual function,and preserved photoceptors.Our findings establish a framework for the preclinical development of PE and motivate further testing of PE for the treatment of inherited retinal diseases caused by various mutations.Kaiqin She Yi Liu Qinyu Zhao Xiu Jin Yiliu Yang Jing Su Ruiting Li Li Song Jianlu Xiao Shaohua Yao Fang Lu Yuquan Wei Yang Yang 2023Signal Transduction and Targeted Therapy2023,8,3:1
10Comparison of caspase-3 activation in tumor cells upon treatment of chemotherapeutic drugs using capillary electrophoresis显示文摘Caspases play important roles in cell apoptosis.Meas-urement of the dynamics of caspase activation in tumor cells not only facilitates understanding of the molecular mechanisms of apoptosis but also contributes to the development,screening,and evaluation of anticancer drugs that target apoptotic pathways.The fluorescence resonance energy transfer(FRET)technique provides a valuable approach for defining the dynamics of apop-tosis with high spatio-temporal resolution.However,FRET generally functions in the single-cell level and becomes ineffective when applied in the high throughput detection of caspase activation.In the cur-rent study,a FRET sensor was combined with capillary electrophoresis(CE)to achieve a high throughput method for cellular caspase detection.The FRET-based CE system is composed of a homemade CE system and a laser source for detecting the dynamics of caspase-3 in various cells expressing sensors of caspase-3 that have been treated with anticancer drugs,such as cell cycle-independent drug cisplatin and specific cell cycle drugs camptothecin and etoposide,as well as their combination with tumor necrosis factor(TNF).A posi-tive correlation between the caspase-3 activation ve-locity and drug concentration was observed when the cells were treated with cisplatin,but cells induced by camptothecin and etoposide did not show any apparent correlation with their concentrations.Moreover,differ-ent types of cells presented distinct sensitivities under the same drug treatment,and the combination treat-ment of TNF and anticancer drugs significantly accel-erated the caspase-3 activation process.Its high throughput capability and detection sensitivity make the FRET-based CE system a useful tool for investi-gating the mechanisms of anticancer drugs and anti-cancer drug screening.Shuang Sha Honglin Jin Xiao Li Jie Yang Ruiting Ai Jinling Lu 2012Protein & Cell2012,3,5:1
11Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 12Improving antibody affinity through in vitro mutagenesis in complementarity determining regions显示文摘High-affinity antibodies are widely used in diagnostics and for the treatment of human diseases.However,most antibodies are isolated from semi-synthetic libraries by phage display and do not possess in vivo affinity maturation,which is triggered by antigen immunization.It is therefore necessary to engineer the affinity of these antibodies by way of in vitro assaying.In this study,we optimized the affinity of two human monoclonal antibodies which were isolated by phage display in a previous related study.For the 42A1 antibody,which targets the liver cancer antigen glypican-3,the variant T57H in the second complementarity-determining region of the heavy chain(CDR-H2)exhibited a 2.6-fold improvement in affinity,as well as enhanced cell-binding activity.For the I4A3 antibody to severe acute respiratory syndrome coronavirus 2,beneficial single mutations in CDR-H2 and CDR-H3 were randomly combined to select the best synergistic mutations.Among these,the mutation S53P-S98T improved binding affinity(about 3.7 fold)and the neutralizing activity(about 12 fold)compared to the parent antibody.Taken together,single mutations of key residues in antibody CDRs were enough to increase binding affinity with improved antibody functions.The mutagenic combination of key residues in different CDRs creates additive enhancements.Therefore,this study provides a safe and effective in vitro strategy for optimizing antibody affinity.Wei Ye Xiaoyu Liu Ruiting He Liming Gou Ming Lu Gang Yang Jiaqi Wen Xufei Wang Fang Liu Sujuan Ma Weifeng Qian Shaochang Jia Tong Ding Luan Sun Wei Gao 2022The Journal of Biomedical Research2022,36,3:0
13Extracorporeal membrane oxygenation in adult patients with sepsis and septic shock: Why, how, when, and for whom显示文摘Sepsis and septic shock remain the leading causes of death in intensive care units.Some patients with sepsis fail to respond to routine treatment and rapidly progress to refractory respiratory and circulatory failure,necessitating extracorporeal membrane oxygenation(ECMO).However,the role of ECMO in adult patients with sepsis has not been fully established.According to existing studies,ECMO may be a viable salvage therapy in carefully selected adult patients with sepsis.The choice of venovenous,venoarterial,or hybrid ECMO modes is primarily determined by the patient’s oxygenation and hemodynamics(distributive shock with preserved cardiac output,septic cardiomyopathy(left,right,or biventricular heart failure),or right ventricular failure caused by acute respiratory distress syndrome).Veno-venous ECMO can be used in patients with sepsis and severe acute respiratory distress syndrome when conventional mechanical ventilation fails,and early application of veno-arterial ECMO in patients with sepsis-induced refractory cardiogenic shock may be critical in improving their chances of survival.When ECMO is indicated,the choice of an appropriate mode and determination of the optimal timing of initiation and weaning are critical,particularly in an experienced ECMO center.Furthermore,some special issues,such as ECMO flow,anticoagulation,and antibiotic therapy,should be noted during the management of ECMO support.Hongling Zhang Youdong Xu Xin Huang Shunyin Yang Ruiting Li Yongran Wu Xiaojing Zou Yuan Yu You Shang 2024Journal of Intensive Medicine2024,4,1:0

15Effects of vitamin C on the structural and functional characteristics of wheat gluten显示文摘Vitamin C(VC)can greatly enhance the quality properties of dough and steamed bread in the food field.However,there are few studies on the interaction between VC and gluten,and the mechanism of VC influencing the foaming capacity,emulsifiablity,and dynamic viscosity of gluten is not clear.In this study,different content of VC(0,1%,2%,3%,4%and 5%)was added to gluten to obtain VC-gluten dough samples(CK,GV1,GV2,GV3,GV4 and GV5),and the properties of the VC-gluten dough were determined.Results showed that the increasing VC content lead to increase in the content of high-,medium-and low-molecular weight gluten,suggesting that VC improved the protein solubility of gluten.The G'and G'values of dough containing VC were higher than those of the control with the increase of scanning frequency.The foaming ability of gluten increased with the increase of VC content.Emulsibility of the dough of GV1 to GV 5 were significantly lower than that of the CK.Furthermore,VC could reduce the number of bubbles in gluten protein,when the VC content was less than 3%.In conclusion,VC might reduce the emulsifying characteristic and emulsifying stability of gluten,increase the solubility of gluten,and improve the dynamic viscosity of gluten dough by changing the content of disulfide bond in gluten.This study hopes to not only improve the functional characteristics of gluten protein,but also provide the basis for future development and improvement of VC-enriched health foods.Feng Jia Kaichang Ye Changfu Zhang Shihao Zhang Mingxin Fu Xiao Liu Ruijia Guo Ruiting Yang Huiru Zhang Jinshui Wang 2022Grain & Oil Science and Technology2022,5,2:0
16Modulation of Antiviral Immunity and Therapeutic Efficacy by 25-Hydroxycholesterol in Chronically SIV-Infected, ART-Treated Rhesus Macaques显示文摘Cholesterol-25-hydroxylase(CH25 H)and its enzymatic product 25-hydroxy cholesterol(25 HC)exert broadly antiviral activity including inhibiting HIV-1 infection.However,their antiviral immunity and therapeutic efficacy in a nonhuman primate model are unknown.Here,we report that the regimen of 25 HC combined with antiretroviral therapy(ART),provides profound immunological modulation towards inhibiting viral replication in chronically SIVmac239-infected rhesus macaques(RMs).Compared to the ART alone,this regimen more effectively controlled SIV replication,enhanced SIVspecific cellular immune responses,restored the ratio of CD4/CD8 cells,reversed the hyperactivation state of CD4^(+)T cells,and inhibited the secretion of proinflammatory cytokines by CD4^(^(+))and CD8^(+)T lymphocytes in chronically SIVinfected RMs.Furthermore,the in vivo safety and the preliminary pharmacokinetics of the 25 HC compound were assessed in this RM model.Taken together,these assessments help explain the profound relationship between cholesterol metabolism,immune modulation,and antiviral activities by 25 HC.These results provide insight for developing novel therapeutic drug candidates against HIV-1 infection and other related diseases.Chunxiu Wu Jin Zhao Ruiting Li Fengling Feng Yizi He Yanjun Li Runhan Huang Guangye Li Heng Yang Genhong Cheng Ling Chen Feng Ma Pingchao Li Caijun Sun 2021Virologica Sinica2021,36,5:0
17Synthesis of Bio-based Epoxy Containing Phosphine Oxide as a Reactive Additive Toward Highly Toughened and Fire-retarded Epoxy Resins显示文摘The integration of high mechanical toughness,impact strength as well as excellent flame-retardant properties toward epoxy resins(EPs)have always been a dilemma.The inadequate overall performance of EPs severely restricts their sustainable utilization in engineering aspects over long-term.Herein,a new bio-based agent(diglycidyl ether of magnolol phosphine oxide,referred as DGEMP)derived from magnolol(classified as lignan),extracted from natural plants Magnolia officinalis,was successfully synthesized and further employed as a flameretardant reactive additive to diglycidyl ether of bisphenol A(DGEBA).As demonstration,the composite resin,DGEBA/15DGEMP(15 wt%DGEMP),achieved an Underwriters Laboratories-94 V-0 rating with a high limiting oxygen index(LOI)value(41.5%).In cone calorimeter tests,it showed that heat release and smoke production were effectively inhibited during combustion,wherein the peak heat release rate(PHRR)value of DGEBA/15DGEMP was reduced by 50%compared to neat DGEBA.Additionally,it exhibited a superior tensile strength(82.8 MPa),toughness(5.11MJ/m^(3))and impact strength(36.5 k J/m^(2)),much higher than that of neat DGEBA(49.7 MPa,2.05 MJ/m^(3)and 20.9 k J/m^(2)).Thus,it is highly anticipated that DGEMP imparts significantly improved mechanical and fire-retarded properties to conventional EPs,which holds a great potential to address the pressing challenges in EP thermosets industry.Chunxiang Wei Tianyu Gao Yu Xu Wenjie Yang Guangjian Dai Ruiting Li SanE Zhu Richard K.K.Yuen Wei Yang Hongdian Lu 2023Chinese Journal of Polymer Science2023,41,11:0
