2Genetic diversity in Chinese modern wheat varieties revealed by microsatellite markers显示文摘Genetic diversity of 1680 modern varieties in Chinese candidate core collections was analyzed at 78 SSR loci by fluorescence detection system. A total of 1336 alleles were detected, of which 1253 alleles could be annotated into 71 loci. For these 71 loci, the alleles ranged from 4 to 44 with an average of 17.6, and the PIC values changed from 0.19 to 0.89 with an average of 0.69. (1) In the three genomes of wheat, the average genetic richness was B>A>D, and the genetic diversity indexes were B>D>A. (2) Among the seven homoeologous groups, the average genetic richness was 2=7>3>4>6>5>1, and the genetic diversity indexes were 7>3>2>4>6>5>1. As a whole, group 7 possessed the highest genetic diversity, while groups 1 and 5 were the lowest. (3) In the 21 wheat chromosomes, 7A, 3B and 2D possessed much higher genetic diversity, while 2A, 1B, 4D, 5D and 1D were the lowest. (4) The highest average genetic diversity index existed in varieties bred in the 1950s, and then it declined continually. However, the change tendency of genetic diversity among decades was not greatly sharp. This was further illustrated by changes of the average genetic distance between varieties. In the 1950s it was the largest (0.731). Since the 1960s, it has decreased gradually (0.711, 0.706, 0.696, 0.695). The genetic base of modern varieties is becoming narrower and narrower. 4Data Analytics and Machine Learning for Smart Process Manufacturing: Recent Advances and Perspectives in the Big Data Era显示文摘Safe, ef cient, and sustainable operations and control are primary objectives in industrial manufacturing processes. State-of-the-art technologies heavily rely on human intervention, thereby showing apparent limitations in practice. The burgeoning era of big data is in uencing the process industries tremendously, providing unprecedented opportunities to achieve smart manufacturing. This kind of manufacturing requires machines to not only be capable of relieving humans from intensive physical work, but also be effective in taking on intellectual labor and even producing innovations on their own. To attain this goal, data analytics and machine learning are indispensable. In this paper, we review recent advances in data analytics and machine learning applied to the monitoring, control, and optimization of industrial processes, paying particular attention to the interpretability and functionality of machine learning mod- els. 8Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis.The symptoms of COVID-19 range from mild to severe,but the physiological changes associated with COVID-19 are barely understood.In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms.We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients,indicating that the development of COVID-19 affected their whole-body metabolism.In particular,malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction,respectively.It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms.And,more importantly,guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases.In addition,dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19.Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou 2020National Science Review2020,7,7:9
9Research on 3D Target Pose Tracking and Modeling显示文摘This paper tackles pose tracking and model refinement, one of the fundamental work for 3D photogrammetry. The researches belong to the videometrics, an interdisciplinewhich combines computer vision, digital image processing, photogrammetry and optical measurement. Related works are summarized briefly in this paper. This paper studies the problem of pose tracking for target with 3D model. For the target with accurate 3D model, line model based pose tracking methods are proposed for target which is rich in line features. Experimental results indicate that the proposed methods track the target pose accurately. Normal distance iterative reweighted least squares and distance image iterative least squares methods are proposed to process more general targets. This paper adopts bundle adjustment to tackle pose tracking in image sequence for target with inaccurate 3D line model. The proposed method optimizes the model line parameters and the pose parameters simultaneously. The model line orientation, position and mean angle error, mean position error of the pose are 0.3°,3.5 mm and 0.12°,20.1 mm in simulation experiments of satellite pose tracking. Line features are used to track target pose with unknown 3D model through image sequence. The model line parameters and pose parameters are optimized under the framework of SFM. 15Study of ciprofloxacin removal by biochar obtained from used tea leaves显示文摘In this study,used tea leaves(UTLs) were pyrolyzed to obtain used tea-leaf biochar(UTC),and then the UTC was used as an adsorbent to remove ciprofloxacin(CIP) from aqueous solutions.Batch experiments were conducted to investigate the CIP adsorption performance and mechanism.The results showed that the CIP-adsorbing ability first increased and then declined as the UTC pyrolysis temperature increased.The UTC obtained at 450°C presented excellent CIP-absorbing ability at p H 6 and 40°C.The maximum monolayer adsorption capacity was 238.10 mg/g based on the Langmuir isotherm model.The pseudo-second-order kinetic equation agreed well with the CIP adsorption process,which was controlled by both external boundary layer diffusion and intra-particle diffusion.The characterization analysis revealed that the \OH groups,C_C bonds of aromatic rings,C\H groups in aromatic rings and phenolic C\O bonds play vital roles in the CIP adsorption process,and that the N\C,N\O,O\C_O and C\OH groups of UTC were consumed in large quantities.π–π interactions,hydrogen bonding and electrostatic attraction are inferred as the main adsorption mechanisms.The present work provides not only a feasible and promising approach for UTLs utilization but also a potential adsorbent material for removing high concentrations of CIP from aqueous solutions.Jie Li Guangwei Yu Lanjia Pan Chunxing Li Futian You Shengyu Xie Yin Wang Jianli Ma Xiaofu Shang 2018Journal of Environmental Sciences2018,30,11:5

19High-throughput screening identifies established drugs as SARS-CoV-2 PLpro inhibitors显示文摘A new coronavirus(SARS-CoV-2)has been identified as the etiologic agent for the COVID-19 outbreak.Currently,effective treatment options remain very limited for this disease;therefore,there is an urgent need to identify new anti-COVID-19 agents.In this study,we screened over 6,000 compounds that included approved drugs,drug candidates in clinical trials,and pharmacologically active compounds to identify leads that target the SARS-CoV-2 papain-like protease(PLpro).Together with main protease(Mpro),PLpro is responsible for processing the viral replicase polyprotein into functional units.There-fore,it is an attractive target for antiviral drug develop-ment.Here we discovered four compounds,YM155,cryptotanshinone,tanshinone I and GRL0617 that inhibit SARS-CoV-2 PLpro with IC50 values ranging from 1.39 to 5.63 pmol/L.These compounds also exhibit strong antiviral activities in cell-based assays.YM155,an anti-cancer drug candidate in clinical trials,has the most potent antiviral activity with an EC50 value of 170 nmol/L.In addition,we have determined the crystal structures of this enzyme and its complex with YM155,revealing a unique binding mode.YM155 simultaneously targets three'hot'spots on PLpro,including the substrate-binding pocket,the interferon stimulating gene product 15(ISG15)binding site and zinc finger motif.Our results demonstrate the efficacy of this screening and repur-posing strategy,which has led to the discovery of new drug leads with clinical potential for COVID-19 treatments.Yao Zhao Xiaoyu Du Yinkai Duan Xiaoyan Pan Yifang Sun Tian You Lin Han Zhenming Jin Weijuan Shang Jing Yu Hangtian Guo Qianying Liu Yan Wu Chao Peng Jun Wang Chenghao Zhu Xiuna Yang Kailin Yang Ying Lei Luke W.Guddat Wenqing Xu Gengfu Xiao Lei Sun Leike Zhang Zihe Rao Haitao Yang 2021Protein & Cell2021,12,11:4
20Anti-BCMA CAR T-cell therapy CT103A in relapsed or refractory AQP4-IgG seropositive neuromyelitis optica spectrum disorders:phase 1 trial interim results显示文摘Chimeric antigen receptor(CAR)T-cell therapy that targets B-cell maturation antigen(BCMA)have great potentials in autoimmune diseases and could be novel therapeutics for relapsed/refractory neuromyelitis optica spectrum disorder(NMOSD).To evaluate the safety and efficacy of the CT103A,a self-developed BCMA-targeting CAR construct against BCMA,in patients with AQP4-IgG seropositive NMOSD,an ongoing,investigator-initiated,open-label,single-arm,phase 1 clinical trial is conducted at our center.In total,12 patients were administered with a CAR-BCMA infusion.Ten of the 12 patients dosed were women(83.3%),with a median age of 49.5 years(range,30-67).were The most common events of grade 3 or higher were hematologic toxic effects.Seven patients(58%)developed infections,but no grade 4 infections occurred.Cytokine release syndrome was reported in all patients with only events of grade 1 or 2 observed.During the follow-up of a median 5.5 months,11 patients had no relapse;all patients generally reported improvement in disabilities and quality-of-life outcomes;11 patients’AQP-4 antibodies in serum showed a downward trend by the cutoff date.CAR T-cell expansion was associated with responses,and persisted more than 6 months post-infusion in 17%of the patients.In summary,CAR T-cell therapy shows a manageable safety profile and therapeutic potentials for patients with relapsed/refractory AQP4-IgG seropositive NMOSD.Another expansion phase is currently underway to determine the safety and efficacy of CAR T-BCMA infusion in patients with other neuro-inflammatory diseases.Chuan Qin Dai-Shi Tian Luo-Qi Zhou Ke Shang Liang Huang Ming-Hao Dong Yun-Fan You Jun Xiao Ying Xiong Wen Wang Hao Pang Jing-Jing Guo Song-Bai Cai Di Wang Chun-Rui Li Min Zhang Bi-Tao Bu Wei Wang 2023Signal Transduction and Targeted Therapy2023,8,2:4
