| 2 | Isolation and characterization of glacier VMY22, a novel lytic cold-active bacteriophage of Bacillus cereus显示文摘As a unique ecological system with low temperature and low nutrient levels, glaciers are considered a 'living fossil' for the research of evolution. In this work, a lytic cold-active bacteriophage designated VMY22 against Bacillus cereus MYB41-22 was isolated from Mingyong Glacier in China, and its characteristics were studied. Electron microscopy revealed that VMY22 has an icosahedral head(59.2 nm in length, 31.9 nm in width) and a tail(43.2 nm in length). Bacteriophage VMY22 was classified as a Podoviridae with an approximate genome size of 18 to 20 kb. A one-step growth curve revealed that the latent and the burst periods were 70 and 70 min, respectively, with an average burst size of 78 bacteriophage particles per infected cell. The pH and thermal stability of bacteriophage VMY22 were also investigated. The maximum stability of the bacteriophage was observed to be at pH 8.0 and it was comparatively stable at p H 5.0–9.0. | 3 | Target-triggered inhibiting oxidase-mimicking activity of platinum nanoparticles for ultrasensitive colorimetric detection of silver ion显示文摘The development of efficient methods for the detection of hazardous and toxic elements is extremely important for environmental security and public health. In this work, we developed a facile colorimetric assaying system for Ag+ detection in aqueous solution. Chitosan-stabilized platinum nanoparticles(ChPtNPs) were synthesized and severed as an artificial oxidase to catalyze the oxidation of the substrate3,30,5,50-tetramethylbenzidine(TMB) and generate color signal. In the presence of Ag+, due to the strong metallophilic interactions between Ag+ and Pt2+ on the surface of Ch-PtNPs, Ag+ can weaken the affinity to the substrates and inactivate the catalytic activity of Ch-PtNPs, leading to decreased absorbance signal to varying degrees depending on Ag+ amount. Combing the specific binding between Ch-PtNPs and Ag+ with signal amplification procedure based on the Ch-PtNPs-catalyzed TMB oxidation, a sensitive,selective, simple, cost-effective, and rapid detection method for Ag+ can be realized. Ag+ ions in tap and lake waters have been successfully detected. | 16 | Immunogenicity and protective efficacy of an inactivated SFTS vaccine candidate in mice显示文摘Severe fever with thrombocytopenia syndrome(SFTS),caused by a novel identified bunyavirus SFTS virus(SFTSV),was an emerging viral infectious disease that was firstly reported in China.There are no licensed vaccines and therapeutics against SFTSV currently.B‐Propiolactone(BPL)inactivated whole virions of SFTSV strain AH12 were prepared as experimental vaccine in different antigen dose with or without Al(OH)3 adjuvant.The experimental SFTS vaccine was a satisfying immunogen,which could efficiently trigger the development of high levels of SFTSV NP‐specific IgG antibodies and neutralizing antibodies against SFTSV Strain HB29 in BALB/c and C57/BL6 mice,and could induce SFTS virus‐specific cellular immune responses to a certain extent.A single dose of vaccine was immunogenically insufficient in BALB/c mice;the second and third dose resulted in significant boost in antibody response.The use of Al(OH)3 adjuvant resulted in higher antibody titers.The mediate‐dose of vaccine could induce as high and equivalent level of antibody titer as that of high‐dose.The experimental SFTS vaccine in mediate‐and high antigen dose with adjuvant resulted in solid protection of C57/BL6 mice against wild‐type SFTSV challenge with markedly accelerated virus clearance from blood and spleen compared with controls.The experimental SFTS vaccine prepared in this study could efficiently elicit virus specific humoral and cellular immune responses in both BALB/c and C57/BL6 mice,and could protect C57/BL6 mice against SFTS virus challenge.These results supplied evidence that inactivated vaccine was a promising vaccine candidate for the prevention of SFTSV infection. | Aqian Li Xinxian Dai Lei Chen Lin Liu Chuan Li Yang Liu Wei Wu Xiaoxia Huang Jiandong Li Shiwen Wang Mifang Liang Xiuling Li Dexin Li | 2022 | Biosafety and Health2022,4,1: | 0 |

Colonies were circular, bulgy, opaque, moist and white, with a 1-2 mm diameter. Its sequence showed 97.46% and96.91% homology with Sporolactobacillus vineae SL153 T and S. putidus QC81-06 T, respectively.
