| 1 | Effect of biofilm formation by clinical isolates of Helicobacter pylori on the efflux-mediated resistance to commonly used antibiotics显示文摘AIM To evaluate the role of biofilm formation on the resistance of Helicobacter pylori(H. pylori) to commonly prescribed antibiotics, the expression rates of resistance genes in biofilm-forming and planktonic cells were compared.METHODS A collection of 33 H. pylori isolates from children and adult patients with chronic infection were taken for the present study. The isolates were screened for biofilm formation ability, as well as for polymerase chain reaction(PCR) reaction with HP1165 and hp1165 efflux pump genes. Susceptibilities of the selected strains to antibiotic and differences between susceptibilities of planktonic and biofilm-forming cell populations were determined. Quantitative real-time PCR(qP CR) analysis was performed using 16 S r RNA gene as a H. pylori-specific primer, and two efflux pumps-specific primers, hp1165 and hefA.RESULTS The strains were resistant to amoxicillin, metronidazole, and erythromycin, except for one strain, but they were all susceptible to tetracycline. Minimum bactericidalconcentrations of antibiotics in the biofilm-forming cells were significantly higher than those of planktonic cells. qP CR demonstrated that the expression of efflux pump genes was significantly higher in the biofilm-forming cells as compared to the planktonic ones.CONCLUSION The present work demonstrated an association between H. pylori biofilm formation and decreased susceptibility to all the antibiotics tested. | 2 | Combination of Model-based Observer and Support Vector Machines for Fault Detection of Wind Turbines显示文摘Support vector machines and a Kalman-like observer are used for fault detection and isolation in a variable speed horizontalaxis wind turbine composed of three blades and a full converter. The support vector approach is data-based and is therefore robust to process knowledge. It is based on structural risk minimization which enhances generalization even with small training data set and it allows for process nonlinearity by using flexible kernels. In this work, a radial basis function is used as the kernel. Different parts of the process are investigated including actuators and sensors faults. With duplicated sensors, sensor faults in blade pitch positions,generator and rotor speeds can be detected. Faults of type stuck measurements can be detected in 2 sampling periods. The detection time of offset/scaled measurements depends on the severity of the fault and on the process dynamics when the fault occurs. The converter torque actuator fault can be detected within 2 sampling periods. Faults in the actuators of the pitch systems represents a higher difficulty for fault detection which is due to the fact that such faults only affect the transitory state(which is very fast) but not the final stationary state. Therefore, two methods are considered and compared for fault detection and isolation of this fault: support vector machines and a Kalman-like observer. Advantages and disadvantages of each method are discussed. On one hand, support vector machines training of transitory states would require a big amount of data in different situations, but the fault detection and isolation results are robust to variations in the input/operating point. On the other hand, the observer is model-based, and therefore does not require training, and it allows identification of the fault level, which is interesting for fault reconfiguration. But the observability of the system is ensured under specific conditions, related to the dynamics of the inputs and outputs. | 3 | Performances of local poultry breed fed black soldier fly larvae reared on horse manure显示文摘In poultry,feed based on maggots,like larvae of black soldier fly(Hermetia illucens) is an attractive option to substitute current ingredients which are expensive and often in direct or indirect competition with human food.Little information is currently available on the utility of these larvae in poultry feed,so goals of this study were to determine whether larvae could be reared on horse manure under traditional farming conditions and to evaluate the growth performances of a local poultry fed these larvae and the fatty acids profiles of their meat.After freezing and thawing,larvae were introduced in the feed of Ardennaise chickens between 30 and 80 days of age.Birds in the control group received a commercial standard feed,while those in the treatment group received the same commercial feed in which 8% was substituted with whole fresh larvae corresponding to 2% on a dry matter basis.Means ± standard errors of larval length and weight were 20.67 ± 2.21 mm and 0.14 ± 0.02 g,respectively.Mean larval percentages of dry matter and of substances extractable in diethyl ether were 24.6% and 23.1%,respectively.Larval fatty acids profiles were predominantly composed of lauric acid(28.1%) and palmitic acid(22.0%).Least squares means of weekly weights of chicken,adjusted for the effects of sex,replication and initial weights,were significantly higher(P < 0.05) by 77.03 ± 53.37 g in larvae-fed than in control chickens.All other measurements were not statistically different between larvae-fed and control chicken,including fatty acid profiles,protein content and w6/w3 ratio. | Nassim Moula Marie-Louise Scippo Caroline Douny Guy Degand Edwin Dawans Jean-Francois Cabaraux Jean-Luc Hornick Rudy C.Medigo Pascal Leroy Frederic Francis Johann Detilleux | 2018 | Animal Nutrition2018,,1: | 7 |
| 4 | High tacrolimus intra-patient variability is associated with graft rejection,and de novo donor-specific antibodies occurrence after liver transplantation显示文摘AIM To investigate the role of tacrolimus intra-patient variability(IPV) in adult liver-transplant recipients.METHODS We retrospectively assessed tacrolimus variability in a cohort of liver-transplant recipients and analyzed its effect on the occurrence of graft rejection and de novo donor-specific antibodies(dn DSAs), as well as graft survival during the first 2 years posttransplantation. Between 02/08 and 06/2015, 116 patients that received tacrolimus plus mycophenolate mofetil(with or without steroids) were included. RESULTS Twenty-two patients(18.5%) experienced at least one acute-rejection episode(BPAR). Predictive factors for a BPAR were a tacrolimus IPV of > 35% [OR = 3.07 95%CI(1.14-8.24), P = 0.03] or > 40% [OR = 4.16(1.38-12.50), P = 0.01), and a tacrolimus trough level of < 5 ng/mL [OR=3.68(1.3-10.4), P =0.014]. Thirteen patients(11.2%) developed at least one dn DSA during the follow-up. Tacrolimus IPV [coded as a continuous variable: OR = 1.1, 95%CI(1.0-1.12), P = 0.006] of > 35% [OR = 4.83, 95%CI(1.39-16.72), P = 0.01] and > 40% [OR = 9.73, 95%CI(2.65-35.76), P = 0.001] were identified as predictors to detect dn DSAs. IPV did not impact on patient-or graft-survival rates during the follow-up. | 5 | Early plasmapheresis and rituximab for acute humoral rejection after ABO-compatible liver transplantation显示文摘Acute humoral rejection (AHR) is uncommon after ABO-compatible liver transplantation. Herein, we report two cases of AHR treated with plasmapheresis and rituximab in two ABO-compatible liver-transplant patients with preformed anti-human leukocyte antigen donor-specif ic antibodies. Patient 1 experienced a biopsy-proven AHR at day 10 post-transplant. She was treated by steroid pulses, and OKT3. Because of persisting signs of biopsy-proven AHR at day 26, she was treated by plasmapheresis and rituximab. Liver enzyme levels did not improve, and she died on day 41. Patient 2 experienced a biopsy-proven AHR on day 10 post-transplant. She was treated by steroid pulses, plasmapheresis, and rituximab. Liver enzymes returned to within normal range 18 d after diagnosis. Liver biopsies, at 3 and 9 mo post-transplant, showed complete resolution of AHR. We conclude that plasmapheresis should be started as soon as AHR is diagnosed, and be associated with a B-cell depleting agent. | 6 | Lens thickness assessment : anterior segment optical coherence tomography versus A-scan ultrasonography显示文摘AIM: To assess lens thickness measurements with anterior segment-optical coherence tomography(AS-OCT)in comparison with A-scan ultrasonography(A-scan US).·METHODS: There were 218 adult subjects(218 eyes)aged 59.2 ±9.2y enrolled in this prospective cross-sectional study. Forty-three eyes had open angles and 175 eyes had narrow angles. Routine ophthalmic exam was performed and nuclear opacity was graded using the Lens Opacities Classification System Ⅲ(LOCS Ⅲ). Lens thickness was measured by AS-OCT(Visante OCT, Carl Zeiss Meditec, Dublin, CA, USA). The highest quality image was selected for each eye and lens thickness was calculated using Image J software. Lens thickness was also measured by A-scan US.·RESULTS: Interclass correlations showed a value of99.7% for intra-visit measurements and 95.3% for inter-visit measurements. The mean lens thickness measured by AS-OCT was not significantly different from that of A-scan US(4.861 ±0.404 vs 4.866 ±0.351 mm, P =0.74).Lens thickness values obtained from the two instruments were highly correlated overall(Pearson correlation coefficient =0.81,P <0.001), and in all LOCS Ⅲ specific subgroups except in grade 5 of nuclear opacity. Bland-Altman analysis revealed a 95% limit of agreement from-0.45 to 0.46 mm. Lens thickness difference between the two instruments became smaller as the lens thickness increased and AS-OCT yielded smaller values than A-scan US in thicker lens(β=-0.29, P <0.001)· CONCLUSION: AS-OCT-derived lens thickness measurement is valid and comparable to the results obtained by A-scan US.
