| 1 | Recent advances in cell sheet technology for bone and cartilage regeneration: from preparation to application显示文摘Bone defects caused by trauma,tumour resection,infection and congenital deformities,together with articular cartilage defects and cartilage–subchondral bone complex defects caused by trauma and degenerative diseases,remain great challenges for clinicians.Novel strategies utilising cell sheet technology to enhance bone and cartilage regeneration are being developed.The cell sheet technology has shown great clinical potential in regenerative medicine due to its effective preservation of cell–cell connections and extracellular matrix and its scaffold-free nature.This review will first introduce several widely used cell sheet preparation systems,including traditional approaches and recent improvements,as well as their advantages and shortcomings.Recent advances in utilising cell sheet technology to regenerate bone or cartilage defects and bone–cartilage complex defects will be reviewed.The key challenges and future research directions for the application of cell sheet technology in bone and cartilage regeneration will also be discussed. | Yuezhi Lu Wenjie Zhang Jie Wang Guangzheng Yang Shi Yin Tingting Tang Chunhua Yu Xinquan Jiang | 2019 | International Journal of Oral Science2019,11,2: | 14 |
| 2 | The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy. | Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu | 2021 | Cancer Communications2021,41,1: | 5 |
| 3 | Behavior features of heavy metals in the Haikou Bay waters显示文摘Dissolved Cu, Ph, Zn and Cd in the Haikou Bay waters were measured to be respectively in the range concentrations of 0.47-1.16 μg/dm^3, 0. 94-- 2. 36μg/dm^3, 1.28-4.83 μg/dm^3 and 0. 005-0.072μg/dm^3; with respectively average values of 0.78μg/dm^3, 1.36μg/dm^3, 3.14 g/dm^3 and 0. 03 μg/dm^3. Dissolved Cu and Zn concentrations are relatively high at the stations near the Longkun Road Outfall for domestic sewage, the Xiuying Outfall for industry waste water and the Haidian Island Estuary, but dissolved Pb and Cd concentrations are low in these stations. The values in Other stations are comparatively homogenous. Vertical dissolved Cu, Pb and Zn concentrations at the bottom layer are higher than at the surface layer, but dissolved Cd concentration appears to be on the opposite. The measurement results of Cu, Pb, Zn and Cd in suspended particle show that particulate matters in the Haikou Bay seawater play a role in purifying heavy metals. The study on strong complexed form and non-liable form of dissolved copper show that the ratio of strong complexed form and dissolved form is about 85%, and non-liable form is very low with a value lower than 5 nmol/dm^3. | 5 | Efficacy of water fraction from Dioscorea cirrhosa on oxidative stress and apoptosis in H9c2 cardiomyocytes induced by H2O2显示文摘OBJECTIVE:To investigate the efficacy of water fraction from Dioscorea cirrhosa(WF)on oxidative damage and apoptosis of cardiomyocytes induced by H2O2,and to study its mechanism.METHODS:Cell viability was measured by the MST assay kit.The content of malondialdehyde(MDA),release of lactate dehydrogenase(LDH)and activity of catalase(CAT)and superoxide dismutase(SOD)were detected by biochemical kit.The content of reactive oxygen species(ROS)was assessed by nonfluorescent probe 2′,7′-dichlorofluorescin diacetate(DCFH-DA).JC-1 was used to analyze the mitochondrial membrane potential(mtΔΨ)and Annexin-V-FITC/PI staining was applied to assess apoptosis of H9c2 by flow cytometry.Moreover,the expression of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X(Bax),caspase-3,caspase-9,cleaved-caspase-3 and cleaved-caspase-9 proteins was determined by western blot analysis.RESULTS:WF increased cell viability and decreased LDH leakage in H9c2 cells exposed to H2O2.WF treatment decreased ROS and MDA level,enhanced SOD and CAT activities,improved mtΔΨand inhibited apoptosis.Western blot analysis demonstrated that the ratio of Bcl-2/Bax was increased and the expression cleaved-caspase-3,caspase-3,cleaved-caspase-9 and caspase-9 were decreased in group treated with WF.CONCLUSION:WF protects H9c2 myocardial cells on H2O2-induced oxidative stress and apoptosis by scavenging ROS,improving antioxidant capacity,protecting mitochondrial and regulating the proteins expression related to apoptosis. | LIU Chunhua PAN Jie WANG Aimin LAN Yanyu LI Yongjun LU Yuan SUN Jia WANG Yonglin LIU Ting LU Dingyan YOU Jingrui | 2021 | Journal of Traditional Chinese Medicine2021,41,1: | 4 |
| 6 | Treatment of Enzymolysis Wastewater by Centrifugation- Coagulation- Fenton Reagent Oxidation- Adsorption Process显示文摘[Objective]The study aimed to discuss the optimal conditions for the treatment of enzymolysis wastewater by centrifugation-coagulation-Fenton reagent oxidation-adsorption process.[Method]According to the water-quality characteristics of wastewater from a heparin sodium production factory of Jiangsu Province,enzymolysis wastewater was segregated from intestinal lavage wastewater and treated through centrifugation-coagulation-Fenton reagent oxidation-adsorption process,and the optimal technical parameters were determined.[Result]After enzymolysis wastewater was centrifuged at a speed of 4 000 rpm,0.6 g/L CTS as the coagulant was added to the supernatant.Hereafter,pH of the coagulated effluent was adjusted to 3,and then 1.5%(V/V)H2O2was added to the coagulated effluent;a certain amount of ferrous sulfate(n H2O2∶n FeSO4·7H2O=8∶1)was added to the mixture;the reaction conducted for 30 min,and then solution pH was adjusted to about9.Finally,the oxidized effluent flowed through a resin red until the adsorptive capacity reached 240 BV,and COD of the effluent water was lower than 100 mg/L,meeting the Grade-I standard of Comprehensive Discharge Standard of Sewage(GB8978-1996).[Conclusion]The research could provide a new process for the treatment of enzymolysis wastewater. | Jian Zhai Chunhua Jiang Guoqing Cao Xiuguo Lu | 2013 | Meteorological and Environmental Research2013,4,9: | 3 |

| 10 | Effects of Microbial Inoculums on Soil Enzyme Activity and Microbial Diversity in a Reclaimed Mining Area显示文摘Through a pot experiment,effects of various microbial inoculums on soil microbial diversity and enzyme activity in a typical reclaimed mining area in Shanxi Province were discussed based on quantitative analysis of PLFA,soil urease,phosphatase and sucrase activity. The results showed that the application of microbial inoculums increased microorganism quantity in rhizosphere of rape by 2. 3%-66. 4%,and quantities of bacteria,gram-positive bacteria,gram-negative bacteria,fungi and actinomycetes in the treatments with microbial inoculums were significantly higher than the contrast( P <0. 05),while there was no obvious change in protist quantity.
