3Sequencing of Cultivated Peanut, Arachis hypogaea, Yields Insights into Genome Evolution and Oil Improvement显示文摘Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. 4Structure and function of interleukin-17 family cytokines显示文摘The recently identified interleukin-17(IL-17)cytokines family,which comprises six members in mammals(IL-17A–F),plays essential roles in the host immunity against infectious diseases and chronic inflammatory diseases.The three-dimensional structures containing IL-17A or IL-17F have become available and revealed the unique structural features of IL-17s as well as their receptors.Molecular modeling in this review shows that IL-17s may adopt a“cysteine knot”fold commonly seen in nerve growth factor(NGF)and other neurotrophins.Further modeling analysis unmasks a signature interaction feature of the IL-17F/IL-17RA complex,where a small loop of IL-17RA slots into the deep groove of the interface of IL-17F homodimer.This is quite different from the interaction between the best known four-helix cytokines and their cognate receptors.On the other hand,structure of IL-17A and its monoclonal antibody(CAT-2200)shows that,albeit that the antigenic epitope of IL-17A resides outside of the IL-17A homodimer interface,its physical proximity to the receptor binding groove may explain that antibody blockage would be achieved by interfering with the ligand-receptor interaction.This review is to summarize the advance in understanding the structure and function of IL-17 family cytokines,focusing mainly on IL-17A,IL-17F and IL-17E,in the hope of gaining better knowledge of immunotherapeutic strategies against various inflammatory diseases.Xiaoping Zhang Pornpimon Angkasekwinai Chen Dong Hong Tang 2011Protein & Cell2011,2,1:17
5IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6

7Asymmetric distribution of cytokinins determines root hydrotropism in Arabidopsis thaliana显示文摘The phenomenon of plant root tips sensing moisture gradient in soil and growing towards higher water potential is designated as root hydrotropism,which is critical for plants to survive when water is a limited factor.Molecular mechanisms regulating such a fundamental process,however,are largely unknown.Here we report our identification that cytokinins are key signaling molecules directing root growth orientation in a hydrostimulation(moisture gradient)condition.Lower water potential side of the root tip shows more cytokinin response relative to the higher water potential side.Consequently,two cytokinin downstream type-A response regulators,ARR16 and ARR17,were found to be up-regulated at the lower water potential side,causing increased cell division in the meristem zone,which allows the root to bend towards higher water potential side.Genetic analyses indicated that various cytokinin biosynthesis and signaling mutants,including the arr16 arr17 double mutant,are significantly less responsive to hydrostimulation.Consistently,treatments with chemical inhibitors interfering with either cytokinin biosynthesis or cell division completely abolished root hydrotropic response.Asymmetrically induced expression of ARR16 or ARR17 effectively led to root bending in both wild-type and miz1,a previously known hydrotropism-defective mutant.These data demonstrate that asymmetric cytokinin distribution is a primary determinant governing root hydrotropism.Jinke Chang Xiaopeng Li Weihao Fu Jiawen Wang Yueyuan Yong Hongyong Shi Zhaojun Ding Hong Kui Xiaoping Gou Kai He Jia Li 2019Cell Research2019,29,12:4

9Quantifying 3D cell-matrix interactions during mitosis and the effect of anticancer drugs on the interactions显示文摘The mechanical force between cells and the extracellular microenvironment is crucial to many physiological processes such as cancer metastasis and stem cell differentiation. Mitosis plays an essential role in all these processes and thus an in-depth understanding of forces during mitosis gains insight into disease diagnosis and disease treatment. Here, we develop a traction force microscope method based on monolayer fluorescent beads for measuring the weak traction force (tens of Pa) of mitotic cells in three dimensions. We quantify traction forces of human ovarian granulosa (KGN) cells exerted on the extracellular matrix throughout the entire cell cycle in three dimensions. Our measurements reveal how forces vary during the cell cycle, especially during cell division. Furthermore, we study the effect of paclitaxel (PTX) and nocodazole (NDZ) on mitotic KGN cells through the measurement of traction forces. Our results show that mitotic cells with high concentrations of PTX exert a larger force than those with high concentrations of NDZ, which proved to be caused by changes in the structure and number of microtubules. These findings reveal the key functions of microtubule in generating traction forces during cell mitosis and explain how dividing cells regulate themselves in response to anti-mitosis drugs. 10Performance analysis of microcantilever array sensing显示文摘Array sensing is increasingly important in the development of microcantilever(MC) sensors, and response consistency is the foundation for MC array sensing. In the present work, we investigated the response consistency of MC array sensing. The responses of two types of commercially available MC arrays were studied under conditions of temperature change, solution replacement and biochemical molecular interaction. For the thermal response, the deflections of both arrays were found to be proportional to temperature, and the responses of the MCs in both arrays were consistent with each other. The thermal response sensitivity for each MC during temperature increase and decrease also showed good consistency. Moreover, the MC array showed good consistency for the response induced by solution replacement. Finally, we also demonstrated that the MC array had good consistency in biochemical detection, exemplified by aflatoxin antibody-anti gen binding. 11An Ultra-microporous Carbon Material Boosting Integrated Capacitance for Cellulose-Based Supercapacitors显示文摘A breakthrough in advancing power density and stability of carbon-based supercapacitors is trapped by inefficient pore structures of electrode materials.Herein,an ultramicroporous carbon with ultrahigh integrated capacitance fabricated via one-step carbonization/activation of dense bacterial cellulose(BC)precursor followed by nitrogen/sulfur dual doping is reported.The microporous carbon possesses highly concentrated micropores(~2 nm)and a considerable amount of sub-micropores(<1 nm).The unique porous structure provides high specific surface area(1554 m^2 g^-1)and packing density(1.18 g cm^-3).The synergistic effects from the particular porous structure and optimal doping effectively enhance ion storage and ion/electron transport.As a result,the remarkable specific capacitances,including ultrahigh gravimetric and volumetric capacitances(430 F g^-1 and 507 F cm^-3 at 0.5 A g^-1),and excellent cycling and rate stability even at a high current density of 10 A g^-1(327 F g^-1 and 385 F cm^-3)are realized.Via compositing the porous carbon and BC skeleton,a robust all-solid-state cellulose-based supercapacitor presents super high areal energy density(~0.77 mWh cm^-2),volumetric energy density(~17.8 W L^-1),and excellent cyclic stability.Chenfeng Ding Tianyi Liu Xiaodong Yan Lingbo Huang Seungkon Ryu Jinle Lan Yunhua Yu Wei?Hong Zhong Xiaoping Yang 2020Nano-Micro Letters2020,12,5:2
12Efficacy of Williams LifeSkills Training in improving psychological health of Chinese male juvenile violent offenders:a randomized controlled study显示文摘This randomized controlled study was conducted to evaluate the efficacy of Williams Life Skills Training(WLST) as a means ofimproving the psychological health of Chinese male juvenile violent offenders.Sixty-six participants were assigned randomly to receive the usual intervention plus 8 weeks of WLST(study group,n = 33) or only the usual intervention(control group,n = 33). We found that the study group exhibited significantly decreased State-Trait Anxiety Inventory(STAI X-1,X-2) STAX2 scores and Trait Coping Style Questionnaire(TCSQ) negative scores,and increased Interpersonal Support Evaluation List(ISEL) tangible scores from baseline to 9 weeks later(P <0.01). In addition,a between-group difference in changes of TCSQ negative score was observed at the end of week 9(P <0.05). 13Human Bop is a novel BH3-only member of the Bcl-2 protein family显示文摘One group of Bcl-2 protein family,which shares only the BH3 domain(BH3-only),is critically involved in the regulation of programmed cell death.Herein we demonstrated a novel human BH3-only protein(designated as Bop)which could induce apoptosis in a BH3 domain-dependent manner.Further analysis indicated that Bop mainly localized to mitochondria and used its BH3 domain to contact the loop regions of voltage dependent anion channel 1(VDAC1)in the outer mitochondrial membrane.In addition,purified Bop protein induced the loss of mitochondrial transmembrane potential(ΔΨm)and the release of cytochrome c.Furthermore,Bop used its BH3 domain to contact pro-survival Bcl-2 family members(Bcl-2,Bcl-XL,Mcl-1,A1 and Bcl-w),which could inhibit Bop-induced apoptosis.Bop would be constrained by pro-survival Bcl-2 proteins in resting cells,because Bop became released from phosphorylated Bcl-2 induced by microtubule-interfering agent like vincristine(VCR).Indeed,knockdown experiments indicated that Bop was partially required for VCR induced cell death.Finally,Bop might need to function through Bak and Bax,likely by releasing Bak from Bcl-XL sequestration.In conclusion,Bop may be a novel BH3-only factor that can engage with the regulatory network of Bcl-2 family members to process intrinsic apoptotic signaling.Xiaoping Zhang Changjiang Weng Yuan Li Xiaoyan Wang Chunsun Jiang Xuemei Li Youli Xu Quan Chen Lei Pan Hong Tang 2012Protein & Cell2012,3,10:2

15Nano-TiO_2 Decorated Radial-Like Mesoporous Silica: Preparation,Characterization, and Adsorption-Photodegradation Behavior显示文摘Supported nanocrystalline Ti_O2 with a diameter of 15–30 nm was prepared from previously synthesized radial mesoporous silica(RMS) by a post-synthesis method. In addition, their adsorption-photocatalytic activity toward the degradation of methylene blue(MB) was determined. RMS was tailor-made with the main template of CTAB and the Si_O2 precursor of TEOS through a facile self-assembly process. The structural, morphological and textural properties of the well-designed Ti_O2/RMS samples were characterized.The RMS structure was retained after loading Ti_O2, but its surface area and pore diameter decreased as a result of partial pore blocking. The removal activity of MB for Ti_O2/RMS was significantly higher than that of commercial Ti_O2 nanoparticles. The optimal Ti_O2 loading(20 wt%) on the support could achieve the complete removal of MB within 70 min. 16Susceptibility of dairy cows to subacute ruminal acidosis is reflected in both prepartum and postpartum bacteria as well as odd-and branched-chain fatty acids in feces显示文摘Background:The transition period is a challenging period for high-producing dairy cattle.Cows in early lactation are considered as a group at risk of subacute ruminal acidosis(SARA).Variability in SARA susceptibility in early lactation is hypothesized to be reflected in fecal characteristics such as fecal pH,dry matter content,volatile and odd-and branched-chain fatty acids(VFA and OBCFA,respectively),as well as fecal microbiota.This was investigated with 38 periparturient dairy cows,which were classified into four groups differing in median and mean time of reticular pH below 6 as well as area under the curve of pH below 6.Furthermore,we investigated whether fecal differences were already obvious during a period prior to the SARA risk(prepartum).Results:Variation in reticular pH during a 3-week postpartum period was not associated with differences in fecal pH and VFA concentration.In the postpartum period,the copy number of fecal bacteria and methanogens of unsusceptible(UN)cows was higher than moderately susceptible(MS)or susceptible(SU)cows,while the genera Ruminococcus and Prevotellacea_UCG-001 were proportionally less abundant in UN compared with SU cows.Nevertheless,only a minor reduction was observed in iso-BCFA proportions in fecal fatty acids of SU cows,particularly iso-C15:0and iso-C16:0,compared with UN cows.Consistent with the bacterial changes postpartum,the lower abundance of Ruminococcus was already observed in the prepartum fecal bacterial communities of UN cows,whereas Lachnospiraceae_UCG-001 was increased.Nevertheless,no differences were observed in the prepartum fecal VFA or OBCFA profiles among the groups.Prepartum fecal bacterial communities of cows were clustered into two distinct clusters with 70%of the SU cows belonging to cluster 1,in which they represented 60%of the animals.Conclusions:Inter-animal variation in postpartum SARA susceptibility was reflected in post-and prepartum fecal bacterial communities.Differences in prepartum fecal bacterial communities could alert for susceptibility to develop SARA postpartum.Our results generated knowledge on the association between fecal bacteria and SARA development which could be further explored in a prevention strategy.Hong Yang Stijn Heirbaut Xiaoping Jing Nympha De Neve Leen Vandaele Jeyamalar Jeyanathan Veerle Fievez 2023Journal of Animal Science and Biotechnology2023,14,1:1
