1Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual.Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang 2020Protein & Cell2020,11,4:44

3Heterologous biosynthesis of natural product naringenin by co-culture engineering显示文摘Co-culture engineering is an emerging approach for microbial biosynthesis of a variety of biochemicals.In this study,E.coli-E.coli co-cultures were developed for heterologous biosynthesis of the natural product naringenin.The co-cultures were composed of two independent E.coli strains dedicated to functional expression of different portions of the biosynthetic pathway,respectively.The co-culture biosynthesis was optimized by investigating the effect of carbon source,E.coli strain selection,timing of IPTG induction and the inoculation ratio between the co-culture strains.Compared with the monoculture strategy,the utilization of the designed co-cultures significantly improved the naringenin production,largely due to the reduction of metabolic stress,employment of proper hosts for improving pathway enzyme activities,and flexible adjustment of the relative biosynthetic strength between the coculture strains.The findings of this study extend the applicability of co-culture engineering in complex natural product biosynthesis.Vijaydev Ganesan Zhenghong Li Xiaonan Wang Haoran Zhang 2017Synthetic and Systems Biotechnology2017,2,3:13

5First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale.The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement.We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument.Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes.The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results,and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11
6The endocrine regulation network of growth hormone synthesis and secretion in fish: Emphasis on the signal integration in somatotropes显示文摘In teleosts, growth hormone (GH) production is governed by multiple neuroendocrine factors from the hypothalamus and other regulators from the pituitary and peripheral organs. Exploring the principles followed by pituitary somatotropes when differentiating and integrating the signals from these regulators at the cellular and intracellular level is essential for understanding the endocrine regulation network of growth hormone synthesis and secretion in fish. 7Effects of tourmaline on nitrogen removal performance and biofilm structures in the sequencing batch biofilm reactor显示文摘The effects of tourmaline on nitrogen removal performance and biofilm structures were comparatively investigated in two identical laboratory-scale sequencing batch biofilm reactors(SBBRs)(denoted SBBR1 and SBBR2) at different nitrogen loading rates(NLRs) varying from(0.24 ± 0.01) to(1.26 ± 0.02) g N/(L·day). SBBR1 was operated in parallel with SBBR2, but SBBR1 was filled with polyurethane foam loaded tourmaline(TPU) carriers and another(SBBR2) filled with polyurethane foam(PU) carriers. Results obtained from this study showed that the excellent and stable performance of SBBR1 was obtained. Ammonia nitrogen removal and total nitrogen removal were higher in SBBR1 than that in SBBR2 with increase of NLR. At an NLR of(0.24 ± 0.01) g N/(L·day), the majority of the spherical and elliptical bacteria were surrounded by the extracellular polymeric substance(EPS) and bacillus or filamentous bacteria in two SBBRs biofilms. When NLR increased to(1.26 ± 0.02) g N/(L·day), the clusters were more obvious in the SBBR1 biofilm than that in the SBBR2 biofilm. Bacteria in SBBR1 were inclined to synthesis more EPS, and the formed EPS could protect the bacteria from free ammonia(FA) under extreme condition NLR(1.26 ± 0.02) g N/(L·day). 8Microscopic structures of ionic liquids 1-ethyl-3-methylimidazolium tetrafluoroborate in water probed by the relative chemical shift显示文摘The relative chemical shifts (△δ) △δwere put forward to investigate the microscopic structure of 1-ethyl-3-methyl-imidazolium tetrafluoroborate (EmimBF4) during the dilution process with water.The concentration-dependent △δ(C2)H-(C4)H,△δ(C2)H-(C5)H and △δ(C4)H-(C5)H were analyzed.The results reveal that the variations of the microscopic structures of three aromatic protons are inconsistent.The strength of the H-bond between water and three aromatic protons follows the order:(C2)H···O > (C4)H···O > (C5)H···O.The concentration-dependent △δ(C6)H-(C7)H and △δ(C6)H-(C8)H indicate the formation of the H-bonds of (Calkyl)H···O is impossible,and more water is located around (C6)H than around (C7)H or (C8)H.The concentration-dependent △δ(C2)H-(C4)H and △δ(C2)H-(C5)H both increase rapidly when xwater > 0.9 or so,suggesting the ionic pairs of EmimBF4 are dissociated rapidly.The turning points of concentration-dependent △δ(C2)H-(C4)H and △δ(C2)H-(C5)H indicate that some physical properties of the EmimBF4/water mixtures also change at the corresponding concentration point.The microscopic structures of EmimBF4 in water could be clearly detected by the relative chemical shifts.XU YingJie GAO Yan ZHANG LiQun YAO Jia WANG CongMin LI HaoRan 2010Science China Chemistry2010,53,7:8
9Development of ecohydrological assessment tool and its application显示文摘The development of Hydro-Informatic Modelling System (HIMS) provides an integrated platform for hydrological simulation. To extend the application of HIMS, an ecohydrological modeling system named ecohydrological assessment tool (EcoHAT) has been developed. Integrating parameter-management tools, RS (remote sensing) inversion tools, module-design tools and GIS analysis tools, the EcoHAT provides an integrated tool to simulate ecohydrological processes on regional scale, which develops a new method on sustainable use of water. EcoHAT has been applied to several case studies, such as, the Yellow River Basin, the acid deposition area in Guizhou province and the riparian catchment of Guanting reservoir in Beijing. 10SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19.Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan 2021Cell Research2021,31,12:6

12Geochronology, Geochemistry, and Hf Isotopic Compositions of Monzogranites and Mafic-Ultramafic Complexes in the Maxingdawannan Area, Eastern Kunlun Orogen, Western China: Implications for Magma Sources, Geodynamic Setting, and Petrogenesis显示文摘This paper presents zircon U-Pb-Hf isotopic compositions and whole-rock geochemical data for monzogranites and mafic-ultramafic complexes of the Maxingdawannan area in the western end of the east Kunlun orogenic belt, western China. The data are used to determine the ages, petrogenesis, magma sources, and geodynamic setting of the studied rocks. U-Pb zircon dating indicates that monzogranites and gabbros of the complexes were emplaced at 399 and 397 Ma, respectively. The monzogranites are shoshonitic, with high SiO2, Al2 O3 and total-alkali contents, and low TFeO, MgO, TiO2 and P2 O5 contents. The mafic-ultramafic complexes are characterized by low Si O2 contents. The monzogranites display enrichment in light rare-earth elements(LREE) and large-ion lithophile elements(LILE), depletion in heavy REEs(HREE) and high-field-strength elements(HFSE), and negative Eu anomalies(Eu/Eu*=0.36–0.48). The mafic-ultramafic complexes are also enriched in LREEs and LILEs, and depleted in HREEs and HFSEs, with weak Eu anomalies(Eu/Eu*=0.84–1.16). Zircon εHf(t) values for the monzogranites and mafic-ultramafic complexes range from-6.68 to 1.11 and-1.81 to 6.29, with zircon model ages of 1 812–1 319 Ma(TDM2) and 1 087–769 Ma(TDM1), respectively. Hf isotopic data indicate that primary magmas of the monzogranites are originated from partial melting of ancient lower crust during the Paleo-Mesoproterozoic, with a juvenile-crust component. Primitive magmas of the mafic-ultramafic complexes are likely originated from a depleted-mantle source modified by slab-derived fluids and contaminated by crustal components. 13N-doped carbon quantum dots as fluorescent probes for highly selective and sensitive detection of Fe^3+ ions显示文摘To investigate the effect of nitrogen on the photoluminescence properties of carbon quantum dots (CO Ds), N-doped carbon quantum dots (N-CQDs)were synthesized by one-step hydrothermal treatment using biomass tar as the carbon precursor.As an inevitable organic pollutant,the unsaturated bonds in biomass tar,such as carboxylic acids,aldehydes,and aromatics,are favorable for formation of the graphitic carbon lattice.The obtained N-CQDs are spherical with an average particle size of 2.64nm and the crystal lattice spacing is 0.25nm,corresponding to the (100)facet of graphitic carbon.The N-CQDs emit bright blue photoluminescence under 365nm ultraviolet light,and they have excellent water solubility and stability with a high quantum yield of 26.1%.Coordination between the functional groups on the N-CQD surface and Fe^3+ ions is promoted because of the improved electronic properties and surface chemical reactivity caused by N atoms,leading to a significant fluorescence quenching effect of the N-CQDs in the presence of Fe^3+ions with high selectivity and sensitivity.There is a linear relationship between In (Fo/F)and the Fe^3+ concentration in the N-CQD concentration range 0.06-1400μmol/L with a detection limit of 60nmol/L, showing that the N-CQ.Ds have great potential as a fluorescent probe for Fe^3+detection.Xiangyi Deng Yali Feng Haoran Li Zhuwei Du Qing Teng Hongjun Wang 2018Particuology2018,16,6:5
14Marine Carbon-Sulfur Biogeochemical Cycles during the Steptoean Positive Carbon Isotope Excursion(SPICE) in the Jiangnan Basin, South China显示文摘Global occurrences of Steptoean Positive Carbon Isotope Excursion(SPICE) during Late Cambrian recorded a significant perturbation in marine carbon cycle, and might have had profound impacts on the biological evolution. In previous studies, SPICE has been reported from the Jiangnan slope belt in South China. To evaluate the bathymetric extent of SPICE, we investigate the limestone samples from the upper Qingxi Formation in the Shaijiang Section in the Jiangnan Basin. Our results show the positive excursions for both carbonate carbon(δ^(13)C) and organic carbon(δ^(13)C_(org)) isotopes, as well as the concurrent positive shifts in sulfur isotopes of carbonate associated sulfate(CAS, δ^(34)S_(CAS)) and pyrite(δ^(34)S_(pyrite)), unequivocally indicating the presence of SPICE in the Jiangnan Basin. A 4‰ increase in δ^(13)C_(carb) of the Qingxi limestone implies the increase of the relative flux of organic carbon burial by a factor of two. Concurrent positive excursions in δ^(34)S_(CAS) and δ^(34)S_(pyrite) have been attributed to the enhanced pyrite burial in oceans with extremely low concentration and spatially heterogeneous isotopic composition of seawater sulfate. Here, we propose that the seawater sulfur isotopic heterogeneity can be generated by volatile organic sulfur compound(VOSC, such as methanethiol and dimethyl sulfide) formation in sulfidic continental margins that were widespread during SPICE. Emission of 32S-enriched VOSC in atmosphere, followed by lateral transportation and aerobic oxidation in atmosphere, and precipitation in open oceans result in a net flux of ^(32)S from continental margins to open oceans, elevating δ^(34)S of seawater sulfate in continental margins. 18Chemoselective hydrogenation of phenol to cyclohexanol using heterogenized cobalt oxide catalysts显示文摘Cyclohexanol is an important intermediate in the synthesis of Nylon-6 and plasticizers. In this work,cobalt oxide nanoparticles(NPs) supported on porous carbon(Co O_x@CN) were fabricated by one-pot method and the hybrids could efficiently and selectively hydrogenate phenol to cyclohexanol with a high yield of 98%. The high catalytic performance of Co Ox@CN was associate with the high surface area(340 m^2/g) and uniformly dispersed NPs. Furthurmore, by detailed analysing the relationship between catalytic activity and catalysts composition, it clearly indicated that the Co_3O_4 in Co Ox@CN played an important role for the adsorption and activation of phenol and the in situ gernerated Co was responsible for hydrogen adsorption and dissociation. 19Optimization design of airfoils under atmospheric icing conditions for UAV显示文摘Natural ice accretion on the lifting surface of an aircraft is detrimental to its aerodynamic performance, as it changes the effective streamlined body. The main focus of this work considers the optimization design of airfoils under atmospheric icing conditions for the Unmanned Aerial Vehicle(UAV). The ice formation process is simulated by the Eulerian approach and the three-dimensional Myers model. A three-equation turbulence model is implemented to accurately predict the stall performance of the iced airfoil. In recognition of the real atmospheric variability in the icing parameters, the medium volume diameter of supercooled water droplets is treated as an uncertainty with an assumed probability density function. A technique of polynomial chaos expansion is used to propagate the input uncertainty through the deterministic system. The numerical results show that the multipoint/multiobjective optimization strategy can efficiently improve both the ice tolerance and the cruise performance of an airfoil. 20Three Tiaobu Feishen therapies protect human alveolar epithelial cells against cigarette smoking and tumor necrosis factor-mlear factor-kappa B pathwayα-induced inflamation by nuc显示文摘OBJECTIVE: To investigate the efficacy of Tiaobu Feishen formulae(TBFS), including Bufei Jianpi formula(BJF), Bufei Yishen formula(BYF), and Yiqi Zishen formula(YZF), on inflammatory response, protease-anti-protease imbalance and collagen deposition in rats.METHODS: In present work, we used an in vitro model of cigarette smoking extract(CSE)-and tumor necrosis factor-α(TNF-α)-induced A549 cellsto examine the efficacy of BJF, BYF and YZF on the production of inflammatory cytokines, including TNF-α and interleukin(IL)-8, IL-6, matrix metalloproteinases(MMP)-9, and IL-10 in CSE or TNF-ls. And their related transcripα-induced A549 celtion factors and signaling pathway were also analyzed.RESULTS: The results showed that BJF, BYF and YZF could significantly decrease the expression levels of the pro-inflammatory cytokines induced by CSE or TNF-α. Furthermore, BJF, BYF and YZF could suppress CSE-or TNF-α-induced activation of nuclear factor-kappa B(NF-κB) transcription factors and its corresponding pathways. Taken together, these data implied that BJF, BYF and YZF effectively inhibited CSE-or TNF-α-induced inflammatory response in alveolar epithelial cell, which was due to their inhibition effect on NF-κB pathways.CONCLUSION: Our findings suggest that the Tiaobu Feishen therapies may protect human alveolar epithelial cells against cigarette smoking and TNF-α-induced inflammation.
