| 1 | Transplantation of Human Bone Marrow Mesenchymal Stem Cell Ameliorates the Autoimmune Pathogenesis in MRL/lpr Mice显示文摘Recent evidence indicates that mesenchymal stem cells (MSC) possess immunosuppressive properties both in vitro and in vivo. We previously demonstrated the functional abnormality of bone marrow derived MSC in patients with systemic lupus erythematosus (SLE). In this study, we aimed to investigate whether transplantation of human bone marrow derived MSC affects the autoimmune pathogenesis in MRL/lpr mice. We found that human MSC from healthy donors reduced the proliferation of T lymphocytes from MRL/lpr mice in a dose-dependent fashion. Two weeks after in vivo transfer of MSC, we detected significantly reduced serum levels of anti ds-DNA antibodies and 24 hour proteinuria in MRL/lpr mice as compared with control groups without MSC transplantation. Moreover, flow cytometric analysis revealed markedly reduced number of CD4+ T cells while increased Th1 subpopulation in MSC group and MSC + CTX group when compared with controls. Histopathological examination showed significantly reduced renal pathology in MSC-treated mice. Immunohistochemical studies further revealed reduced expression of TGF-β, FN, VEGF and the deposition of complement C3 in renal tissue after MSC and MSC + CTX treatment. | 7 | Clinical characteristics and effectiveness of antiviral agents in hospitalized children with infectious mononucleosis in China:A multicenter retrospective study显示文摘Importance:The clinical characteristics of infectious mononucleosis(IM)in Chinese children have not been evaluated in multicenter studies,and the effectiveness of antiviral treatment are controversial.Objective:To investigate the clinical characteristics of Chinese children with IM and current status of antiviral therapy for affected patients.Methods:Hospitalized patients with IM were enrolled between 2018 and 2020 in five children’s hospitals in China.The clinical characteristics were compared among four age groups:<3 years,3-<6 years,6-<10 years,and≥10 years.The clinical characteristics of IM and effectiveness of antiviral therapy were compared among patients receiving acyclovir(ACV),ganciclovir(GCV),and no antiviral therapy(i.e.,non-antiviral group).Results:In total,499 patients were analyzed;most patients were 3-<6 years of age.The most common symptoms and signs included fever(100%),lymphadenopathy(98.6%),pharyngitis(86.4%),eyelid edema(76.8%),and snoring(72.9%).There were significant differences in rash,hepatomegaly,and liver dysfunction among the four age groups.Patients aged<3 years had a lower incidence of liver dysfunction and a higher incidence of rash.Among the 499 patients,50.1%were treated with GCV,26.3%were treated with ACV,and 23.6%received no antiviral therapy.Compared with the non-antiviral group,patients in the ACV and GCV groups had longer durations of fever(P<0.001).There were no significant differences in the incidences of complications among the three treatment groups.Interpretation:The incidence of IM in Chinese children peaked at 3-<6 years of age.Clinical features of IM varied according to age.Patients receiving antiviral therapy exhibited more serious clinical manifestations than did patients without antiviral therapy.The effectiveness of antiviral therapy for IM requires further analysis. | Huili Hu Huiling Deng Jing Bi Yi Xu Shuangjie Li Yue Xie Xinrong Sun Dongmeng Wang Xufang Li Wenxian Ouyang Bing Hu Yufeng Zhang He Tang Chunxiao Fang Hui Zhang Lingyun Guo Chen Wang Tianyi Wang Fengxia Yang Tao Jiang Zhengde Xie Gang Liu | 2021 | Pediatric Investigation2021,5,3: | 12 |

| 13 | Neural Mechanisms of Mental Fatigue Revisited: New Insights from the Brain Connectome显示文摘Maintaining sustained attention during a prolonged cognitive task often comes at a cost: high levels of mental fatigue. Heuristically, mental fatigue refers to a feeling of tiredness or exhaustion, and a disengagement from the task at hand;it manifests as impaired cognitive and behavioral performance. In order to effectively reduce the undesirable yet preventable consequences of mental fatigue in many real-world workspaces, a better understanding of the underlying neural mechanisms is needed, and continuous efforts have been devoted to this topic. In comparison with conventional univariate approaches, which are widely utilized in fatigue studies, convergent evidence has shown that multivariate functional connectivity analysis may lead to richer information about mental fatigue. In fact, mental fatigue is increasingly thought to be related to the deviated reorganization of functional connectivity among brain regions in recent studies. In addition, graph theoretical analysis has shed new light on quantitatively assessing the reorganization of the brain functional networks that are modulated by mental fatigue. This review article begins with a brief introduction to neuroimaging studies on mental fatigue and the brain connectome, followed by a thorough overview of connectome studies on mental fatigue. In this study, we constructed a recombinant adeno-associated virus(rAAV) vector expressing the human HIF-1α gene(rAAV-HIF-1α), and tested the assumption that rAAV-HIF-1α represses hippocampal neuronal apoptosis induced by amyloid-beta protein. Our results confirmed that rAAV-HIF-1α significantly reduces apoptosis induced by amyloid-beta protein in primary cultured hippocampal neurons. Direct intracerebral rAAV-HIF-1α administration also induced robust and prolonged HIF-1α production in rat hippocampus. Single rAAV-HIF-1α administration resulted in decreased apoptosis of hippocampal neurons in an Alzheimer's disease rat model established by intracerebroventricular injection of aggregated amyloid-beta protein(25–35). Existing AI approaches have failed to convert varied user sketches into artistically beautiful paintings while preserving their semantic concepts. To bridge this gap, we have developed Smart Paint, a co-creative drawing system based on generative adversarial networks(GANs), enabling a machine and a human being to collaborate in cartoon landscape painting. Smart Paint trains a GAN using triples of cartoon images, their corresponding semantic label maps, and edge detection maps. The machine can then simultaneously understand the cartoon style and semantics, along with the spatial relationships among the objects in the landscape images. The trained system receives a sketch as a semantic label map input, and automatically synthesizes its edge map for stable handling of varied sketches. It then outputs a creative and fine painting with the appropriate style corresponding to the human’s sketch. | 20 | Modeling of microstructure evolution of AZ80 magnesium alloy during hot working process using a unified internal state variable method显示文摘In this paper, a unified internal state variable(ISV) model for predicting microstructure evolution during hot working process of AZ80 magnesium alloy was developed. A novel aspect of the proposed model is that the interactive effects of material hardening, recovery and dynamic recrystallization(DRX) on the characteristic deformation behavior were considered by incorporating the evolution laws of viscoplastic flow, dislocation activities, DRX nucleation and boundary migration in a coupled manner. The model parameters were calibrated based on the experimental data analysis and genetic algorithm(GA) based objective optimization. The predicted flow stress, DRX fraction and average grain size match well with experimental results. The proposed model was embedded in the finite element(FE) software DEFORM-3 D via user defined subroutine to simulate the hot compression and equal channel angular extrusion(ECAE) processes.
