1Pollution characteristics and health risk assessment of benzene homologues in ambient air in the northeastern urban area of Beijing, China显示文摘Ambient benzene homologues were measured at a site in the northeastern urban area of Beijing, China, from August 24 to September 4, 2012 by SUMMA canister sampling followed by laboratory determination using cryogenic cold trap pre-concentration-GC-MS/FID, and their health risks were also assessed. Daily total benzene homologues ranged from 0.99 to 49.71 μg/m3with an average of 11.98 μg/m3. Benzene homologues showed higher concentrations in the morning and evening than that at noontime. Comparison with previous studies revealed a trend of decrease for ambient benzene homologues probably due to the efective emission control in Beijing in recent years. Vehicular exhaust was the main source while volatilization of paints and solvents also made substantial contributions. Health risk assessment showed that BTEX(benzene, toluene, ethylbenzene, o-xylene, m-xylene and p-xylene) and styrene had no appreciable adverse non-cancer health risks for the exposed population, while benzene has potential cancer risk of 1.34E-05. 2The calcium uniporter regulates the permeability transition pore in isolated cortical mitochondria显示文摘To investigate the influence of the mitochondrial calcium uniporter on the mitochondrial permeability transition pore, the present study observed mitochondrial morphology in cortical neurons isolated from adult rats using transmission electron microscopy, and confirmed the morphology and activity of isolated mitochondria by detecting succinic dehydrogenase and monoamine oxidase, two mitochondrial enzymes. Isolated mitochondria were treated with either ruthenium red, an inhibitor of the uniporter, spermine, an activator of the uniporter, or in combination with cyclosporin A, an inhibitor of the mitochondrial permeability transition pore. Results showed that ruthenium red inhibited CaCl2-induced mitochondrial permeability transition pore opening, spermine enhanced opening, and cyclosporin A attenuated the effects of spermine. 3A complex structure of arrestin-2 bound to a G protein-coupled receptor显示文摘Arrestins comprise a family of signal regulators of G-protein-coupled receptors(GPCRs),which include arrestins 1 to 4.While arrestins 1 and 4 are visual arrestins dedicated to rhodopsin,arrestins 2 and 3(Arr2 and Arr3)areβ-arrestins known to regulate many nonvisual GPCRs.The dynamic and promiscuous coupling of Arr2 to nonvisual GPCRs has posed technical challenges to tackle the basis of arrestin binding to GPCRs.Here we report the structure of Arr2 in complex with neurotensin receptor 1(NTSR1),which reveals an overall assembly that is strikingly different from the visual arrestin–rhodopsin complex by a 90°rotation of Arr2 relative to the receptor.In this new configuration,intracellular loop 3(ICL3)and transmembrane helix 6(TM6)of the receptor are oriented toward the N-terminal domain of the arrestin,making it possible for GPCRs that lack the C-terminal tail to couple Arr2 through their ICL3.Molecular dynamics simulation and crosslinking data further support the assembly of the Arr2‒NTSR1 complex.Sequence analysis and homology modeling suggest that the Arr2‒NTSR1 complex structure may provide an alternative template for modeling arrestin–GPCR interactions.Wanchao Yin Zhihai Li Mingliang Jin Yu-Ling Yin Parker Wde Waal Kuntal Pal Yanting Yin Xiang Gao Yuanzheng He Jing Gao Xiaoxi Wang Yan Zhang Hu Zhou Karsten Melcher Yi Jiang Yao Cong X.Edward Zhou Xuekui Yu H.Eric Xu 2019Cell Research2019,29,12:4

5Effect of Different Sowing Dates in South Henan 's Rice-growing Areas on the Growth and Yield of Ratoon Rice显示文摘In order to determine the optimal sowing date of ratoon rice in South Henan's rice-growing areas,this paper performs a comparative analysis of rice growth process,seedling quality and yield of first season rice and ratoon rice under different sowing date treatments. The results show that under climatic conditions( 2014),by using dry seedling cultivation in a small plastic shed,the growth of seedling sown on February21 was affected,while the sowing treatments from March 1 to April 11 can breed normal seedlings,and in this period,the maturity period of first season rice was delayed with prolonged sowing date,and ratoon rice yield declined with prolonged sowing date( total production of rice sown on 11 March reaching a peak). 6MXR7 facilitates liver cancer metastasis via epithelial-mesenchymal transition显示文摘MXR7 is a cell-surface protein and highly expressed in hepatocellular carcinoma(HCC). The aim of this study is to determine the expression profile of MXR7 in HCC and investigate the influence of MXR7 on invasion and metastasis of HCC cells. For this purpose, immunohistochemical assay was used to identify the differential expression of MXR7 in 94 HCC specimens. Expression of MXR7 in 4 pairs of HCC and portal vein tumor thrombus(PVTT) was also tested. The motility of HCC cells were characterized by transwell migration and matrigel invasion assays. In vivo metastasis potential was determined via tail vein injection assay.Moreover, compared with noninvasive HCC tumors or human HCC cell lines with low metastatic potential, invasive HCC samples and HCC cell lines with high metastatic potential exhibited higher MXR7 expression. Furthermore, forced expression of MXR7 in SMMC-7721 promoted cell proliferation, migration and invasion in vitro and accelerated tumor growth and metastasis in vivo. Conversely, knockdown of MXR7 expression in HuH7 cells inhibited proliferation and motility of cells. Mechanically,overexpression of MXR7 promoted epithelial-mesenchymal transition(EMT) progress, and MXR7 depletion repressed the EMT phenotype. 10Stability of inner dump slope under coal pillar support:case study in an open‑pit coal mine显示文摘The stability of an inner dump slope was investigated under the efect of coal pillar support considering the development position of dumping.Based on the instability mechanism and load distribution characteristics of the supporting coal pillar,the three-dimensional mechanical efects of the supporting coal pillar are characterized.Using the two-dimensional equivalent principle and the residual thrust method,the stability of the inner dump slope was analyzed under the efect of pillar support at diferent dump development positions.The quantitative efects of various factors on the inner dump slope stability were revealed,and the coal pillar shape parameters were optimized through numerical simulations.The results indicate that the slope stability coefcient is linearly related to the top width and height of the coal pillar,slope angle,and base inclination angle,and has an exponential relation with the coal pillar strike length and slope height increment.There are quadratic and absolute value relations with the coal pillar outer and the inner bottom angle,respectively.The top width of the coal pillar in the inner dump of Shengli East No.2 open-pit coal mine should be at a level of+824 m,and the optimal top width and height are 15 and 36.7 m,respectively.The instability mechanism of the supporting and retaining coal pillar obtained by numerical simulations and the stability of the inner dump are in good agreement with the theoretical analysis.Our results provide a theoretical basis for the design,treatment,and safe implementation of similar open-pit mine slope engineering.Guanghe Li Xiaoxu Yang Dong Wang Yanting Wang Xiangyu Yu 2022International Journal of Coal Science & Technology2022,9,2:1

12Efficient bright γ-ray vortex emission from a laser-illuminated light-fan-in-channel target显示文摘X/γ-rays have many potential applications in laboratory astrophysics and particle physics.Although several methods have been proposed for generating electron,positron,and X/γ-photon beams with angular momentum(AM),the generation of ultra-intense brilliant γ-rays is still challenging.Here,we present an all-optical scheme to generate a high-energy γ-photon beam with large beam angular momentum(BAM),small divergence,and high brilliance.In the first stage,a circularly polarized laser pulse with intensity of 10^(22) W/cm^(2) irradiates a micro-channel target,drags out electrons from the channel wall,and accelerates them to high energies via the longitudinal electric fields.During the process,the laser transfers its spin angular momentum(SAM)to the electrons’orbital angular momentum(OAM).In the second stage,the drive pulse is reflected by the attached fan-foil and a vortex laser pulse is thus formed.In the third stage,the energetic electrons collide head-on with the reflected vortex pulse and transfer their AM to the γ-photons via nonlinear Compton scattering.Three-dimensional particle-in-cell simulations show that the peak brilliance of the γ-ray beam is∼10^(22) photons·s^(-1)·mm^(-2)·mrad^(-2) per 0.1% bandwidth at 1 MeV with a peak instantaneous power of 25 TW and averaged BAM of 10^(6)h/photon.The AM conversion efficiency from laser to the γ-photons is unprecedentedly 0.67%.Hao Zhang Jie Zhao Yanting Hu Qianni Li Yu Lu Yue Cao Debin Zou Zhengming Sheng Francesco Pegoraro Paul McKenna Fuqiu Shao Tongpu Yu 2021High Power Laser Science and Engineering2021,9,3:1

16Laser chirp controlled relativistic few-cycle mid-infrared pulse generation显示文摘Relativistic few-cycle mid-infrared(mid-IR)pulses are unique tools for strong-field physics and ultrafast science,but are difficult to generate with traditional nonlinear optical methods.Here,we propose a scheme to generate such pulses with high efficiency via plasma-based frequency modulation with a negatively chirped laser pulse(NCLP).The NCLP is rapidly compressed longitudinally due to dispersion and plasma etching,and its central frequency is downshifted via photon deceleration due to the enhanced laser intensity and plasma density modulations.Simulation results show that few-cycle mid-IR pulses with the maximum center wavelength of 7.9µm and pulse intensity of a_(MIR)=2.9 can be generated under a proper chirp parameter.Further,the maximum energy conversion efficiency can approach 5.0%.Such a relativistic mid-IR source is promising for a wide range of applications.Dongao Li Guobo Zhang Jie Zhao Yanting Hu Yu Lu Hao Zhang Qianni Li Dongze Zhang Rong Sha Fuqiu Shao Zhengming Sheng Tongpu Yu 2023High Power Laser Science and Engineering2023,11,5:0
17Genome degradation promotes Salmonella pathoadaptation by remodeling fimbriae-mediated proinflammatory response显示文摘Understanding changes in pathogen behavior(e.g.increased virulence,a shift in transmission channel)is critical for the public health management of emerging infectious diseases.Genome degradation via gene depletion or inactivation is recognized as a pathoadaptive feature of the pathogen evolving with the host.However,little is known about the exact role of genome degradation in affecting pathogenic behavior,and the underlying molecular detail has yet to be examined.Using large-scale global avian-restricted Salmonella genomes spanning more than a century,we projected the genetic diversity of Salmonella Pullorum(bvSP)by showing increasingly antimicrobial-resistant ST92 prevalent in Chinese flocks.The phylogenomic analysis identified three lineages in bvSP,with an enhancement of virulence in the two recently emerged lineages(L2/L3),as evidenced in chicken and embryo infection assays.Notably,the ancestor L1 lineage resembles the Salmonella serovars with higher metabolic flexibilities and more robust environmental tolerance,indicating stepwise evolutionary trajectories towards avian-restricted lineages.Pan-genome analysis pinpointed fimbrial degradation from a virulent lineage.The later engineeredfim-deletion mutant,and all other five fimbrial systems,revealed behavior switching that restricted horizontal fecal-oral transmission but boosted virulence in chicks.By depleting fimbrial appendages,bvSP established persistent replication with less proinflammation in chick macrophages and adopted vertical transovarial transmission,accompanied by ever-increasing intensification in the poultry industry.Together,we uncovered a previously unseen paradigm for remodeling bacterial surface appendages that supplements virulence-enhanced evolution with increased vertical transmission.Xiao Zhou Xiamei Kang Jiaqi Chen Yan Song Chenghao Jia Lin Teng Yanting Tang Zhijie Jiang Xianqi Peng Xiaoxi Tao Yiwei Xu Linlin Huang Xuebin Xu Yaohui Xu Tengfei Zhang Shenye Yu Jiansen Gong Shaohui Wang Yuqing Liu Guoqiang Zhu Corinna Kehrenberg Franois-Xavier Weill Paul Barrow Yan Li Guoping Zhao Min Yue 2023National Science Review2023,10,10:0
18Synergistic effects of AAGL and anti-PD-1 on hepatocellular carcinoma through lymphocyte recruitment to the liver显示文摘Objective:Therapy for hepatocellular carcinoma(HCC)is a major challenge,and targeted therapies provide only a modest benefit in terms of overall survival.Treatment with antibodies to programmed cell death protein 1(PD-1)/PD-L1 can restore the functions of tumor-infiltrating T cells in HCC and has shown clinical efficacy in 20%of patients with advanced HCC.Novel approaches are urgently needed to treat HCC and to augment the efficacy of immunotherapy.Methods:Tumor-bearing mice were treated with Agrocybe aegerita galectin(AAGL)alone or in combination with anti-PD-1,and the tumor sizes and lifespans of mice were determined.Transcriptome analysis,cytokine analysis,flow cytometry analysis of the number and proportion of immune cell subsets in the liver and spleen,and molecular and cellular analyses of tumors were used to define the underlying mechanisms.Results:AAGL significantly inhibited the growth of liver tumors in a dose-dependent manner.Furthermore,AAGL increased the expression of multiple cytokines and chemokines in tumor-bearing mouse livers;this effect was associated with the activation and migration of T cells and macrophages,in agreement with the in vitro results.Importantly,the aggregation of T cells and macrophages induced by AAGL in tumor-bearing mouse livers clearly enhanced the response to PD-1 blockade immunotherapy.Conclusions:The results showed that AAGL induced the activation and migration of lymphocytes to the liver,and that the combination of AAGL and anti-PD-1 may be a promising strategy for HCC treatment.Xiangdong Ye Xueqing Wang Wenhui Yu Qing Yang Yan Li Yanxia Jin Yanting Su Jiaqi Song Bo Xu Hui Sun 2021Cancer Biology & Medicine2021,18,4:0
19Enhancing health-promoting isothiocyanates in Chinese kale sprouts via manipulating BoESP显示文摘Glucosinolates(GSLs)are a group of sulfur-containing secondarymetabolites,which are abundant in Brassica vegetables.GSL breakdown products(GBPs),especially isothiocyanates(ITCs)benefit human health.Chinese kale is a native Brassica vegetable in China,and its sprouts are rich in GSLs and nutritional substances.ITCs are the predominant GBPs while alternative products are formed in the presence of specifier proteins.However,fewer ITCs are formed in the sprouts.Epithiospecifier(ESP)promotes the formation of epithionitriles at the expense of ITCs in Arabidopsis,but a systematic study of different isoforms of ESPs in most vegetables is still missing.In this study,changes in the content of GBPs and the precursor GSLs,as well as thiols per plant were monitored during sprout development.The proportions of epithionitriles and ITCs in total GBPs were found to be increased and decreased,respectively.RNA-seq showed enhanced expression of numerous genes involved in GSLs biosynthesis and degradation,as well as sulfur assimilation in sprouts compared to seeds.Four copies of BoESPs were isolated and BoESP2 was the most abundant isoform.Generally,transcription of BoESPs showed a strong response to abscisic acid and gibberellin,and consequently epithionitriles increased under these treatments.Knockdown of BoESP2 expression through virus-induced gene silencing system could effectively increase total ITCs and decrease total epithionitriles.Overall,dynamic GSL metabolic f lux exists in the sprouting period,and the expression of BoESPs determines the pattern of GBPs,suggesting that improving the health-promoting ITCs in Chinese kale sprouts through manipulating BoESPs by metabolic engineering is feasible.Huiying Miao Chuchu Xia Shunhao Yu Jiansheng Wang Yanting Zhao Qiaomei Wang 2023Horticulture Research2023,10,4:0

Because microRNAs regulate the expression of hundreds of target genes, it is not surprising that they have profoundly important functions in brain development and pathological processes. For example, miR-124 plays an important role in inducing and maintaining neuronal identity through targeting at least two anti-neural factors. MicroRNAs have also been implicated in brain disorders, including brain tumors and neurodegenerative diseases.
