Pollution characteristics and health risk assessment of benzene homologues in ambient air in the northeastern urban area of Beijing, China显示文摘Ambient benzene homologues were measured at a site in the northeastern urban area of Beijing, China, from August 24 to September 4, 2012 by SUMMA canister sampling followed by laboratory determination using cryogenic cold trap pre-concentration-GC-MS/FID, and their health risks were also assessed. Daily total benzene homologues ranged from 0.99 to 49.71 μg/m3with an average of 11.98 μg/m3. Benzene homologues showed higher concentrations in the morning and evening than that at noontime. Comparison with previous studies revealed a trend of decrease for ambient benzene homologues probably due to the efective emission control in Beijing in recent years. Vehicular exhaust was the main source while volatilization of paints and solvents also made substantial contributions. Health risk assessment showed that BTEX(benzene, toluene, ethylbenzene, o-xylene, m-xylene and p-xylene) and styrene had no appreciable adverse non-cancer health risks for the exposed population, while benzene has potential cancer risk of 1.34E-05. Available data from cities in China all implied that benzene imposes relatively higher cancer risk on the exposed populations and therefore strict control measures should be taken to further lower ambient benzene levels in China.Lei Li Hong Li Xinmin Zhang Li Wang Linghong Xu Xuezhong Wang Yanting Yu Yujie Zhang Guan Cao 2014Journal of Environmental Sciences2014,26,1:40
Effectiveness of Jinying capsule on pelvic inflammatory disease in patients with symptom pattern of damp and heat accumulation: a double-blinded, multicenter, randomized, placebo-controlled clinical trial显示文摘OBJECTIVE: To evaluate the therapeutic effectiveness and safety of Jinying capsule on pelvic inflammatory disease(PID) in patients with symptoms identified as the pattern of damp and heat accumulation in terms of Traditional Chinese Medicine(TCM).METHODS: We conducted a double-blinded, multicenter, randomized, placebo-controlled clinical trial which included 155 patients diagnosed with PID and identified as symptom pattern of damp and heat accumulation. They were randomly divided into experimental group(n = 78) and control group(n = 77) according to a random number table. The treatment lasted for a period of 28 d. The experimental group was given Jinying capsules and oral levofloxacin plus oral metronidazole for first 7 d.They continued with Jinying capsules and levofloxacin placebo and metronidazole placebo for another 7 d. For the remaining 14 d, they continued with Jinying capsules only. Whereas, the control group was treated with oral levofloxacin and metronidazole and Jinying capsule placebo for the first 14 d in the same way as the experimental group and then continued with Jinying capsule placebo only for the remaining 14 d. The clinical efficacy was assessed using McCormack scale, TCM symptom pattern scores, physicochemical indexes including white blood cell and erythrocyte sedimentation rate, C-reaction protein, smear of vaginal discharge, and pelvic ultrasound.RESULTS: Comparing McCormack scale between both groups after treatment, the difference in curative effect between both groups was significant (P = 0.0269). The cure rate of the experimental group and control group is 76.32% and 59.46% respectively at week 4. Comparing TCM symptom pattern scores between both groups before and after treatment, the differences in total effective rate were both significant(P < 0.05). The curative effect rate of experimental group is 2.63% and 13.70% of the control group at week 1(P = 0.0131), and73.33% of the experimental group and 56.94% of the control group at week 4(P = 0.0369). No significant differences were found between the two groups on the Physicochemical indexes(all P >0.05). No adverse events or reactions occurred in the experimental groups.CONCLUSION: Jinying capsule can reduce the dosage of antibiotics needed for PID treatment, and improve the symptoms in PID patients.Yang Yan Lu Yao Zhou Deping Pei Fenglan Li Qin Chen Shuqiong Wang Qiufeng Wang Yanting Jin Zhe Liu Yanxia 2020Journal of Traditional Chinese Medicine2020,40,3:10

CuFe2O4@GO nanocomposite as an effective and recoverable catalyst of peroxymonosulfate activation for degradation of aqueous dye pollutants显示文摘Recently, heterogeneous activation of peroxymonosulfate(PMS) to oxidatively degrade organic pollutants has been a hotspot. In the present work, copper ferrite-graphite oxide hybrid(CuFe2 O4@GO)was prepared and used as catalyst to activate PMS for degradation of methylene blue(MB) in aqueous solution.A high degradation efficiency(93.3%) was achieved at the experimental conditions of20 mg/L MB,200 mg/L CuFe2 O4@GO,0.8 mmol/L PMS,and 25℃temperature.Moreover,CuFe2 O4@GO showed an excellent reusability and stability. The effects of various operational parameters including pollutant type, solution pH,catalyst dosage,PMS dosage, pollutant concentration, temperature, natural organic matter(NOM),and inorganic anions on the catalytic degradation process were comprehensively investigated and elucidated. The further mechanistic study revealed the Cu(Ⅱ)/Cu(Ⅰ) redox couple on CuFe2 O4@GO played the dominant role in PMS activation, where both hydroxyl and sulfate radicals were generated and proceeded the degradation of pollutants. In general,CuFe2 O4@GO is a promising heterocatalyst for PMS-based advanced oxidation processes(AOPs) in wastewater treatment.Xiaoman Lei Menghan You Fei Pan Min Liu Peng Yang Dongsheng Xia Qiang Li Yanting Wang Jie Fu 2019Chinese Chemical Letters2019,30,12:8
Cloning of Ln Gene Through Combined Approach of Map-based Cloning and Association Study in Soybean显示文摘Increasing yield is one of the most important goals in crop breeding.Soybean(Glycine max L.Merr.),one of the most economically important leguminous seed crops, provides the majority of plant proteins, and more than a quarter of the world's food and animal feed(Graham and Vance,2003). The yield of soybean is finally determined by the number of seeds per unit area, which affected by many characters, such as height, branching number, photosynthesis, seed size, seed number. The number of seeds per pod is taken for one of the critical components that related to yield(You et al.,1995),and it has long been considered in soybean production(Takahashi,Chao Fang Weiyu Li Guiquan Li Zheng Wang Zhengkui Zhou Yanming Ma Yanting Shen Congcong Li Yunshuai Wu Baoge Zhu Weicai Yang Zhixi Tian 2013Journal of Genetics and Genomics2013,40,2:7
Atorvastatin attenuates surgery-induced BBB disruption and cognitive impairment partly by suppressing NF-κB pathway and NLRP3 inflammasome activation in aged mice显示文摘In clinic, perioperative neurocognitive disorder is becoming a common complication of surgery in old patients.Neuroinflammation and blood-brain barrier(BBB)disruption are important contributors for cognitive impairment.Atorvastatin,as a strong HMG-CoA reductase inhibitor, has been widely used in clinic. However,it remains unclear whether atorvastatin could prevent anesthesia and surgery-induced BBB disruption and cognitive injury by its anti-inflammatory property. In this study, aged C57BL/6J mice were used to address this question.Initially, the mice were subject to atorvastatin treatment for 7 days(10 mg/kg).After a simple laparotomy under 1.5%isoflurane anesthesia,Morris water maze was performed to assess spatial learning and memory.Western blot analysis, immunohistochemistry, and enzyme-linked immunosorbent assay were used to examine the inflammatory response,BBB integrity, and cell apoptosis.Terminal-deoxynucleotidyl transferase mediated nick end labeling assay was used to assess cell apoptosis. The fluorescein sodium and transmission electron microscopy were used to detect the permeability and structure of BBB.The results showed that anesthesia and surgery significantly injured hippocampal-dependent learning and memory, which was ameliorated by atorvastatin.Atorvastatin could also reverse the surgery-induced increase of systemic and hippocampal cytokines, including IL-1β,TNF-α,and IL-6,accompanied by inhibiting the nuclear factor kappa-B(NF-κB)pathway and Nucleotide-Binding Oligomerization Domain,or Leucine Rich Repeat and Pyrin Domain Containing 3(NLRP3)inflammasome activation,as well as hippocampal neuronal apoptosis. In addition, surgery triggered an increase of BBB permeability, paralleled by a decrease of the ZO-1,occludin,and Claudin 5 proteins in the hippocampus. However, atorvastatin treatment could protect the BBB integrity from the impact of surgery,by up-regulating the expressions of ZO-1,occludin,and Claudin 5.These findings suggest that atorvastatin exhibits neuroprotective effects on cognition in aged mice undergoing surgery.Pengfei Liu Quansheng Gao Lei Guan Yanting Hu Jingwen Jiang Teng Gao Weixuan Sheng Xinying Xue Hui Qiao Tianzuo Li 2021Acta Biochimica et Biophysica Sinica2021,53,5:6
Design and baseline data of a populationbased metabonomics study of eye diseases in eastern China: the Yueqing Ocular Diseases Investigation显示文摘Background:China is undergoing a massive transition toward an urban and industrial economy. These changes will restructure the demographics and economy which will eventually influence the future patterns of disease. The risk factors of vision-impairing eye diseases remain ambiguous and poorly understood.Metabolomics is an ideal tool to understand and shed light on the ocular disease mechanisms for earlier treatment. This article aims to describe the design, methodology and baseline data of the Yueqing Ocular Diseases Investigation(YODI),a developed county population-based study to determine the prevalence and primary causes of visual impairment;also with metabonomics analysis we aimed to identify, predict and suggest some preventive biomarkers that cause blindness. Methods:A population-based,cross-sectional study.Randomized clustering sampling was used to identify adults aged 50 years and older in Xiangyang Town,Yueqing county-level City. The interviews covered demographic, behavioral, ocular risk factors and mental health state. The ocular examination included visual acuity, autorefraction, intraocular pressure, anterior and posterior segment examinations, fundus photography, retinal tomography and angiography, and visual field testing.Anthropometric measurements included height and weight, waist and hip circumference, blood pressure, pulse rate, electrocardiogram, and abdominal ultrasound scan.A venous blood sample was collected for laboratory tests and metabonomics studies. Results:Of the 5319 individuals recruited for the YODI,4769(89.7%)subjects were enrolled for analyses. The median age was 62.0 years, and 45.6%were male. The educational level of illiteracy or semi-illiteracy,primary,middle and high school or above was 29.8%,45.5%,20.1%,and 3.3%,respectively.Majority of the participants were female, younger, and less educated when compared with nonparticipants. The average body mass index and waist-hip ratios were 24.4±3.4 kg/m^(2) and 0.9±0.1 respectively.Blood sample collection reached a sample size of 1909(479 from subjects with selfreported diabetes and 1430 from one-third of the 4290 subjects without self-reported diabetes). Conclusions:The YODI provides population-based data with a high response rate(89.7%)on the prevalence and primary causes of major vision-impairing eye diseases in developed county areas in eastern China.Metabonomics analysis from YODI will provide further association of metabolic characteristics with the visual impairment eye diseases. The risk prediction model could be created and has the potential to be generalized to developed eastern areas in China for prevention.Yuxuan Deng Yuanbo Liang Sigeng Lin Liang Wen Jin Li Yue Zhou Meixiao Shen Jingwei Zheng Kemi Feng Yanting Sun Kwapong Willaim Robert Jia Qu Fan Lu 2020Eye and Vision2020,7,1:4
Research progress in protein post-translational modification显示文摘Protein post-translational modification plays an important role in organism. It makes the protein obtain more complicated structures, perfect functions, more accurate regulations and more spe-cific operations. The most common protein post- translational modifications include ubiquitylation, phosphorylation, glycosylation, lipodation, methyla-tion, and acetylation and so on. Ubiquitylation plays an essential role in cellular functions such as cellular differentiation, apoptosis, DNA repair, antigen proc-essing, and stress response. Phosphorylation is re-lated to physiological and pathological processes including cellular signal conduction, nervous activity, muscle contraction and proliferation, development and differentiation of cells. Protein glycosylation is of great importance for many cell processes like im-munoprotection, virus replication, cell growth, and occurrence of inflammation and so on. Lipodation is vital to signal conduction. Histone methylation and acetylation are responsible for transcription regula-tion. In vivo, different post-translational modifications do not occur isolatedly, but influence each other’s function and cooperate with each other. Under-standing what influences the post-translational modi-fications will help to uncover cellular processes and protein network in molecular level and finally direct more precise drug design targeting molecules. Post-translational modification mimics are set to dominate the next wave of protein therapeutics and become powerful medicinal tools in the 21st century.HU Jia GUO Yanting LI Yanme 2006Chinese Science Bulletin2006,51,6:4
The calcium uniporter regulates the permeability transition pore in isolated cortical mitochondria显示文摘To investigate the influence of the mitochondrial calcium uniporter on the mitochondrial permeability transition pore, the present study observed mitochondrial morphology in cortical neurons isolated from adult rats using transmission electron microscopy, and confirmed the morphology and activity of isolated mitochondria by detecting succinic dehydrogenase and monoamine oxidase, two mitochondrial enzymes. Isolated mitochondria were treated with either ruthenium red, an inhibitor of the uniporter, spermine, an activator of the uniporter, or in combination with cyclosporin A, an inhibitor of the mitochondrial permeability transition pore. Results showed that ruthenium red inhibited CaCl2-induced mitochondrial permeability transition pore opening, spermine enhanced opening, and cyclosporin A attenuated the effects of spermine. Results demonstrated that the mitochondrial calcium uniporter plays a role in regulating the mitochondrial permeability transition pore in mitochondria isolated from the rat brain cortex.Ning Yu Shilei Wang Peng Wang Yu Li Shuhong Li Li Wang Hongbing Chen Yanting Wang 2012Neural Regeneration Research2012,7,2:4
A complex structure of arrestin-2 bound to a G protein-coupled receptor显示文摘Arrestins comprise a family of signal regulators of G-protein-coupled receptors(GPCRs),which include arrestins 1 to 4.While arrestins 1 and 4 are visual arrestins dedicated to rhodopsin, arrestins 2 and 3(Arr2 and Arr3)areβ-arrestins known to regulate many nonvisual GPCRs.The dynamic and promiscuous coupling of Arr2 to nonvisual GPCRs has posed technical challenges to tackle the basis of arrestin binding to GPCRs.Here we report the structure of Arr2 in complex with neurotensin receptor 1(NTSR1),which reveals an overall assembly that is strikingly different from the visual arrestin–rhodopsin complex by a 90°rotation of Arr2 relative to the receptor. In this new configuration, intracellular loop 3(ICL3)and transmembrane helix 6(TM6)of the receptor are oriented toward the N-terminal domain of the arrestin, making it possible for GPCRs that lack the C-terminal tail to couple Arr2 through their ICL3.Molecular dynamics simulation and crosslinking data further support the assembly of the Arr2‒NTSR1 complex.Sequence analysis and homology modeling suggest that the Arr2‒NTSR1 complex structure may provide an alternative template for modeling arrestin–GPCR interactions.Wanchao Yin Zhihai Li Mingliang Jin Yu-Ling Yin Parker Wde Waal Kuntal Pal Yanting Yin Xiang Gao Yuanzheng He Jing Gao Xiaoxi Wang Yan Zhang Hu Zhou Karsten Melcher Yi Jiang Yao Cong X.Edward Zhou Xuekui Yu H.Eric Xu 2019Cell Research2019,29,12:4
Mechanism underlying the retarded nuclear translocation of androgen receptor splice variants显示文摘As shown in our previous study, two alternatively spliced androgen receptor(AR) variants, which are exclusively expressed in the granulosa cells of patients with polycystic ovary syndrome, exhibit retarded nuclear translocation compared with wild-type AR. However, researchers have not yet determined whether these abnormalities correlate with heat shock protein 90(HSP90)and importin α(the former is a generally accepted co-chaperone of AR, and the latter is a component of classical nuclear import complexes). Here, these two variants were mainly retained in cytoplasm with HSP90 and importin α in the presence of dihydrotestosterone(DHT), and their levels in nucleus were significantly reduced, according to the immunofluorescence staining. The binding affinity of two AR variants for importin α was consistently decreased, while it was increased in WT-AR following DHT stimulation, leading to reduced nuclear import, particularly for the insertion-AR(Ins-AR). However, the binding affinities of two AR variants for HSP90 were increased in the absence of DHT compared with WT-AR, which functioned to maintain spatial structural stability, particularly for the deletion-AR(Del-AR). Therefore, the retarded nuclear translocation of two AR variants is associated with HSP90 and importin α, and the abnormal binding affinities for them play critical roles in this process.Ye Liu Yinyu Wang Fangfang Wang Jiexue Pan Jingjing Xu Jingyi Li Chengliang Zhou Guolian Ding Yanting Wu Xinmei Liu Jianzhong Sheng Hefeng Huang 2019Science China(Life Sciences)2019,62,2:4
Synthesis, characteristics of hierarchical EU-1 zeolite for xylene isomerization probe reaction显示文摘Introduced a method of synthesizing hierarchical EU-1 zeolite with organosilanes as additive, and studied the influences of following different kinds of organosilanes on the synthesis of hierarchical EU-1 zeolite: γ-glycidoxy propyl trimethoxy silane(GPTMS), N-β-(aminoethyl)-γ-aminopropyl methyl dimethoxyl silane(APAEDMS),and N-(β-aminoethyl)-γ-aminopropyl dimethoxyl(ethyoxyl) silane(TMPED). The hierarchical EU-1 samples were characterized by XRD, SEM, N_2 adsorption, FT-IR and NH_3-TPD to analyze the crystallinity, morphology, surface area, pore size distribution and acidity. The results showed that hierarchical EU-1 zeolites were successfully synthesized; organosilanes have great influence on crystal morphology of EU-1 zeolites; the exterior surface area of hierarchical EU-1 zeolite, which synthesized with organosilanes(APAEDMS) adding into synthesis system, increased by 62.1% and mesopore volume increased by 129.1% compared with conventional EU-1 zeolites, thus can reduce the diffusional restriction markedly in catalytic reaction. The catalytic performance of hierarchical EU-1zeolites were evaluated in m-xylene isomerization on fixed bed reactor. The catalytic data showed that the isomerization activity PX/X of the hierarchical EU-1 zeolites reached around 24.09% in theoretical thermodynamic equilibrium from 23.83%, and the selectivity of C_8 aromatic hydrocarbon increased from 75.16% to 84.87%. 14Platelet-mediated adhesion facilitates leukocyte sequestration in hypoxia-reoxygenated microvessels显示文摘Leukocyte transendothelial migration and sequestration are two distinct outcomes following leukocyte adhesion to endothelium during ischemia-reperfusion injury, in which platelets may play a pivotal role. In the present study, we established an in vitro hypoxia-reoxygenation model to mimic ischemia-reperfusion injury and found platelet pre-incubation significantly increased leukocyte adhesion to endothelial cells after hyoxia-reoxygenation(over 67%). Blockade of endothelial-cell-expressed adhesion molecules inhibited leukocyte direct adhesion to endothelial cells, while platelet-mediated leukocyte adhesion was suppressed by blockade of platelet-expressed adhesion molecules. Further experiments revealed platelets acted as a bridge to mediate leukocyte adhesion, and platelet-mediated adhesion was the predominant pattern in the presence of platelets. However, platelet pre-incubation significantly suppressed leukocyte transendothelial migration after hypoxia-reoxygenation(over 31%), which could be aggravated by blockade of endothelial-cell-expressed adhesion molecules, but alleviated by blockade of platelet-expressed adhesion molecules. This would indicate that platelet-mediated adhesion disrupted leukocyte transendothelial migration. An in vivo mesenteric ischemia-reperfusion model demonstrated leukocyte transfusion alone caused mild leukocyte adhesion to reperfused vessels and subsequent leukocyte infiltration, while simultaneous leukocyte and platelet transfusion led to massive leukocyte adhesion and sequestration within reperfused microvessels. Our studies revealed platelets enhanced leukocyte adhesion to endothelial cells, but suppressed leukocyte transendothelial migration. Overall, this leads to leukocyte sequestration in hypoxia-reoxygenated microvessels.Senfeng Zheng Yanting Cao Wenjian Zhang Honglin Liu Jia You Yiqing Yin Jinning Lou Chenghui Li 2016Science China(Life Sciences)2016,59,3:3
Characterization of Electroless Ni-P Deposits on Surface of AZ91D Magnesium Alloy显示文摘Characterization of electroless nickel deposits on magnesium alloys was investigated by X-ray diffraction(XRD),scanning electron microscope(SEM),energy dispersive spectrometer(EDS)and electrochemical potentiodynamic polarization analysis. The result of XRD showed that the electroless nickel deposits on magnesium alloys were composed mostly of amorphous Ni-P and partially of microcrystalline nickel. The phosphorus content in the Ni-P deposits was 10.8%by the calculation of EDS.SEM micrographs showed that morphology of electroless nickel deposits on magnesium alloys was a typical cellular structure, and the growth of cellular structure was influenced by surface condition of substrate.Zinc layer between substrate and electroless nickel deposits was not found by the results of SEM and EDS,which indicated that the zinc layer on the surface of magnesium alloy was very thin, and the zinc would dissolved while being immersed in electroless nickel plating solution. The thin zinc layer was almost dissolved completely before Ni-P plating. The result of electrochemical potentiodynamic polarization analysis showed that the corrosion potential Ecorr of electroless nickel deposits in 3.5%NaCl solution was about-0.53 V,which was 1 V higher than that of magnesium alloys substrate. And corrosion current density of Ni-P deposits in 3.5%NaCl solution was much smaller than that of magnesium alloys substrate at the same potential value, which indicated that corrosion resistance of electroless Ni-P deposits was excellent.Discoloration time of concentrated nitric acid test for electroless nickel deposits on magnesium alloys was 445 s,which also indicated excellent corrosion resistance.Li Jiansan Guo Yanting Liu Junquan 2012稀有金属材料与工程2012,41,S1:3
SPDEF suppresses head and neck squamous cell carcinoma progression by transcriptionally activating NR4A1显示文摘SAM pointed domain containing E26 transformation-specific transcription factor(SPDEF)plays dual roles in the initiation and development of human malignancies. However, the biological role of SPDEF in head and neck squamous cell carcinoma(HNSCC)remains unclear. In this study, the expression level of SPDEF and its correlation with the clinical parameters of patients with HNSCC were determined using TCGA-HNSC,GSE65858,and our own clinical cohorts.CCK8,colony formation, cell cycle analysis, and a xenograft tumor growth model were used to determine the molecular functions of SPDEF in HNSCC.ChIP-qPCR,dual luciferase reporter assay, and rescue experiments were conducted to explore the potential molecular mechanism of SPDEF in HNSCC.Compared with normal epithelial tissues,SPDEF was significantly downregulated in HNSCC tissues.Patients with HNSCC with low SPDEF mRNA levels exhibited poor clinical outcomes.Restoring SPDEF inhibited HNSCC cell viability and colony formation and induced G0/G1 cell cycle arrest, while silencing SPDEF promoted cell proliferation in vitro. The xenograft tumor growth model showed that tumors with SPDEF overexpression had slower growth rates, smaller volumes, and lower weights.SPDEF could directly bind to the promoter region of NR4A1 and promoted its transcription, inducing the suppression of AKT,MAPK,and NF-κB signaling pathways. Moreover, silencing NR4A1 blocked the suppressive effect of SPDEF in HNSCC cells. Here,we demonstrate that SPDEF acts as a tumor suppressor by transcriptionally activating NR4A1 in HNSCC.Our findings provide novel insights into the molecular mechanism of SPDEF in tumorigenesis and a novel potential therapeutic target for HNSCC.Yanting Wang Xianyue Ren Weiyu Li Ruoyan Cao Suyang Liu Laibo Jiang Bin Cheng Juan Xia 2021International Journal of Oral Science2021,13,4:3
Key Recovery Against 3DES in CPU Smart Card Based on Improved Correlation Power Analysis显示文摘The security of CPU smart cards, which are widely used throughout China,is currently being threatened by side-channel analysis.Typical countermeasures to side-channel analysis involve adding noise and filtering the power consumption signal. In this paper,we integrate appropriate preprocessing methods with an improved attack strategy to generate a key recovery solution to the shortcomings of these countermeasures. Our proposed attack strategy improves the attack result by combining information leaked from two adjacent clock cycles.Using our laboratory-based power analysis system,we verified the proposed key recovery solution by performing a successful correlation power analysis on a Triple Data Encryption Standard(3DES)hardware module in a real-life 32-bit CPU smart card. All 112 key bits of the 3DES were recovered with about 80 000 power traces.Yanting Ren Liji Wu Hexin Li Xiangyu Li Xiangmin Zhang An Wang Hongyi Chen 2016Tsinghua Science and Technology2016,21,2:3
PINK1 kinase dysfunction triggers neurodegeneration in the primate brain without impacting mitochondrial homeostasis显示文摘In vitro studies have established the prevalent theory that the mitochondrial kinase PINK1 protects neurodegeneration by removing damaged mitochondria in Parkinson's disease(PD).However, difficulty in detecting endogenous PINK1 protein in rodent brains and cell lines has prevented the rigorous investigation of the in vivo role of PINK1.Here we report that PINK1 kinase form is selectively expressed in the human and monkey brains.CRISPR/Cas9-mediated deficiency of PINK1 causes similar neurodegeneration in the brains of fetal and adult monkeys as well as cultured monkey neurons without affecting mitochondrial protein expression and morphology.Importantly,PINK1 mutations in the primate brain and human cells reduce protein phosphorylation that is important for neuronal function and survival. Our findings suggest that PINK1 kinase activity rather than its mitochondrial function is essential for the neuronal survival in the primate brains and that its kinase dysfunction could be involved in the pathogenesis of PD.Weili Yang Xiangyu Guo Zhuchi Tu Xiusheng Chen Rui Han Yanting Liu Sen Yan Qi Wang Zhifu Wang Xianxian Zhao Yunpeng Zhang Xin Xiong Huiming Yang Peng Yin Huida Wan Xingxing Chen Jifeng Guo Xiao-Xin Yan Lujian Liao Shihua Li Xiao-Jiang Li 2022Protein & Cell2022,13,1:3
