| 2 | Oncofetal antigen glypican-3 as a promising early diagnostic marker for hepatocellular carcinoma显示文摘BACKGROUND:Hepatocellular carcinoma (HCC) is characterized by a multi-cause,multi-stage and multi-focus process of tumor progression.Its prognosis is poor and early diagnosis is of utmost importance.This study was undertaken to investigate the dynamic expression of oncofetal antigen glypican-3 (GPC-3) and GPC-3 mRNA in hepatocarcinogenesis and to explore their early diagnostic value for HCC.METHODS:A hepatoma model was induced in male Sprague-Dawley rats with 0.05% 2-fluorenylacetamide and confirmed by hematoxylin and eosin staining and gamma-glutamyltransferase (GGT) expression.Total RNA was purified and transcribed into cDNA by reverse transcription.Fragments of the GPC-3 gene were amplified by nested RT-PCR,and confirmed by sequencing.GPC-3 was analyzed by immunohistochemistry,Western blotting or ELISA.RESULTS:Positive GPC-3 expression showed as brown granule-like staining localized in the cytoplasm.Histological examination of hepatocytes revealed three morphological stages of granule-like degeneration,atypical hyperplasia (precancerous),and cancer formation,with a progressive increase of liver total RNA and GGT expression.The incidence of liver GPC-3 mRNA and GPC-3,and serum GPC-3 was 100%,100% and 77.8% in the HCC group,100%,100%,and 66.7% in the precancerous group,83.3%,83.3%,and 38.9% in the degeneration group,and no expression in the liver or blood of the control group,respectively.There was a positive correlation between liver GPC-3 mRNA and total RNA level (r=0.475,P<0.05) or liver GPC-3 (r=1.0,P<0.001) or serum GPC-3 (r= 0.994,P<0.001).CONCLUSION:Abnormal oncofetal antigen GPC-3 and GPC-3 mRNA expression in hepatocarcinogenesis may be promising molecular markers for early diagnosis of HCC. | Research Center of Clinical Medicine,Affiliated Hospital of Nantong University,Nantong 226001,China (Yao M,Yao DF,Qiu LW,Wu W,Sai WL,Yang JL and Zhang HJ) Department of Oncology,Yancheng First People’s Hospital,Yancheng 224001,China (Bian YZ) Department of Oncology,Second Affiliated Hospital,Nanjing Medical University,Nanjing 210011,China (Zhang CG) | 2011 | Hepatobiliary & Pancreatic Diseases International2011,10,3: | 51 |
| 3 | Tibetan Plateau climate dynamics: recent research progress and outlook显示文摘This paper reviews progress in the study of Tibetan Plateau(TP) climate dynamics over the past decade.Several theoretical frameworks, including thermal adaptation and the TP sensible heat(SH) driving air-pump, have been developed to identify the mechanisms responsible for the circulation anomaly produced by thermal forcing of the TP. Numerical simulations demonstrate that the thermal efects of large-scale orography, including the Tibetan and Iranian Plateaus(TIP), are crucial for the formation of the East Asian and South Asian summer monsoons(SASM) because the surface SH of the TIP is the major driver of the water vapor transport required for the genesis of the north branch of the SASM. he large-scale orography of the TP afects the Asian climate through thermal forcing in spring and summer, and mechanical forcing in winter. he TP forcing can also inluence the Asian summer monsoon(ASM) onset over the Bay of Bengal(BOB) by enhancing the BOB warm pool at the surface and by modulating the South Asian High(SAH) in the upper troposphere. On intra-seasonal timescales, the TP thermal forcing signiicantly modulates spring rainfall in southern China and generates the biweekly oscillation of the SAH in summer. Despite climate warming, the atmospheric heat source over the TP, particularly the spring SH,exhibits a clear weakening trend from the 1980 s to 2000 s. his weakening of the spring SH contributed to the anomalous ‘dry in the north' and ‘wet in the south' rainfall patern observed over East China. | 4 | Environmental issues from coal mining and their solutions显示文摘The environmental challenges from coal mining include coal mine accidents,land subsidence,damage to the water environment,mining waste disposal and air pollution.These are either environmental pollution or landscape change.A conceptual framework for solving mine environmental issues is proposed.Clean processes,or remediation measures,are designed to address environmental pollution.Restoration measures are proposed to handle landscape change.The total methane drainage from 56 Chinese high methane concentration coal mines is about 101.94 million cubic meters.Of this methane,19.32 million,35.58 million and 6.97 million cubic meters are utilized for electricity generation,civil fuel supplies and other industrial purposes,respectively.About 39% of the methane is emitted into the atmosphere.The production of coal mining wastes can be decreased 10% by reuse of mining wastes as underground fills,or by using the waste as fuel for power plants or for raw material to make bricks or other infrastructure materials.The proper use of mined land must be decided in terms of local physical and socio-economical conditions.In European countries more than 50% of previously mined lands are reclaimed as forest or grass lands.However,in China more than 70% of the mined lands are reclaimed for agricultural purposes because the large population and a shortage of farmlands make this necessary.Reconstruction of rural communities or native residential improvement is one environmental problem arising from mining.We suggest two ways to reconstruct a farmer's house in China. | BIAN, Zhengfu INYANG, Hilary I DANIELS, John L OTTO, Frank STRUTHERS, Sue | 2010 | Mining Science and Technology2010,20,2: | 47 |

| 8 | A Positioning System based on Communication Satellites and the Chinese Area Positioning System(CAPS)显示文摘The Chinese Area Positioning System(CAPS) is a positioning system based on satellite communication that is fundamentally different from the 3'G'(GPS,GLONASS and GALILEO) systems.The latter use special-purpose navigation satellites to broadcast navi-gation information generated on-board to users,while the CAPS transfers ground-generated navigation information to users via the communication satellite.In order to achieve accurate Positioning,Velocity and Time(PVT),the CAPS employs the following strategies to over-come the three main obstacles caused by using the communication satellite:(a) by real-time following-up frequency stabilization to achieve stable frequency;(b) by using a single carrier in the transponder with 36 MHz band-width to gain sufficient power;(c) by incorporating Decommissioned Geostationary Orbit communication satellite(DGEO),barometric pressure and Inclined Geostationary Orbit communication satellite(IGSO) to achieve the 3-D posi-tioning.Furthermore,the abundant transponders available on DGEO can be used to realize the large capacity of communication as well as the integrated navigation and communication.With the communication functions incorporated,five new functions appear in the CAPS:(1) combination of navigation and communication;(2) combination of navigation and high accu-racy orbit measurement;(3) combination of navigation message and wide/local area differen-tial processing;(4) combination of the switching of satellites,frequencies and codes;and(5) combination of the navigation message and the barometric altimetry.The CAPS is thereby labelled a PVT5C system of high accuracy.In order to validate the working principle and the performance of the CAPS,a trial system was established in the course of two years at a cost of about 20 million dollars.The trial constellation consists of two GEO satellites located at E87.5° and E110.5°,two DGEOs located at E130° and E142°,as well as barometric altimetry as a virtual satellite.Static and dynamic performance tests were completed for the Eastern,the Western,the Northern,the Southern and the Middle regions of China.The evaluation results are as follows:(1) land static test,plane accuracy range:C/A code,15~25m;P code,5~10 meters;altitude accuracy range,1~3 m;(2) land dynamic test,plane accuracy range,C/A code,15~25m;P code,8~10m;(3) velocity accuracy,C/A code,0.13~0.3ms-1,P code,0.15~0.17ms-1;(4) timing accuracy,C/A code,160ns,P code,13 ns;(5) timing compared accuracy of Two Way Satellite Time and Frequency Transfer(TWSTFT),average accuracy,0.068ns;(6) random error of the satellite ranging,10.7 mm;(7) orbit determination accuracy,better than 2 m.The above stated random error is 1σ error.At present,this system is used as a preliminary operational system and a complete system with 3 GEO,3 DGEO and 3 IGSO is being established. | Guo-Xiang Ai Hu-Li Shi Hai-Tao Wu Yi-Hua Yan Yu-Jing Bian Yong-Hui Hu Zhi-Gang Li Ji Guo Xian-De Cai | 2008 | Chinese Journal of Astronomy and Astrophysics2008,8,6: | 33 |
| 9 | Effect of preoperative transcatheter arterial chemoembolization on angiogenesis of hepatocellular carcinoma cells显示文摘AIM:To evaluate the effects of four types of preoperative transcatheter arterial chemoembolization(TACE)on angiogenesis of hepatocellular carcinoma(HCC)cells. METHODS:A total of 136 patients with HCC underwent liver resection.One to five courses of TACE prior to liver resection were performed in 79 patients(TACE group),in which one to four courses of chemotherapy alone were performed in 11 patients(group A);one to five courses of chemotherapy combined with iodized oil were performed in 33 patients(group B);one to three courses of chemotherapy combined with iodized oil and gelatin sponge were performed in 23 patients(group C); one to three courses of chemotherapy combined with iodized oil,ethanol and gelatin sponge were performed in 12 patients(group D).The other 57 patients only received liver resection(non-TACE group).The microves- sels were marked by CD31.The expression of CD31 and vascular endothelial growth factor(VEGF)protein were detected by immunohistochemical methods. RESULTS:The mean microvessel density(MVD)in HCC cells was significantly higher in groups A,B,C and D than in the non-TACE group(P<0.05).The expression of VEGF protein in HCC cells were significantly higher in groups A,B,C and D than in the non-TACE group(P<0.05).MVD and the expression of VEGFprotein were positively correlated.Mean MVD and the expression of VEGF protein were closely related to the number of courses of TACE and the interval of TACE. | 13 | Efficacy and safety of Xiangsha Liujunzi granules for functional dyspepsia: A multi-center randomized double-blind placebo-controlled clinical study显示文摘AIM To assess the efficacy and safety of a Chinese herbal medicine(CHM), Xiangsha Liujunzi granules, in the treatment of patients with functional dyspepsia(FD). METHODS We performed a randomized, double-blind, placebocontrolled trial with patients from three centers. Two hundred and sixteen subjects diagnosed with FD according to ROME Ⅲ criteria and confirmed by upper gastrointestinal endoscopy and spleen-deficiency and Qi-stagnation syndrome were selected to receive Xiangsha Liujunzi granules or placebo for 4 wk in a 2:1 ratio by blocked randomization. The subjects also received follow-up after the 4-wk intervention. Herbal or placebo granules were dissolved in 300 m L of water. Participants in both groups were administered 130 m L(45 ℃) three times a day. Participants were evaluated prior to and following 4 wk of the intervention in terms of changes in the postprandial discomfort severity scale(PDSS) score, clinical global impression(CGI) scale score, hospital anxiety and depression scale(HADS) score, traditional Chinese medicine symptoms score(SS), scores of various domains of the 36-item short form health survey(SF-36), gastric emptying(GE) and any observed adverse effects. RESULTS Compared with the placebo group, patients in the CHM group showed significant improvements in the scores of PDSS, HADS, SS, SF-36 and CGI scale(P < 0.05 or P < 0.01). | 14 | Correlation of human epidermal growth factor receptor 2 expression with clinicopathological characteristics and prognosis in gastric cancer显示文摘AIM:To investigate human epidermal growth factor receptor 2(HER2) gene amplification and protein expression in Chinese patients with resectable gastric cancer and the association with clinicopathological characteristics and survival.METHODS:One hundred and ninety-seven gastric cancer patients who underwent curative surgery procedures were enrolled into this study.HER2 gene amplification and protein expression were examined using fluorescence in-situ hybridization(FISH) and immunohistochemistry(IHC) analysis on formalin-fixed paraffinembedded gastric cancer samples from all patients.For scoring,Hofmann's HER2 gastric cancer scoring system was adopted.All cases showing IHC3+ or FISH positiv-ity were defined as HER2 positive.Patient clinicopathological data and survival information were collected.Finally,χ 2 statistical analysis was performed to analyze the HER2 positivity rate amongst the subgroups with different clinicopathological characteristics including;gender,age,tumor location,Lauren classification,differentiation,TNM staging,depth of invasion,lymph node metastases and distant metastasis.The probability of survival for different subgroups with different clinicopathological characteristics was calculated using the Kaplan-Meier method and survival curves plotted using log rank inspection.RESULTS:According to Hofmann's HER2 gastric cancer scoring criteria,31 cases(15.74%) were identified as HER2 gene amplified and 19 cases(9.64%) were scored as strongly positive for HER2 membrane staining(3+),25 cases(12.69%) were moderately positive(2+) and 153 cases(77.66%) were HER2 negative(0/1+).The concordance rate between IHC and FISH analyses was 88.83%(175/197).Thirty-six cases were defined as positive for HER2 gene amplification and/or protein expression,with 24 of these cases being eligible for Herceptin treatment according to United States recommendations,and 29 of these cases eligible according to EU recommendations.Highly consistent results were detected between IHC3+,IHC0/1 and FISH(73.68% and 95.42%),but low consistency was observed between IHC2+ and FISH(40.00%).The positivity rates in intestinal type and well-differentiated gastric cancer were higher than those in diffuse/mixed type and poorly-differentiated gastric cancer respectively(28.57% vs 13.43%,P = 0.0103;37.25% vs 11.64%,P < 0.0001),but were not correlated with gender,age,tumor location or TNM stage,depth of invasion,lymph node metastases and distant metastasis.In poorly-differentiated gastric cancer patients,those without lymph node metastasis showed a higher HER2 positivity rate than those with lymph node metastasis(26.47% vs 7.14%,P = 0.0021).This association was not present in thosepatients with well-differentiated gastric cancer(28.57% vs 43.33%,P = 0.2832).Within our patient cohort,26 cases were lost to follow-up.The median survival time for the remaining 171 patients was 18 mo.The median survival times of the HER2 positive and negative groups were 17 and 18.5 mo respectively.Overall survival was not significantly different between HER2-positive and negative groups(χ 2 = 0.9157,P = 0.3386),but in patients presenting well-differentiated tumors,the overall survival of the HER2-positive group was significantly worse than that of the HER2-negative group(P = 0.0123).In contrast,patients with poorly differentiated and diffuse/mixed subtype gastric cancers showed no significant differences in overall survival associated with HER2.Furthermore,the median survival time of the HER2 positive group did not show any statistically significant differences when compared to the subgroups of gender,age,tumor location,TNM classification,lymph node metastases and distant metastasis.CONCLUSION:Patients with intestinal type gastric cancer(GC),well-differentiated GC and poorly-differentiated GC without lymph node metastasis,may all represent suitable candidates for targeted therapy using Herceptin. | Chao He Xue-Yi Bian Xing-Zhi Ni Dan-Ping Shen Yan-Ying Shen Hua Liu Zhi-Yong Shen Qiang Liu | 2013 | World Journal of Gastroenterology2013,19,14: | 28 |
| 15 | Extending the CONSORT Statement to moxibustion显示文摘The STandards for Reporting Interventions in Clinical Trials Of Moxibustion (STRICTOM), in the form of a checklist and descriptions of checklist items, were designed to improve reporting of moxibustion trials, and thereby facilitating their interpretation and replication. The STRICTOM checklist included 7 items and 16 sub-items. These set out reporting guidelines for the moxibustion rationale, details of moxibustion, treatment regimen, other components of treatment, treatment provider background, control and comparator interventions, and precaution measures. In addition, there were descriptions of each item and examples of good reporting. It is intended that the STRICTOM can be used in conjunction with the main CONSORT Statement, extensions for nonpharmacologic treatment and pragmatic trials, and thereby raise the quality of reporting of clinical trials of moxibustion. | 17 | Hydrogen sulfide regulates abiotic stress tolerance and biotic stress resistance in Arabidopsis显示文摘Hydrogen sulfide(H_2S) is an important gaseous molecule in various plant developmental processes and plant stress responses.In this study,the transgenic Arabidopsis thaliana plants with modulated expressions of two cysteine desulfhydrases,and exogenous H_2S donor(sodium hydrosulfide,NaHS) and H_2S scavenger(hypotaurine,HT) pre-treated plants were used to dissect the involvement of H_2S in plant stress responses.The cysteine desulfhydrases overexpressing plants and NaHS pre-treated plants exhibited higher endogenous H_2S level and improved abiotic stress tolerance and biotic stress resistance,while cysteine desulfhydrases knockdown plants and HT pre-treated plants displayed lower endogenous H_2S level and decreased stress resistance.Moreover,H_2S upregulated the transcripts of multiple abiotic and biotic stress-related genes,and inhibited reactive oxygen species(ROS) accumulation.Interestingly,MIR393-mediated auxin signaling including MIR393a/b and their target genes(TIR1,AFB1,AFB2,and AFB3) was transcriptionally regulated by H_2S,and was related with H_2S-induced antibacterial resistance.Moreover,H_2S regulated 50 carbon metabolites including amino acids,organic acids,sugars,sugar alcohols,and aromatic amines.Taken together,these results indicated that cysteine desulfhydrase and H_2S conferred abiotic stress tolerance and biotic stress resistance,via affecting the stress-related gene expressions,ROS metabolism,metabolic homeostasis,and MIR393-targeted auxin receptors. | Haitao Shi Tiantian Ye Ning Han Hongwu Bian Xiaodong Liu Zhulong Chan | 2015 | Journal of Integrative Plant Biology2015,57,7: | 26 |
| 18 | Impact of psychological stress on irritable bowel syndrome显示文摘Psychological stress is an important factor for the development of irritable bowel syndrome(IBS). More and more clinical and experimental evidence showed that IBS is a combination of irritable bowel and irritable brain. In the present review we discuss the potential role of psychological stress in the pathogenesis of IBS and provide comprehensive approaches in clinical treatment. Evidence from clinical and experimental studies showed that psychological stresses have marked impact on intestinal sensitivity, motility, secretion and permeability, and the underlying mechanism has a close correlation with mucosal immune activation, alterations in central nervous system, peripheral neurons and gastrointestinal microbiota. Stress-induced alterations in neuro-endocrine-immune pathways acts on the gut-brain axis and microbiota-gut-brain axis, and cause symptom flare-ups or exaggeration in IBS. IBS is a stresssensitive disorder, therefore, the treatment of IBS should focus on managing stress and stress-induced responses. Now, non-pharmacological approaches and pharmacological strategies that target on stress-related alterations, such as antidepressants, antipsychotics, miscellaneous agents, 5-HT synthesis inhibitors, selective 5-HT reuptake inhibitors, and specific 5-HT receptor antagonists or agonists have shown a critical role in IBS management. | 19 | QTLs for Sugar Content of Stalk in Sweet Sorghum(Sorghum bicolor L.Moench)显示文摘High sugar content of sorghum stalk is an important factor in the sorghum silage production. To identify the genomic regions controlling sugar content and to develop molecular markers linked to sugar content in sweet sorghum, we used an F2:3 segregating population consisting of 207 individuals derived from a cross between a high sugar content inbred line, Early Folger, and a normal inbred line, N32B, for genetic linkage mapping and quantitative trait locus (QTL) analysis. We constructed a genetic linkage map spanning 983.5 cM based on a total of 327 markers comprising 31 restriction fragment length polymorphism (RFLP) markers, 254 amplified fragment length polymorphism (AFLP) markers, and 42 simple sequence repeat (SSR) markers. In the 20 linkage groups detected, 98.2% of markers aligned to the 10 linkage groups of sorghum. Variations in sugar content at different growth stages and among internodes suggested that the sugar content of middle internodes is stable and suitable for measuring at early dough stage. The broad sense heritability (hB2) of sugar content was 0.64 and 0.62 estimated from the data of F3 families and each parent in 2003 and 2004. We identified one and two QTLs accounting for 22.2 to 25.0% of phenotypic variance using simple interval mapping method in 2003 and 2004, respectively. These two QTLs showed a negative additive effect, and over-dominance effect. A QTL on LG-D was detected in both two years.
