| 1 | A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide. | Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen | 2019 | Molecular Plant2019,12,5: | 18 |
| 2 | Chromosome-scale genome assembly provides insights into the evolution and flavor synthesis of passion fruit (Passiflora edulis Sims)显示文摘Passion fruit(Passiflora edulis Sims)is an economically valuable fruit that is cultivated in tropical and subtropical regions of the world.Here,we report an~1341.7Mb chromosome-scale genome assembly of passion fruit,with 98.91%(~1327.18Mb)of the assembly assigned to nine pseudochromosomes.The genome includes 23,171 protein-coding genes,and most of the assembled sequences are repetitive sequences,with long-terminal repeats(LTRs)being the most abundant.Phylogenetic analysis revealed that passion fruit diverged after Brassicaceae and before Euphorbiaceae.Ks analysis showed that two whole-genome duplication events occurred in passion fruit at 65 MYA and 12 MYA,which may have contributed to its large genome size.An integrated analysis of genomic,transcriptomic,and metabolomic data showed that‘alpha-linolenic acid metabolism’,‘metabolic pathways’,and‘secondary metabolic pathways’were the main pathways involved in the synthesis of important volatile organic compounds(VOCs)in passion fruit,and this analysis identified some candidate genes,including GDP-fucose Transporter 1-like,Tetratricopeptide repeat protein 33,protein NETWORKED 4B isoform X1,and Golgin Subfamily A member 6-like protein 22.In addition,we identified 13 important gene families in fatty acid pathways and eight important gene families in terpene pathways.Gene family analysis showed that the ACX,ADH,ALDH,and HPL gene families,especially ACX13/14/15/20,ADH13/26/33,ALDH1/4/21,and HPL4/6,were the key genes for ester synthesis,while the TPS gene family,especially PeTPS2/3/4/24,was the key gene family for terpene synthesis.This work provides insights into genome evolution and flavor trait biology and offers valuable resources for the improved cultivation of passion fruit. | Zhiqiang Xia Dongmei Huang Shengkui Zhang Wenquan Wang Funing Ma Bin Wu Yi Xu Bingqiang Xu Di Chen Meiling Zou Huanyu Xu Xincheng Zhou Rulin Zhan Shun Song | 2021 | Horticulture Research2021,8,1: | 15 |

| 6 | The effects of altered BMP4 signaling in first branchialarch- derived murine embryonic orofacial tissues显示文摘The first branchial arch(BA1),which is derived from cranial neural crest(CNC)cells,gives rise to various orofacial tissues.Cre mice are widely used for the determination of CNC and exploration of gene functions in orofacial development.However,there is a lack of Cre mice specifically marked BA1’s cells.Pax2-Cre allele was previously generated and has been widely used in the field of inner ear development.Here,by compounding Pax2-Cre and R26R-mTmG mice,we found a specific expression pattern of Pax2^(+)cells that marked BA1’s mesenchymal cells and the BA1-derivatives.Compared to Pax2-Cre;R26R-mTmG allele,GFP^(+)cells were abundantly found both in BA1 and second branchial arch in Wnt1-Cre;R26R-mTmG mice.As BMP4 signaling is required for orofacial development,we over-activated Bmp4 by using Pax2-Cre;pMes-BMP4 strain.Interestingly,our results showed bilateral hyperplasia between the upper and lower teeth.We also compare the phenotypes of Wnt1-Cre;pMes-BMP4 and Pax2-Cre;pMes-BMP4 strains and found severe deformation of molar buds,palate,and maxilla-mandibular bony structures in Wnt1-Cre;pMes-BMP4 mice;however,the morphology of these orofacial organs were comparable between controls and Pax2-Cre;pMes-BMP4 mice except for bilateral hyperplastic tissues.We further explore the properties of the hyperplastic tissue and found it is not derived from Runx2^(+)cells but expresses Msx1,and probably caused by abnormal cell proliferation and altered expression pattern of p-Smad1/5/8.In sum,our findings suggest altering BMP4 signaling in BA1-specific cell lineage may lead to unique phenotypes in orofacial regions,further hinting that Pax2-Cre mice could be a new model for genetic manipulation of BA1-derived organogenesis in the orofacial region. | Jue Xu Meiling Chen Yanan Yan Qiaoxue Zhao Meiying Shao Zhen Huang | 2021 | International Journal of Oral Science2021,13,4: | 3 |
| 7 | Vacuum carburization of 12Cr2Ni4A low carbon alloy steel with lanthanum and cerium ion implantation显示文摘As bearing parts, 12 Cr2 Ni4 A is expected to have high hardness and excellent fatigue strength, so carburizing is employed to improve the inherit properties of 12 Cr2 Ni4 A. However, the traditional carburizing is limited by poor microstructure distribution and low rate of carburizing. The rare earth ion implantation is known to help improving the properties of tribology, corrosion resistance and oxidation resistance of metal. In this article, the RE implantation is employed to assist the carburizing. Lanthanum and cerium ion implantations are initially used to assist 12 Cr2 Ni4 A low pressure vacuum carburization.The microstructure, content of retained austenite, hardness, thickness of layer and carbon diffusion were analyzed by optical microscopy(OM), scanning electron microscopy(SEM), X-ray diffraction(XRD) and Rockwell/Vickers hardness tester, respectively. It was shown that lanthanum and cerium implantations can improve structure of the vacuum carburizing layer, and enhance the uniformity of carbon element distribution on the carburized surface. Meanwhile the RE implantation plays a positive role in promoting the surface hardness and carburized rate. The lanthanum element has more significant effect on surface hardness and content of retained austenite than cerium element. | 14 | Experimental Wave Attenuation Study over Flexible Plants on a Submerged Slope显示文摘Using plants is a kind of environmentally-friendly coastal protection to attenuate wave energy. In this paper, a set of experiments were conducted to investigate the wave attenuation performance using flexible grasses on a submerged slope, and the wave attenuation coefficient for these experiments was calculated for different still water depths, slope and grass configurations. It was found that the slope plays a significant role in wave attenuation. The wave attenuation coefficient increases with increasing relative row number and relative density. For a small relative row number, the two configurations from the slope top to its toe and from the slope toe to its top performed equally to a large extent. For a medium relative row number, the configuration from the slope toe to its top performed more poorly than that from the slope top to its toe; however, it performed better than that from the slope top to its toe for a high relative row number. With a single row of grasses close to the slope top from the slope toe, the wave attenuation coefficient shows double peaks. With increasing grass rows or still water depth, the grass location corresponding to the maximum wave attenuation coefficient is close to the slope top.
