3Chromosome-scale genome assembly provides insights into the evolution and flavor synthesis of passion fruit (Passiflora edulis Sims)显示文摘Passion fruit(Passiflora edulis Sims)is an economically valuable fruit that is cultivated in tropical and subtropical regions of the world.Here,we report an~1341.7Mb chromosome-scale genome assembly of passion fruit,with 98.91%(~1327.18Mb)of the assembly assigned to nine pseudochromosomes.The genome includes 23,171 protein-coding genes,and most of the assembled sequences are repetitive sequences,with long-terminal repeats(LTRs)being the most abundant.Phylogenetic analysis revealed that passion fruit diverged after Brassicaceae and before Euphorbiaceae.Ks analysis showed that two whole-genome duplication events occurred in passion fruit at 65 MYA and 12 MYA,which may have contributed to its large genome size.An integrated analysis of genomic,transcriptomic,and metabolomic data showed that‘alpha-linolenic acid metabolism’,‘metabolic pathways’,and‘secondary metabolic pathways’were the main pathways involved in the synthesis of important volatile organic compounds(VOCs)in passion fruit,and this analysis identified some candidate genes,including GDP-fucose Transporter 1-like,Tetratricopeptide repeat protein 33,protein NETWORKED 4B isoform X1,and Golgin Subfamily A member 6-like protein 22.In addition,we identified 13 important gene families in fatty acid pathways and eight important gene families in terpene pathways.Gene family analysis showed that the ACX,ADH,ALDH,and HPL gene families,especially ACX13/14/15/20,ADH13/26/33,ALDH1/4/21,and HPL4/6,were the key genes for ester synthesis,while the TPS gene family,especially PeTPS2/3/4/24,was the key gene family for terpene synthesis.This work provides insights into genome evolution and flavor trait biology and offers valuable resources for the improved cultivation of passion fruit.Zhiqiang Xia Dongmei Huang Shengkui Zhang Wenquan Wang Funing Ma Bin Wu Yi Xu Bingqiang Xu Di Chen Meiling Zou Huanyu Xu Xincheng Zhou Rulin Zhan Shun Song 2021Horticulture Research2021,8,1:15
4Original position statistic distribution analysis study of low alloy steel continuous casting billet显示文摘The homogeneity of low alloy steels continuous casting billet obtained under different technological conditions has been investigated by original position statistic distribution analysis technique. On the basis of systematic analysis of ten thousands primary optical signals at the corresponding original positions, the quantitative statistic distribution information of each element was obtained. The biggest degrees of segregation of low alloy steel continuous casting billet were calculated accurately according to the quantitative distribution maps of the contents. It was suggested that the weight ratio in a certain content range was used to judge the homogeneity of the materials, and the two models —— the total weight ratio of contents (the degree of statistic homogeneity, H) within the permissive content range (C0±R) and the median value confidence extension ratio (the degree of statistic segregation, S) at 95% of confidence limit of weight ratio —— were put forward. The two models reflect the composition and state distribution regularity of the metal materials in a large region. 5The apricot(Prunus armeniaca L.)genome elucidates Rosaceae evolution and beta-carotenoid synthesis显示文摘Apricots,scientifically known as Prunus armeniaca L,are drupes that resemble and are closely related to peaches or plums.As one of the top consumed fruits,apricots are widely grown worldwide except in Antarctica.A high-quality reference genome for apricot is still unavailable,which has become a handicap that has dramatically limited the elucidation of the associations of phenotypes with the genetic background,evolutionary diversity,and population diversity in apricot.DNA from P.armeniaca was used to generate a standard,size-selected library with an average DNA fragment size of~20 kb.The library was run on Sequel SMRT Cells,generating a total of 16.54 Gb of PacBio subreads(N50=13.55 kb).The high-quality P.armeniaca reference genome presented here was assembled using long-read single-molecule sequencing at approximately 70×coverage and 171×Illumina reads(40.46 Gb),combined with a genetic map for chromosome scaffolding.The assembled genome size was 221.9 Mb,with a contig NG50 size of 1.02 Mb.Scaffolds covering 92.88%of the assembled genome were anchored on eight chromosomes.Benchmarking Universal Single-Copy Orthologs analysis showed 98.0%complete genes.We predicted 30,436 protein-coding genes,and 38.28%of the genome was predicted to be repetitive.We found 981 contracted gene families,1324 expanded gene families and 2300 apricot-specific genes.The differentially expressed gene(DEG)analysis indicated that a change in the expression of the 9-cis-epoxycarotenoid dioxygenase(NCED)gene but not lycopene beta-cyclase(LcyB)gene results in a lowβ-carotenoid content in the white cultivar“Dabaixing”.This complete and highly contiguous P.armeniaca reference genome will be of help for future studies of resistance to plum pox virus(PPV)and the identification and characterization of important agronomic genes and breeding strategies in apricot.Fengchao Jiang Junhuan Zhang Sen Wang Li Yang Yingfeng Luo Shenghan Gao Meiling Zhang Shuangyang Wu Songnian Hu Haoyuan Sun Yuzhu Wang 2019Horticulture Research2019,6,1:9
6Akt Inhibitor Perifosine Prevents Epileptogenesis in a Rat Model of Temporal Lobe Epilepsy显示文摘Accumulating data have revealed that abnormal activity of the mTOR(mammalian target of rapamycin)pathway plays an important role in epileptogenesis triggered by various factors. We previously reported that pretreatment with perifosine, an inhibitor of Akt(also called protein kinase B), abolishes the rapamycin-induced paradoxical increase of S6 phosphorylation in a rat model induced by kainic acid(KA). Since Akt is an upstream target in the mTOR signaling pathway, we set out to determine whether perifosine has a preventive effect on epileptogenesis. Here, we explored the effect of perifosine on the model of temporal epilepsy induced by KA in rats and found that pretreatment with perifosine had no effect on the severity or duration of the KA-induced status epilepticus. However, perifosine almost completely inhibited the activation of p-Akt and p-S6 both acutely and chronically following the KA-induced status epilepticus.Perifosine pretreatment suppressed the KA-induced neuronal death and mossy fiber sprouting. The frequency of spontaneous seizures was markedly decreased in rats pretreated with perifosine. Accordingly, rats pretreated with perifosine showed mild impairment in cognitive functions. 7Frequency-domain diagonal extension imaging显示文摘The pixel size of a charge-coupled device(CCD)camera plays a major role in the image resolution,and the square pixels are attributed to the physical anisotropy of the sampling frequency.We synthesize the high sampling frequency directions from multiple frames acquired with different angles to enhance the resolution by 1.4×over conventional CCD orthogonal sampling.To directly demonstrate the improvement of frequency-domain diagonal extension(FDDE)microscopy,lens-free microscopy is used,as its resolution is dominantly determined by the pixel size.We demonstrate the resolution enhancement with a mouse skin histological specimen and a clinical blood smear sample.Further,FDDE is extended to lens-based photography with an ISO 12233 resolution target.This method paves a new way for enhancing the image resolution for a variety of imaging techniques in which the resolution is primarily limited by the sampling pixel size,for example,microscopy,photography,and spectroscopy.Shan Jiang Meiling Guan Jiamin Wu Guocheng Fang Xinzhu Xu Dayong Jin Zhen Liu Kebin Shi Fan Bai Shu Wang Peng Xi 2020Advanced Photonics2020,2,3:3
8Genetic Analysis and Mapping of the Purple Gene in Purple Heading Chinese Cabbage显示文摘To analyze genetic linkage and map purple gene(Br Pur) controlling purple inner leaves trait of Chinese cabbage, a F2 population was constructed by selfing a F1 plant from homozygous purple heading line ‘14S839' and homozygous orange heading line ‘14S162'. The phenotype investigation of head color showed that the segregation ratio of purple to non-purple individuals was consistent with the expected ratio of 3:1,which indicated that purple inner leaves trait is controlled by a single dominant gene. A total of 297 SSRs in whole genome of Chinese cabbage were tested by modified Bulked Segregant Analysis(BSA) method. Two linked markers flanking Br Pur, A710 and A714, were obtained and Br Pur was mapped on linkage group A07 based on the sequence of these two markers. Subsequently, two new markers flanking Br Pur, CL-12 and B214-87, were developed based on two known anthocyanins-related genes, Br4CL3 and Bra004214. 14Nonhematopoietic erythropoietin derivative protects cardiomyocytes from hypoxia/reoxygenation-induced apoptosis显示文摘Objective:Carbamylated EPO(CEPO)is a derivative of erythropoietin(EPO)by subjecting it to carbamylation.It does not stimulate erythropoiesis,but effectively protects tissue from injury.The present study was to investigate the effect of CEPO treatment using in vitro models of hypoxia/reoxygenation(H/R).Methods:Cardiomyocytes were exposed to hypoxia(95% N2 and 5% CO2)for 1 hour followed by 4 hours of reoxygenation(95% O2 and 5% CO2).CEPO was administered after hypoxia,just before reoxygenation.The apoptotic cardiomyocytes were determined by flow cytometry.The level of protein was assessed by western blot analysis.Results:CEPO treatment significantly decreased the apoptotic cardiomyocytes by 54.20% compared with H/R group.Western blot analysis showed that CEPO administration increased the level of Bcl-2(an antiapoptotic protein)by 62.22% compared with H/R group.Conclusion:Acute administration of CEPO protected cardiomyocytes from H/R-induced apoptosis.CEPO protected cardiomyocytes with a concomitant upregulation of Bcl-2 after H/R injury.Xuan Xu Xiaohong Shan Zhijuan Cao Meiling Wu Qi Chen Yuehua Li 2008Journal of Nanjing Medical University2008,22,2:2
15Microstructure and heat treatment for Ni3Al-based single crystal alloy with different crystal orientations显示文摘Ni3Al-based single crystal alloy IC6SX with different crystal orientations were prepared by seed crystal method.The microstructure and heat treatment of the alloy were investigated.The results showed that the microstructure of the alloy was in dendrite structure,and the crystal orientation had significant effect on the dendrite morphology of this alloy.The precipitated phases of(MoNi)6C and NiMo appeared in the microstructure of the three alloys with different crystal orientations during solidification process.Compared with other two alloys,the volume fractions of precipitated phases of both(MoNi)6C and NiMo was the most in the alloy with[111]orientation and the least in the alloy with[001]orientation.The solidus and liquidus temperatures of the alloy IC6SX tested by differential scanning calorimetry(DSC)were 1356℃and 1387℃,respectively.Meanwhile,the effect of different solution temperatures on the microstructure of the alloy with different orientations was studied.The results showed that the precipitated phases of(MoNi)6C and NiMo were eliminated with the solid solution treatment under the condition of 1300℃/10h.However,the incipient melting of the alloys occurred due to the dissolution of low melting point phases.As the temperature dropped to 1280℃,the area of incipient melting in the alloy with different orientations decreased gradually.However,there was no incipient melting appearing in the three alloys with different orientations when the solution treatment temperature dropped to 1260℃.Liwu Jiang Xuezheng Dou Meiling Wu Min Cai 2020Progress in Natural Science:Materials International2020,30,4:2
16Termination of Transcription of LAT Increases the Amounts of ICP0 mRNA but Does Not Alter the Course of HSV-1 Infection in Latently Infected Murine Ganglia显示文摘On entering sensory ganglia,herpes simplex viruses 1(HSV-1)establishes a latent infection with the synthesis of a latency associated transcript(LAT)or initiates productive infection with expression of a set of immediate early viral proteins.The precise mechanisms how expression of a genes is suppressed during the latency are unknown.One mechanism that has been proposed is illustrated in the case of ICP0,a key immediate early viral regulatory protein.Specifically,the 2 kb LAT intron is complementary to the 30 terminal portion of ICP0 m RNA.To test the hypothesis that accumulation of LAT negatively affects the accumulation of ICP0 m RNA,we inserted a DNA fragment encoding two poly(A)sequences into LAT to early terminate LAT transcript without interrupting the complementary sequence of ICP0 transcript(named as SR1603).Comparisons of the parent(SR1601)and mutant(SR1603)HSV-1 viruses showed the following:Neurons harboring latent SR1603 virus accumulated equivalent amounts of viral DNA but higher amounts of ICP0 m RNA and lower amounts of LAT,when compared to neurons harboring the SR1601 virus.One notable difference between the two viruses is that viral RNA accumulation in explanted ganglia harboring SR1603 virus initiated significantly sooner than that in neurons harboring SR1601 virus,suggesting that ICP0 may act as an activator of viral gene expression in permissive cells.Collectively,these data suggest that increased ICP0 m RNA by suppressed LAT did not affect the establishment of latency in latently infected murine ganglia.Haifang Jiang Jiaming Wu Xianjie Liu Ruitao Lu Manling Zhou Meiling Chen Yonghong Liu Grace Guoying Zhou Wenmin Fu 2021Virologica Sinica2021,36,2:2
